Synergistic Effect in Mosquitocidal Activity of Crystal Proteins from Bacillus thuringiensis NT0423 Transformed with cryIVD Gene (cryIVD 유전자로 형질전환된 Bacillus thuringiensis NT0423 균주 내독소 단백질의 모기 유충에 대한 독성의 상승효과)

  • Bacillus thuringiensis NT0423 produces quite a typical bipyramidal crystals of a common major band of ca. 130 kDa, and has dual specificity against Lepidoptera and Diptera. To enforce the Diptera-toxicity of B. thuringiensis NT0423, cryND gene was transformed 30 B. thuringiensis NT0423. The transfonnant B. thuringiensis PT1227 was obtained from introduction of pCGl0 into B. thuringiensis NT0423 by electroporation. The result showed that cryND and resident crystal protein genes in transformant were stably expressed with its own shape. Furthermore, the toxicity of B. thuringiensis PT1227 against Diptera was highly enforced, suggesting that the enforced toxicity of B. thuringiensis PT1227 was due to synergistic effect of both introduced and resident crystal proteins in transformant.

Effects of Temperature during Moist Heat Treatment on Ruminal Degradability and Intestinal Digestibility of Protein and Amino Acids in Hempseed Cake

  • The objective of this study was to evaluate ruminal degradability and intestinal digestibility of crude protein (CP) and amino acids (AA) in hempseed cake (HC) that were moist heat treated at different temperatures. Samples of cold-pressed HC were autoclaved for 30 min at 110, 120 or $130^{\circ}C$, and a sample of untreated HC was used as the control. Ruminal degradability of CP was estimated, using the in situ Dacron bag technique; intestinal CP digestibility was estimated for the 16 h in situ residue using a three-step in vitro procedure. AA content was determined for the HC samples (heat treated and untreated) of the intact feed, the 16 h in situ residue and the residue after the three-step procedure. There was a linear increase in RUP (p = 0.001) and intestinal digestibility of RUP (p = 0.003) with increasing temperature during heat treatment. The $130^{\circ}C$ treatment increased RUP from 259 to 629 g/kg CP, while intestinal digestibility increased from 176 to 730 g/kg RUP, compared to the control. Hence, the intestinal available dietary CP increased more than eight times. Increasing temperatures during heat treatment resulted in linear decreases in ruminal degradability of total AA (p = 0.006) and individual AA (p<0.05) and an increase in intestinal digestibility that could be explained both by a linear and a quadratic model for total AA and most individual AA (p<0.05). The $130^{\circ}C$ treatment decreased ruminal degradability of total AA from 837 to 471 g/kg, while intestinal digestibility increased from 267 to 813 g/kg of rumen undegradable AA, compared with the control. There were differences between ruminal AA degradability and between intestinal AA digestibility within all individual HC treatments (p<0.001). It is concluded that moist heat treatment at $130^{\circ}C$ did not overprotect the CP of HC and could be used to shift the site of CP and AA digestion from the rumen to the small intestine. This may increase the value of HC as a protein supplement for ruminants.

Differences in Clinical Practice Stress, Sleep Quality, and Daytime Sleepiness before and after Nursing Students (간호 대학생의 실습 전, 후 임상실습 스트레스, 수면의 질 및 주간 졸림증의 차이)

  • This study identified the differences and relationships between clinical practice stress, sleep quality, and daytime sleep disorder before and after clinical practice of college nursing students. The subjects were 130 students in the third year of nursing science. The results showed that the average clinical stress of the study subjects was 3.15 out of 5 points. Out of the external factors for clinical practice stress, the score for the task was 3.64. There was a significant difference in gender (t=-3.88, p<).001), subjective health status (F=5.78, p=.004), residence (F=3.20, p=.026). Nursing students showed lower sleep quality (80.0% before practice, 70.0% after practice), and those with daytime sleepiness (26.9% before practice, 42.3% after practice) were more than those of general college students. There was a difference between sleep quality (χ2=4.06, p=.004) and daytime sleepiness (χ2=6.08, p=.009) before and after clinical practice. There was also a negative correlation between daytime sleepiness and sleep quality (r=-.259, p<0.01). Therefore, strategies to reduce clinical practice stress and improve the quality of sleep are needed for the physical and mental health of nursing college students. The quality of nursing can be improved in the future by helping nursing students adapt to the clinical practice of changing nursing education field.

  • Effective communication flow is a fundamental for the success of any construction projects. The key role of communications among project parties is even more critical in the case of Design-Build (DB) projects. Previous research has shown that these projects have an environment where integration between different project team members and schedule constraints increase channels of communication, and require faster communication flows. This paper summarizes the findings from research analyzing organizational structures and communication flow of the Texas State Highway 130 (SH130) project. Awarded in 2002, the SH130 project, totaling $1.3 billion USD, constitutes the "pilot" application for the DB approach in the state of Texas. A set of observations pertaining to team organization and communication issues in the design-build environment is included.

  • In previous paper, it was reported that antibiotic substance such as tetracycline and streptomycin were produced by S'. albus subsp. and S'. globosus. And increase of mycelial growth of two strains, antibiotic production, and changes of pH range are extended to approximately 110-130 hrs in fermenting medium, there-after they decreased with culture period exception of pH range. Two Streptomyces spp. required commonly 4-5% starch as carbon sources and 1.5-2.0% soybean meal as nitrogen sources. However, 0.005-0.01M potassium phosphate dibasic, calcium carbonate (6mg/ml in S.albus subsp. and 2mg/ml in S. globosus), 0.01-0.03M, magnesium sulgate and 0.01M ferric chloride showed as optimal concentration for the growth of 2 strains. Mineral compoments such as zinc, manganese, cobalt, sodium and copper at the level of 10/sup -4/ -10/sup -6/M were observed. Especially, zinc ion showed toxicity to the growth of 2 strains at 0.005M. In relation with pH, there is a little difference in mycelial growth with cultural initial pH.
