Characteristics of Growth Response and Nitrogen Fixation of Meiilotus suaveolens Seedlings treated with Al (알루미늄 독성에 의한 전동싸리 유식물의 질소고정과 생장반응의 특성)

Accumulation of metals In each organ of M. suaveojens was Increased UC the lowering of pH. Al contents In leaf and root treat- ed with 30ppm Al at pH 4.2 on the 28th day after treatment were 8 and 11 folds higher than those of control, while the contents with 100ppm AA were 21 and 24 folds as compared to control. The significent inhibition in Al contents was induced by 100ppm AA and Ca at pH 6.5. Increased metals inhibited higher at pH 4.2 and the growth inhibition due to Al was reversed by the addition of Ca, suggesting that growth correlates to the pH value. Chlorophyll contents in leaves increased during growth stave were inhibited by Al treatments. The biomass was decreased UD the lowering of pH and the increase of concentrations. 100ppm Ca treatment resulted in 5.1-5.9% increase of the biomass as compared to that of 100ppm Al. Specific nitrogen fuation activities In nodules In the 100ppm Al at pH 4.2 and pH 6.5 were reduced to 35. 2% and 52.2% of control, on the 28th day after treatment, while the combination of Al and Ca induced reduced by 10% as compared to control with the treatment of 100ppm AA at pH 4.2 due to the inhibition bination of 100ppm Al and Ca and lowering of pH.

  • This study was investigated to evaluate the effect of the sodium ion and pH on toxicity of dinitrophenol at high concentrations (0.41 to 0.54 mM), over a sodium concentration range of 0.1 mM to 107 mM and over a pH range of 5 to 9. The concentration of sodium ions in the activated sludge mixed liquor seemed to have very little effect on dinitrophenol toxicity. However, lack of sodium in the growth media resulted in a reduction of the dinitrophenol degradation rate by bacterial isolate from the activated sludge culture, which has been identified as Nocardia asteroides. Dinitrophenol inhibition was found to be strongly dependent on mixed liquor pH. The dinitrophenol degradation rate was highest in the pH range of 6.95 to 7.84; at pH 5.94 degradation of 75 mg/L dinitrophenol was significantly inhibited; at pH < 5.77, dinitrophenol degradation was completely inhibited after approximately 30% of the dinitrophenol was degraded.

  • The low-pH biofiltration system in laboratory experiments demonstrate defective performance for treating H2S. When leachate pH was in the range of 1.5 to 4, the biofilters in three different media removed H2S wi th efficiencies greater than 99% while it was treated as a single contaminant. The posibility of using a single-stage low pH biofilter depends on its performance in treating VOCs. During Phase 2, a single-stage biofilter was effective for treating mixtures of H2S and toluene with toluene concentrations below 20ppm and leachate pH between 2 and 3.5. Biofiltration of xylene was ineffective when pH was lower than 1.5. The treatment system acclimated most slowly to benzene, and treatment of benzene was apparently subject to some competive inhibition from xylene and toluene. However. co-treatment was possible after some acclimation time. Xylene was not easily treated, with higher elimination capacities and no sign of competitive inhibition.

Fermentation Characteristics and anti-Helicobacter pylori Activity of Aqueous Broccoli Fermented by Lactobacillus plantarum MG208

  • Helicobacter pylori infection causes gastrointestinal diseases such as chronic gastritis, peptic ulcers, and may lead to gastric cancer. Several studies have reported that lactobacilli present on broccoli show inhibitory activity against H. pylori. Here, we evaluated aqueous broccoli, fermented by Lactobacillus plantarum MG208, for its fermentation characteristics and anti-H. pylori activities including antibacterial activity, growth inhibition, anti-adhesion, and urease inhibition. The results indicated that the fermentation characteristics changed significantly depending on the amount of aqueous broccoli used for fermentation (p <0.05). There was no significant difference between the samples before fermentation (p >0.05). However, a significant concentration-dependent difference was noted in antibacterial activity and urease inhibition (p <0.05) following the addition of aqueous broccoli. Growth inhibition in the 10 mg/mL sample was significantly higher as compared to the negative control and similar to that with amoxicillin (positive control) (p <0.05). Anti-adhesion activity of aqueous broccoli was also significantly different (p <0.05) from the negative control. Therefore, aqueous broccoli fermented by L. plantarum MG208 could prove useful as a functional diet for protection of the gastric environment against H. pylori infection.

Inhibitory Effect on Tyrosinase and Xanthine Oxidase, and Nitrite Scavenging Activities of Schizandrae Fructus Extract by Gamma Irradiation (감마선 조사한 오미자 추출물의 아질산염 소거능, Tyrosinase와 Xanthine Oxidase 저해 효과)

  • This study was performed to investigate the effects of tyrosinase inhibition, xanthine oxidase inhibition and nitrite-scavenging effects of gamma-irradiated Schizandrae Fructus extracts from different solvents. Schizandrae Fructus was extracted by hot water, ethanol, acetone and methanol, and the extracts were irradiated 10, 20 and 30 kGy with gamma rays. All extracts from Schizandrae Fructus showed inhibition effect against tyrosinase. Tyrosinase inhibition effect of Schizandrae Fructus were higher in solvent extracts than hot water extracts by irradiation. The Schizandrae Fructus extracts had a higher inhibitory effect against xanthine oxidase, and the effect was not changed by irradiation. Nitrite scavenging activity, which was measured at various pH conditions (1.2, 3.0, 6.0), was the highest in Schizandrae Fructus extracts at pH 1.2 and 3.0. Hot water extracts provided higher nitrite scavenging effect than those of the methanol, ethanol and acetone extracts. Gamma-Irradiation may not influence on biological activites of the extracts when irradiated up to 30 kGy.

Effect of Alcohol and Tobacco on Sucrose Fermentation by Streptococcus mutans (알콜과 담배가 Streptococcus mutans의 자당발효에 미치는 영향에 관한 연구)

  • Streptococcus mutans 10449 was cultured in sucrose-containing BHI broth with ethyl alcohol of different concentration from 1% to 18%, The pH of culture media was from pH 7.00 to pH 5.00. Tobacco smoke and tobacco extract were also used. Ethyl acohol began to inhibit sucrose fermentation by S. mutans at 2% and completely inhibited it between 9% and 18%. The lower the pH of media was, the stronger the inhibition of ethyl alcohol became. 9% Ethyl alcohol completely inhibited sucrose fermentation by S. mutans below pH 5.50, Inhibition by tobacco extract was obvious, but it did not inhibit the growth of S. mutans also. Therefore, the increase of caries activity in drinkers and smokers could be the result of indirect effect of alcohol and tobacco by oral ecology, behavior, or systematic course, rather than the result of direct effect of alcohol and tobacco to plaque bacteria and their metabolism.

Characteristics of the Algal Growth inhibition Substances Produced by Alteromonas sp. SR-14 (Alteromonas sp. SR-14가 생산하는 조류증식 저해 물질의 특성)

  • In previous reports, the authors isolated the algicidal marine bacterium, Alteromonas sp. SR-14 and demonstrated its growth inhibition of diatom, Chaetoceros calcitrans (C. calcitrans). In this paper, we studied the effects of cell free culture filtrate of Alteromonas sp. SR-14 on the growth of C. calcitrans, and the characteristics of the algal growth inhibition substance. The culture filtrate of Alteromonas sp. SR-14 grown in peptone broth showed growth inhibition activity against C. calcitrans. The reasonable culture conditions of the bacterium for producing of algal growth inhibition substances were $15~20^{\circ}$ in temperature, 7.0-9.0 in pH and $23~30{\textperthousand}$ in salinity, respectively. The algal growth inhibition activity of culture filtrate was increased from stationary phase in growth curve of Alteromonas sp. SR-14. The molecular weights of algal growth inhibition substances produced by Alteromonas sp. SR-14 were ranged about from 3 KDa to 12 KDa. Among the substances, less than 10 KDa fraction were stable by heating at $100^{\circ}$ for 10 minutes, while more than 10 KDa fraction were heat labile. According to the experimental results, the algal growth inhibition substance produced by the bacterium was not a single compound.

  • Effects of the ultrasonic treatment on plant represents antifeeding activity and chitin synthesis inhibition activity against diamondback moth (Plutella xylostella L.) by a leaf-dipping method at a concentration of 5,000ppm. Methanol extracts of leaves of the ultrasonic treatment on plant was sequentially partitioned with n-hexane, chloroform, ethlyacetate, butanol and water fractions, and bioassayed. Antifeeding activities were observed in n-hexane, chloroform and ethlyacetate fractions. N-hexane fraction has 94.6% of strongest antifeeding activity against P. xylostella. From the n-hexane sequentially partitioned showed H6 and H8 fractions of antifeeding activities against P. xylostella. And chitin synthesis inhibition activity against P. xylostella was obtained the methaol, n-hexane and water fractions.

Quality Deterioration Suppressing Effects of pH Regulators during Heat Treatment of Concentrated Blueberry Puree (농축 블루베리 퓌레 열처리 시 pH 조절제의 품질 저하 억제 효과)

  • Purpose: In this study, we aimed to minimize quality degradation of concentrated blueberry puree by thermal treatment at $90^{\circ}C$ for 5 min, with citric acid (CA) and acidic sodium metaphosphate (ASM) as pH modifiers. Methods: The color values, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were evaluated for quality parameters of concentrated blueberry puree. Results: The highest values of color value, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were 1% CA+0.5% ASM followed by 1% CA and control. The quality parameters of concentrated blueberry puree were significantly different between control and 1% CA+0.5% ASM treatment (p<0.05). Conclusion: During thermal treatment, quality degradation of concentrated blueberry puree was reduced by 1% CA+0.5% ASM treatment, which is expected for new acidulants.

$^{31}p$ Nuclear Magnetic Resonance Studies of Acetic Acid Inhibition of Ethanol Production by Strains of Zymomonas mobilis

  • In vivo $^31p$ Nuclear Magnetic Resonance ($^31p$NMR) and metabolic studies were carried out on an acetic acid tolerant mutant, Zymomonas mobilis $ZM4/Ac^R$, and compared to those of the parent strain, Z. mobilis ZM4, to evaluate possible mechanisms of acetic acid resistance. This investigation was initiated to determine whether or not the mutant strain might be used as a suitable recombinant host far ethanol production from lignocellulose hydrolysates containing various inhibitory compounds. $ZM4/Ac^R$ showed multiple resistance to other lignocellulosic toxic compounds such as syringaldehyde, furfural, hydroxymethyl furfural, vanillin, and vanillic acid. The mutant strain was resistant to higher concentrations of ethanol or lower pH in the presence of sodium acetate, compared to ZM4 which showed more additive inhibition. in vivo $^31p$ NMR studies revealed that intracellular acidification and de-energization were two mechanisms by which acetic acid exerted its inhibitory effect. For $ZM4/Ac^R$, the internal pH and the energy status were less affected by sodium acetate compared to the parent strain. This resistance to pH change and de-energization caused by acetic acid is a possible explanation for the development of resistance by this strain.
