Physicochemical Stabilities of Carthamins from Safflower Petals as Food Colorants (홍화 Carthamin의 식품색소로서의 안정성)

  • The physical and chemical stabilities of carthamin of red pigment from safflower petals were investigated at various conditions of pHs, temperatures, inorganic ions, sugars, light and polysaccharides. The half-life values at pH 3.0, 5.0, 9.0 and 11.0 were 5.3, 5.0, 11.0 and 45.0 h, respectively, at $25^{\circ}C$. Therefore, carthamin is unstable at acidic condition. Carthamin was red, orange and yellow at acidic, neutral and alkaline solutions, respectively. At pH 3.0, carthamin was thermally unstable and the half lives were 3.62, 9.05 and 48.2min at $90^{\circ}C$, $70^{\circ}C$ and $50^{\circ}C$, respectively. Among various inorganic ions, $Al^{3+}$ stabilized carthamin at acidic condition. At pH 5.0, carboxymethylcellulose prolonged the half-lives of carthamin at $25{\sim}90^{\circ}C$. Carthamin was very sensitive to light (20,000 lux) and the half-life was 2.32 min at pH 3.0.

Preparation and Characterization of PE Liposomes Containing Antibody (항체를 포함하는 Phosphatidylethanolamine 리포좀의 제조와 그 특성)

  • A target-sensitive liposome was prepared by using a dioleoyl-phosphatidylethanolamine(DOPE) and a palmitic acid coupled antibody(p-IgG). For the preparation of stable PE-liposomes, the key factors such as antibody modification method with palmitic acrid, molar ratio of p-IgG to lipid and the amount of various additives, were examined. The optimum molar ratio of p-IgG to lipid was found to be $2.5{\times}10^{-4}$ and the final concentration of deoxycholate for the stable liposome formation was about 0.09%. Two kinds of target-sensitive liposomes, containing polyclonal anti-SRBC(Sheep Red Blood Cell)-antibody and monoclonal anti-${\beta}$-HCG(Human Chorionic Gonadotropin)-antibody, were successfully prepared. The destabilization of liposomes was examined by measuring the release of calcein entrapped in the liposome vesicles. Calcein was released only when the liposomes were contacted with the specific target cells. The calcein release with non-specific target cells was negligible. From this result, it is clear that p-IgG is indispensible for the maintenance of stable PE-liposome and the calcein release is mainly due to the specific interactions between the liposomes containing antibody and the target cells containing antigen.

Comparison of Electrocoagulation and Chemical Coagulation in Removal on Water Treatment (정수처리에서 전기응집과 화학응집의 처리효율 비교)

  • Electrocoagulation has been suggested as a promising alternative to conventional coagulation. The process is characterized by reduced sludge production, no requirement for chemical use, and ease of operation. However, this coagulation has scarcely been studied in water purifying process. This study was performed several batch experiments to compare turbidity removal between electrocoagulation and chemical coagulation. In addition, characteristics of floe were evaluated with zeta potential and particle size distributions. Electrocoagulation showed a relatively higher removal of turbidity (approximately 5%) with the same aluminum amount than conventional chemical coagulation. In addition, turbidity removal by electrocoagulation was less sensitive to pH and was greater for more extensive pH range than chemical coagulation. The results of zeta potential and floc size distributions illustrated that electrocoagulation provided the preferable conditions for coagulation such as zeta potential close to zero millivolt and increased portions of particles in the range of 40 and $100{\mu}m$.

  • The design and development of fluorescent chemosensors have recently been intensively explored for sensitive and specific detection of environmentally and biologically relevant metal ions in aqueous solution and living cells. Herein, we report the photophysical results of rhodamine B based fluorogenic and chromogenic receptor for selective copper detection in the complete organic or mixed aqueous-organic media at neutral pH under ambient condition. The ligand exhibited the remarkable increment in the fluorescence emission and UV-visible absorption signal intensities at 587 and 547 nm, respectively, on induction of copper ion while the ligand solution remain completely silent on addition of varieties of other metal ions.

  • In this study, real-time and label-free methods for monitoring the enzymatic reaction of urease, which releases ammonia through the hydrolysis of urea in an aqueous solution, were developed using a liquid crystal (LC)-based pH sensor. Nematic liquid crystal 4-cyano-4'-pentylbiphenyl (5CB), doped with 4'-pentyl-biphenyl-4-carboxylic acid (PBA), exhibited a shift in optical appearance from bright to dark when it was in contact with ammonia generated from the enzymatic reaction between urease and urea. This optical change was attributed to the anchoring transitions of LCs caused by hydrophobic interactions between the tails of deprotonted PBA ($PBA^-$) molecules and the LCs at the aqueous/LC interface. This novel technique holds great promise for the sensitive detection of urease along with its substrates and inhibitors.

  • Casein was chemically modified with acetic, succinic, and maleic anhydride and changes in functional pro-perties were evaluated as affected by the degree of modification. Chemical modification resulted in casein with unique functional properties depending upon the type of anhydrid used and the degree of modification. It was possible to control heat coagulation, calcium precipitability, forming and emulsion capacity and stability. At pH 4.5 heat coagulation was 0% in the case 74.1% acetylated casein; on the contrary, succinylation and maleyation resulted in highly heat sensitive protein. Foaming properties were improved markedly by suc-cinylation and maleylation at pH 4.5. However, emulsifying properties were enhanced only by maleylation.

  • Isocitrate lyase from crude extract of Saccharomycopsis lipolytica ATCC44601 and MX9-11RX8 temperature-sensitive mutant was purified about 54 times and 87 times, respectively by ammonium sulfate fractionation, Toyo peal HW-55F gel filtration and DEAE-Cellulose ion exchange chromatography, The molecular weight of the purified isocitrate lyase from this yeast was estimated to be 230, 000 by gel filtration on Sephadex G-200, and SDS-polyacrylamide Eel electrophoresis showed that the enzyme consisted of four identical or similar subunits with a molecular weight of 59, 000 and the enzyme showed optimum activity at pH 6.9.

  • Lactic acid bacteria in Kimchi fermentation were tested for inhibitory activity against Gram positive bacteria and Gram negative bacteria. The Lactobacillus brevis (KCCM 35464) was found to produce a antimicrobial substance. It showed relatively wide range of inhibition spectrum against gram positive and gram negative bacteria and maintained the inhibitory activity between pH 4.0 and pH 9.0. The antimicrobial substance was obtained in the stationary growth phase and was purified by gel chromatography. The inhibitory effect of the antimicrobial substance on sensitive bacterial strains was determined by filter paper test. The activity of antimicrobial substa- nce was stable at 75$\circ$C. On the basis of its electrophoretic pattern is SDS-PAGE, antimicrobial substance appeared as a single band of 59 KDalton.

  • The s-type pyocin was purified from the lysate of the mitomycin C-induced Pseudomonas aeruginosa 90-2-2205 cells by the other of ammonium sulfate precipitation, DEAE-Sephadex A-50 chromatography and Sephadex G-200 gel filtration. The purity was confirmed by the polyacry-lamide gel electrophoresis. The molecular weight of the purified pyocin was estimated 180, 000 by gel filtration. The pyocin was analyzed to be a complex of some polypeptides by the SDS-PAGE. The pyocin was stable by heat treatment and at pH 6-7.5 by adding 10-3% gelatin and 0.2M NaCl to the 10mM Tris-HCl buffer (pH7.5). Its killing action against the sensitive cells was assumably a single hit process.
