Sampling Error Variation due to Rainfall Seasonality

  • In this study, we characterized the variation of sampling errors using the Waymire-Gupta-rodriguez-Iturbe multi-dimensional rainfall model (WGR model). The parameters used for this study are those derived by Jung et al. (2000) for the Han River Basin using a genetic algorithm technique. The sampling error problems considering in this study are those far using raingauge network, satellite observation and also for both combined. The characterization of sampling errors was done for each month and also for the downstream plain area and the upstream mountain area, separately. As results of the study we conclude: (1) The pattern of sampling errors estimated are obviously different from the seasonal pattern of mentally rainfall amounts. This result may be understood from the fact that the sampling error is estimated not simply by considering the rainfall amounts, but by considering all the mechanisms controlling the rainfall propagation along with its generation and decay. As the major mechanism of moisture source to the Korean Peninsula is obviously different each month, it seems rather norma1 to provide different pattern of sampling errors from that of monthly rainfall amounts. (2) The sampling errors estimated for the upstream mountain area is about twice higher than those for the down stream plain area. It is believed to be because of the higher variability of rainfall in the upstream mountain area than in the down stream plain area.

한국산 참굴(Crassostrea gigas) 미토콘드리아 DNA의 유전적 분석 (Genetic Analysis of Mitochondrial DNA from Korean Oysters, Crassostrea gigas)

  • 한국산 참굴의 유전적 특성을 조사하기 위하여 한국의 지역별 참굴을 대상으로 mtDNA 제한효소 절편분석과 클로닝을 수행하였다. 지역별로 각각 20개체의 mtDNA에 대하여 8가지 제한효소를 사용하여 DNA 절편양상을 분석한 결과 서해안산 참굴에서는 개체간 차이가 없는 단일 양상이었으며 남해안산의 경우는 두 가지 양상을 보였으며 차이는 HindIII 절단양상에서만 나타났다. 그 중 소수 개체들에서 나타난 양상은 서해안산 개체들에서의 양상과 동일하여 남해안에 서해안산 참굴이 유입되어 혼재하는 것으로 추정되었다. 한국산 참굴의 mtDNA를 대장균 E. coli HB101에서 클로닝하여 유전적 분석을 용이하도록 하였다. 전체 mtDNA를 제한효소를 사용하여 세부분으로 나누어 pUC19 유전자운반체에 클로닝하였다. 클로닝된 재조합 DNA를 제한효소들로 절단하여 한국산 참굴 mtDNA의 제한효소지도를 작성하였다. 남해안산과 서해안산 참굴 mtDNA에서 HindIII 적단 양상이 다르게 나타남을 확인하였고 이는 남해안산이나 서해안산에서 염기치환의 돌연변이에 의한 것으로 사료되었다.

  • 우리 나라 자생 짚신나물(Agrimonia pilosa Ledeb.) 수집종의 분포, 자생지의 환경 및 자생지에서의 형태적 특성조사를 실시하고, 자생종 짚신나물와 도입 짚신나물 간의 형태적 특성 및 유연관계를 연구한 결과는 다음과 같다. 1. 수집된 종 중에서 하동지역의 화개면 쌍계사와 청암면 청학동에서 수집된 2종은 다른 수집종에 비하여 유전적 거리지수가 가장 가까운 수치에서 나타나 근연관계로 나타났으며, 충북 영동읍 비탄재와 경기도 강화읍 수집종간에는 유전적 거리지수는 최대값이 같은 것으로 보아 유연관계가 있는 것으로 판단된다. 2. 경남거제에서 수집(No. 7)된 것과 충북 보은에서 수집(No. 21)된 2종이 유전적으로 가장 가깝게 나타나 지역적 차이에 의한 진화에도 다양함을 보이고 있어 수집지역의 확대와 이에 따른 체계적인 선발을 통해서 우량종의 선발도 가능할 것으로 판단된다 3.증폭된 DNA band pattern에 있어서 절편크기는 300∼21000p로 PCR로 증폭된 종 상호간의 1-F값은 평균 0.624였고, 최소값 0.365, 최대값 0.827이었으며, 2개 군으로 분리되어 유전적으로 상당한 차이가 있는 것으로 나타났다.

국내(國內) 가시오갈피 군락(群落)의 유전변이(遺傳變異) 분석(分析) (Genetic Variation of some Patches of Eleutherococcus senticosus (Rupr. & Maxim) Maxim. in Korea)

  • 가시오갈피 군락의 유전변이를 파악하기 위하여 국내산 8개 군락과 러시아산, 중국산 각 1개 군락등 총 10개 군락, 67개체에 대한 ISSR(inter-simple sequence repeats) 표지자 분석을 실시하였다. 국내산 가시오갈피의 ISSR 표지자 분석에서 76%의 다형성 증폭산물 (primer당 평균 11.5개)을 확인했으며, 가시오갈피 수집종에 대한 RAPD 표지자 분석 (Kim등(等), 1998)의 다형성 비율 57%(primer당 평균 5.7 개) 보다 높게 나타나서, ISSR 표지자 분석이 RAPD 표지자 분석보다 많은 정보를 제공할 수 있었다. 국내 가시오갈피 8개 군락의 유전변이분석에서 군락간 유전변이가 62.8%로 매우 높게 나타났는데, 이러한 결과는 자생지의 좁은 면적과 무성번식에 의한 군락 유지기작에 기인하는 것으로 생각된다. 또한 본 연구의 제한된 시료수가 군락간 높은 유전변이를 설명하는 요인이 될 수도 있지만, 가시오갈피 군락의 번식특성과 분포양상을 고려할 때 유전적 해석에 미치는 영향은 미미한 것으로 생각된다. 유연관계분석에서는 자생지의 지리적 위치와 군락의 유전적 연관성을 찾을 수 없었으며 이러한 경향은 AMOVA 결과 및 주성분분석에서도 확인할 수 있었다. 따라서 국내 가시오갈피의 유전자원보존을 위해서는 자생지 보호와 더불어 현지의(ex situ)보존에서는 군락당 소수 개체를 다수의 군락에서 선발하는 군락 위주의 선발이 효과적일 것으로 생각된다.

Ganoderma lucidum의 계통분류(系統分類) (Classification of Strains of Ganoderma lucidum)

  • Ganoerma lucidum의 인공재배(人工栽培)로 얻은 자실체(子實體)의 형태적(形態摘) 특성(特性)을 기초(基礎)로 계통(系統)을 분류(分類)하고 전기영동특성(電氣泳動特性)과의 관련성(關聯性)을 검토하였다. 영지균은 버섯의 형태(形態)와 윤문(輪紋)의 유무에 따라 4계통(系統)으로 분류(分類)되었으며 그중 편각 윤문형(輪紋形)이 가장 많았고 전형적 녹각형은 소수였다. 자실체(子實體)의 모양 즉 편각과 녹각, 갓 표면(表面)의 윤문(輪紋) 유무(有無)는 유전성(遺傳性)이었다. 영지균사(菌絲) 및 자실체(子實體)의 전기영동(電氣泳動)에 의한 단백질(蛋白質), Peroxidase, Esterase 및 Leucine Amino Peptidase Isozyme Pattern을 조사(調査)한 결과(結果) 한국자생영지균(韓國自生靈芝菌)은 유전형질(遺傳形質)에 다양성(多樣性)을 보였다. 그중 Esterase Isozyme Pattern은 편각과 녹각간에 차이가 컸고 동(同) 계통(系統)간에는 높은 유사도(類似度)를 보여 계통분류(系統分類) 결과(結果)와 일치하였다.

Regional Variations of Cellular Slime Molds Referred to Ribosomal DNA

  • Regional variations of Dictyostelid cellular slime molds were examined using molecular data. The intertranscribed spacer regions including the 5.8S ribosomal DNA of 2 species(D. purpureum, P. violaceum) of Cellular Slime Molds were sequenced and analyzed. Among 13 strains of D. purpureum and 12 strains of P. violaceum analyzed, each two strains were obtained from ATCC and the others were isolated from the forest soils in Korea. The sequences of the 5.8S ribosomal DNA were conserved among the strains of the same species, but unexpectedly highly variable among species. A high level of genetic diversity was found which was best resolved at the genus/species level as well as the family level by sequence data from the ITS 1 and ITS 2 regions. According to the sequence alignments by CLUSTAL X and the phylogeographic analyses by PAUP, 12 strains of P. violaceum were divided into three groups among which there were no difference of the morphological characteristics. Among 13 strains of D. purpureum, genetic variations were related to two morphological types, the temperate and subtropical type. There was no variation pattern according to geography in Korea, but there were some variations between Korea and other countries.

Assessing the Performance of Pongamia pinnata (l.) Pierre under Ex-situ Condition in Karnataka

  • Pongamia (Pongamia pinnata L.) as a source of non-edible oil, is potential tree species for biodiesel production. For several reasons, both technical and economical, the potential of P. pinnata is far from being realized. The exploitation of genetic diversity for crop improvement has been the major driving force for the exploration and ex situ/in situ conservation of plant genetic resources. However, P. pinnata improvement for high oil and seed production is not achieved because of unsystematic way of tree improvement. Performance of P. pinnata planted by Karnataka Forest Department was assessed based on yield potential by collecting 157 clones out of 264 clones established by Karnataka Forest Department research wing under different research circles/ranges. It was evident that the all the seed and pod traits were significantly different. Further, selection of superior germplasm based on oil and pod/seed parameters was achieved by application of Mahalanobis statistics and Tocher's technique. On the basis of D2 values for all possible 253 pairs of populations the 157 genotypes were grouped into 28 clusters. The clustering pattern showed that geographical diversity is not necessarily related to genetic diversity. Cluster means indicated a wide range of variation for all the pod and seed traits. The best cluster having total oil content of more than 34.9% with 100 seed weight of above 125 g viz. Cluster I, II, III, IX, XV, XIX, XXI, XXIII, XXVI and XXVII were selected for clonal propagation.

Genetic and Phenetic Differentiation among Three Forms of Korean Salamander Hynobius Ieechii

  • Genetic and morphological variations of Korean Hynobius leechii were surveyed. Starch gel electrophoresis was used to study genetic variation at 23 loci. H. leechii is genetically divided into three groups, each of which is named as Form-A, Form-B, and Form-C. These forms are clearly separated with 4-5 fixed or nearly fixed allelic differences. At a sympatry near Jangseong, Jollanam-do, a significant deviation from random mating between Form-A and Form-B was observed. Therefore, they are considered as two distinct taxi of specific rank. Form-A is widely distributed in the inland of Korea, whereas Form-B is distributed in the islands or near the coastal regions of Western and Southern Korea. Form-B had, in addition, a significant morphological difference from Form-A. On the other hand, Form-C was restricted to an area near Kori atomic power plant, Kyongsangnam-do, and the genetic similarities between this form and Form-A and Form-B were 0.742 and 0.736, respectively. Moreover, Form-C had a unique color pattern, short body and small coccyx. Thus, it seems reasonable to assign this form as another new species.

Computational analysis of SARS-CoV-2, SARS-CoV, and MERS-CoV genome using MEGA

  • The novel coronavirus pandemic that has originated from China and spread throughout the world in three months. Genome of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) predecessor, severe acute respiratory syndrome coronavirus (SARS-CoV) and Middle East respiratory syndrome coronavirus (MERS-CoV) play an important role in understanding the concept of genetic variation. In this paper, the genomic data accessed from National Center for Biotechnology Information (NCBI) through Molecular Evolutionary Genetic Analysis (MEGA) for statistical analysis. Firstly, the Bayesian information criterion (BIC) and Akaike information criterion (AICc) are used to evaluate the best substitution pattern. Secondly, the maximum likelihood method used to estimate of transition/transversions (R) through Kimura-2, Tamura-3, Hasegawa-Kishino-Yano, and Tamura-Nei nucleotide substitutions model. Thirdly and finally nucleotide frequencies computed based on genomic data of NCBI. The results indicate that general times reversible model has the lowest BIC and AICc score 347,394 and 347,287, respectively. The transition/transversions bias for nucleotide substitutions models varies from 0.56 to 0.59 in MEGA output. The average nitrogenous bases frequency of U, C, A, and G are 31.74, 19.48, 28.04, and 20.74, respectively in percentages. Overall the genomic data analysis of SARS-CoV-2, SARS-CoV, and MERS-CoV highlights the close genetic relationship.
