Immunological Relationships among Fungal and Plant Phenylalanine Ammonia-lyases and Bacterial Histidine Ammonia-lyase (진균과 식물의 Phenylalanine Ammonia-lyase 그리고 세균의 Histidine Ammonia-lyase 간의 면역학적 관계 분석)

  • Phenylalanine ammonia-lyase (PAL) from the maize pathogen Ustilago maydis was analysed immunologically to obtain insights into the structural relationships between plant PAL and fungal PAL and between PAL and histidine ammonia-lyase (HAL). Cross-reactivity was found among all the PAL proteins from different species tested, using antibodies raised against both plant and fungal PALs. Both anti-Alfalfa and anti-popular PAL antibodies strongly recognized plant PALs but only weakly recognized fungal PALs. Antibodies raised against U. maydis PAL only weakly recognized the Rhodotorula glutinis yeast PAL. The anti-U. maydis PAL antibodies showed low affinity for the plant PALs but they bound strongly to Pseudomonas bacterial HAL. Significant cross-reactivity between the two plant PAL antibodies and the bacterial HAL was also observed. Both the anti-Ustilago PAL and the anti-poplar PAL antibodies displayed similar enzyme inhibition patterns, including moderate inhibition of bacterial HAL activity. However, the bacterial HAL antibody inhibited only Ustilago PAL. The PAL and HAL antibodies tested showed no inhibition against yeast PAL. This is first report on the immunological relationships between PAL and HAL.

Complete Nucleotide Sequence Analysis and Structural Comparison of 3 members of Tomato Phenylalanine ammonia-lyase gene (토마토에서 분리한 3종류의 Phenylalanine ammonia-lyase gene에 대한 염기서열 및 특성비교)

  • Phenylalanine ammonia-lyase (PAL; EC 4, 3, 1, 5) genomic clones were isolated from tomato(Lycopersicon esculentum L.) genomic DNA libraries using tomato PAL5 cDNA sequences as probes. The nucleotide sequences of tPAL1, tPAL4 and tPAL5 were compared. tPAL5 contains an open reading frame encoding a polypeptide of 722 amino acids, interrupted by a 710 bp intron in the codon for the amino acid 139. tPAL1 encodes a polypeptide of 249 amino acids which is much shorter than tPAL5 gene due to a premature stop codon and does not contain an intron. tPAL4 encodes a polypeptide of 357 amino acids, interrupted by a 305 bp intron in the codon for the amino acid 138. Premature stop codons observed in tPAL1 and tPAL4 gene produce a short polypeptide rather than a normal polypeptide (722 aa). tPALl shows 87.2% homology with tPAL4 and 85.3% homology with tPAL5 gene whereas tPAL4 showes 91.4% homology with tPAL5 at nucleotide level. In general, phylogenetic analysis showed that genes isolated from tomato, potato, and sweet potato were belong to the same group and another dicot plants such as parsley, bean, soybean, pea and alfalfa formed another group. PAL genes isolated from rice and yeast showed very low homology with other PAL genes and formed the other group.

A Design of 8VSB Transmission System for Use in PAL Standard (PAL 표준 지역에 적합한 8VSB 송수신 시스템 설계)

  • 8VSB transmission system of American standard and OFDM system of European standard are to be international standards for the transmission of digital terrestrial television. When countries using PAL standard adopt the American 8VSB system which is designed under the circumstance of NTSC system, one of strong interference is the co-channel PAL interference coming from analog PAL signal. In this paper, in order to solve this problem, we classified seven types of PAL system into three groups according to the bandwidth and a new PAL-8VSB system is proposed to reject the co-channel PAL interference by using modified comb filters suitable for each group. By computer simulations of the proposed PAL-8VSB system with the co-channel PAL interference it is confirmed that the proposed comb filter results in the improvement of DIU ratio by about 9 dB.

  • We observed the structure of phenylalanine ammonia-lyase gene (PAL) which is one of the best studied plant defense-related genes responding to pathogen infection by producing suberin, lignin, and phytoalexins. In tomato, at least 5 different genetic loci have been identified by genomic southern blot hybridization and nucleotide sequence analyses of partially cloned gene fragments (Lee et al. 1992). However, our results suggest that two other isoforms designated as PAL X1 and PAL X2 are located on the chromosome in tomato plant. Furthermore, the preliminary results obtained from southern blot hybridization analyses of subcloned fragment digested with several restriction endonuclease indicated that PAL X1 and PAL X2 clones contain at least two copies of PAL gene and partial nucleotide sequence analyses of each subcloned fragment with the same primer taken from known nucleotide sequence of PAL5 gene indicated that they are located side by side on the same chromosome.

  • The PAL (Pohang Accelerator Laboratory) is persuading to construct a SASE-XFEL facility (PAL XFEL) that supplies coherent X-rays. The bright and stable electron beam is essential for the PAL XEL. The electron beams has to have an emittance of 1.0 mm-mrad, a peak current of 3 kA, and a low energy spread of 1.0 MeV. In order to provide reasonably stable SASE output, the RF stability of 0.02% rms is required for both RF phase and amplitude. This is a technologically challenging issue for PAL XFEL. An inverter technology is to be applied to charge the PFN of a new modulator. Therefore, a new inverter system should provide very stable charging performances. This paper presents the development of an ultra stable klystron-modulator with an inverter power supply.

Using physical activity levels to estimate energy requirements of female athletes

  • [Purpose] The goal of this study was to review data on physical activity level (PAL), a crucial index for determining estimated energy requirement (EER), calculated as total energy expenditure (TEE, assessed with doubly labeled water [DLW]) divided by resting metabolic rate (RMR, PAL = TEE/RMR) in female athletes and to understand the methods of assessing athletes' EERs in the field. [Methods] For the PAL data review among female athletes, we conducted a PubMed search of the available literature related to the DLW method. DLW studies measuring TEE and RMR were included for the present review. [Results] Briefly, the mean PAL was 1.71 for collegiate swimmers with moderate training, which was relatively low, but the mean PAL was 3.0 for elite swimmers during summer training camp. This shows that PAL can largely vary even within the same sport depending on the amount of training, and the differences in PAL were remarkable depending on the sport. Aside from the DLW method, there is currently no research tool related to athletes' EERs that can be used in the field. Phenylalanine Ammonia-Lyase Gene (NtPAL4) Induced by Abiotic Stresses in Tobacco (Nicotiana tabacum)

  • Phenylalanine ammonia-lyase (PAL), a key enzyme of the phenylpropanoid biosynthesis pathway, is activated by a number of developmental and environmental cues. The coding region of the NtPAL4 gene was 2,154 bp in length, and its deduced protein was composed of 717 amino acids. Sequence analysis of NtPAL4 cDNA from tobacco (Nicotiana tabacum L.) revealed high structural similarity to PAL genes of other plant species. The NtPAL4 gene exists as a single copy in the tobacco plant, and its transcripts were strongly expressed in flowers and leaves. NtPAL4 expression was significantly induced in response to NaCl, mannitol, and cold treatments, but it was not induced by abscisic acid (ABA). NtPAL4 expression decreased gradually after treatment with ABA and $H_2O_2$; however, NtPAL4 transcripts accumulated after treatment with methyl viologen (MV). Our results suggest that the NtPAL4 gene may function in response to abiotic stresses.
