제약회사 폐수처리장 방류수 중 항생제 내성 Enterobacteria에 관한 연구 (Study on Antibiotic Resistant Enterobacteria in Pharmaceutical Effluent)

  • Objectives: This study aims to examine the concentration, diversity, and antibiotic characteristics of penicillin G resistant enterobacteria present in pharmaceutical effluent. Methods: Water sampling was performed from a pharmaceutical company in Gyeonggi-do Province, Korea in March 2015. Water samples were plated in triplicate on tryptic soy agar plates with 32 mg/L of penicillin G. Penicillin G resistant enterobacteria were selected from the effluent and were subjected to 16S rRNA analysis for penicillin G resistant species determination. Identified resistant strains were tested for resistance to various antibiotics. Results: Penicillin G resistant enterobacteria were present at 6.2% as to culturable heterotrophic bacteria. Identified penicillin G resistant enterobacteria exhibited resistance to more than 10 of the antibiotics studied. These resistant bacteria are gram negative and are closely related to pathogenic species. Conclusion: Multi-antibiotic resistant bacteria in the effluent suggest a need for disinfection and advanced oxidation processes for pharmaceutical effluents.

  • Rapid, real-time detection of pathogenic microorganisms is an emerging and quickly evolving field of research, especially with regard to microorganisms that pose a major threat to public health. Herein, a new method that uses bioimpedance and solid culture medium for the real-time detection of microorganisms is introduced. We fabricated a new impedimetric biosensor by integrating solid media and two plane electrodes attached on two facing sides of an acryl well. During bioelectrical impedance analysis, the solid medium showed the characteristics of a homogenous conductive material. In a real-time impedance measurement, our solid-medium biosensor could monitor bacterial growth in situ with a detection time of ${\sim}4$ hrs. Our data indicate that the solid-medium biosensor is useful for detecting airborne microorganisms, thereby providing a new analytical tool for impedance microbiology.

Cochlosoma Infection in a Turkey in Iran

  • Cochlosoma sp. infection was identified in a single case among 60 stunted diarrheic native turkey poults, Meleagris galopavo. A large number of the flagellated parasites was found free or within the intervillous spaces of the jejunum, ileum and cecum. Moderate enteritis was associated with the parasites. In TEM studies of the parasagittal sections of the parasite, a prominent ventral sucker like disc and flagella emerging from an opening on the ventrodorsal surface of the pyriform uninuclear parasite were found. The morphological characteristics of this protozoan match with those described for Cochlosoma anatis. The parasite could be considered as an intestinal pathogenic protozoan causing stunting and diarrhea in turkeys in Iran.

  • In 1991, the leaves and roots brown rot disease of scarlet kafir lily were found in Taejon and Seoul. The symptoms were appeared as dark-brown and water soaked on leaves. The discolored area of the leaves become halo. The roots revealed blight gray and water soaked. The pathogenic bacteria were isolated from the diseased leaves of the scarlet kafir lily were identified as Erwinia cypripedii on the bais of bacterial characteristics. E. cypripedii is first described bacteria which cause the disease on scarlet kafir lily in Korea. Therefore, we would like to propose to the name of scarlet kafir lily disease caused by E. cypripedii as“bacterial brown-rot of scarlet kafir lily”hereafter.

  • In 1991, the leaf brown spot of ivy-aureus (Scindapsus aureus) was found in Taejon and Seoul, Korea. The symptoms were appeared as dark-brown spots. The lesions were often surrounded by yellowish halos. These spots were enlarged to circular or elliptical in shape and dark-brown to black in color with slightly elevated in margin and sunken in center. The pathogenic bacteria were isolated from the diseased leaf of ivy-aureus were identified as Erwinia chrysanthemi on the basis of bacterial characteristics therefore, we would like to propose to the name of ivy-aureus disease caused by E. chrysanthemi as“bacterial brown rot of ivy-aureus”hereafter.

Occurrence of Bacterial Soft Rot of Lily Bulb Caused by Pectobacterium carotovorum subsp. carotovorum and Pseudomonas marginalis in Korea

  • Soft rot symptom was observed on lily bulb in the fields and at a low temperature storage house from 1999 to 2000 in Korea. The small dark-brown lesion appeared on the bulb, and enlarged and developed into the inner scales of the bulb. The bulb became water soaked and gave out unpleasant odor. Two different pathogenic bacteria were isolated from infected tissues. The causal bacteria were identified as Pectobacterium carotovorum subsp. carotovorum (Erwinia carotovora subsp. carotovora) and Pseudomonas marginalis based on bacteriological characteristics. Pathogenicity of the bacteria was proven by Koch's postulations. This is the first report of bacterial soft rot of lily bulb in Korea caused by the two bacteria.

Occurrence of Target Leaf Spot of Red and White Clovers Caused by Stemphylium sarciniforme in Korea

  • A terget leaf spot of red and white clovers was observed during 1998-2000 growing seasons in several fields of Chungnam and Chungbuk provinces in Korea. Lesions were circular to oval in outline, brown to dark brown, ranging from a pinpoint to 3-4 mm in diameter, often concentrically zonate and presenting a target effect. A fungus associated with the disease was identified as Stemphylium sarciniforme based on the morphological characteristics of the conidiophores and conidia. The fungus was pathogenic on red and white clovers in the inoculation test. This is the first record of a terget leaf spot of red and white clovers in Korea.

  • Pathogenic fungal species associated with apple anthracnose were studied with diseased fruits collected over two major apple growing areas from 1986 to 1988. Colletotrichum acutatum Simmonds, c. gloeosporioides(Penz.) Sacc. and Glomerella cingulata(Stonem.) Spauld. & Schrenk were identified based on their mycological characteristics. C. acutatum could be easily differentiated from C. gloeosporioides by producing their fusiform conidia and developing apricot∼salmon colony colors, non-forming an ascigerous stages. Growth rate of C. acutatum were slow in plate culture. The three species of anthracnose fungi tested caused apple rot. Symptoms produced by C. gloeosporioides were larger than those of the others. C. gloeosporioides and G. cingulata produced rot lesions on apples that appeared became brown and later black, but C. acutatum produced rot lesions appeared yellowish brown and later reddish brown. G. cingulata has the major species responsible for apple anthracnose, epidemics in Korea.

First Report of Bacterial Wilt Caused by Ralstonia solanacearum Biovar 2 Race 1 on Tomato in Egypt

  • This study aims to isolate and identify the causal pathogen of tomato bacterial wilt in Egypt. In 2008, tomato plants showing typical symptoms of bacterial wilt disease with no foliar yellowing were observed in Minia, Assiut and Sohag governorates, Egypt. When cut stems of symptomatic plants were submerged in water, whitish ooze was evident and longitudinal sections showed a brown discoloration in the vascular tissues. Bacteria were isolated on triphenyl tetrazolium chloride medium and fifteen isolates shown typical morphological and cultural characteristics were confirmed as Ralstonia solanacearum biovar 2 race 1. Pathogenicity tests showed that all isolates proved to be pathogenic to tomato plants, varied from 52 to 97% wilting. This is the first report of R. solanacearum biovar 2 race 1 causing bacterial wilt in tomato crop in Egypt.

Occurrence of Damping-off Caused by Pythium spinosum on Cucumis melo in Korea

  • In 2010 and 2012, damping-off symptoms were found on melon seedlings grown in Yeongam and Suncheon, Korea. Water-soaked and discolored lesions appeared on the lower stems of the infected plants. The diseased plants became wilted and stunted, and eventually collapsed. On the basis of morphological characteristics and molecular analyses of internal transcribed spacer (ITS) rDNA and cytochrome oxidase II (cox2) sequences, the causal organism was identified as Pythium spinosum. The isolates were pathogenic to melon under pot conditions. To our knowledge, this is the first report of P. spinosum causing damping-off on melon in Korea.
