Microscopic Observation of Moso bamboo(Phyllostachys pubescens Mazel) with Various Ages (맹종죽의 죽령별 조직 현미경적 관찰)

  • To obtain the basic information for the use of bamboo, the anatomical characteristics of moso bamboo shoots, and 20-day-old, 60-day-old, one-year-old, and two-year-old moso bamboo were observed by using polarizing microscope. The cross section showed that the bamboo shoots consists of the early stage of cell formation, atactostele, parenchyma, and bundle sheath was formed in 20-day-old moso bamboo, and all cells completely formed in 60-day-old moso bamboo. The tissues in one-year-old moso bamboo appeared to be completely matured. On the tangential surface, no atactostele was observed in the bamboo shoot, but metaxylem, parenchyma, and bundle sheath were found. atactostele and parenchyma appeared to be formed between 20 and 60 days. All tissues and cells were totally formed and the cell wall of parenchyma were completely matured after one year. The metaxylem in bamboo shoots was observed to have pits, which means that it could be the tissues first formed of all bamboo cells.

  • Morpho-histogenesis of fruit sculpture and dehiscence in Thespesia populnea is described. The fruit wall is differentiated into epicarp, mesocarp and endocarp. The epicarp is stony, rind-like, 30 to 35 layers thick and derived from outer epidermis, sub-epidermis and ground parenchyma of the ovary wall. The spherical and/of tangentially elongated, thick walled cells of epicarp are interspersed with radial bands of sclereids. The mesocarp is a product of the inner zone of ground parenchyma. At maturity 20 to 25 layers of thin walled parenchyma of mesocarp appear sinuous of disorgnized. The innermost 1 to 3 layers of ground parenchyma and sub-epidermis and inner epidermis form 35 to 40 layers thick endocarp. Due to the differentiation of fibrous tissue in the projection of median plane of carpel wall and a complete ring of fibrous zone in the endocarp, the dry capsule of Thespesia populnea dehises partially in loculicidal fashion.

Effect of Ensiling with Acremonium Cellulase, Lactic Acid Bacterial and Formic Acid on Tissue Structure of Timothy and Alfalfa

  • The changes of tissue structure in timothy and alfalfa during ensiling process with silage additives; lactic acid bacteria, cellulase and formic acid, were observed with a video microscope. Stem samples were obtained from the second internode, and cut to divide into 2 pieces. One piece was for observation of ensiled material and the other was for silage. The latter piece was put into a nylon cloth bag, and ensiled with grass for 50 days in a small experimental silo Lignification of the plant tissues was checked by acid phloroglucinol. Natural silage fermentation resulted in some degradation of less lignified parenchyma in both plant species. However, lignified sclerenchyma and vascular bundles remained intact. The cellulase enhanced the degradation of parenchyma tissue, while the formic acid suppressed the degradation. The effect of lactobacillus was small. The percentage of remained cross sectional area of stem and the loss of NDF and ADF by silage fermentation confirmed the observation. High negative correlations were obtained between the remained area and loss of fibrous components during silage fermentation in both plants, and between the loss of fibrous components and in vitro dry matter digestibility in timothy but not in alfalfa.

  • The papillary tubercles (PTs) developed on the splenic capsule of normal Landrace pigs were collected and their histological structures were observed with light microscope. The results were summarized as follows : 1. The external features of the PTs were smooth spherical or oval form protruded on the splenic capsules. On cross section of PTs, the shapes were predominantly round or elliptical single follicular form, and were often multifollicular and irregular form in some PTs. 2. The PTs were interposed into the splenic capsule. Therefore the peripheral boundary of PT was consisted of splenic capsular tissue and this tissue was covered with mesothelium, The basal tissues of PT were consisted of thick connective tissue and smooth muscle of splenic capsule, and capsular foramen for transport tract between splenic parenchyma and the PT was found at the center of the basal boundary of PT. 3. The basal region of PT was composed of parenchyma and this tissue was the splenic red pulp but the central and peripheral regions of PT contained much more erythrocytes than in the splenic parenchymae. 4. The splenic parenchymae adjoining to PT contained more erythrocytes than in other splenic parenchymal regions and parallel fixed cells directed to the capsular foramen.

  • Haploid cell obta-ined from microspores of Solanum nigrum were cultured on two kinds of medium, "Callus-inducing medium" and "Differentiation medium", in order to conduct histological studies of callus and examine differentiation of plantlets. On the callus-inducing medium the calli grew rapidly. The bulk of callus mass was light brown colored "Wet callus" covered on the surface with thin layers of rough and gleaming "White callus". The wet callus was consisted of parenchyma and meristematic tissues, while the white callus had no meristematic tissues. Large parenchyma cells, by successive divisions, became multicellular or poly nucleate cells which developed later to be meristematic tissues. The calli embedded on the differentiation medium quickly turned to dark brown color. Plantlets, however, came out later from these blackened callus mass. In the callus sectioned about ten weeks after imbedding on the differentiation medium, radially elongated tissue, concentric tissue, epidermis, tracheid-like structure, and plant jprimordia were observed.ure, and plant jprimordia were observed.

  • Developmental pulmonary abnormalities are known as rare condition. diagnosis was made at autopsy in the early cases reported, however, as diagnostic aids such as X-ray, bronchography, bronchoscopy and exploratory thoracotomy have come into use, the condition is being discovered more often recently in living persons, and it appears to occur with sufficient frequency to merit consideration in the differential diagnosis of certain chest conditions. According to Schneider and Boyden there are three main types of this abnormality: [1] Agenesis, in which there is complete absence of one or both lungs; there is no trace of bronchial or vascular supply or of parenchymal tissue. [2] Aplasia, in which there is suppression of all but a rudimentary bronchus which ends in a blind pouch; there are no vessels or parenchyma. [3] Hypoplasia, in which the bronchus is fully formed but is reduced in size and ends in a _ flesh structure which usually lies within the mediastinum. Rudimentary pulmonary parenchyma may be present around the bronchial stump and often is the site of cystic malformation. We experienced one case of hypoplastic lung with cystic malformation which was originated from a small aberrant rudimentary bronchus, and the rudimentary bronchus was branched from the right side of tracheal end. The diagnosis was finally confirmed by the histopathological finding. Now, we report this case with a brief review of literatures.

Comparative Anatomy of the Secondary Xylem in the Stem of Malvales Plants in Korea (한국산 아옥목 식물 줄기에서 이기목부인 비교해부)

  • Anatomical study of the secondary xylem in Malvales plants, including four families, five genera and ten species grown in Korea, was carried out to elucidate relationship among genera or families in the order. Wood of Elaeocarpus is diffuse porous and shows angular vessels in radial multiples of 2-14 cells and a few apotracheal or paratracheal parenchyma. Tiliaceous genera have diffuse porous wood, vessels in solitary distribution and apotracheal parenchyma of sinuous scalariform uniseriate band. In the family, Tilia shows angular vessel. noded and tile-like cell in ray and storied tissue but Grewia has circular vessel. Hibiscus shows ring porous wood, circular solitary vessel and biseriate band of apotracheal and paratracheal parenchyma. Firmiana shows ring porous wood, circular solitary vessel and confluent parenchyma. Many starch grains appear in ray and axial parenchyma. Judging from arrangement, shape, length and diameter of vessel element and angle of end wall to vessel axis, and arrangement and shape of axial parenchyma, the lines of specialization in these genera are from primitive Elaeocarpaceae through less primitive Tiliaceae and less advanced Malvaceae to advanced Sterculiaceae.iaceae.

Studies on the Ray Parenchyma of Salicaceae in Korea (한국산(韓國産) 버드나무류(類)의 방사조직(放射組織)에 관한 연구(硏究))

  • These studies were carried out to identify anatomical characteristics of ray parenchyma of xylem tissue of trunk in 6 species of Populus and 5 species of Salix which were grown in Korea. The results of these experiments were summarized as follows: In the type of ray parenchyma, Populus had uniseriate homogeneous ray tissue and Sahx had uniseriate heterogeneous ray tissue. Upright ray cells among uniseriate heterogeneous ray tissue in Salix were subdivided into rectangular type and square type. The minimum and maximum length of procumbent ray cells of Populus ranged $26.84-212.28{\mu}$ and those of Salix were $46.36-170.80{\mu}$. However rectangular type of upright ray cell n Salix were $26.84-70.76{\mu}$ and square type were $17.08-43.92{\mu}$. The minimum and maximum width of procumbent ray cells of Populus ranged $12.20-24.40{\mu}$ but those of Salix were $12.20-26.84{\mu}$. However, rectangular type of upright ray cell in Salix were $9.76-41.48{\mu}$ and square type were $19.52-46.36{\mu}$. The height of ray parenchyma of Populus in tangential section ranged $65.88-414.80{\mu}$ but Salix were $65.88-439.20{\mu}$. Ray parenchyma width of Populus ranged $4.88-24.40{\mu}$ but those of Salix were $7.32-21.96{\mu}$. The number of ray parenchyma cells of Populus in tangential section were 3-26 cell, but Salix were 2-21 cells.

Histological Changes of Tissues and Cell Wall of Rice Straw Influenced by Chemical Pretreatments

  • Sodium hydroxide (SH) or ammonium bicarbonate (AB) were applied to rice straw to investigate the effects on histological change of stem tissue or cell wall before and after in sacco degradation using a scanning electron microscope (SEM) and a transmission electron microscope (TEM). The SEM revealed that, the parenchyma and vascular bundles were distorted by treatment with SH at 30 or 45 g/kg straw dry matter. Faultage between phloem of large vascular bundles and parenchyma occurred with further increasing SH to 60 or 75 g/kg. The cell wall in these stem tissues was crimped when observed by TEM. However, only parenchyma and large vascular tissues were slightly distorted in AB-treated stem. For untreated and AB-treated stems, the initiation of observable ruminal degradation of cell wall was prolonged from 12 h for inner parenchyma to 24 h for sclerenchyma and to 48 h for phloem of small vascular bundles, while the outer epidermis was intact even at 72 h. For SH-treated stem, however, the cell wall from all of the investigated tissues, epidermis, small vascular bundles, sclerenchyma, and parenchyma started to be degraded at 12 h incubation. These results indicate that SH treatment contracts rice straw stem leading to an improvement in rumen degradation, and that the degradation of SH-treated stem is bilateral from inner and outer surface simultaneously.

An Experimental Study on the Changes of Liver Tissue by the Administration of Anticlonorchial Drugs to Rabbits with Clonorchiasis (간(肝)디스토마증(症) 치료약투여후(治療藥投與後) 간장(肝臟) 변화(變化)에 관(關)한 실험적(實驗的) 연구(硏究))

  • An experimental study was done on rabbits to observe the effects of several anticlonorchial drugs on the pathology of the liver infested with Clonorchis sinensis. After two months of infestation with Clonorchis sinensis by giving $400{\sim}500$ metacercariae by mouth, hexachlorophene, chloroquine 2,2' methylenebis (3,4,6 trichlorophenoxy acetic acid) and Hetol were administered orally and follow up macro-and microscopic studies of the liver pathology were done in 2 to 3 days, one month, 2 months and 3 months after the completion of medications. The results obtained in this study are as follows: 1. In both groups which were administered hexachlorophene piperazine 20mg/kg for seven days or 8mg/kg for 18 days, the macroscopic findings of the liver after 3 months revealed only minimal changes of the color and consistency The histopathological findings were the reduction of fibrosis, pseudolobulation, proliferation and adenomatous hyperplasia of bile ducts, and regeneration of liver parenchyma. 2. In groups which were administered chloroquine phosphate 20mg/kg for 18 days or 40mg/kg for 5 days, and also in groups which were administered dithiazanine iodide 30mg/kg for 18 days or 60mg/kg for 5 days, no significant findings of recovery were observed either macroscopically or microscopically. 3. In the group which was given 20mg/kg of 2,2' methylenebis(3,4,6 trichlorophenoxy acetic acid) for 5 days, prominent healing of the damaged tissues was observed after 2 months, revealing the decrease of fibrous tissue, caliber of bile ducts and adenomatous hyperplasia of the epitherial cells of the bile ducts, and regenerationof liver parenchyma. 4. In the group which was given Hetol 200mg/kg for 5 days, swelling, congestion and eddish-brown discoloration of the liver were noted macroscopically after 3 days of completion of drug administration. Hemorrhage, congestion, necrosis and degeneration of the parenchyma were observed microscopically After 10 days, liver appeared almost normal macroscopically, but marked fat degeneration was noted microscopically. After 2 months, the liver was almost normal in gross appearence with only slight atrophy and also marked healing was observed microscopically, i. e. decrease of fibrous tissue and reduction of the previously enlarged bile duct and the regeneration of the liver parenchyma.
