Identification of Antifungal Substances of Lactobacillus sakei subsp. ALI033 and Antifungal Activity against Penicillium brevicompactum Strain FI02

  • This study was performed to investigate the antifungal substances and the antifungal activity against fungi of lactic acid bacteria (LAB) isolated from kimchi. LAB from kimchi in Imsil showed antifungal activity against Penicillium brevicompactum strain FI02. LAB LI031 was identified as Lactobacillus sakei subsp. Antifungal substances contained in L. sakei subsp. ALI033 culture media were unstable at high pH levels. Both, the control and proteinase K and protease treated samples showed clear zones, suggesting that the antifungal substances produced by ALI033 were non-protein substances unaffected by protesases. Both, the control and catalase showed clear zones, suggesting that the antifungal metabolite was not $H_2O_2$. The molecular weights of the antifungal substances were ${\leq}3,000Da$. The organic acid content of crude antifungal substances produced by L. sakei subsp. ALI033 showed high concentrations of lactic acid (502.47 mg/100 g). Therefore, these results suggest that antifungal substance produced by L. sakei subsp. ALI033 is most likely due to its ability in producing organic acid.

Bioactive Metabolites Produced by Pseudonocardia endophytica VUK-10 from Mangrove Sediments: Isolation, Chemical Structure Determination and Bioactivity

  • Chemical investigation of the actinobacterial isolate Pseudonocardia endophytica VUK-10 has led to the segregation of two known bioactive compounds, namely 4-(2-acetamidoethyl) phenyl acetate and 4-((1, 4-dioxooctahydropyrrolo [1, 2-a] pyrazin-3-yl) methyl) phenyl acetate. The strain was isolated from a sediment sample of the Nizampatnam mangrove ecosystem, south coastal Andhra Pradesh, India. The chemical structure of the active compounds was established on the basis of spectroscopic analysis, including 1H NMR and 13C NMR spectroscopies, FTIR, and EIMS. The antimicrobial and cytotoxic activities of the bioactive compounds produced by the strain were tested against opportunistic and pathogenic bacteria and fungi and on MDA-MB-231, OAW, HeLa, and MCF-7 cell lines. The compounds exhibited antimicrobial activities against gram-positive and gram-negative bacteria and fungi and also showed potent cytotoxic activity against MDA-MB-231, OAW, HeLa, and MCF-7 cell lines. This is the first example for this class of bioactive compounds isolated from Pseudonocardia of mangrove origin.

Canna edulis Leaf Extract-Mediated Preparation of Stabilized Silver Nanoparticles: Characterization, Antimicrobial Activity, and Toxicity Studies

  • A novel approach to synthesize silver nanoparticles (AgNPs) using leaf extract of Canna edulis Ker-Gawl. (CELE) under ambient conditions is reported here. The as-prepared AgNPs were analyzed by UV-visible spectroscopy, transmission emission microscopy, X-ray diffraction, Fourier transform-infrared spectroscopy, energy-dispersive analysis of X-ray spectroscopy, zeta potential, and dynamic light scattering. The AgNPs showed excellent antimicrobial activity against various pathogens, including bacteria and various fungi. The biocompatibility of the AgNPs was analyzed in the L929 cell line using NRU and MTT assays. Acridine orange/ethidium bromide staining was used to determine whether the AgNPs had necrotic or apoptotic effects on L929 cells. The concentration of AgNPs required for 50% inhibition of growth of mammalian cells is far more than that required for inhibition of pathogenic microorganisms. Thus, CELE is a candidate for the eco-friendly, clean, cost-effective, and nontoxic synthesis of AgNPs.

벼 잎집무늬마름병균(Rhizoctoniz solani)의 균핵에서 분리한 Bacillus subtilis SJ-2의 식물 병원 곰팡이에 대한 항균 활성 및 항균 물질의 특성 (Antifungal Effects on Plant Pathogenic Fungi and Characteristics of Antifungal Substances Produced by Bacillus subtillis SJ-2 Isolated from Sclerotia of Rhizoctonia solani)

  • 벼 잎집무늬마름병균(Rhizoctonia solani)의 균핵에서 분리한 길항세균 SJ-2를 대치 배양 방법에 의해 벼도열병균(Pyricularia oryzae)외 6종의 식물 병원균에 대한 억제 효과를 조사하였다. 형태 및 생리적 특성을 조사한 결과, 이 균은 Bacillus subtilis로 동정되었다. 이 균은 AM 1(antibiotic medium 1)액체 배지에서 항균 물질을 분비하였으며, 배양 2일째 가장 항균 활성이 높았다. Butyl alcohol로 배양액에서 항균 물질을 조추출하여 100$\mu\textrm{g}$/ml의 농도로 조제한 감자 한천 배지에서 P. oryzae의 15개균에 대한 생육을 조사한 결과 P. oryzae, R. solani, Cochliobolus sativus에 대해서는 100% 생장을 억제하였으며, C. miyabeanus, Alternaria alternata, Botrytis cinerea, Cladosporium fulvum, Colletotrichum gloeosporioides, Fusarium moniliforme, F. oxysporum에는 80% 이상의 억제 효과를 보인 반면 난균강에 속하는 병원균에 대한 효과는 낮게 나타났다. 활성물질을 분리 정제하여 동정한 결과 B. subtilis가 분비하는 항균 물질로 알려진 polypeptides계의 iturins로 밝혀졌다. 이러한 항균 물질은 벼 도열병균(P. oryzae)의 포자 발아 및 발아관의 팽대(swelling)를 야기했으며, 벼 잎집무늬마름병균(R. solani)에는 균사의 용균(lysis)현상을 일으켰다.

High Throughput Screening of Antifungal Metabolites Against Colletotrichum gloeosporioides

  • Colletotrichum gloeosporioides forms an appressorium, a specialized infection structure, to infect its hosts. Among 400 and 600 culture filtrates from fungi and class Actinomycetes, six methanol extracts (A5005, A5314, A5387, A5560, A5597, and A5598) from the class Actinomycetes significantly inhibited appressorium formation in C. gloeosporioides infecting pepper fruits in a dose-dependent manner, while conidial germination was slightly enhanced. Two (A5005 and A5560) of them also exhibited distinctive inhibitory effect on the disease progress of pepper anthracnose. Water fractions of both culture filtrates also specifically inhibited appressorium formation in C. gloeosporioides and pepper anthracnose disease. Inhibition of appressorium formation by culture filtrate of A5005 was partially restored by the exogenous calcium. This results suggests that chemicals within A5005 extents its biological activity through disturbance of intracellular $Ca^{2+}$ regulation during prepenetration morphogenesis by C. gloeosporioides. Together, cell-based and target-oriented screening system used in this study should be applicable for other plant pathogenic fungi prerequisite appressorium formation to infect their hosts.

수확 후 인삼뿌리썩음병의 생물학적 방제 (Biological Control of Postharvest Root Rots of Ginseng)

  • The production of Korean ginseng, one of the most important medicinal root crops, is limited by many factors including soil sickness, root rots in fields as well as during storage prior to consumption. Although much research has been conducted on the diseases in field condition, little information is available on the control of postharvest roots rots. To obtain better management strategy of postharvest root rots in ginseng, biological control using antagonistic bacteria was attempted. Of 208 bacteria obtained form suppressive soil samples, 4 were selected based on the inhibitory effect on mycelial growth of two major causal fungi for postharvest root rots in ginseng, Botrytis cinerea and Fusarium solani. The culture filtrates of these bacterial antagonists greatly inhibited the conidial germination of both pathogenic fungi and produced abnormal morphology such as swollen germ tubes in F. solani and vacuolation of nongerminated conidia in B. cinerea. The population levels of bacterial antagonists on the ginseng roots were gradually increased up to 8 days of incubation. Postharvest root rots of ginseng caused by f. solani and B. cinerea were controlled in dipping tests in the ranges of 60∼80% by antagonistic Bacillus spp. obtained from suppressive soil. These results suggest that biological control using these antagonistic bacteria would be an alternative strategy to control postharvest root rots in ginseng.

Antimicrobial Properties of Cold-Tolerant Eucalyptus Species against Phytopathogenic Fungi and Food-Borne Bacterial Pathogens

  • Mechanol extracts of three cold-tolerant eucalyptus trees-Eucalyptus darlympleana, E. gunnii and E. unigera were screened for antimicrobial activity against twenty two phyto-pathogenic fungi and six food-borne bacterial pathogens. E. unigera showed the antagonistic activity against all the tested pathogens. Among the tested fungal pathogens, Pythium species were highly sensitive to the leaf extracts. Especially, P. vanterpoolii, a causal agent of leaf blight in creeping bentgrass (Agrostis palustris), was completely inhibited by the extracts. The eucalyptus extracts were also effective in inhibiting the fungal growth of Botrytis cinerea and Phomopsis sp. isolated from the lesions of kiwifruit soft rot during post-harvest storage. Escherichia coli O-157 was less sensitive to the inhibition than the other bacterial pathogens tested. It was likely that Gram positive bacteria-Bacillus subtilis and Streptococcus mutans were more sensitive to the eucalyptus extracts than Gram negative bacteria-Escherichia coli, Salmonella enteritidis and Pseudomonas aeruginosa. Our findings suggest that the cold-tolerant eucalyptus species have antimicrobial properties that can serve the development of novel fungitoxic agents or food preservatives.

Biocontrol with Myxococcus sp. KYC 1126 Against Anthracnose in Hot Pepper

  • Antifungal activity of three Myxococcus spp., KYC 1126, 1136, and 2001, was tested in vitro against three phytopathogenic fungi (Botrytis cinerea, Colletotrichum acutatum, and Pyricularia grisea). Spore germination and mycelial growth of the three pathogenic fungi were completely inhibited by bioactive substances from a myxobacterium KYC 1126. In addition, the activity of KYC 1126 was fungicidal, but liquid culture filtrate of KYC 1126 did not affect protoplast reversion in C. acutatum. A bioassay of KYC 1126 filtrate against anthracnose in hot pepper was conducted in the greenhouse and field at 2009 and 2010. The incidence of anthracnose in control seedlings was 74%, but was reduced to 29% after KYC 1126 treatment. The control value with KYC 1126 was 60% while that with the fungicide dithianon was 42%. In the greenhouse, disease incidence with KYC 1126 was consistentely 10-35% lower than with fungicide as a positive control. The control value with KYC 1126 was 13.4% and 41.0%, whereas that with the fungicide was 52.3% and 63% in 2009 and 2010, respectively. Although anti-anthracnose activity of KYC 1126 was not maintained for long time in the field, the bacteriolytic myxobacterium KYC 1126 could be a prospective biocontrol agent.

Control of Soybean Sprout Rot Caused by Pythium deliense in Recirculated Production System

  • A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.

Screening of Cyanobacteria (Blue-Green algae) from Rice Paddy Soil for Anti-fungal Activity against Plant Pathogenic Fungi

  • Soil cyanobacteria isolated from the rice paddy fields of 10 different locations across Korea were evaluated by agar plate diffusion test for antifungal activity. Aqueous, petroleum ether, and methanol extracts from one hundred and forty two cyanobacterial strains belonging to the 14 genera were examined for antifungal properties against seven phytopathogenic fungi causing diseases in hot pepper (Capsicum annuum L). Of total cyanobacteria, nine cyanobacteria (6.34%) exhibited antifungal effects. The nine cyanobacteria selected with positive antifungal activities were two species of Oscillatoria, two of Anabaena, three of Nostoc, one of Nodularia, and one of Calothrix. Alternaria alternata and Botrytis cinerea were inhibited by nine and eight species of cyanobacteria, respectively. Rhizopus stolonifer was suppressed by only methanol extract of Nostoc commune FK-103. In particular, Nostoc commune FK-103 and Oscillatoria tenuis FK-109 showed strong antifungal activities against Phytophthora capsici. Their antifungal activity at the late exponential growth phase is related to the growth temperature and not associated with the growth parameters such as cell biomass and $chlorophyll-{\alpha}$ concentration. The high inhibition levels of antibiotics were 22.5 and 31.8 mm for N. commune FK-103 and O. tenuis FK-109, respectively. The optimal temperature for antibiotic productivity was $35^{\circ}C$.
