Recovery over Time of Production Performance and Biological Functions of Laying Hens after Withdrawal Toxic Levels of Dietary Roxarsone

  • Roxarsone (3-nitro-4-hydroxyphenylarsonic acid) has been used as feed additives in poultry industries to improve production and control coccidiosis. The effect of high dietary levels of Roxarsone (ROX) on the performance and function of internal organs and the kinetics of recovery as well as its after-effects were examined in laying hens. The inclusion rates of ROX were 0, 100, 200, 300, and 400 mg per kg feed. Inclusion up to 200 mg did not show any adverse effects (p>0.05), whereas in the 300 and 400 mg groups, significant effects, particularly in the latter, were observed for three weeks after ROX addition (p<0.05). Recovery of the physical appearance occurred soon after ROX addition was withdrawn. Recovery of performance and internal organs, however, appeared to be dependent on the amount of residual ROX in the body; as the amount of ROX decreased, the toxic effect of ROX also decreased. In the third week after the withdrawal of ROX, complete recovery was observed in the lower dosage groups (100 or 200 mg groups) (p>0.05), whereas in the higher dosage groups (300 or 400 mg groups), recovery took at least five weeks; when complete recovery was observed in egg production and in liver weight (p>0.05). On the other hand, ROX might have damaged the liver and other tissues. The recovery of liver weight was probably due to accumulation of fatty particles rather than repair. It appeared, therefore, there were little after-effects of ROX on the hen's physical appearance, but some internal organs were probably damaged.

Relationship between Thin Film Thickness and Structural Properties of BaTiO3 Thin Films Grown on p-Si Substrates (p-Si 기판에 성장한 BaTiO3 박막의 두께와 구조적 특성과의 관계)

  • In this study, $BaTiO_3$ thin films were grown by RF-magnetron sputtering, and the effects of the thin film thickness on the structural characteristics of $BaTiO_3$ thin films were systematically investigated. Instead of the oxide substrates generally used for the growth of $BaTiO_3$ thin films, p-Si substrates which are widely used in the current semiconductor processing, were used in this study in order to pursue high efficiency in device integration processing. For the crystallization of the grown thin films, annealing was carried out in air, and the annealing temperature was varied from $700^{\circ}C$. The changed thickness was within 200 nm~1200 nm. The XRD results showed that the best crystal quality was obtained for ample thicknesses 700 nm~1200 nm. The SEM analysis revealed that Si/$BaTiO_3$ are good quality interface characteristics within 300 nm when observed thickness. And surface roughness observed of $BaTiO_3$ thin films from AFM measurement are good quality surface characteristics within 300 nm. Depth-profiling analysis through GDS (glow discharge spectrometer) showed that the stoichiometric composition could be maintained. The results obtained in this study clearly revealed $BaTiO_3$ thin films grown on a p-Si substrate such as thin film thickness. The optimum thickness was 300 nm, the thin film was found to have the characteristics of thin film with good electrical properties.

Antimicrobial Activity of Methanol Extract from Rheum tanguticum against Food Hazardous Microorganisms and the Composition of the Extract (식품위해성 미생물에 대한 대황(Rheum tanguticum) 메탄올 추출물의 항균활성 및 성분분석)

  • Daehwang (Rheum tanguticum) was extracted by methanol, and the antimicrobial activities of the extract, against some food hazardous microorganisms, were investigated by the paper disc method. The minimum inhibitory concentration (MIC) of the extract against the microorganisms was determined, and the extract components were analyzed by GC/MS. The Daehwang extract showed the strongest antimicrobial effect against P. aeruginosa. The MICs on S. aureus, P. aeruginosa and S. typhimurium were 250, 300 and 300mg/$m\ell$, respectively. In the methanol extracts from Daehwang, 120 different compounds were separated, with the extraction yield of the phenolic compounds being relatively high (29.85%). Substances with antimicrobial activity, such as 2-methoxy-phenol and 4-vinyl-2-methoxy-phenol were detected in the study.

Health Behavior Motivators about Quit-Smoking in Some Outpatients of University Hospitals (일부 병원 내원 환자의 금연에 관한 건강행위 동기요소)

  • This study was aimed to investigate the motivators of smoking in 300 subjects using convenient extraction in one hospital. After informed consent, the survey that consisted of general characteristics, health behavior, stage of change, health behavior-related-perceived benefit, health behavior-related-perceived barrier, health behavior-related-family support, oral health behavior, subjective oral health and others was carried out. The smoking rate was higher in case of male (sex), high school graduates (education) and 2~3 million Korean won (KRW) (monthly income). The present smokers responded lower oral health behavior (p<0.001), lower health behavior-related-perceived benefit (p=0.021), higher health behavior- related-perceived barrier (p<0.001) and lower stage of change (p<0.001). The oral health related variables were not influenced by smoking. In logistic regression model, odds ratios in smoking were 0.378 in below average (health behavior-related-perceived barrier), 4.746 in below average (stage of change), 12.743 in male (sex) and 3.271 in 2~3 million KRW (monthly income/standard : over 3 million KRW). It was suggested that oral health related manpower should take account of not only smoking-related oral health but also motivators of smoking as to deliver effective no-smoking program.

  • Unlike cationic liposome/DNA complexes, neutralliposomes containing plasmid DNA are stable in blood and does not selectively entrapped in the lung. The objective of this study was to construct neutral liposomes containing plasmid DNA with optimal encapsulation efficiency. Plasmid DNA (pGL2 clone 753,-6 kb) was encapsulated by the freeze/thawing method into liposomes composed of 1-palmitoyl-2-oleyl-sn-glycerol-3-phosphocholine(POPC), didodecyldimethylammonium bromide(DDAB), distaroylphosphatidyl-ethanolamine polyethylene glycol 2000(DSPE-PEG 2000) and DSPE-PEG 2000-maleimide. Radish (Raphanus sativus L. leaf) ethanol extract inhibits protein and mRNA expression of $ErbB_2$ and $ErbB_3$ in MDA-MB-231 human breast cancer cells

  • In this study, we investigated the effects of the ethanol extract of aerial parts of Raphanus sativus L. (ERL) on breast cancer cell proliferation and gene expression associated with cell proliferation and apoptosis in MDA-MB-231 human breast cancer cells. The MDA-MB-231 cells were cultured in the presence or absence of various concentrations (100, 200, or 300 ${\mu}g$/mL) of ERL. ERL significantly decreased cell proliferation after 48 h of incubation (P < 0.05). The protein and mRNA expression of $ErbB_2$ were decreased significantly in a dose-dependent manner (P < 0.05). The protein expression of $ErbB_3$ was decreased significantly at an ERL concentration of 300 ${\mu}g$/mL (P < 0.05), and mRNA expression of $ErbB_3$ was decreased significantly in a dose-dependent manner (P < 0.05). The protein expression of Akt was decreased significantly at the ERL concentration of 200 ${\mu}g$/mL (P < 0.05), and the protein expression of pAkt was decreased significantly in a dose-dependent manner (P < 0.05). The mRNA expression of Akt was decreased significantly at the ERL concentration of 200 ${\mu}g$/mL ERL (P < 0.05). The protein and mRNA expression of Bax were increased significantly at ERL concentrations of 200 ${\mu}g$/mL or higher (P < 0.05). The protein expression of $Bcl_2$ was increased significantly at ERL concentrations of 100 ${\mu}g$/mL or higher (P < 0.05), and mRNA expression of $Bcl_2$ was increased significantly at an ERL concentration of 300 ${\mu}g$/mL (P < 0.05). In conclusion, we suggest that Raphanus sativus, L. inhibits cell proliferation via the ErbB-Akt pathway in MDA-MB-231 cells.

Studies on the Infection Condition and Formaldehyde Inactivation of Human Rotavirus Wa (사람 로타바이러스 Wa의 감염 조건 및 formaldehyde에 의한 inactivation에 대한 연구)

  • Effects of protease, $CaCl_2$ EGTA, polybrene, medium pH, and formaldehyde on the infection and inactivation of human rotavirus Wa were investigated using T-flask culture of monkey kidney MA-104 cells. Rotavirus titer was improved by the addition of trypsin or clostripain. Rotavirus titer was increaeed 8 and 10 time sin the infection medium supplemented with 300 $\mu\textrm{g}$/mL of $CaCl_2$ and in the medium adjusted its pH to 8, respectively. However, addition of EGTA or polybrene to the medium decreased rotavirus titer. Rotavirus titer was reduced to 53-95% of the initial value at 1 hr after formaldehyde treatment. Furthermore, rotavirus was inactivated more than 98% at 12 hrs after formaldehyde treatment.

Red Ginseng Extract Reduced Metastasis of Colon Cancer Cells In Vitro and In Vivo

  • This study investigated the effect of red ginseng extract on metastasis of colon cancer cells in vitro and in vivo. Wound healing migration, cell motility, invasion, and activity, protein expression, and mRNA expression of matrix metalloproteinases (MMPs) were examined in SW480 human colon cancer cells. SW480 cells were cultured with or without $100{\mu}g/L$ PMA in the absence or presence of various concentrations (100, 200, or $300{\mu}g/mL$) of red ginseng extract. Red ginseng extract treatment caused signifi cant suppression of cell motility and invasion (p<0.05) in SW480 cells. Red ginseng extract inhibited MMP-2 and MMP-9 activity and their protein and mRNA expression in a dose-dependent manner (p<0.05) in SW480 cells. For experimental metastasis, BALB/c mice were injected intravenously with CT-26 mouse colon cancer cells in the tail vein, and were orally administered various concentrations (0, 75, 150, or 300 mg/kg body weight) of red ginseng extract for 3 weeks. Numbers of pulmonary nodules were signifi cantly decreased in mice that were fed red ginseng extract (p<0.05). Plasma MMP-2 and MMP-9 activity signifi cantly decreased in response to treatment with red ginseng extract in mice (p<0.05). These data suggest that red ginseng extract may be useful for prevention of cancer invasion and metastasis through inhibition of MMP-2 and MMP-9 pathways.

Correlations of Litter Size and Maternal Serum Progesterone Concentration during Pregnancy with Mammary Gland Growth and Development Indices at Parturition in Javanese Thin-Tail Sheep

  • An experiment was conducted to investigate correlations of litter size and average serum progesterone concentrations during pregnancy with mammary gland growth and development at parturition. Twenty ewes (5, 9, 4, and 2 ewes carrying 0, 1, 2, and 3 lambs, respectively) were used to measure weekly serum progesterone concentration during pregnancy. At parturition, the experimental ewes were slaughtered for determination of mammary gland growth and development at parturition (mammary dry fat-free tissue [DFFT], DNA, RNA, collagen, protein, and glycogen). Correlation of mammary DFFT with litter size and averages serum progesterone concentrations were 0.75 and 0.72, respectively. Litter size or maternal serum progesterone concentrations did not correlate with the mammary DNA concentration. However, litter size or maternal serum progesterone concentrations positively correlated (p < 0.01) with the mammary RNA and protein concentrations, but negatively correlated with the mammary collagen (p < 0.01) and. glycogen (p < 0.05) concentrations. Litter size or maternal serum progesterone positively correlated (p < 0.01) with the total mammary DNA, RNA, collagen, protein and glycogen contents. These results implied that the increased concentrations of progesterone with the increased litter size during pregnancy improved mammary gland growth and development at parturition.
