Seasonal Distribution of Bacterial Populations and Escherichia coli O157 at Hanwoo Cattle Feedlots in Gyeongsangbuk-do (경상북도 한우축사에서의 계절별 세균 및 Escherichia coli O157 분포 변화)

  • The seasonal variation of bacterial populations in the Korean cattle (Hanwoo) feedlots dispersed in the 20 cities of Gyeongsangbuk-do, Korea, was monitored for two years (2006 and 2007) to provide quantitative criteria for good agricultural management. Outside the feedlots, the average falling bacterial populations of the air were $3{\pm}1$ CFU/$cm^2\cdot15$ min (n=63) in a year. Inside the feedlots, the average falling bacterial populations of the air were $8{\pm}1$ CFU/$cm^2\cdot15$ min (n=63) in the spring, $16{\pm}2$ CFU/$cm^2\cdot15$ min (n=69) in the summer, $7{\pm}2$ CFU/$cm^2\cdot15$ min (n=69) in the autumn, and $6{\pm}1$ CFU/$cm^2\cdot15$ min (n=70) in the winter. Without using the summer data, the average falling bacterial population of the air was $7{\pm}1$ CFU/$cm^2\cdot15$ min, which was not statistically significant (P=0.37). The average bacterial populations in the cattle drinking water of the cattle feedlots were $4,710{\pm}780$ CFU/ml (n=65) in the spring, $10,430{\pm}1170$ CFU/ml (n=65) in the summer, $4,820{\pm}700$ CFU/ml (n=64) in the autumn, and $2,510{\pm}530$ CFU/ml (n=64) in the winter. Without using the summer data, the average bacterial population of the drinking water was $4,000{\pm}400$ CFU/ml, which was statistically significant (P=0.027). The average frequency of Escherichia coli O157 inside the feedlots was 5% (n=65) in the spring, 72% (n=65) in the summer, 67% (n=66) in the autumn, and 29% (n=66) in the winter on the basis of soil samples of the year 2007. The results indicate that most of the Escherichia coli O157 strains distributed in the summer and autumn was disappeared in the spring through the cold weather of the winter.

Antimicrobial Activities of Solvent Extracts from Citrus sudachi Juice and Peel (영귤(Citrus sudachi) 과즙과 과피 용매 추출물의 항균 효과)

  • Extracts of Citrus sudachi juice and peel were obtained by several organic solvents such as hexane, acetone, ethyl acetate and methanol. Their antimicrobial spectrum were determined against 14 strains of gram positive and 4 strains of gram negative bacteria by paper disk method and minimal inhibitory concentration (MIC) was also obtained. Antimicrobial activities of solvent extracts from Citrus sudachi juice showed stronger than those of solvent extracts from peel. Acetone extract from juice showed the strongest antimicrobial activity among extracts, but the hexane extract did not show antimicrobial activities on tested target strains. The MIC was differant among tested strains; i.e. 0.5% (v/v) to Bacillus subtilis, 1% to Pseudomonas fragi, 1.5% to Listeria monocytogenes and Escherichia coli O157:H7, 2% to Propionibacterium acnes and Salmonella typhimurium, and 2.5% to Staphylococcus aureus.

Isolation and Characterization of Lactobacillus fermentum YL-3 as a poultry probiotic. (가금류 생균제 개발을 위한 Lactobacillus fermentum YL-3의 분리 및 생리 특성)

  • This study was per-formed to screen lactic acid bacteria poultry for the probiotic use. Among the previously obtained acid tolerant, 139 strains, 111 strains were selected with MRS medium containing 0.3% oxgall. 34 strains of 111 was re-selected by Gram-staining and acid producing ability. These strains was identified by MIDI Sherlock Microbial Identification System. Among the identified 34 strains Lactobacillus fermenum YL-3 was selected for the final pro-biotic use because of the good growth and high survival rate at pH 2.0. 60%, 50% and 40% cells of Lactobacillus fermentum YL-3 survived at pH 3.0, 2.5 and 2.0, respectively. More than $10^{7}$ / CFU/ml survived when exposed with the number of $10^{8}$ CFU/ml at pH 2.0 after 12 hr. L.fermenum YL-3 maintained growth in MRS broth containing 0.3, 0.5, 1.0 and 2.0% oxgall for 24 hr. L.fermenum YL-3 showed an inhibitory effect against pathogenic strains of Sal. enteritidis and E. coli O157:H7. In mixed culture with L.fermenum YL-3 Sal. enteritidis lost ability com-pletely in 15 hrs and E. coil O157:H7 in 16 hrs.

Optical spectroscopy of LMC SNRs to reveal the origin of [P II] knots

  • Observational studies of supernova (SN) feedback are limited. In our galaxy, most supernova remnants (SNRs) are located in the Galactic plane, so there is contamination from foreground/background sources. SNRs located in other galaxies are too far, so we cannot study them in detail. The Large Magellanic Cloud (LMC) is a unique place to study the SN feedback due to their proximity, which makes possible to study the structure of individual SNRs in some detail together with their environment. Recently, we carried out a systematic study of 13 LMC SNRs using [P II] (1.189 ㎛) and [Fe II] (1.257 ㎛) narrowband imaging with SIRIUS/IRSF, four SNRs (SN 1987A, N158A, N157B and N206), show [P II]/[Fe II] ratio much higher than the cosmic abundance. While the high ratio of SN 1987A could be due to enhanced abundance in SN ejecta, we do not have a clear explanation for the other cases. We investigate the [P II] knots found in SNRs N206, N157B and N158A, using optical spectra obtained last November with GMOS-S mounted on Gemini-South telescope. We detected several emission lines (e.g., H I, [O I], He I, [O III], [N II] and [S II]) that are present in all three SNRs, among other lines that are only found in some of them (e.g., [Ne III], [Fe III] and [Fe II]). Various line ratios are measured from the three SNRs, which indicate that the ratios of N157B tend to differ from those of other two SNRs. We will use the abundances of He and N (from the detection of [N II] and He I emission lines), together with velocity measurements to tell whether the origin of the [P II] knots are SN ejecta or CSM/ISM. For this purpose we have built a family of radiative shock with self-consistent pre-ionization using MAPPINGS 5.1.18, with shock velocities in the range of 100 to 475 km/s. We will compare the observed and modeled line fluxes for different depletion factors.

A Study on Highly Dispersed Pt/$Al2O_3$ Catalyst for Preferential CO Oxidation (고분산 담지된 Pt/$Al2O_3$ 촉매의 선택적 CO 산화반응 특성에 관한 연구)

  • 선택적 CO 산화반응(PrOx)에 사용되는 촉매 중 Pt, Ru, Rh 등의 귀금속 계 촉매들은 비귀금속 계 촉매에 비해 활성이 좋은 반면 가격이 비싸다는 경제적인 제한점이 있다. 따라서 소량의 귀금속을 사용하여 높은 활성의 촉매를 제조하고자 활성금속의 고분산 담지 방법에 대한 연구가 이루어지고 있다. 본 연구에서는 담체인 ${\gamma}-Al_2O_3$ 표면에 활성금속인 Pt의 고분산 담지를 위해 증착-침전법(Deposition-precipitation)을 적용하였으며 용액의 pH 변화에 따른 Pt 금속 입자의 분산도에 대한 영향을 살펴보았다. Pt의 함량은 1wt%로 고정하였고 침전제로 NaOH를 사용하여 용액의 pH를 pH 7.5 ~ 10.5로 변화시켰다. 제조된 촉매는 세척 후 $400^{\circ}C$, 3시간 소성 하였다. 제조된 1wt% Pt/$Al_2O_3$ 촉매의 특성분석을 위해 BET, TPR, CO-chemisorption을 수행하였다. PrOx 반응 실험은 GHSV=60,000 $ml/g_{cat}{\cdot}h$, $T=100{\sim}200^{\circ}C$, ${\lambda}$=4 조건에서 수행되었으며 반응 전에 촉매는 $400^{\circ}C$, 3시간 환원 후 사용하였다. 촉매의 특성분석과 PrOx 반응 실험 결과를 통해 촉매가 담체 위에 고분산 되는 최적의 pH를 확인할 수 있었으며, 기존의 함침법으로 제조된 촉매와 성능 비교를 통해 제조방법에 따른 영향을 살펴보았다.

Microbial Detection and Identification Using Biosensors

  • Various biosensors were evaluated for identifying and detecting foodborne pathogens in a rapid and effective manner. First, five strains of Escherichia coli and six strains of Salmonella were identified using Fourier transform infrared spectroscopy and a statistical program. For doing this, lipopolysaccharides (LPSs) and outer membrane proteins (OMPs) were extracted from a cell wall of each bacterial strain. As a result, each strain was identifed at the level of 97% for E. coli and 100% for Salmonella. Second, E. coli O157:H7, S. Enteritidis, and Listeria monocytogenes were identified by multiplex PCR products from four specific genes of each bacteria using a capillary electrophoresis (CE). Also, ground beef for E. coli O157:H7, lettuce for S. Enteritidis, and hot dog for L. monocytogenes were used to determine the possibility of detecting pathogens in foods. Foods inoculated with respective pathogen were cultivated for six hours and multiplex PCR products were obtained and assessed. The minimum detection levels of tested bacteria were <10 cells/g, <10 cells/g, and $10^4$ cells/g for E. coli O157:H7, S. Enteritidis, and L. monocytogenes, respectively. Third, it was possible to detect S. Typhimurium in a pure culture and lettuce by a bioluminescence-based detection assay using both recombinant bacteriophage P22::luxI and a bioluminescent bioreporter. In addition, bacteriophage T4 was quantitatively monitored using E. coli including luxCDABE genes.

Antimicrobial Effects of Organic Acids and Ethanol on Several Foodborne Microorganisms (식중독 미생물에 대한 유기산 및 에탄올의 항균활성 비교연구)

  • The antimicrobial effects of ethanol and organic acids(acetic, citric, lactic. propionic, tartaric acid), either alone or in combination against four foodborne microorganisms (Listeria monocytogenes, Salmonella typhimurium, Staphylococcus aureus and Escherichia coli O157:H7) in tryptic soy broth were determined. Area under the growth curve, minimum generation time, maximum growth rate, and detection time were measured by using automated turbidometer Bioscreen(Labsystem, Finland), for 24 hr at $30^{\circ}C$. All microorganisms were not grown at 7% ethanol in the media. The 0.1% propionic acid showed the strongest inhibitory effects against S. aureus, L. monocytogenes and E. coli O157 : H7 compared with other organic acids, whereas 0.01% organic acids did not show significant inhibitory effect against microorganisms tested (p > 0.01) except S. aureus. The combination of 1% ethanol and 0.01% organic acids were significantly more effective than alone on growth of S. aureus and L. monocytogenes(p < 0.01).

Bactericidal Effect of Combination of Atmospheric Pressure Plasma and Nisin on Meat Products Inoculated with Escherichia coli O157:H7

  • This study was conducted to investigate the bactericidal effect of nisin (Nisin) only, atmospheric pressure plasma (APP) only, and a combination of APP and nisin (APP+Nisin)(APP+Nisin) on beef jerky and sliced ham inoculated with Escherichia coli O157:H7, gram-negative bacteria. The bactericidal effect against E. coli O157:H7 and Listeria monocytogenes was confirmed using a nisin solution at a concentration of 0-100 ppm, and APP+Nisin was tested on beef jerky and sliced ham using 100 ppm nisin. Beef jerky and sliced ham were treated with APP for 5 min and 9 min, respectively. In the bacterial solution, 100 ppm nisin out of 0-100 ppm nisin exhibited the highest bactericidal activity against L. monocytogenes (gram-positive bacteria; p<0.05); however, it did not exhibit bactericidal effects against E. coli O157:H7 (gram-negative bacteria). The APP+Nisin APP+Nisin exhibited a 100% reduction rate in both E. coli O157:H7 and L. monocytogenes compared to the control group, and was more effective than the Nisin. The APP+Nisin decreased the number of colonies formed by 0.80 and 1.96 Log CFU/g for beef jerky and sliced ham, respectively, compared to the control, and exhibited a higher bactericidal effect compared to the Nisin (p<0.05). These results demonstrate the synergistic bactericidal effect of APP and nisin, providing a possible method to improve the limitations of nisin against gram-negative bacteria. In addition, this technology has the potential to be applied to various meats and meat products to control surface microorganisms.

Comparative study of Clostridium perfringens, Salmonella spp. and E. coli focused on characteristics of E. coli O157 isolated from pigs of HACCP- and non-HACCP-accredited swine farms in Korea

  • To determine the prevalence of Escherichia (E.) coli O157 : H7 from pigs after the Hazard Analysis and Critical Control Point (HACCP) system has been applied to Korean swine farm since 2006, 291 fecal samples were tested between May and December in 2008. Four E. coli O157:non-H7 (1.4%) were isolated from 4 different non-HACCP-accredited farms and they didn't have virulent genes which can cause illness for human. Also, Clostridium (C.) perfringens, Salmonella spp. and E. coli enterotoxins were tested using multiplex PCR. The positive rate for these pathogens of non-HACCP-accredited farms was higher than that of HACCP-accredited farms, and especially in case of C. perfringens, E. coli enterotoxins LT and STa, it was statistically significant (p < 0.05). Thus, the early implementation of the HACCP program is expected to greatly contribute to the safety of livestock products as well as food hygiene.

Effect of Ozone Water to Reduce Pathogenic Microorganisms on Chopping Board (도마표면의 병원성미생물 제어를 위한 오존수 처리효과)

  • The efficacy of ozone water in reducing food-borne pathogenic bacteria on High Density Polyethylene (HDPE) and wooden chopping boards in food industry was investigated in this study. 1-5 log reductions of E. coli O157:H7, S. aureus, S. Typhimurium, and B. cereus were observed with increasing concentrations of ozone water. The immersion treatment evidenced superior capability to inactivate food-borne pathogens than washing treatment. The Gram-negative bacteria, such as E. coli O157:H7 and S. Typhimurium, evidenced lower resistance against ozone water than was seen with the gram-positive bacteria, which included S. aureus and B. cereus. The sterilizing effects of ozone water on HDPE chopping boards was superior to that on wooden boards. This result might be utilized to remove food-borne pathogens from food contact surfaces in the food industry.
