창과감초(Glycyrrhiza inflata Batal)는 한약재의 조화를 돕고 해독, 항염증, 항궤양 등의 약리작용으로 한방에서 널리 이용되는 약용식물이다. 본 연구에서는 지방세포와 조골세포에서 감초의 생리활성을 확인하고자 감초에탄올추출물(GBE)을 이용하여 세포분화 촉진여부를 조사하였다. GBE의 세포독성여부를 통해 안전하다고 확인된 $1{\sim}30{\mu}g/mL$의 농도 내에서 실험이 진행되었고 지방세포 분화조건에서 다분화능 세포 C3H10T1/2과 지방전구세포 3T3-L1의 Oil Red O 염색을 통해 GBE의 지방세포분화 촉진효과를 확인할 수 있었다. 또한 지방세포의 핵심전사조절인자인 $PPAR{\gamma}$와 그의 표지유전자 aP2, AdipoQ, $C/EBP{\alpha}$의 발현량 증가를 통해 GBE의 지방세포분화 촉진효과를 재확인하였다. 이와 일관된 결과로서 조골세포 분화조건에서 다분화능 세포 C3H10T1/2과 조골전구세포 MC3T3-E1의 ALP 염색을 통해 GBE의 조골세포분화 촉진효과를 확인하였고 조골세포 표지유전자인 ALP, RUNX2, osterix, collagen의 발현량 증가를 통해 GBE의 조골세포분화 촉진효과를 재확인하였다. 이에 따라 본 연구에서는 GBE의 지방세포와 조골세포 분화효과를 매개하는 감초의 구성성분을 조사하기 위해 GA(glycyrrhizic acid)와 LA(licochalcone A)의 분화촉진 여부를 확인한 결과, GA는 영향을 주지 않으나 LA가 GBE의 세포분화효과를 매개한다고 사료된다. 따라서 본 연구에서는 제2형 당뇨와 그에 수반되는 골질환과 골다공증에 대한 치료 소재로서 GBE와 그의 생리활성을 매개할 수 있는 LA의 가능성을 보았으며 GBE에서 분리되는 다양한 화합물의 동정 및 생리활성 효과, LA와의 상승효과 등의 추후 연구가 기대된다.
본 연구에서는 이온쌍 역상 HPLC/UV를 이용하여 건강한 한국인에서 분리된 말초혈액단핵구(PBMCs)에서 이노신 5'-일인산 탈수소효소(IMPDH)의 활성을 측정하였다. IMPDH는 이노신 5'-일인산(IMP)을 잔토신 5'-일인산(XMP)로 전환시키는 베타-니코틴아마이드 아데닌 디뉴클로티드 수화물(${\beta}-NAD^+$) 의존성 탈수소효소이며, 이것의 활성은 기질인 IMP와 조효소인 $NAD^+$의 존재 하에서 분해한 PBMCs로 부터 생성된 XMP에 해당하는 HPLC 크로마토그램을 정량적으로 분석함으로써 측정하였다. 생성된 XMP는 260 nm에서 검출하였다. 이동상으로는 7 mM tetra-n-butylammonium hydrogen sulfate가 포함된 37 mM potassium dihydrogen phosphate (pH 5.5)와 methanol의 혼합용액(85:15, v/v)을 사용하였으며, 유속은1 mL/min이었다. 정량 범위는 $0.2-50.0\;{\mu}M$이었으며, 이 때 정량 한계(LOQ)는 $0.2\;{\mu}M$이었다. 또한, 본 연구에서 확립된 시험법은 일내 정밀성(0.88-1.47%), 정확성(98.74-99.99%)과 일간 정밀성(0.85-5.24%), 정확성(99.95-101.65%)을 측정하여 검증하였다. 11명의 건강한 한국인에 대한 IMPDH 활성 측정 결과, 18.29-36.60 nmol/h/mg protein(평균값 $27.70{\pm}6.28\;nmol/h/mg$ protein)이었다.
연구배경 : 산소기가 여러 종류의 급성 폐손상에 중요한 역할을 한다는 것은 이미 잘 알려진 사실이다. 생체내에는 여러 항산화 방어기전이 존재하는데, SOD는 두 개의 superoxide radical이 과산화수소와 산소로 dismutation되는 과정을 $10^4$배 촉진시키는 효소로서 산소기에 대한 일차적인 방어기전으로 작용한다. Eukaryotic 세포내에는 두 가지 종류의 SOD가 존재하는데, 하나는 세포질에 위치하고 이중체(dimeric)의 구조를 가지며 구리와 아연을 포함하는 효소(CuZnSOD)이고, 또 하나는 미토콘드리아에 있고 사중체(tetrameric)의 구조를 갖는 망간을 포함하는 효소(MnSOD)이다. 내독소에 의한 백서의 급성 폐손상 모델에서 내독소 투여 후 시간 경과에 따른 백서 폐장의 MnSOD와 CuZnSOD의 유전자 발현을 관찰하여 이를 급성 폐손상의 양상과 비교하고자 본 연구를 시행하였다. 방법 : 백서에 E. coli의 내독소를 투여한 후 0, 1, 2, 4, 6, 12, 18, 그리고 24시간 후에 백서를 희생시켜 폐장을 얻은 후 폐장의 총 RNA를 single step phenol extraction 방법으로 추출하였다(n=3, respectively). 총 RNA를 formaldehyde를 함유한 1.2% agarose gel 에 전기영동하고, gel의 RNA를 nylon membrane으로 transfer시켰다. Nylon membrane을 $^{32}P$로 labeling시킨 MnSOD와 CuZnSOD를 probe로 하여 hybridization하고 autoradiography를 시행하였다. 결과 : 내독소률 투여하고 4시간후부터 MnSOD mRNA가 발현되기 시작하여 6시간에 최고치를 보였고, 약 12시간까지 지속되었으며, 24시간이 경과한 후에는 대조군의 수준으로 감소되었다. CuZnSOD 유전자는 내독소를 투여하고, 1 시간후부터 발현되기 시작하여 24시간까지 지속되었는데, 18시간에 최고치에 도달하였다. 결론 : 이상의 결과는 SOD가 백서에서 내독소에 의해 유발된 급성 폐손상에 대한 방어기전에 관여 할 가능성을 시사하는 것으로 생각된다.
본 연구에서는 양파껍질 추출물의 항균, 항산화활성과 tyrosinase 및 elastase 저해활성을 측정하였다. 피부 상재균에 대한 양파껍질 추출물의 항균활성 측정결과, 황색포도상구균(S. aureus)에 대한 에틸아세테이트 분획의 MIC는 0.06%로 매우 큰 항균활성을 나타내었다. 에틸아세테이트 분획 중 aglycone 분획은 우수한 free raical [1-diphenyl-2-picrylhydrazyl (DPPH)] 소거활성($FSC_{50}=5.05{\mu}g/mL$)을 나타내었다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}-EDTA/H_2O_2$ 계에서 에틸아세테이트 분획과 aglycone 분획의 총항산화능($OSC_{50}$)은 각각 0.05 및 $0.03{\mu}g/mL$이었다. Rose-bengal로 증감된 사람 적혈구의 용혈 실험에서 aglycone 분획은 $25{\mu}g/mL$의 농도에서 매우 큰 세포보호활성(${\tau}_{50}=480.3{\pm}0.2min$)을 나타냈다. 양파껍질 추출물 중 에틸아세테이트 분획과 aglycone 분획은 tyrosinase 저해활성($IC_{50}$)이 각각 9.16, $8.68{\mu}g/mL$이었고, aglycone 분획의 elastase의 저해활성은 $14.12{\mu}g/mL$이었다. 양파껍질 추출물 중 ethyl acetate 분획 0.1% 함유하는 크림의 경표피 수분 손실량은 추출물 함유 크림이 $6.8g/m^2h$로 대조군($8.3g/m^2h$)에 비해 경표피 수분 손실량 감소 효과를 나타내었다. 이상의 결과들은 양파껍질 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히, 태양 자외선에 노출된 피부에서 항산화제로서 그리고 항노화제로서 작용할 수 있음을 가리킨다. 따라서 양파껍질 추출물은 미백이나 항주름관련 새로운 기능성화장품에의 응용이 가능함을 나타낸다.
The human muscarinic acetylcholine receptor (mAChR) subtypes Hml, Hm2 and Hm3 have been expressed in insect cells (Spodoptera frugiperda, Sf9) using the baculovirus expression system. Expression of relevant DNA, transcript and receptor proteins was identified by PCR, Northern blotting and [$^{3}H$]QNB binding, respectively. As assessed by [$^{3}H$]QNB binding sites, yields of muscarinic receptors in membrane preparations in this study were as about 5-20 times high as those in mammalian cells reported in previous studies. The [$^{3}H$]QNB competition binding studies with well-known subtype-selective mAChR antagonists showed that the receptors expressed in Sf9 cells retain the pharmacological characteristics expected for the ml , m2 and m3 muscarinic receptors. The ml-selective antagonist, pirenzepine, displayed a considerably higher affinity for Hml by 110-fold and 35-fold than for Hm2 and Hm3, respectively, The m2-selective methoctramine displayed a significantly higher affinity for Hm2 than for Hml and Hm3 (10- and 26-fold, respectively). p-F-HHSiD exhibited high affinity for Hm3 that is not significantly different from those for Hml, but 66-fold higher than its affinity for Hm2. The functional coupling of the recombinant receptors to second messenger systems was also examined. While both Hml and Hm3 stimulated phosphoinositide hydrolysis upon activation by carba-chol, Hm2 produced no response. On the other hand, activation of mAChRs induced the inhibition of forsko-lin-stimulated cyclic AMP formation in Hm2-expressing cells, whereas the significant dose-dependent increase in or poor response on cyclic AMP formation were produced in Hml or Hm3-expressing cells, respectively. These results indicate the differential coupling of recombinant Hml, Hm2 and Hm3 receptors expressed in SF9 cells to intracellular signalling system.
It has been known that ATM plays a central role in response of cells to ionizing radiation by enhancing DNA repair. We have investigated the feasibility of increasing radiosensitivity of tumor cells with the use of ATM inhibitors such as caffeine, pentoxifylline and wortmannin. Human colorectal cancer RKO.C cells and RKO-ATM cells (RKO cells overexpressing ATM) were used in the present study. The clonogenic cell survival in vitro indicated that RKO-ATM cells were markdely radioresistant than RKO.C cells. Treatment with 3 mM of caffeine significantly increased the radiosensitivity of cells, particulary the RKO-ATM cells, so that the radiosensitivity of RKO.C cells and RKO-ATM cells were almost similar. The radiation induced G2/M arrest in RKO-ATM cells was noticeably longer than that in RKO.C cells and caffeine treatment significantly reduced the length of the radiation induced G2/M arrest in both RKO.C and RKO-ATM cells. Pentoxifylline and wortmannin were also less effective than caffeine to radiosensitize RKO.C or RKO-ATM cells. However, wortmannin was more effective than caffeine against human lung adenocarcinoma A549 cells indicating the efficacy of ATM inhibitor to increase radiosensitivity is cell line dependent. For in vivo study, RKO.C cells were injected s.c. into the hind-leg of BALB/C-nuslc nude mice, and allowed to grow to 130mm3 tumor. The mice were i.p. injected with caffeine solution or saline and the tumors irradiated with 10 Gy of X-rays. The radiation induced growth delay was markedly increased by 1-2 mg/g of caffeine. It was concluded that caffeine increases radiosensitivity of tumor cells by inhibiting ATM kinase function, thereby inhibiting DNA repair, that occurs during the G2/M arrest after radiation.
PURPOSE. The objective of this study was to investigate the biologic effects of enamel matrix derivative (EMD) with different concentrations on cell viability and the genetic expression of human gingival fibroblasts (HGF) to zirconia surfaces. MATERIALS AND METHODS. Immortalized human gingival fibroblasts (HGF) were cultured (1) without EMD, (2) with EMD $25{\mu}g/mL$, and (3) with EMD $100{\mu}g/mL$ on zirconia discs. MTT assay was performed to evaluate the cell proliferation activity and SEM was carried out to examine the cellular morphology and attachment. The mRNA expression of collagen type I, osteopontin, fibronectin, and TGF-${\beta}1$ was evaluated with the real-time polymerase chain reaction (RT-PCR). RESULTS. From MTT assay, HGF showed more proliferation in EMD $25{\mu}g/mL$ group than control and EMD $100{\mu}g/mL$ group (P<.05). HGFs showed more flattened cellular morphology on the experimental groups than on the control group after 4h culture and more cellular attachments were observed on EMD $25{\mu}g/mL$ group and EMD $100{\mu}g/mL$ group after 24h culture. After 48h of culture, cellular attachment was similar in all groups. The mRNA expression of type I collagen increased in a concentration dependent manner. The genetic expression of osteopontin, fibronectin, and TGF-${\beta}1$ was increased at EMD $100{\mu}g/mL$. However, the mRNA expression of proteins associated with cellular attachment was decreased at EMD $25{\mu}g/mL$. CONCLUSION. Through this short term culture of HGF on zirconium discs, we conclude that EMD affects the proliferation, attachment, and cell morphology of HGF cells. Also, EMD stimulates production of extracellular matrix collagen, osteopontin, and TGF-${\beta}1$ in high concentration levels. CLINICAL RELEVANCE. With the use of EMD, protective barrier between attached gingiva and transmucosal zirconia abutment may be enhanced leading to final esthetic results with implants.

In order to discover new types of 5-hydroxytryptamine antagonists, we have devoted our attention to investigating naturally occurring compounds having anti-5HT activity in vitro. Recently, ${\gamma}$-mangostin [1,3,6,7-tetrahydroxy-2,8-bis(3-methyl-2-bytenyl)-9H-xanthen-9-one] from the fruit hull of Garcinia mangostana Linn has been shown to be a selective antagonist for 5-hydroxytryptamine$_{2A}$ receptors in smooth muscle and platelets. It is of interesting that y-mangostin which does not have a nitrogen atom, possesses marked 5-$HT_{2A}$ receptor blocking activity. The present study was undertaken to investigate the effects of ${\gamma}$-mangostin on central 5-HT receptors by using animal behavioural models. Intracerebronventricular injection of ${\gamma}$-mangostin (10-40n mol/mouse) inhibited 5-fluoro-${\alpha}$-methyltryptamin (5-FMT) (45 mg kg$^{-1}$, i.p.)-induced head-twitch response in mice in the presence or absence of citalopram (5-HT-uptake inhibitor). Neither the 5-FMT- nor the 8-hydroxy-2-( di-n-propylamino )tetralin (5-HT$_{1A}$-agonist)-induced 5-HT syndrome (head weaving and hindlimb abduction) was affected by ${\gamma}$-mangostin. The locomotor activity stimulated by 5-FMT through the activation of at-adrenoceptors did not alter in the presence of ${\gamma}$-mangostin. 5-HT-induced inositol phosphates accumulation in mouse brain slices was abolished by ketanserin. ${\gamma}$-Mangostin caused a concentration-dependent inhibition of the inositol phosphates accumulation and the binding of [$^3H$]-spiperone, a specific 5-$HT_{2A}$ receptor antagonist, to mouse brain membranes. Kinetic analysis of the [$^H3$]-spiperone binding revealed that ${\gamma}$-mangostin increased the $_{d}$ value without affecting the $B_{max}$ value, indicating the mode of the competitive nature of the inhibition by ${\gamma}$-mangostin. These results suggest that ${\gamma}$-mangostin inhibits 5-FMT-induced head-twitch response in mice by blocking 5-$HT_{2A}$ receptors not by blocking the release of 5-HT from the central neurone. ${\gamma}$-Mangostin is a promising 5-$HT_{2A}$ receptors antagonist in the central nervous system.m.

The fungal strain EML-DML3PNa1 isolated from leaf of white dogwood (Cornus alba L.) showed strong nematicidal activity with juvenile mortality of 87.6% at a concentration of 20% fermentation broth filtrate at 3 days after treatment. The active fungal strain was identified as Aspergillus oryzae, which belongs to section Flavi, based on the morphological characteristics and sequence analysis of the ITS rDNA, calmodulin (CaM), and β-tubulin (BenA) genes. The strain reduced the pH value to 5.62 after 7 days of incubation. Organic acid analysis revealed the presence of citric acid (515.0 mg/kg), malic acid (506.6 mg/kg), and fumaric acid (21.7 mg/kg). The three organic acids showed moderate nematicidal activities, but the mixture of citric acid, malic acid, and fumaric acid did not exhibit the full nematicidal activity of the culture filtrate of EML- DML3PNa1. Bioassay-guided fractionation coupled with 1H- and 13C-NMR and EI-MS analyses led to identification of kojic acid as the major nematicidal metabolite. Kojic acid exhibited dose-dependent mortality and inhibited the hatchability of M. incognita, showing EC50 values of 195.2 μg/ml and 238.3 μg/ml, respectively, at 72 h post-exposure. These results suggest that A. oryzae EML-DML3PNa1 and kojic acid have potential as a biological control agent against M. incognita.
Zespri green kiwi의 무기질은 K, P, Ca 순으로 많았으며, 아미노산은 glutamic acid, aspartic acid, arginine 순으로 많이 함유되어 있었다. 지방산 함량은 a-linolenic acid, oleic acid 순으로 많이 함유되어 있었으며, 유리당은 fructose 함량이 가장 높았다. Vitamin C와 ${\beta}$-carotene 함량은 각각 29.21 mg/100 g, $1.35{\mu}g/100g$이었다. Total phenolics는 n-hexane 분획물이 11.83 mg GAE/g으로 가장 높았으며, ABTS radical 소거활성 및 지질 과산화 억제활성에서는 농도 의존적으로 in vitro 항산화 활성이 증가하는 경향을 보였다. 또한 oxidative stress에 대한 신경세포 보호효과 역시 농도의존적 경향을 보여주었다. 본 연구 결과를 종합해볼 때, 생리활성 소재로서의 phenolics를 함유한 kiwi fruit (A.deliciosa)는 in vitro 항산화 활성과 함께 신경세포 보호효과를 기초로 한 고부가 가치 건강지향식품 소재로서의 활용가치가 높다고 판단된다.

통해 사전에 공지합니다.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
