High Resolution Melting Analysis for Epidermal Growth Factor Receptor Mutations in Formalin-fixed Paraffin-embedded Tissue and Plasma Free DNA from Non-small Cell Lung Cancer Patients

  • Background:The aim of the research was to explore a cost effective, fast, easy to perform, and sensitive method for epidermal growth factor receptor (EGFR) mutation testing. Methods: High resolution melting analysis (HRM) was introduced to evaluate the efficacy of the analysis for dectecting EGFR mutations in exons 18 to 21 using formalin-fixed paraffin-embedded (FFPE) tissues and plasma free DNA from 120 patients. Results: The total EGFR mutation rate was 37.5% (45/120) detected by direct sequencing. There were 48 mutations in 120 FFPE tissues assessed by HRM. For plasma free DNA, the EGFR mutation rate was 25.8% (31/120). The sensitivity of HRM assays in FFPE samples was 100% by HRM. There was a low false-positive mutation rate but a high false-negative rate in plasma free DNA detected by HRM. Conclusions: Our results show that HRM analysis has the advantage of small tumor sample need. HRM applied with plasma free DNA showed a high false-negative rate but a low false-positive rate. Further research into appropriate methods and analysis needs to be performed before HRM for plasma free DNA could be accepted as an option in diagnostic or screening settings.

  • Mycoplasma hyopneumoniae is among the most prevalent and important infectious agents associated with porcine respiratory disease complex. The airway dagame caused by M. hyopneumoniae adversely affect the pulmonary host defense mechanisms and may lead to secondary bacterial infections. Culture is considered to be the "gold standard" for diagnosis but this is a very slow and labor-intensive procedure. Isolation of M. hyopneumoniae is complicated by its fastidious nature and extremely slow growth. Thirty days of incubation may be necessary to detect the organism in primary broth cultures. The purposes of the study were (ⅰ) to develop nested PCR for the detection of M. hyopneumoniae for the detection of M. hyopneumoniae DNA in the formalin-fixed, paraffin-embedded lung tissues from experimentally and naturally infected pigs and (ⅱ) to compare the utility of nested PCR with in situ hybridization.   • This paper has dealt with the heat storage characteristics of gel type micro size latent heat storage material slurry. The heat release operation to the gel type micro size latent heat storage material slurry was carried out using hot air bubbles by direct contact heat exchange. This experiment was carried out using phase change material of n-paraffin so the heat release amount is higher than cold water system. The parameters of this experiment were concentration of latent heat phase change material, height of heat release bath and inlet velocity of hot air. The main results obtained are as follows : (1) The effect of concentration of latent heat phase change material dispersed with water is very affective to the direct contact heat exchange between hot air and gel type micro size latent heat storage material slurry. (2) It is clarified that the most effective concentration of latent heat phase change material dispersed with water exists around 20mass% at this type of direct heat exchange model experiment.

경유 대체연료로서 수첨바이오디젤의 윤활 특성 연구 (Lubricity Characterization of Hydrogenated Biodiesel as an Alternative Diesel Fuel)

  • Paraffin bio-based hydrotreated biodiesel(HBD) is originated from vegetable oil(the process can also be applied to animal fat) with the the chemical structure $C_nH_{2n+2}$. In the number of process of the oil or fat, the hydrogenation is significantly important to create a bio-based diesel fuel. This study is focused on lubricity characteristics of BTL diesel blends to use alternative diesel fuel in Korea. The BTL diesel are blended the different volume ratios (HBD 5(5 vol.% HBD - 95 vol.% diesel), HBD 10, HBD 20, HBD 30, HBD 40 and HBD 50. HBD with paraffin compounds showed a very high centane number, low sulfur content and free aromatic compound. Especially, the wear scar of HBD showed poor lubricity compared to automotive diesel due to the fuel composition, low sulfur content and free aromatic compound. Also, the lubricity specification of automotive diesel with different six HBD blends is within the limit by the Korean standards. Finally, HBD as an alternative diesel fuel is challengeable in transportation sector of Korea.

Improving the Surface Roughness of SL Parts Using a Coating and Grinding Process

  • Rapid prototyping (RP) technology can fabricate any 3D physical model regardless of geometric complexity using the layered manufacturing (LM) process. Stereolithography (SL) is the best-known example of RP technology. In general, the surface quality of a raw SL-generated part is unsatisfactory for industrial purposes due to the step artefact created by the LM process. Despite of the increased number of applications for SL parts, this side effect limits their uses. In order to improve their surface quality, additional post-machining finishing, such as traditional grinding, is required, but post-machining is time consuming and can reduce the geometric accuracy of a part. Therefore, this study proposes a post-machining technology combining coating and grinding processes to improve the surface quality of SL parts. Paraffin wax and pulp are used as the coating and grinding materials. By grinding the coating wax only up to the boundary of the part, the surface smoothness can be improved without damaging the surface. Finally, moulding and casting experiments were performed to confirm the suitability of the SL parts finished using the proposed process with rapid tooling (RT) techniques.

  • 원자력 발전소에서 발생한 시멘트 또는 파라핀 고화체 중의 핵종분석을 하기 위해서는 시료의 용액화가 필요하다. 이를 위하여 산 침출법과 극초단파 산분해법을 이용하여 SRM(standard reference material)을 용액화한 다음 AAS와 ICP-AES를 사용하여 용액 속의 원소들을 분석 비교하였다. 완전 용액화가 가능한 극초단파 산분해법으로 처리한 결과와 일치하는 산 침출조건을 제안하여 많은 양의 시료를 한 번에 처리할 수 있는 산 침출법으로 방사성 고화체 시료 중의 비휘발성핵종분석을 위한 고화체 용액화 방법을 확립하였다. 방사성 고화체의 산 침출시마다 Re을 첨가하여 시료 전처리 단계에서의 회수율은 $80\%$ 이상으로 나타났다.

  • The heat transfer experiment in a latent heat storage tank as a solar energy storage system for the hot water supply was carried out. The latent heat storage tank was consisted of triple - tube type ; Outer shell for hot water from solar collector, PCM storage vessel in the middle of the tank and inside tube for hot water recovery. The heat storage tank has the dimension of 60 cm long and 34 cm outside diameter. Paraffin wax(m.p = 55.4C) and sodium acetate trihydrate(m.p = 58 C) were employed as the PCM this study. Experimental variables were inlet temperature and flow rate of the hot water for heat storage stage and cold water for heat recovery stage. Temperature profiles, heat transfer coefficient and the efficiency of heat storage$(Q/Q_{max})$ and heat recovery $(Q/Q_{max})$ were determined for the paraffin wax and inorganic salt respectively.

  • The heat transfer experiment in a pilot scale latent heat storage tank as a solar energy storage system for the hot water supply was carried out. The latent heat storage tank was consisted of three parts; Outer shell for hot water from solar collector, PCM storage vessel in the middle of the tank and immersed coil in the PCM vessel for hot water recovery.   • Americium-beryllium (Am-Be; n, ${\gamma}$) is a neutron emitting source used in various research fields such as chemistry, physics, geology, archaeology, medicine, and environmental monitoring, as well as in the forensic sciences. It is a mobile source of neutron activity (20 Ci), yielding a small thermal neutron flux that is water moderated. The aim of this study is to develop a model to increase the neutron thermal flux of a source such as Am-Be. This study achieved multiple advantageous results: primarily, it will help us perform neutron activation analysis. Next, it will give us the opportunity to produce radio-elements with short half-lives. Am-Be single and multisource (5 sources) experiments were performed within an irradiation facility with a paraffin moderator. The resulting models mainly increase the thermal neutron flux compared to the traditional method with water moderator.

  • A simple mathematical model for facilitated mass transport through a fixed site carrier membrane was derived by assuming an instantaneous, microscopic concentration (activity) fluctuation. The current model demonstrates that the facilitation factor depends on the extent of concentration fluctuation, the time scale ratios of diffusion to chemical reaction and the ratio of the carrier concentration to the solute solubility in matrix. The model was examined against the experimental data on oxygen transport in membranes containing metallo-porphyrin carriers, and the agreement was exceptional (within 10% error). The basic concept of this approach was applied to separate olefin from olefin/paraffin mixtures. A proprietaty carrier, developed here, resulted that the selectivity of propylene over propane was more than 120 and the propylene permeance exceed 40 gpu.
