Expression and Purification of an ACE-Inhibitory Peptide Multimer from Synthetic DNA in Escherichia coli

  • An angiotensin I-converting enzyme (EC 3.4.15.1) (ACE), which can convert inactive angiotensin I into angiotensin II, a vasoconstrictor, is one of the key enzymes in controlling hypertension. It is suggested that the inhibition of ACE prevents hypertension, and many inhibitory peptides have already been reported. In the current study, oligonucleotides encoding ACE inhibitory peptides (IY, VKY) were chemically synthesized and designed to be multimerised due to isoschizomer sites (BamHI, BglII). The cloned gene named AP3 was multimerised up to 6 times in pBluescript and expressed in BL2l containing pGEX-KG. The fusion protein (GST-AP3) was easily purified with a high recovery by an affinity resin, yielding 38 mg of synthetic AP3 from a 1-1 culture. The digestion of AP3 by chymotrypsin exhibited an $IC_50$ value of $18.53{\mu}M$. In conclusion, the present experiment indicated that AP3 could be used as a dietary antihypertensive drug, since the potent ACE inhibitory activity of AP3 could be activated by chymotrypsin in human intestine.

Nano-Calcium Ameliorates Ovariectomy-Induced Bone Loss in Female Rats

  • In this study, we examined the effects of organic types of calcium derived from oyster shell (OS-Ca) and nano-calcium (Nano-Ca) on the bio-availability and physiological responses associated with bone health in ovariectomised rats. Increased body weight, which is one of the physiological effects of ovary removal, was significantly recovered by Nano-Ca treatment (p<0.05). The reduced calcium level in the liver in ovariectomised rat was increased significantly with OS-Ca and Nano-Ca treatment (p<0.05), suggesting improved calcium bio-availability. Alkaline phosphatase (ALP), osteocalcin, and deoxypyridinoline (DPD) were analysed as biochemical markers of bone metabolism and health in the presence or absence of OSCa and Nano-Ca. ALP, osteocalcin, and DPD levels increased following ovary removal and tended to decrease after treatment with Nano-Ca, indicating that Nano-Ca induces favourable bone metabolism. This result was reflected in the recovery of bone mineral density (BMD) and bone mineral content (BMC) of the femur after Nano-Ca treatment following ovary removal. Taken together, our data show that the tested calcium treatments, especially using Nano-Ca, enhanced the bioavailability or absorption of calcium and positively affected bone metabolism in ovariectomised rats.

HPLC를 이용한 생체 시료중의 새로운 플라보노이드 유도체인 DA-6034의 분석 (Analysis of DA-6034, a New Flavonoid Derivative in Biological Fluids by HPLC)

  • A high performance liquid chromatographic method was developed for the determination of DA-6034 in biological fluids using internal standard. Plasma containing DA-6034 and inter nal standard was extracted by liquid-liquid extraction at an acidic pH. After evaporation of the organic layer, the drug and internal standard were reconstituted with mobile phase and injected into the column. They were separated by high performance liquid chromatography on inertsil ODS II column at 334 nm. The detection limit of DA-6034 in plasma was 0.02 ${\mu}$g/ml. In this method, the range of recovery and coefficients of variation were 96-110% and 0.40-3.78%, respectively. There was no interference from endogenous substances. Urine and bile were analysed using the deproteinization method and the detection limit of DA-6034 was 1${\mu}$g/l.

저산소 허혈 뇌 손상을 유발시킨 미성숙 흰쥐에서 마우스 골수 기원 중간엽 줄기 세포 이식 후 기능 회복 (Functional recovery after transplantation of mouse bone marrow-derived mesenchymal stem cells for hypoxic-ischemic brain injury in immature rats)

  • 목 적 : HIE의 치료법으로 줄기 세포가 대안으로 떠오르고 있다. mMSC가 성인 동물 모델에서의 뇌졸중 및 퇴행성 뇌질환에 유의한 기능 회복을 보인다는 보고가 많이 있으나 미성숙 동물 모델에서의 연구 보고는 거의 없는 상태이다. 이에 HIE가 유발된 미성숙뇌에 mMSC를 투여하여 기능 회복에 대한 효과를 평가하고자 본 연구를 시행하였다. 방 법 : 생후 7일된 수컷 Sprague-Dawley 흰쥐를 sham 대조군, 뇌 손상 대조군, 고용량 mMSC 이식군 및 저용량 mMSC 이식군으로 나누었으며 저산소 허혈 뇌 손상 유도 2주 후에 mMSC를 병변부 국소 이식을 시행하였다. 세포 이식 2, 4, 6 및 8주째에 개방장 시험을 시행하여 운동 기능 회복 정도를 평가하였고, 이후 1주일 동안 Morris 수중 미로 시험을 3개 부문으로 시행하여 학습 및 기억력 회복 정도를 평가하였다. 결 과 : 개방장 시험 결과 네 군간에서 통계적으로 유의한 차이는 없었다(F=0.412, P=0.745). 공간 획득 검사에서 네 군간 평균 탈출 시간에 서로 차이가 있었고(F=380.319, P<0.01), 고용량 mMSC 이식군 및 sham 대조군은 뇌 손상 대조군에 비해 검사 2일째부터 5일째까지 각각 평균 탈출 시간이 감소하였으며(P<0.05), 시간이 갈수록 더 유의하게 차이를 보였다(F=16.034, P<0.01). 참조 기억 검사에서는 네 군간 차이는 없었으며 시각 검사에서는 다섯번째 시행 검사에서만 고용량 mMSC 이식군과 뇌 손상 대조군간에 통계적으로 유의한 차이가 있었다(P<0.05). 결 론 : 일부 검사에서만 고용량 mMSC 이식의 효과가 뚜렷하였고 그 외의 검사에는 통계적으로 유의한 결과는 보이지는 않았으나 산술적인 호전을 보이는 것을 확인할 수 있었다. 향후 최적의 효과를 보이는 줄기 세포의 농도와 이식 시기를 결정하는 연구가 필요할 것으로 보이며 HIE에서 mMSC가 대안적인 치료 수단으로 이용될 수 있다고 생각된다.

Hematopoietic stem cell transplantation in children with acute leukemia: similar outcomes in recipients of umbilical cord blood versus marrow or peripheral blood stem cells from related or unrelated donors

  • Purpose: This study compared outcomes in children with acute leukemia who underwent transplantations with umbilical cord blood (UCB), bone marrow, or peripheral blood stem cells from a human leukocyte antigen (HLA)-matched related donor (MRD) or an unrelated donor (URD). Methods: This retrospective study included consecutive acute leukemia patients who underwent their first allogeneic hematopoietic stem cell transplantation (HSCT) at Samsung Medical Center between 2005 and 2010. Patients received stem cells from MRD (n=33), URD (n=46), or UCB (n=41). Results: Neutrophil and platelet recovery were significantly longer after HSCT with UCB than with MRD or URD ($p$ <0.01 for both). In multivariate analysis using the MRD group as a reference, the URD group had a significantly higher risk of grade III to IV acute graft-versus-host disease (GVHD; relative risk [RR], 15.2; 95% confidence interval [CI], 1.2 to 186.2; $p$=0.03) and extensive chronic GVHD (RR, 6.9; 95% CI, 1.9 to 25.2; $p$ <0.01). For all 3 donor types, 5-year event-free survival (EFS) and overall survival were similar. Extensive chronic GVHD was associated with fewer relapses (RR, 0.1; 95% CI, 0.04 to 0.6; $p$ <0.01). Multivariate analysis showed that lower EFS was associated with advanced disease at transplantation (RR, 3.2; 95% CI, 1.3 to 7.8; $p$ <0.01) and total body irradiation (RR, 2.1; 95% CI, 1.0 to 4.3; $p$=0.04). Conclusion: Survival after UCB transplantation was similar to survival after MRD and URD transplantation. For patients lacking an HLA matched donor, the use of UCB is a suitable alternative.

Comparison of Mechanical Stability between Fibular Free Flap Reconstruction versus Locking Mandibular Reconstruction Plate Fixation

  • Background: The fibular free flap has been used as the standard methods of segmental mandibular reconstruction. The objective of mandibular reconstruction not only includes restored continuity of the mandible but also the recovery of optimal function. This paper emphasizes the advantage of the fibular free flap reconstruction over that of locking mandibular reconstruction plate fixation. Methods: The hospital charts of all patients (n=20) who had a mandibular reconstruction between 1994 and 2013 were retrospectively reviewed. Eight patients had plateonly fixation of the mandible, and the remaining 12 had vascularized fibular free flap reconstruction. Complications and outcomes were reviewed and compared between the 2 groups via statistical analysis. Results: Overall complication rates were significantly lower in the fibular flap group (8.3%) than in the plate fixation group (87.5%; p =0.001). Most (7/8) patients in the plate fixation group had experienced plate-related late complications, including plate fracture or exposure. In the fibular flap group, no complications were observed, except for a single case of donor-site wound dehiscence (1/12). Conclusion: The fibular free flap provides a more stable support and additional soft tissue support for the plate, thereby minimizing the risk of plate-related complications. Fibular free flap is the most reliable option for mandibular reconstruction, and we believe that the flap should be performed primarily whenever possible.

Transcriptome analysis of Panax ginseng response to high light stress

  • Background: Ginseng (Panax ginseng Meyer) is an essential source of pharmaceuticals and functional foods. Ginseng productivity has been compromised by high light (HL) stress, which is one of the major abiotic stresses during the ginseng cultivation period. The genetic improvement for HL tolerance in ginseng could be facilitated by analyzing its genetic and molecular characteristics associated with HL stress. Methods: Genome-wide analysis of gene expression was performed under HL and recovery conditions in 1-year-old Korean ginseng (P. ginseng cv. Chunpoong) using the Illumina HiSeq platform. After de novo assembly of transcripts, we performed expression profiling and identified differentially expressed genes (DEGs). Furthermore, putative functions of identified DEGs were explored using Gene Ontology terms and Kyoto Encyclopedia of Genes and Genome pathway enrichment analysis. Results: A total of 438 highly expressed DEGs in response to HL stress were identified and selected from 29,184 representative transcripts. Among the DEGs, 326 and 114 transcripts were upregulated and downregulated, respectively. Based on the functional analysis, most upregulated and a significant number of downregulated transcripts were related to stress responses and cellular metabolic processes, respectively. Conclusion: Transcriptome profiling could be a strategy to comprehensively elucidate the genetic and molecular mechanisms of HL tolerance and susceptibility. This study would provide a foundation for developing breeding and metabolic engineering strategies to improve the environmental stress tolerance of ginseng.

잎 단백질(蛋白質)(Leaf Protein Concentrates)의 개발(開發)에 관한 연구(硏究) -I. 잎 단백질(蛋白質)의 추출조건(抽出條件)에 대한 검토(檢討)- (Development of Leaf Protein Concentrates I. Studies on the Isolation of Leaf Protein Concentrates)

  • 69종(種)의 각종 식물체(植物體)를 대상으로 하여 이것을 마쇄(磨碎)한 slurry로부터 총질소(總窒素)의 추출상황(抽?狀況)을 조사(調査)하고, 추출액(抽出液)으로 부터의 LPC의 제조방법(製造方法)을 검토(?討)하여 다음과 같은 결과(結果)를 얻었다. 1. 원료(原料)의 총질소(總窒素)의 함량(含量)이 클수록 총질소(總窒素)의 추출비(抽出比)는 커지며, 원료(原料)로부터의 추출(抽出)은 적어도 2회추출(回抽出)까지 필요(必要)하다. 원료엽체(原料葉體)로부터의 1차(次) 추출액(抽出液) $(E_1;\;%)$, 2次 抽出液$(E_2;\;%)$와 (T; %)사이에는 $E_1=0.8168T,\;E_2=0.1830T$의 1차(次) 관계식(關係式)이 성립(成立)한다. 2. pH 처리(處理)에 의한 LPC 생성(生成)에 있어서 pH 3 이하(以下)에서는 생성(生成)된 LPC의 색택(色擇)이 갈변(褐變)되고 제품(製品)의 탄력성(彈力性)이 커지며, 그 정도는 pH가 낮아질 수록 현저(顯著)해진다. pH 처리(處理)에 의한 LPC 생성(生成)의 지적(至適) pH 는 $3.5{\sim}4.5$ 라고 할 수 있다. 3 추출액(抽出液)으로 부터의 LPC의 생성(生成)은 TCA 처리(處理)> pH 4> pH 3> 열처리(熱處理)의 순서(順序)가 되며, TCA 처리(處理)에 비(比)하여 pH 4 처리(處理)는 10%감(減), pH 3 처리(處理)는 11.4%감(減), 열처리(熱處理)는 14.8%감(減)이었다. 4. pH 처리법(處理法)은 열처리법(熱處理法) 보다 LPC의 생성량(生成量)이 다소 높으나 대량처리시(大量處理時)의 pH 조절(調節)의 복잡성(複雜性)과 생성제품(生成製品)의 품질(品質)로 보아 열처리(熱處理)에 의한 LPC의 생성방법(生成方法)이 효과적(效果的)이고 간편(簡便)하다.
