소나무속내 속생 잎의 수가 1개인 종류와 한 개체에서 2~3개의 속생 잎 수를 갖는 종류의 기원을 밝히고자 ITS DNA 지역의 염기서열을 조사하였다. 또한 속생 잎 수 변이가 출현하는 지역에서 생육하는 소나무, 리기다소나무 그리고 잣나무 등의 동일지역 염기서열을 비교 조사하였다. 확인된 ITS1, 5.8S 그리고 ITS2 DNA 등 3지역의 총 길이는 종류에 따라서 580~584 염기이었으며, ITS1 지역에서 가장 변이가 크게 나타났다. 5.8S 지역은 잣나무의 2개 염기 치환을 제외하면 조사된 모든 종류에서 일치하였다. 조사된 일부 ITS1 지역은 5.8S 위쪽으로 종에 따라 181~185 염기이며, 1개 또는 2~3개의 속생 잎 수를 갖는 변이들은 소나무와 동일한 염기서열로 확인되었다. ITS2 지역은 모두 237 염기이며, 소나무와 잎 변이들의 염기서열은 일치하였다. 확인된 염기서열을 이용하여 유집분석을 수행한 결과는 소나무와 속생 잎 수 변이들이 유사도 100%로 유집되었다. 따라서 조사된 속생 잎 수 변이들은 소나무의 속생 잎 수 변이로 최종 판별되었다.
본 논문은 영한 기계번역을 위한 예제기반 기계번역에서 예제 문장의 비교를 위한 척도에 관한 것으로 주어진 질의 문장과 가장 유사한 예제 문장을 찾아내는데 사용되는 유사성 척도를 제안한다. 제안하는 척도는 편집거리 알고리즘에 기반을 둔 것으로 표면어가 일치하지 않는 단어에 대해 기본적으로 단어의 표제어 정보와 품사 정보를 이용하여 유사도를 계산한다. 편집거리 척도는 비교 단위의 순서에 의존적이기는 하지만 순서만 일치하면 동일한 유사성 기여도를 갖는 것으로 판단하기 때문에 완전 문맥을 반영하지는 못한다. 따라서 본 논문에서는 완전 문맥 반영을 위해 추가적으로 이들 정보 외에 일치하는 단위 정보를 갖는 연속된 단어들에 대해 연속 정보를 반영한 문맥 가중치를 제안한다. 또한 비유사성 정도를 의미하는 척도인 편집거리 척도를 유사성 척도로 변경하고, 문맥 가중치가 적용된 척도를 문장 비교에 적용하기 위하여 정규화를 수행하며, 이를 통하여 유사도에 따른 순위를 결정한다. 또한 언어적 정보를 이용한 기존 방법류들에 대한 일반화를 시도하였으며, 문맥 가중치가 적용된 척도의 우수성을 증명하기 위해 일반화된 기존 방법류들과의 비교 실험을 수행하였다.
배추의 약(anther) 발달에 관여하는 유전자를 분리, 특성을 구명하고, 궁극적인 목표로는 특이 발현을 조절하는 프로모터를 분리 작물의 웅성불임 유기 및 임성화복에 응용하고자 본 연구를 수행하였다. 배추 약 cDNA 유전자 은행으로부터 차별화 선별(differential screening)에 의해 얻어진 15개의 약특이 유전자들의 부분 염기서열 결정 후 기존에 보고된 유전자들과 상동성 검색을 통해 그 기능을 추정해 본 결과 cDNA clone BAN52는 polygalacturonase, BAN84는 ascorbate oxidase, BAN101은 $H^+-translocating$ ATPase, BAN2는 pectin esterase 유전자와 상동성을 보였으나, 그 외 유전자들은 상동성이 없어 그 기능을 추정할 수 없었다. 상기 유전자들의 발현시기를 꽃 발달 시기별로 조사한 결과 대부분의 유전자들이 (BAN5, 10, 33, 52, 57, 102, 103, 215, 229) 꽃봉오리 2.1mm 내외에서 발현이 시작하여 성숙화분까지 그 발현양이 증가하였고, BAN32, 54, 62, 84, 101는 꽃봉오리 3.9mm인 약발달 후기부터 나타났고, BAN87 유전자는 그 양은 적지만 약 발달 초기부터 후기까지 발현하였다.
조류 인플루엔자바이러스(AIV) H5N1 아형을 Real-time PCR법을 이용하여 가장 빠르게 진단할 수 있는 방법을 개발하였다. 검색 대상의 염기서열은 AIV H5N1 아형의 hemagglutinin 유전자 중 가장 상동성이 높은 387 bp의 부위를 선택하였고, 실험의 안전을 위하여 인공합섬의 방법으로 제작하였다. Microchip을 기반으로 한Real-time PCR법을 사용하였으며, 총PCR 반응액의 양을 $1{\mu}l$로, PCR 과정 중 각 단계, 즉 해리, 접합, 신장의 시간을 각1초, 1초, 3초로 하여 총 실험시간을 단축하였다. 진단을 위한 실험과정에서 PCR 및 융점분식에 소요된 최단 시간은 12분28초였으며, 민감도측정에서 최소2.4개의 hemaggutinin 유전자를 기질로 하여 목적한 특이 189 bp의 PCR 산물을 증폭할 수 있었기에, 본 연구에서는 이런 초고속 PCR 실험방식을 Quick Real-time PCR이라 명명하였다. 이 결과들은 가금류 및 사람에게 전파된 AIV H5N1아형의 진단에 적용될 수 있을 뿐 아니라, PCR이 사용되는 다른 신속검색법에도 널리 적용 될 수 있을 것으로 기대한다.
In linear discriminant analysis there are two important properties concerning the effectiveness of discriminant function modeling. The first is the separability of the discriminant function for different classes. The separability reaches its optimum by maximizing the ratio of between-class to within-class variance. The second is the stability of the discriminant function against noises present in the measurement variables. One can optimize the stability by exploring the discriminant variates in a principal variation subspace, i. e., the directions that account for a majority of the total variation of the data. An unstable discriminant function will exhibit inflated variance in the prediction of future unclassified objects, exposed to a significantly increased risk of erroneous prediction. Therefore, an ideal discriminant function should not only separate different classes with a minimum misclassification rate for the training set, but also possess a good stability such that the prediction variance for unclassified objects can be as small as possible. In other words, an optimal classifier should find a balance between the separability and the stability. This is of special significance for multivariate spectroscopy-based classification where multicollinearity always leads to discriminant directions located in low-spread subspaces. A new regularized discriminant analysis technique, the principal discriminant variate (PDV) method, has been developed for handling effectively multicollinear data commonly encountered in multivariate spectroscopy-based classification. The motivation behind this method is to seek a sequence of discriminant directions that not only optimize the separability between different classes, but also account for a maximized variation present in the data. Three different formulations for the PDV methods are suggested, and an effective computing procedure is proposed for a PDV method. Near-infrared (NIR) spectra of blood plasma samples from mastitic and healthy cows have been used to evaluate the behavior of the PDV method in comparison with principal component analysis (PCA), discriminant partial least squares (DPLS), soft independent modeling of class analogies (SIMCA) and Fisher linear discriminant analysis (FLDA). Results obtained demonstrate that the PDV method exhibits improved stability in prediction without significant loss of separability. The NIR spectra of blood plasma samples from mastitic and healthy cows are clearly discriminated between by the PDV method. Moreover, the proposed method provides superior performance to PCA, DPLS, SIMCA and FLDA, indicating that PDV is a promising tool in discriminant analysis of spectra-characterized samples with only small compositional difference, thereby providing a useful means for spectroscopy-based clinic applications.

불로초의 계통 분류학적 조사를 위해 불로초속 중 8종(Ganoderma lucidum, G. tsugae, G. pfeifferi, G. resinaceum, G. australe-applanatum, G. oregonense, G. neo-japonicum, G. applanatum) 12균주와 out-group 균주로는 Inonotus xeranticus의 rDNA ITS II를 PCR로 증폭하여 염기서열을 비교 조사하였다. 불로초 속 중 ITS II 영역의 길이는 $247{\sim}257\;bp$로 분포하였고 종간 상동성은$70{\sim}100%$ 로 조사되었다. 계통도를 분석한 결과는 5개의 군(cluster)을 형성하였고 G. tsugae는 각각의 G. lucidum과 하나의 clade를 이루어 두 종은 서로 매우 밀접하게 진화된 것으로 사료되었다. 본 조사에서는 국내에서 재배되고 있는 불로초(G. lucidum)는 그 발생지 및 국외 ITS염기서얼 자료와 비교 및 분석할 때 G. tsugae로 하는 것이 타당할 것으로 사료되었다.
한국 마늘에 감염된 바이러스의 종류와 병 발생 메카니즘을 구명하기 위하여, 마늘 바이러스 cDNA clone들을 분리하였다. 24개 cDNA clone들의 부분적인 염기 서열을 결정하였고, 이 중 poly(A) tail을 가진 5개 clone들의 염기 서열을 결정하였다. 이를 이미 알려진 다른 식물 바이러스와 비교했을 때, clone V9은 일차구조가 carlavirus와 유사성을 보이므로 GLV cDNA clone으로 여겨진다. Northern blot 결과로부터 GLV genome의 크기는 8.5 knt이고, poly(A) tail을 가지고 있다는 것을 알 수 있었다. clone V9의 3' 말단부분에는 바이러스 복제과정에서 cis-acting element로 작용한다고 여겨지는 hexanucleotide motif(5'-ACCUAA)가 존재한다. 또한 carlavirus의 껍질 단백질 subgenomic RNA의 5' 말단에 보존되어 있는 5'-TTAGGT도 나타난다. 이들은 모두 carlavirus의 특징들이다. 껍질 단백질 유전자를 pRSET-A 발현 벡터에 재조합하고, E. coli BL21에서 발현시켰다. 발현된 껍질 단백질을 $Ni^{2+}$ NTA affinity chromatography에 의해 정제하였다. 껍질 단백질을 토끼에 주사하여 항체를 만든 후, immunoblot을 한 결과 GLV 껍질 단백질에 해당하는 24 kDa polypeptide가 인지되었다. 또한 다양한 마늘 품종에 대해서 immunoblot을 한 결과, GLV 껍질 단백질의 크기와 GLV의 감염정도가 마늘 품종에 따라서 차이가 있다는 것을 알 수 있었다.

Gabapentin, 1-(aminomethyl) cyclohexaneacetic acid, is a amino acid derivative, and is clinically effective in the treatment of neuropathic pain and partial seizures of epilepsy as a complementary therapy. The purpose of the present study was to evaluate the bioequivalence of two gabapentin tablets, $Neurontin^{R}$ tablet 800 mg (Pfizer Pharmaceuticals Co., Ltd.) and Gabapenin tablet 800 mg (Hanmi Pharm. Co., Ltd.), according to the guidelines of the Korea Food and Drug Administration (KFDA). The release of gabapentin from the two gabapentin formulations in vitro was tested using KP VIII Apparatus II method with 0.06 M HCI dissolution media. Twenty six healthy male subjects, $23.85{\pm}2.24$ years in age and $69.40{\pm}11.11$ kg in body weight, were divided into two groups and a randomized $2{\times}2$ crossover study was employed. After a single tablet containing 800 mg as gabapentin was orally administered, blood samples were taken at predetermined time intervals and the concentrations of gabapentin in serum were determined using HPLC with fluorescence detector. The dissolution profiles of two formulations were similar in the tested dissolution media. The pharmacokinetic parameters such as $AUC_{t}$, $C_{max}$ and $T_{max}$ were calculated, and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_{t}$, $C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the reference drug, $Neurontin^{R}$, were 1.28%, 0.63% and 0.62% for $AUC_{t}$, $C_{max}$ and $T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log 0.8 to log 1.25 (e.g., $log0.9097{\sim}log1.1598$ and $log0.8919{\sim}log1.1262$ for $AUC_{t}$ and $C_{max}$, respectively). Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Gabapenin tablet 800 mg was bioequivalent to $Neurontin^{R}$ tablet 800 mg.
