Studies on the Edwardsiellosis of cultured flounder, Paralichthys oliuaceus;- Characteristics, pathgenicity and control of Edwardsiella tarda - (양식넙치(Paralichthys olivaceus)의 복수증에 관한 연구 - Edwardsiella tarde의 일부 특성과 병원성 및 대책 -)

  • Characteristics, pathogenicity and control of the causative organisms isolated front diseased cultured flounder, Paralichthys olivaceus were studied. The causative organisms were identified as E tarda by biochemical and biophysical characteristics. Also, it strains were named as E tarda KBF-1 and E tarda KMF-1, and optimal pH of E tarda KBF-1 and E tarda KMF-1 were 8.0, and optimal concentration of NaCl. E tarda KBF-1 was 0% and E tarda KMF-1 was 1%. In the pathogenicity test, 0~10 of the flounders of artificially infected group(E tarda KBF-1) with $1.0{\times}10^7$ cfu/fish were died within 60 hrs, but 0~9 flounders infected group with 41.0{\times}10^6$ cfu/fish were died within 60 hrs. Also, 0~10 flounders infected group(E tarda KMF-1) with $1.0{\times}10^7$ cfu/fish were died within 36 hrs, while 0~7 flounders infected with $1.0{\times}10^6$ cfu/fish were died within 60 hrs. Drug sensitivity of E tarda KBF-1 strain was resistant to AM, CF and N, and intermediate to E, K and S, and sensitivity to C, G, SxT and FF. But E tarda KMF-1 strain was resistant to CF, E and V, and intermediate to AM, C, N and SxT, and sensitivity to GM and FF.

  • This study was carried out for the identification of several fungi isolated from some different geographic regional soil samples and evaluation of its pathgenicity to the mice. isolates, The results were isolated from 200 soil samples and 11 strains from 25 pigeon excreta. 2. Seventeen of Cryptococcus neoformans, eight of Candida albicans and Microporum gypseum, six of M. coorei, and three of Sporothrix schencrii were identified group. 3. An association of C. neoformans with avian habitats was found. Ten of the 11 isolates came from areas frequently by pigeons such as pigeon nest and pigeon excreta. 4. The inflammatory reaction produced in mice that were inoculated with C. neoformans and C. albicans intraperitoneal route.

Comparison of Pepper Anthracnose Pathogens from Korea and China

  • Anthracnose of pepper casued by Colletotrichum spphas been a great problems for pepper production in Korea and China. Especially Colletotrichum gloeosporioides was found predominantly over cultivation areas during infection periods and caused severe rots on bath unripe and ripe fruits that resulted in major yield losses. In this study, comparison of Colletrichum spp.isolated from Korea and China in morphology and pathgenicity, and RAPD-PCR analysis were conducted. Based on morphological characteristics, the pathogen isolates, K1 and C1, K2 and C2, and K3 and C3 were identified as Colletotrichum gloeosporioides (G) type, C. gloeosporioides (R) type and C. coccodes, respectively. in pathogenicity test, K1 and C1, and K2 and C2 were found to attack mainly fruits and to be the most virulent among isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates did not show any difference. Results of the RAPD-PCR analyses indicate the varying levels of molecular diversity within and between Colletotrichum spp.of Korea and China. The similarities between K1 and C1, K2 and C2, and K3 and C3 were 85.71%, 71.43% and 50.0% respectively.

Pathogenicity of Spodoptera exigua Nuclear Polyhedrosis Virus and Cross Infection of Baculoviruses to the Beet Armyworm, S. exigua (Lepidoptera: Noctuidae) (파밤나방 핵다각체병바이러스의 병원성과 곤충간상바이러스의 파밤나방에 대한 교차감염에 관한 연구)

  • Pathogenicity of Spodoptera exigua nuclear polyhedrosis virus (SeMNPV) against the host insect and 8 species of lepidopterous insects and cross infection of baculoviruses to third instar of S. exigua larvae were studied to determine as a biocontrol agent for S. exigua. The median lethal concentrations($LC_{50}$)of the SeMNPV to egg mass was $2.855\times10^5$ PIBs/ml and higher than that to the larvae of S. exigua. Mortality of the SeMNPV in third ins tar larvae was more increased than that in first and fifth instar of S. exigua larvae by 1.16 and 4.11 times, respectively. The median lethal times($LT_{50}$) to $1.56\times10^6$ PIBs/ml was in the range of 4.25 to 5.04 days. Infectivity of the SeMNPV against eight species of lepidopterous insects was showed only in the host insect, S. exigua. Autographa cali/ornica MNPV, Mamestra barassicae MNPV, and Trichoplusia ni MNPV were cross-infected to third instar of S. exigua larvae among ten of baculoviruses tested.

Pathgenicity on Ginseng and Sequence Assays of Ilyonectria radicicola Isolated from Chestnut Rhizosphere Soils (밤나무 근권토양에서 분리한 Ilyonectria radicicola 균주의 인삼에 대한 병원성 및 유전적 분석)

  • Background: A soil-borne pathogenic fungus, Ilyonectria radicicola (Cylindrocarpon destructans) causes root rot on ginseng (Panax ginseng C. A. Meyer) and is known to attack many other plants. The Nectria/Neonectria radicicola complex has been renamed as the I. radicicola complex after analysis of its multi-gene relatedness and morphological characteristics. The fungi in this complex have been reclassified into 16 species under the genus Ilyonectria based on characteristics analysis Methods and Results: To obtain useful data from the Korean ginseng root rot, I. radicicola was isolated from the rhizosphere soils of the chestnut tree. They were identified through a pathogenicity test and a survey of the morphological features. The existence of I. radicicola in soil samples was confirmed by PCR detections using nested PCR with species-specific primer sets. These were subsequenctly isolated on semi-selective media from PCR-positive soils. Genetic analysis of the I. radicicola complex containing these pathogens was done by comparing the DNA sequences of the histone h3 region. These isolates originating from the rhizosphere soils of chestnut constituted a clade with other closely related species or I. radicicola isolates originating from ginseng or other host plants, respectively. Additionally, the pathogenicity tests to analyze the characteristics of these I. radicicola isolates revealed that they caused weakly virulent root rot on ginseng. Conclusions: This is the first study reporting that I. radicicola isolates from chestnut rhizosphere soils can attack ginseng plant in Korea. Thus, these results are expected to provide informations in the selection of suitable fields for ginseng cultivation.

Expression control of ssaJ and ssaK of SPI2 in Salmonella typhimurium (Salmonella typhimurium에서 SPI2의 ssaK와 ssaJ의 발현조절)

  • Salmonella Pathgenicity Island 2 plays an important role in Salmonella pathogenicity, especially invasion into host cell. We have investigated the effect of various environmental factors, such as oxygen level, osmolarity, pH, carbon starvation and glycerol addition on the expression of SPI2. For this research, we constructed the reporter plasmids, in which the promoter-less lac operons are fused with the regulatory regions (including promoter) of ssaJ and ssaK, major genes in SPI2. The study using the reporters showed that low oxygen, low osmolarity, or weak alkali conditions increased the expression levels of ssaJ and ssaK and when these three conditions exist simultaneously, the expression levels of ssaJ and ssaK are the highest. However carbon starvation and glycerol addition did not affect the expression of ssaJ and ssaK. These environmental effects on the expression levels of ssaJ and ssaK are the same in three Salmonella typhimurium wild types, LT2, UK1, and SL1344. In addition, we confirmed that the mutation in hilA, a regulatory gene encoding a transcriptional activator of SPI1, had no effect on the expression of ssaJ and ssaK. Thus, these results strongly suggest that the expressions of SPI2 and SPI1 are regulated by different control systems.

Studies on the Host Range of Colletotrichum dematium Isolated from Anthracnose of Pepper and Toxic Metabolites Produced by the Pathogen (고추 탄저병균(炭疽病菌) Colletotrichum dematium의 기주범위(寄主範圍) 및 대사독소(代謝毒素)에 관(關)한 연구(硏究))

  • This studies were conducted to investigate pathgenicity and host range of Colletotrichum dematium isolated from anthracnose of pepper, and phytoxicity of its culture filtrate and the partially purified toxin. The results obtained were as follows. 1. Investigation on the host range of C. dematium has revealed that pepper as well as soybean, tomato, spinach, and beet were highly susceptible, egg plant and water melon were moderately susceptible and stone leek was slightly suceptible, but no symptoms were produced on carrot, tabacco, cucumber and melon. 2. The culture filtrates of C. dematium in Czapeck dox liquid media were toxic to leaves of pepper and caused necrosis and wilting of the plant. The toxicity of culture filtrates was most active at 15 days after fungal growth in Czapeck dox liquid media and the toxin productivity in still culture was higher than that in shaking culture. 3. The partially purified toxic substance was isolated from the culture filtrates by the acetone precipitation method. When cuttings of various pepper cultivars were placed in the toxin solutions, suceptible cultivars and resistant cultivars were equally toxic and showed necrosis and wilting of the leaves. 4. Several other plants such as soybean, tomato and carrot were also affected with the toxin solution by shoot cutting bioassay and showed veinal necrosis, leaf spots and wilting of the shoots. 5. The acetone precipitation toxin affected seed germination of pepper, cucumber, sesame and egg plant and inhibited the growth of root and hypocotyl of the seedlings.
