Relation of BAALC and ERG Gene Expression with Overall Survival in Acute Myeloid Leukemia Cases

  • Background: The objectives of this study were to evaluate the expression of brain and acute leukemia, cytoplasmic (BAALC) gene and erythroblast transformation-specific related gene (ERG) in de novo cases of acute myeloid leukemia (AML) and identify roles in disease progression and outcome. Materials and Methods: This study included 50 newly diagnosed AML patients, along with 10 apparently healthy normal controls. BAALC and ERG expression was detected in the bone marrow of both patients and controls using real-time RT-PCR. Results: BAALC and ERG expression was detected in 52% of cases but not in any controls. There was a statistically significant correlation between BAALC and ERG gene expression and age (p-value=0.004 and 0.019, respectively). No statistical significance was noted for sex, lymphadenopathy, hepatomegaly, splenomegaly, other hematological findings, immunophenotyping and FAB sub-classification except for ERG gene and FAB (p-value=0.058). A statistical significant correlation was found between response to treatment with ERG expression (p-value=0.028) and age (p-value=0.014). A statistically significant variation in overall survival was evident with patient age, BM blast cells, FAB subgroups, BAALC and ERG expression (p-value=<0.001, 0.045, 0.041, <0.008 and 0.025 respectively). Conclusions: Our results suggest that BAALC and ERG genes are specific significant molecular markers in AML disease progression, response to treatment and survival.

  • Platelet-derived growth factor (PDGF) is a critical regulator of mesenchymal cell migration and proliferation. The vital functions of PDGFs for angiogenesis, as well as development of kidney, brain, cardiovascular system and pulmonary alveoli during embryogenesis, have been well demonstrated by gene knock-out approaches. Clinical studies reveal that aberrant expression of PDGF and its receptor is often associated with a variety of disorders including atherosclerosis, fibroproliferative diseases of lungs, kidneys and joints, and neoplasia. PDGF contributes to cancer development and progression by both autocrine and paracrine signaling mechanisms. In this review article, important features of the PDGF isoforms and their cell surface receptor subunits are discussed, with regards to signal transduction, PDGF-isoform specific cellular response, and involvement in angiogenesis, and tumorstromal interactions.

  • An important recent advance in the field of plant-microbe interactions has been the cloning of genes that confer resistance to specific viruses, bacteria, fungi or insects. Disease resistance (R) genes encode proteins with predicted structural motifs consistent with them having roles in signal recognition and transduction. Plant disease resistance is the result of an innate host defense mechanism, which relies on the ability of plant to recognize pathogen invasion and efficiently mount defense responses. In tomato, resistance to the pathogen Pseudomonas syringae pv. tomato is mediated by the specific recognition between the tomato serine/threonine kinase Pto and bacterial protein AvrPto or AvrPtoB. This recognition event initiates signaling events that lead to defense responses including an oxidative burst, the hypersensitive response (HR), and expression of pathogenesis- related genes.

나일틸라피아, Oreochromis niloticus의 비특이적 면역반응에 대한 생약재 투여 효과 (Effect of Dietary Herb Medical Stuff on the Non-specific Immune Response of Nile Tilapia, Oreochromis niloticus)

  • 연구는 어류에 각종 생약재를 투여하였을 때 나타나는 방어 능력의 변화와 질병에 대한 예방 효과를 밝히고자 수행되었다. 실험의 내용은 인삼(Panax ginseng), 구기자(Lycium chinense), 하수오(Polygonum multiflorum), 오미자(Schizandra chinensis)의 첨가 사료를 나일틸라피아에 일정 기간 투여한 후 실험어의 성장, 비특이적 방어 능력 및 생리학적 기능에 미치는 영향을 조사한 것이다. 실험에 사용한 각종 생약재 중에서 체중의 증가는 구기자 3% 투여구가 가장 높게 나타났다. 보체의 살균 능력은 모든 실험구에서 살균력이 12주째에 증강되었고 보체의 용혈 능력도 모든 생약재 투여구에서 대조구 보다 높고 특히 인삼 2% 투여구에서 가장 높게 나타났다. 그러나 라이소자임의 용균능력과 신장의 부착 식세포 활성 산소 생성 능력은 구기자 3% 투여구가 가장 높았다. 그리고 나일틸라피아에 병원성을 지닌 Edwardsiella tarda 균주로 공격 실험한 결과 구기자 3% 투여구가 생존율이 85%로 가상 높았다. Hematocrit, hemoglobin, total protein, glucose, GOT, GPT와 같은 혈액의 생리학적 지표 성분을 분석한 결과 생약재를 첨가한 사료를 먹인 실험어는 대조구와 차이가 없으며 생리적 기능 면에서 나쁜 영향을 주지 않는 것으로 조사되었다. 따라서 인삼, 구기자, 하수오, 오미자 등과 같은 생약재는 그 종류에 따라 나일틸라피아의 성장률, 각종 비특이적 면역력을 증강시키는 능력과 병원성 세균에 대한 저항력의 증강 효과에 차이가 있지만 장기간 투여하여도 생리 기능에 문제를 야기하지 않는 것을 알 수 있었다. 모든 실험 결과를 종합해 보면, 실험에 사용한 각종 생약재 중에서 구기자는 가장 경제적이며, 각종 면역 기능 증강 효과가 뛰어나므로 새로운 사료내 첨가물로서의 충분한 가치가 있으며, 양식 어가에 많은 도움을 줄 것으로 생각된다.

A pathogen-induced osmotin-like protein gene, CAOSMl, from pepper: Differential expression and in situ localization in pepper tissues during pathogen infection and abiotic stresses

  • An osmotin-like protein (CAOSMl) gene was isolated from pepper leaves infected with the avirulent strain Bv5-4a of Xmthomonas campestris pv. vesicatoria. The cDNA encodes a polypeptide of 250 amino acids with a molecular mass of 27, 361 Da. Its amino acid sequence is highly homologous to various osmotin-like proteins from other plant species. The CAOSMl gene expression was organ- and tissue-specifically regulated In pepper plants. The CAOSMl mRNA was intensely localized in the endodermis area of root tissue and in the phloem cells of vascular bundles of red fruit tissue, but not in leaf, stem, and green fruit tissues of healthy pepper plants. Infection by X. c. pv vesintoria, Colletotrichum coccodes, or Phytopkhora capsici iinduced CAOSMl transcription in the leaf or stem tissues. Expression of the CAOSMl gene was somewhat higher in the incompatible than the compatible interactions of pathogens with pepper. The CAOSMl mRNA was prevalently localized in the phloem cells of the vascular bundle of leaf tissues infected by C. coccodes. The CAOSMl gene was activated in leaf tissues by treatment with ethylene, methyl jasmonate, high salinity, cold acclimation and mechanical wounding, but not by abscisic acid (ABA) and drought. These results indicate that the pepper CAOSMl protein functions in response to Pathogens and some abiotic stresses in pepper plants

감송향(甘松香) 물추출물이 U937 백혈병 세포의 분화유도에 미치는 영향 (Effect of Nardostachys chinensis on Induction of Differentiation in U937 Monomyelocytic Cells)

  • Nardostachyts chinensis (N. chinensis) belonging to the family Valerianaceae has been used to elicit stomachic and sedative effects. The MAPKs are serine/threonine kinases involved in the regulation of various cellular responses, such as cell proliferation, differentiation and apoptosis. The PKC also plays a key role in regulating the response of hematopoietic cells to both physiological and pathological inducers of proliferation and differentiation. This study investigated the signaling pathways on the U937 cell differentiation induced by N. chinensis. N. chinensis induced the differentiation of U937 cells, as shown by increased of differentiation surface antigen CD11b. Activation of ERK increased time-dependently in differentiation of U937 cells induced by N. chinensis, but activations of JNK and p38 were unaffected. Inhibitor of ERK (PD98059) significantly reduced CD11b expression induced by N. chinensis in U937 cells. In addition, N. chinensis increased protein level of PKC ${\beta}$I and PKC ${\beta}$II isoforms, but the protein level of PKC ${\alpha}$ and PKC ${\gamma}$was constant. PKC inhibitors (GF 109203X and H-7) inhibited U937 cell differentiation and the ERK activation induced by N. chinensis. These results indicated that PKC and ERK may be involved in U937 cell differentiation induced by N. chinensis.

Genetic and Expression Analysis of the SIRT1 Gene in Gastric Cancers

  • Purpose: Silent mating-type information regulation 2 homologue 1 (SIRT1) is a nicotinamide adenine dinucleotide-dependent deacetylase. SIRT1 plays an important role in the regulation of cell death/survival and stress response in mammals. The aim of this study was to investigate whether the SIRT1 gene is involved in the development or progression of gastric cancers. Materials and Methods: SIRT1 and p53 genes in 86 gastric cancers were examined for genetic alterations by PCR-single strand conformation polymorphism sequencing, as well as SIRT1 protein expression in 170 gastric cancers by immunohistochemistry. Results: In the genetic analysis, we found SIRT1 and p53 mutations in two and 12 cases, respectively. Two missense mutations, c.599 C>T (T200I) and c.1258 G>A (E420K), were detected in the SIRT1 gene coding region. The SIRT1 and p53 mutation were found in mutually exclusive gastric cancers. The immunohistochemistry revealed that SIRT1 overexpression was found in 95 (55.9%) of 170 gastric cancers. Altered SIRT1 expression was not statistically associated with clinicopathological parameters, including tumor differentiation, location, lymph node metastasis, or p53 expression. Two cases with an SIRT1 mutation showed increased SIRT1 expression. Conclusions: These results suggest that genetic alterations and overexpression of the SIRT1 gene may contribute to gastric cancer development.

Modulation of Antibody Responses against Gnathostoma spinigerum in Mice Immunized with Crude Antigen Formulated in CpG Oligonucleotide and Montanide ISA720

  • This study aimed to investigate the antibody responses in mice immunized with Gnathostoma spinigerum crude antigen (GsAg) incorporated with the combined adjuvant, a synthetic oligonucleotide containing unmethylated CpG motif (CpG ODN 1826) and a stable water in oil emulsion (Montanide ISA720). Mice immunized with GsAg and combined adjuvant produced all antibody classes and subclasses to GsAg except IgA. IgG2a/2b/3 but not IgG1 subclasses were enhanced by immunization with CpG ODN 1826 when compared with the control groups immunized with non-CpG ODN and Montanide ISA or only with Montanide ISA, suggesting a biased induction of a Th1-type response by CpG ODN. After challenge infection with live G. spinigerum larvae, the levels of IgG2a/2b/3 antibody subclasses decreased immediately and continuously, while the IgG1 subclass remained at high levels. This also corresponded to a continuous decrease of the IgG2a/IgG1 ratio after infection. Only IgM and IgG1 antibodies, but not IgG2a/2b/3, were significantly produced in adjuvant control groups after infection. These findings suggest that G. spinigerum infection potently induces a Th2-type biased response.

Nonlinear Regression Analysis to Determine Infection Models of Colletotrichum acutatum Causing Anthracnose of Chili Pepper Using Logistic Equation

  • A logistic model for describing combined effects of both temperature and wetness period on appressorium formation was developed using laboratory data on percent appressorium formation of Colletotrichum acutatum. In addition, the possible use of the logistic model for forecasting infection risks was also evaluated as compared with a first-order linear model. A simplified equilibrium model for enzymatic reactions was applied to obtain a temperature function for asymptote parameter (A) of logistic model. For the position (B) and the rate (k) parameters, a reciprocal model was used to calculate the respective temperature functions. The nonlinear logistic model described successfully the response of appressorium formation to the combined effects of temperature and wetness period. Especially the temperature function for asymptote parameter A reflected the response of upper limit of appressorium formation to temperature, which showed the typical temperature response of enzymatic reactions in the cells. By having both temperature and wetness period as independent variables, the nonlinear logistic model can be used to determine the length of wetness periods required for certain levels of appressorium formation under different temperature conditions. The infection model derived from the nonlinear logistic model can be used to calculate infection risks using hourly temperature and wetness period data monitored by automated weather stations in the fields. Compared with the nonlinear infection model, the linear infection model always predicted a shorter wetness period for appressorium formation, and resulted in significantly under- and over-estimation of response at low and high temperatures, respectively.

Identification and Expression Profiles of Six Transcripts Encoding Carboxylesterase Protein in Vitis flexuosa Infected with Pathogens

  • Plants protect themselves from pathogen attacks via several mechanisms, including hypersensitive cell death. Recognition of pathogen attack by the plant resistance gene triggers expression of carboxylesterase genes associated with hypersensitive response. We identified six transcripts of carboxylesterase genes, Vitis flexuosa carboxylesterase 5585 (VfCXE5585), Vf-CXE12827, VfCXE13132, VfCXE17159, VfCXE18231, and VfCXE47674, which showed different expression patterns upon transcriptome analysis of V. flexuosa inoculated with Elsinoe ampelina. The lengths of genes ranged from 1,098 to 1,629 bp, and their encoded proteins consisted of 309 to 335 amino acids. The predicted amino acid sequences showed hydrolase like domains in all six transcripts and contained two conserved motifs, GXSXG of serine hydrolase characteristics and HGGGF related to the carboxylesterase family. The deduced amino acid sequence also contained a potential catalytic triad consisted of serine, aspartic acid and histidine. Of the six transcripts, Vf-CXE12827 showed upregulated expression against E. ampelina at all time points. Three genes (VfCXE5585, VfCXE12827, and VfCXE13132) showed upregulation, while others (VfCXE17159, VfCXE18231, and VfCXE47674) were down regulated in grapevines infected with Botrytis cinerea. All transcripts showed upregulated expression against Rhizobium vitis at early and later time points except VfCXE12827, and were downregulated for up to 48 hours post inoculation (hpi) after upregulation at 1 hpi in response to R. vitis infection. All tested genes showed high and differential expression in response to pathogens, indicating that they all may play a role in defense pathways during pathogen infection in grapevines.
