Prevalence of porcine parvovirus in pigs with postweaning multisystemic wasting syndrome in Jeju Island

  • Postweaning multisystemic wasting syndrome (PMWS), which was first identified in western Canada in 1991 and more recently in the United States, Europe and Asia, is an emerging disease in pigs. Porcine circovirus type 2 (PCV-2) is the primary infectious viral agent causing PMWS, but the full expression of the disease may require the presence of other agents. It is reported that there is apparent synergism between PCV-2 and porcine parvovirus (PPV) in increasing the severity of the clinical signs and lesions of PMWS. From January 2006 to May 2008, a total of the 154 lymph node samples were collected from 4~12 weeks old pigs which had been submitted to the College of Veterinary Medicine, Jeju National University, Korea. These pigs were diagnosed as PMWS on the basis of clinical and pathological examination from 48 commercial herds in Jeju Island. Based on the immunohistochemistry, porcine parvovirus was detected in 69 cases (44.8%) from 154 weaned or grower pigs. PPV antigens were detected in the cytoplasm of histiocytic cells multifocally infiltrated in the cortex and paracortex of lymph nodes. The results of this study clarify that PPV is prevalent in pigs with PMWS on Jeju Island. Therefore PPV is one of the most important co-agents in the development of naturally acquired PMWS. This study may be helpful to the control of this disease and to epidemiological aspects.

  • From 1988 to 1989, 8 strains of canine parvovirus-2 (CPV-2) were isolated from the fecal specimens from the dogs that were clinically diagnosed as canine parvoviral enteritis in the veterinary hospitals located in the regions of Taejon and Chungbuk province. Studios on biological and physicochemical properties for the isolates were carried out. The results obtained by experiments are summarized as follows. 1. Among 62 fecal samples collected from the dogs with enteric diseases, 24 (38.7%) showed the haemagglutinating activity against porcine erythrocyte, ranging from 16 to 16, 384 of HA titers. 2. When 8 fecal specimens with high HA titer over 1, 000 were inocultated into CRFX cells, intranuclear inclusion bodies were obseverd in all of eight specimens, of w)lick three specimens showed cytoplasmic inclusions concurrently with the intranuclear inclusion bodies. 3. In study on species-specificity of haemagglutinating activity of the isolates, TJ-89-1 and TJ-89-2, it was found that the isolates revealed the highest haemagglutinating activity with porcine erythrocytes, showing the relatively lower haemagglutination titers with the erythrocytes from cat and rabbit. None of erythrocytes from other animals reacted with the isolates. 4. By the cross-haemagglutination inhibition test of the Isolates with reference viruses and sera, the Isolates were evidently identified as the strains of canine parvovirus-2. 5. In Physicochemical property test, it was evident that the isolates were stable in, lipid solvent, pH and heat treatment at $56^{\circ}C$ for 30 min. and contain DNA genome. 6. When seven puppies were inoculated intraorally with the isolate at HA titer of 8, 192, all of the puppies showed the symptoms of anorexia, vomiting, diarrhea and died at the 5th to 10th days post inoculation(pi). The fecal samples from all of the puppies revealed significantly high HA titers afterward the 5th days pi. Body temperature and the number of total leucocytes were slightly increased at the early stage of infection. but extremely decreased at the stage of collapse. HI titers of the sera started to increase at the 2nd to 3rd days pi reaching 512 to 1, 024 at the 4th to 5th day pi.

  • Importance: Canine parvovirus enteritis (CPE) is a contagious viral disease of dogs caused by the canine parvovirus-2 (CPV-2) associated with high morbidity and mortality rates. CPV-2 has a high global evolutionary rate. Molecular characterization of CPV-2 and understanding its epidemiology are essential for controlling CPV-2 infections. Objective: This study examined the risk factors and survival outcomes of dogs infected with CPV-2. Molecular characterization of CPV-2 genotypes circulating in Egypt was performed to determine the evolution of CPV-2 nationally and globally. Methods: An age-matched case-control study was conducted on 47 control and 47 CPV-infected dogs. Conditional logistic regression analysis examined the association between the potential risk factors and CPE in dogs. Survival analysis was performed to determine the survival pattern of the infected dogs. Thirteen fecal samples from infected dogs were collected to confirm the CPV genotype by CPV-2 VP2 gene sequencing, assembly of nucleotide sequences, and phylogenic analysis. Results: Unvaccinated and roamer dogs had eight and 2.3 times higher risks of CPV infection than vaccinated dogs and non-roamer dogs, respectively. The risk of death from CPE was high among dogs without routine visits to veterinary clinics and among non-roamer dogs. Molecular characterization of CPV-2 confirmed its genotype identity and relationship with the CPV-2 c and b clade types. Conclusions and Relevance: This study highlights the potential factors for CPE control, especially vaccination and preventing dogs from roaming freely outside houses. Isolated CPV genotypes are closely related to southern Asian genotypes, suggesting a substantial opportunity for global transmission.

국내 성견의 개 파보바이러스에 대한 항체가 조사 (Serum Parvovirus Antibody Titers among Healthy Adult Dogs in Korea)

  • This study was performed to monitor the level of serum canine parvovirus(CPV) antibody titers in adult dogs throughout the Korea from January 2003 to April 2004. A total of clinically healthy 885 dogs between 1 year and 17 years old were included in this study. Serum antibody titers against CPV were measured by means of hemagglutination inhibition(HI) titers at the time dogs were brought to the hospital for revaccination. Most of dogs had been primarily vaccinated or previously revaccinated. Dogs were grouped by age, breed, sex, and primary vaccination and revaccination to determine whether these factors were associated with antibody titers. Serum CPV titers ${\geq}80$ were considered protective. Protective antibodies against CPV were present in 95.0% of the population. Breed, age, and primary vaccination and revaccination were not significantly associated with serum CPV antibody titers. But sex was significantly associated with CPV antibody titers. The results of this study have shown that there is a need to reconsider the annual revaccination strategy against CPV infection.

한국에서 사육중인 모돈 및 30, 60, 90일령 돼지의 돼지파보바이러스에 대한 혈청학적 역학조사 (Sero-prevalence against porcine parvovirus in sows and 30-, 60-, 90-day-old pigs in Korea)

  • A total of 701 swine sera from 55 swine farms (Mar, 1998 through Feb, 2001) were nation-widely collected for the presence of antibody to porcine parvovirus (PPV) in sows and 30-, 60-, 90-day-old pigs. Sero-prevalence by haemagglutination inhibition assay with guinea pig red blood cells was investigated on the basis of year, region and season, respectively. In general, there was no significant difference with gradual decrease of passive immunity for the sero-prevalence to PPV in sows and 30-, 60-, 90-day-old pigs for the period of 1999 and 2000. However, regional variation was observed in the provinces of Kyonggi, Choongnam and Kyungnam, Natural infection of the virus in 90-day-old pigs was increased during the fall and the winter. Thus, it seems that the natural infection of PPV in growing pigs may be attributed to the increased outbreak of postweaning multisystemic wasting syndorme in co-infection with porcine circoviruses.

  • To elucidate the molecular genetical properties of the canine parvoviruses isolated from the diseased puppies in the regions of Kyunggi and Chungnam provinces, the replicative form (RF) DNA of four field isolates were compared with those of two attenuated vaccine strains and a reference strains of CPV by restriction endonuclease analysis (REA). REA by Hinf I showed that three CPV isolates except CPV-V15 had an identical banding pattern with two vaccine strains, one standard strain and feline panleukopenia virus (FPLV). In CPV-V15 strain the fourth fragment of DNA with 800 bp was deleted. REA by Bgl II and Pst I indicated that CPV-V15 and FPLV had a bigger second fragment than those of the other strains of CPV. Meanwhile REA by Bam HI revealed that all the field isolates and vaccine strains used in this experiment showed similar banding patterns.

In cytopathological studies with CRFK cells, intranuclear inclusion bodies were observed in all of eight specimens with the high HA titer over 1,000, of which three specimens showed cytoplasmic inclusions concurrently with the intranuclear inclusion bodies. It was found that the isolates revealed the highest haemagglutinating activity with porcine erythrocytes and the relatively lower haemagglutination titers with the erythrocytes from cat and rabbit. None of erythrocytes from the other animals reacted with the isolates. By the cross-haemagglutination inhibition test for the isolates with the reference viruses and sera, the isolates were evidently identified as the strains of CPV-2. In physicochemical property test, the isolates were stable in lipid solvent, pH and heat treatment at $56^{\circ}C$ for 30 min, and showed the virus particle size less than 25 nm, containing a DNA genome.

Development of Nested PCR Primer Set for the Specific and Highly Sensitive Detection of Human Parvovirus B19

  • For the specific detection of human Parvovirus B19 (HuPaV-B19), we designed ten specific PCR primers from 3,800~4,500 nucleotides of HuPaV-B19 complete genome (NC_000883.2). Seventeen candidate PCR primer sets for specific detecting HuPaV-B19 were constructed. In specific reaction of HuPaV-B19, seventeen PCR primer sets showed specific band, however five PCR primer sets were selected basis of band intensity, amplicon size and location. In non-specific reaction with seven reference viruses, four PCR primer sets showed non-specific band, however one PCR primer set is not. Detection sensitivity of final selective PCR primer set was $100fg/{\mu}L$ for 112 minute, and PCR amplicon is 539 base pairs (bp). In addition, nested PCR primer set was developed, for detection HuPaV-B19 from a low concentration of contaminated samples. Selection of nested PCR primer set was basis of sensitivity and groundwater sample tests. Detection sensitivity of final selective PCR and nested PCR primer sets for the detection of HuPaV-B19 were $100fg/{\mu}L$ and $100ag/{\mu}L$ basis of HuPaV-B19 plasmid, it was able to rapid and highly sensitive detection of HuPaV-B19 than previous reports. We expect developed PCR primer set in this study will used for detection of HuPaV-B19 in various samples.

  • The present study was conducted to characterize maternal antibody status which haemagglutination inhibition(HI) titers against canine parvovirus(CPV) in the 15 puppies delivered from 3 dams. The range of HI titers of 5 puppies delivered from a mother dog(A) with HI titer of 1 : 1,024 were 1 : 16~1 : 64 at 1 day old before suckling, 1 : 512~1 : 1,024 at 2 days old after suckling, 1 : 512~1 : 2,048 at 1 week old, 1 : 256~l : 1,024 at 2 weeks old, 1 : 128~l : 512 at 3 weeks old, 1 : 128~l : 256 at 4 weeks old, 1 : 32~1 : 128 at 5 weeks old, 1 : 16~1 : 64 at 6 weeks old, 1 : 16~1 : 64 at 7 weeks old, and 1 : 16~l : 32 at 8 weeks old. After vaccination with DHPPL to canine parvovirus in 60 days and 80 days old puppies, 1 : 8~l : 32 at 9 weeks old, 1 :16~1 : 128 at 10 weeks old, 1 : 32~1 : 256 at 11 weeks old, 1 : 16~1 : 256 at 12 weeks old, 1 : 128~1 : 256 at 13 weeks old, 1 : 64~l : 512 at 14 weeks old, and 1 : 128~1 : 512 at 15 weeks old. The HI titers of 3 puppies delivered from a mother dog(B) with HI titer of 1 : 512 were 1 : 16 at 1 day old before suckling, 1 : 256~1 : 512 at 2 days old after suckling, 1 : 512 at 1 week old, 1 : 128~1 : 256 at 2 weeks old, 1 : 64~1 : 128 at 3 weeks old, 1 : 64~1 : 128 at 4 weeks old, 1 : 128 at 5 weeks old, 1 : 64~1 : 128 at 6 weeks old, 1 : 16 at 7 weeks old, and 1 : 8 at 8 weeks old. The HI titers of 7 puppies delivered from mother dog(C) with Hl titer 1 : 1,024 were 1 : 512~1 : 1,024 at 2 days old after suckling, 1 : 256~1 : 1,024 at 1 week old, 1 : 256~l : 1,024 at 2 weeks old, 1 : 64~1 : 512 at 3 weeks old, 1 : 64~1 : 512 at 4weeks old, 1 : 8~l : 64 at 5 weeks old, 1 : 8~1 : 64 at 6weeks old, 1 : 8~1 : 32 at 7 weeks old, and < : 8~1 : 8 at 8 weeks old. Antibody to CPV was transferred mainly from mother to progeny through the colostrum and the transferred maternal antibody was in proportion to the HI titer of the mother As the HI titer of maternal antibody in puppies was low, puppies have a rapid immune response and a massive rise in HI titer to vaccination against CPV compared with puppies haying high level of maternal antibody.

Prevalence of Canine Influenza Infection in Pet Dogs and Canine Parvovirus Infection in Street Dogs of Bangladesh

  • A cross-sectional study was conducted to investigate the prevalence of canine influenza (CI) infection in pet dogs and canine parvovirus (CPV) infection in street dogs of different age and sex by collecting rectal and nasal swab samples from three districts, Dhaka, Mymensingh and Sirajgonj, in Bangladesh using a RapiGEN$^{(R)}$ Canine Influenza Virus Ag Test kit and RapiGEN$^{(R)}$ Canine Parvovirus Ag Test Kit. Out of 114 rectal swabs and 115 nasal swab samples, the overall prevalence of CI and CPV was found to be 11.30% and 32.45%, respectively. The prevalence of parvovirus infection was found to be significantly higher in puppies and dogs 6 months of age (50.0%) than those > 24 months of age (p = 0.005). The prevalence was also higher in males (34.42%) than females (30.18%). The prevalence of CI was higher (30.43%) in dogs up to 6 months of age (p = 0.011) than 6-12 month (7.93%) and 12-18 month (6.66%) old dogs. Moreover, the prevalence of CI was found to be higher in males (16.10%) than females (5.66%). The prevalence of CPV infection also varied significantly in different study areas (p = 0.0029), with 12.72%, 12.5% and 7.14% of dogs found to be CI positive in Dhaka, Mymensingh and Sirajgonj, respectively. Overall, the highest prevalence of CI was found in local breeds (6.08%) followed by German shepherds and Keeshonds (1.73%), and Bloodhounds and Terriers (0.86%). Additionally, there were more positive CI found in unvaccinated dogs (14.81%) than vaccinated (2.94%) dogs. Dogs with flu-like symptoms were more positive (19.23%) for CI relative to those without flu-like symptoms (4.76%) samples. Overall, the results of this study indicate that canine vaccination should be initiated to prevent the occurrence of diseases and that regular monitoring should be continued in Bangladesh.
