Development of the Pneumatic Manipulator of Gait Rehabilitation Robot using Fuzzy Control (퍼지제어기를 이용한 보행재활로봇의 공압식 조작기 개발)

  • Stable and comfortable walking supports, which can reduce the body weight load partially, are needed for the recovering patients from neurologic disease and orthopedic procedures. In this paper, the development of a manipulator of rehabilitation robot for the patients with walking disabilities are studied. A force controller using pneumatic actuators is designed and implemented to the human friendly rehabilitation robot considering the safety of patients, reliability of the system, effectiveness of the unloading control and economic maintenance of the system. The mechanism of the unloading manipulator is devised to improve the sensibility for the movement of the patients such as direction and velocity. For the unloading force control, fuzzy control algorithm is adopted to reduce the partial body weight and suppress the unwanted fluctuation of the body weight load to the weak legs due to the unnatural working of the patients with walking disabilities. The effectiveness of the force control is experimentally demonstrated.

Comparison of Size-Exclusion Chromatography and Flow Field-Flow Fractionation for Separation of Whey Proteins

  • Whey protein (WP) is a mixture of proteins, and is of high nutritional values. WP has become an important source of functional ingredients in various health-promoting foods. In this study, size-exclusion chromatography (SEC) and asymmetrical flow field-flow fractionation (AsFlFFF) were used for separation and analysis of whey proteins. It was found that a lab-prepared WP from raw milk is mostly of ${\beta}$-lactoglobulin with small amount of higher molecular weight components, while a commercial whey protein isolate (WPI) powder contains relatively larger amount of components other than ${\beta}$-lactoglobulin, including IgG and protein aggregates. Results suggest that AsFlFFF provides higher resolution for the major whey proteins than SEC in their normal operation conditions. AsFlFFF could differentiate the BSA and Albumin, despite a small difference in their molecular weights, and also was able to separate much smaller amount of aggregates from monomers. It is noted that SEC was able to show the presence of low molecular weight components other than the major whey proteins in the WP samples, which AsFlFFF could not show, probably due to the partial loss of those low molecular weight species through the membrane.

Effects of Hexavalent Chromium on Mitochondrial Respiration of Rat Kidney and Function of Low-Molecular-Weight, Chromium-Binding Substances ($Cr^{6+}$가 흰쥐 신장세포내 미토콘드리아의 호흡에 미치는 영향과 저분자 크롬결합물질의 작용)

  • To investigate the effects of hexavalent chromium on mitochondrial respiration of rat kidney, various hexavalent chromium concentrations were treated, then respiration and electron transfer enzyme activities were measured. Ultrastructural changes at state IV respiration of mitochondria were also observed. Then, to investigate protective role against hexavalent chromium in the body, low-molecular-weight, chromium-binding substances (LMCr) were purified from livers of rabbits 2hr after intravenously administrated with sodium dichromate at a dose of 74mg per kg body weight. And then, respiration rates of mitochondria treated with LMCr, hexavalent chromium containing 0.7mM chromium were measured. Hexavalent chromium decreased state IV respiration rates and electron transfer enzyme activities of mitochondria, and increased labile membrane and swelling. And partial inhibitions of condensed to orthodox conformational change were observed. Respiration rates of mitochondria treated with LMCr containing 0.7mM chromium did not differ from that of the non-treated mitochondria. But respiration rates of 0.7mM hexavalent chromium-treated mitochondria decreased by 42%, compared to non-treated mitochondria. These results suggest that LMCr may play an important role in detoxification of toxic hexavalent chromium.

Purification and Characterization of Phytoferritin

  • Ferritins from germinated pumpkin seeds were isolated by ammonium sulfate precipitation (0.55 saturation), ion-exchange chromatography on DEAE-cellulose, and gel filtration chromatographies on Sephacryl S-300 and Sephadex G-100. Pumpkin ferritin contains less iron than soybean ferritin. Pumpkin ferritin cross-reacted with anti-soybean ferritin antiserum made in rabbit, and showed two distinct antibody reactive bands, both of equal intensity. The pumpkin ferritins corresponding to the two bands were separable by centrifugation in a sucrose gradient (20~50%). The molecular weights of the native pumpkin ferritins based on the estimation of sucrose gradient centrifugation, gel filtration on Sephacryl S-300 and non-denaturing polyacrylamide gel electrophoresis appeared to be: 530~580 KD (the large molecular weight pumpkin ferritin) and 330-360 KD (the small molecular weight pumpkin ferritin) The large molecular weight pumpkin ferritin contains less iron. Both pumpkin ferritins cross-reacted with anti-soybean ferritin antibody with a spur formation suggesting partial antigenic recognition.

Effect of Methanethiol Administration on the Erythrocyte Damage in Rats (흰쥐에 Methanethiol 투여가 적혈구막 손상에 미치는 영향)

  • To evaluate an effect of methanethiol on a cause of erythrocyte membrane damage in rats, methanethiol was given at 11.25 rag/100 g body weight, and after 4 hr, the animals were sacrifled, the activities of Na$^+$/K$^+$ ATPase, protein contents in partial purified erythrocyte membrane and erythrocyte indices were determined Concomitantly, in vitro, effect of methanethiol on the erythrocyte fragility, Na$^+$/K$^+$ ATPase activity and its kinetics in various concentration of substrate from the preincubated erythrocyte membrane with methanethiol were demonstrated. The spleen weight per body weight (%) and MCV of erythrocyte in methanethiol-treated rats were more increased than those in the control group. The Na$^+$/K$^+$ ATPase activities in erythrocyte membrane were more decreased in methanethiol-treated rats than those in the control group. The apply of 0.05 ng rat whole blood to the 0.24 mg/ng of methanethiol solution in isotonic condition showed the complete hemolysis. The Na$^+$/K$^+$ ATPase activity in preincubated erythrocyte membrane with methanethiol at 37$\circ$C showed the dual effect and the K$_m$ value of Na$^+$/K$^+$ ATPase was higher in the preincubated erythrocyte membrane with methanethiol than that in the preincubated erythrocyte membrane omitted the methanethiol. These results suggest that the methanethiol may induce the damage of rat's erythrocyte membrane due to a change in substrate binding affinity of Na$^+$/K$^+$ ATPase.

  • The pine bark from mechanical pulp industry was cooked with bisulfite liquor, and the chemical and physical properties of the spent liquor after cooking for 90 minutes, which allowed partial defibration were determined. Lignosulfonate or lignin-related materials in the spent liquor have a molecular weight of approximately 3,000, which is lower than that of commercial lignosulfonate (C-LS) prepared from wood. The sulfonic equivalent weight calculated by colloid titration was 307 for C-LS, 238~239 for spent liquor. The contents of lignosulfonic acid and phenolic hydroxyl groups on $C_9$ unit of lignin in the spent liquor were 0.98~0.99, and 1.51~1.66, respectively. These values were remarkably higher than those of C-LS. Especially the content of phenolic hydroxyl group was almost twice that of C-LS. The viscosity of spent liquor after freeze drying was very low compared to C-LS. The dispersing ability of spent liquor was almost equivalent to or more than that of C-LS.

Total Ankle Arthroplasty Management and Rehabilitation (족관절 인공관절 치환술 후 관리 및 재활)

  • Although total ankle arthroplasty (TAA) has increased considerably in the past ten years, reflecting improvements in implant design and survivorship, the clinical outcomes have been less satisfactory than total hip or total knee arthroplasties. Several issues under debate include postoperative management and rehabilitation in TAA. Especially, there is no consensus or evidence for the most appropriate postoperative management and rehabilitation for patients undergoing TAA. This study was therefore undertaken to suggest appropriate postoperative management and rehabilitation in TAA, after reviewing published articles and focusing on the following topics: prehabilitation, hospital stay, immobilization type and duration, weight-bearing management, pharmacological treatment, and adopted rehabilitation protocols. In previous studies, the postoperative management and rehabilitation proposed depended on the surgeon's preference, the patient's characteristics, and the associated surgical procedures performed after TAA. Nonetheless, our research indicates the best approach is to include a prehabilitation program, immobilization in the early postoperative stage (2~4 weeks), range of motion exercise with partial weight-bearing ambulation, followed by full weight-bearing ambulation after six weeks. Further studies are required to develop a standardized rehabilitation protocol and improve the overall quality of care after TAA.

Substitution of Fish Meal by Mealworm (Tenebrio molitor) in Practical Diets for Pacific White Shrimp, Litopenaeus vannamei

  • Mealworm (Tenebrio molitor) was evaluated to investigate the effect of partial or total replacement of fish meal in diets for white shrimp, Litopenaeus vannamei. Experimental groups of shrimp with average initial body weight (2.43 ± 0.54 g) were fed each with 4 isonitrogeneous (38% crude protein) diets formulated to include 0, 25, 50 and 100% (diets 1 to 4, respectively) of fish meal substituted with mealworm. After eight weeks of feeding trials, shrimp fed with diet 3 and 4 revealed the highest values for live weight gain(8.01 ± 2.51 and 7.93 ± 1.12), specific growth rates (2.70 ± 1.12 and 2.59 ± 0.51) as well as better feed conversion ratio (2.69 ± 0.09 and 2.72 ± 0.19) compared to the control group with statistically significant manner (p<0.05). Survival range was 98% in all the treatments. An increase in weight gain and other growth associated parameters was observed with higher replacement. These results clearly indicate that 50% and 100% of fish meal protein in shrimp diet can be replaced by mealworm not only without any adverse effect but also the effect of promoting growth performance.

Effects of Carcass Traits on Auction Price in Hanwoo (한우의 경락가격에 대한 요인별 기여도 분석)

  • The aim of this study was to estimate the effect of sex, slaughtered season and year on carcass traits, and the contribution of carcass traits to auction and total prices. The data used in this study were the carcass grade of Hanwoo (4,231 heads) slaughtered from 2008 to 2010. Carcass traits were affected significantly (p<0.01) by sex, slaughtered season and year. The R-Square of auction prices, analyzed the contribution of causes using the squared semi-partial correlation, was 0.55. The contributions of auction prices of backfat thickness, eye muscle area, carcass weight and marbling score were 2.66, 0.32, 3.48 and 93.54%, respectively. The R-Square of total price was 0.82, and the contributions of marbling score and carcass weight were 46.25% and 52.38%.

Effects of Curculiginis Rhizoma on anti-thrombotic activity (선모(仙茅) 열수(熱水) 추출물(抽出物)의 항혈전(抗血栓) 효능 연구)

  • Objectives : An aim of study is to investigate effects of curculiginis rhizoma in vitro (factor Xa (FXa) inhibitor assay, prothrombinase assay, prothrombin time (PT) assay, activated partial thromboplastin time (aPTT) assay) and in vivo experiment (blood clotting time, thromboxane B2 content assay in serum and weight of thrombus by AV-shunt rat model). Methods : We gained a human serum and used serum in vitro study such as factor X activity (FXa) inhibition, prothrombinase inhibition, prothrombin time (PT) and activated partial thromboplastin time. Fifteen SD rats were divided into three groups (intact control group and two experimental group treated with extract of Curculiginis Rhizoma(ECR)). Rats were orally administrated DW (intact control group), 600 mg/kg concertration of ECR and 200 mg/kg concertration of ECR. After one hour, we anesthetized rats and made arteriovenous (AV) shunt rat models to study weights of thrombus, took a hole blood to study content of thromboxane B2 and blood clotting time. Results : In vitro, ECR increased a inhibitory activity of FXa, prothrombinase and aPTT compared than intact control group. Especially ECR made significant increase of FXa and prothrombinase inhibitory activity (p<0.05, p<0.01). And PT were increased in ECR control group compared with intact control group. In vivo, a blood clotting time of experiment group treated with ECR 600 mg/kg were significantly increased compared with that of intact control group (p<0.05) and content of thromboxane B2 was significantly decreased in group treated with ECR 600 mg/kg in seum. The weight of thrombus were significantly reduced in group treated with ECR 600 mg/kg compared with intact control group (p<0.05). But in vivo experiment study, those of group treated with ECR 200 mg/kg were reduced compared with those of intact control group without statistical significance. Conclusions : ECR has a antithromboic activity in internal course with inhibitory activity of FXa and prothrombinase in vitro, it required to research more study for effective compounds.
