Design Criteria to Miniaturize the Single Use Functional Respiratory Air Flow Tube

  • Respiratory tubes with a length of 35mm and diameters of 10, 15, and 20mm were made for experimental purpose, and both the static$(P_s)\;and\;dynamic(P_D)$ pressures were simultaneously measured for steady flow rates ranging 1-12//sec. Least squares analysis resulted successful fitting of $P_s\;and\;P_D$ data with quadratic equations with correlation coefficients higher than 0.99(P<0.0001). The spirometric measurement standards of the American Thoracic Society(ATS) were applied to $P_s$ data, which demonstrated the smallest tube diameter of 15mm to satisfy the ATS standards. The maximum $P_D$ value of the velocity type transducer(the functional single use respiratory air flow tube) with the diameter of 15mm was estimated to be approximately $75cmH_2O$, implying more than 7 times larger sensitivity than the widely used pneumotachometers. These results showed that the velocity type respiratory air flow transducer is a unique device accomplishing miniaturization with the sensitivity increased, thus would be of great advantage to develop portable medical devices.

  • In this study, we fabricated the dual gate (DG) ion-sensitive field-effect-transistor (ISFET) with flexible polyimide (PI) extended gate (EG). The DG ISFETs significantly enhanced the sensitivity of pH in electrolytes from 60 mV/pH to 1152.17 mV/pH and effectively improved the drift and hysteresis phenomenon. This is attributed to the capacitive coupling effect between top gate and bottom gate insulators of the channel in silicon-on-transistor (SOI) metal-oxide-semiconductor (MOS) FETs. Accordingly, it is expected that the PI-EG based DG-ISFETs is promising technology for high-performance flexible biosensor applications.

A Study on the SiO2Sensing Layer Used in ISFET (ISFET용 SiO2 감응박막에 관한 연구)

  • A study on the oxidation of SiO2 sensing layer was done at 950, 1000, 105$0^{\circ}C$ under dry O2 atmosphere. The rate determining step around the oxide layer thickness, 1000$\AA$ was different with the oxidation temperature, as follows ; ⅰ) linear growth at 95$0^{\circ}C$ and ⅱ) parabolic growth at 100$0^{\circ}C$ and 105$0^{\circ}C$. The flatness of SiO2 film was observed within $\pm$1% and surface state charge density was reduced by annealing in N2 atmosphere. Finally, pH sensitivity of SiO2 film, in the range of pH 3-9, was 20mV/pH.

Spectral Properties of a pH Responsive Water Soluble Spironaphthoxazine and Its Multi-Switching Property

  • A water soluble spironaphthoxazine (SPO) was synthesized, and its spectral properties were determined. Under UV irradiation, colorless SPO shows intensive blue color while the intensity of its initial fluorescence decreased. In addition, SPO also exhibited high sensitivity to pH stimuli both in colorimetry and fluorometry distinguishing from the spectral appearance observed under UV irradiation. Further, integrating these two optical characteristics a three-state switching system can be established, and all interconversions can be observed by naked-eye.

  • Al2O3 film was chemically deposited by pyrolytic decom,positio of the Al-tri-isopropoxide/N2 system at 350$^{\circ}C$, 30 and 1.86torr. FTIR analysis showed a deposited film was a hydrated alumina and transformed to an anhydrous one after heat treatment(1hr, >800$^{\circ}C$ or 4hr, >500$^{\circ}C$) in N2 atmosphere. This transformation influenced on the CV-hysteresis of Si-Al2O3 structure. Also, a pH sensitivity of EIS(Electrolyte-Insulator-Semiconductor)structure using Si-Al2O3/SiO2 film was 50mV/pH in the range of pH 3 to 7.

  • Helicobacter pylori (H. pylori) survives in acidic and fluctuating pH conditions of the stomach. The pH effect on H. pylori proteins is important for the advanced understanding of its evolution and viability, although this bacterium has the molecular machinery that neutralizes the acidic condition. HP1492 is known as a conserved NifU-like protein from H. pylori. NifU is a nitrogen fixation protein that mediates the transfer of iron-sulfur (Fe-S) cluster to iron-sulfur proteins like ferredoxin. Commonly, the monomeric reduced state of NifU can be converted to the dimeric oxidized state by intermolecular disulfide bond formation. Because it remains unclear that HP1492 actually behaves as known NifU protein, we first found that this protein can adopt both oxidized and reduced forms using size exclusion chromatography. Circular dichroism experiment showed that HP1492 is relatively well-structured at pH 6.5, compared to other pH conditions. On the basis of the backbone resonance assignment of HP1492, we further characterized the residues that are sensitive to pH using NMR spectroscopy. These residues showing large chemical shift changes could be mapped onto the secondary structure of the protein. Our results could provide the foundation for structural and biophysical studies on a wide spectrum of NifU proteins.

Characterization of the Membrane-bound Adenosine Triphosphatase from Corn Roots (옥수수 뿌리로부터 분리한 Membrane-bound ATPase의 특성에 관한 연구)

  • The membrane-bound ATPases were separated on sucrose gradient from corn roots and characterized by pH optima, sensitivity to monovalent salt, Km and Vmax. The pH optima for the activity of all the ATPases associated with 13, 000g pellet and 13, 000~80, 000g pellet were 5 and 9, respectively. The ATPases in Fractions B and C of the 13, 000 g pellet were more active at pH 5 than pH 9. While, in the case of Fractions D, E and F, they were reverse. The activities of the ATPase in Fractions A and C of the 13, 000~80, 000 g pellet were greater at pH 5 than pH 9. On the other hand, the ATPases in Fractions B, D, E, and F were more active at pH 9 than pH 5. The optimum concentraction of ATP for the assay was about 3 to 5 mM. The Km's for the membrane-bound ATPases in 13, 000g pellet and in 13, 000~80, 000 g pellet were 0.25 mM. While Vmax values for 13, 000g pellet were from 8.0 to 12.5 $\mu$M Pi/mg protein/hr. according to pH values, those for 13, 000~80, 000 g pellet were from 35.7 to 55.6 $\mu$M Pi/mg protein/hr. Activities of the membrane-bound ATPases in both 13, 000 g pellet and 13, 000~80, 000 g pellet were stimulated with increasing the concentration of $K^+$.

Comparison of Soil Physicochemical Properties According to the Sensitivity of Forest Soil to Acidification in the Republic of Korea (우리나라 산림토양의 산성화 민감도평가와 그에 따른 토양 이화학적 특성 비교분석)

  • The sensitivity of forest soil to acidification in the Republic of Korea (ROK) was evaluated based on pHH2O, cation exchange capacity (CEC), and base saturation (BS). Sensitivity to acidification was categorized into three grades: adequate level (AL, pH ≧ 4.2, CEC ≧ 15cmol/kg, BS ≧ 15%), caution level (CL, at least one indicator is below AL), and severe Level (SL, all three indicators are below AL). Soil samples were collected from the 65 stationary monitoring plots (40 × 40 ㎢), distributed throughout ROK. Only 19% of soil samples were classified as AL, while 66% and 15% were CL and SL, respectively. The median of pHH2O, CEC, BS, and Ca/Al indicator in AL soils was pH 4.64, 20.7cmol/kg, 29%, and 6.3, respectively. Moreover, BCex (K+, Mg2+, Ca2+) and available phosphorus (AP) concentration compared with a threshold value and molar ratio of BCex and AP to total nitrogen (TN) was high. This indicates that AL soils have a good nutrient condition. The molar Ca/Al ratio, an indicator for toxicity of exchangeable aluminum (Alex), was more than 1, indicating no negative impact of Alex on plant growth. On the contrary, the median of pHH2O, CEC, and BS in SL soils was pH 4.02, 13.2cmol/kg, and 10%, respectively. The Ca/Al index was less than 0.6, which indicates that negative impacts of Alex on plants were high. Furthermore, both the concentration of BCex in SL soils and the BCex/TN ratio were the lowest among the three acidity degrees. This shows that SLsoils can be degraded by soil acidification compared with less acidic soils.

Changes in the Antibacterial Activity of Green Tea Extracts in Various pH of Culture Broth against Staphylococcus aureus and Salmonella typhimurium (배지의 pH에 따른 녹차추출물의 Staphylococcus aureus 와 Salmonella typhimurium에 따른 항균작용)

  • Water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activity. The sensitivity of Staphylococcus aureus and Salmonella typhimurium to the green tea extracts in various pH of culture broth was tested. Tryptic soy broth(TSB) containing 0∼2%(w/v) of green tea extracts was adjusted to pH 5.0∼7.0 and inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacteria. The plate counting method and clear zone test were used to determine inhibitory effect of green tea extracts. Green tea extracts completely inhibited the growth of S. aureus at 0.5% level and bactercidal at 0.5∼1.0% level of GTW and GTE at pH 5.0∼7.0. Green tea extracts were bactercidal to S. typhimurium at 1.5∼2.0% level of GTW and 1.0∼2.0% level of GTE at pH 7.0. Sal. typhimurium was more resistant than S. aureus. in same concentration of green tea extracts at same pH. The resistance of S. aureus and Sal. typhimurium was increased with decreasing pH of culture broth. The morphology of S. aureus cells treated with green tea extracts showed damage of cell wall and cytoplasmic membrane. Severely damaged cells of S. aureus lost electron dense material and cytoplasm. Green tea extracts stimulated autolysis and cell death of S. aureus. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.
