Fourteen phenolic compounds(benzoic acid, p-hydroxybenzoic acid, protocatechuic acid, vanillic acid, syringic acid, gallic acid, caffeic acid, ferulic acid, (+)-catechin, quercetin, rutin, catechol, chlorogenic acid and L-ascorbic acid) were examined for their effects on the anticarigenic activity. Among tested samples, catechol was significantly inhibited the S. mutans, exhibiting an clear zone 18.5-19.5mm by 10 mg/disc level. The minimal inhibition concentration(MIC) of the phenolic compounds for Streptococcus mutans, M1 and M2 strain were determined as 2,000 ppm, whereas catechol was 1,000 ppm. The activity of glucosyltransferase(GTase) was significantly inhibited by catechol, at 10 ppm(58.7%), 50 ppm(60.7%) and 100 ppm(88.4%) and 500 ppm(89.6%), respectively. Among them, catechol showed most significant anticariogenic activity as well as inhibition of GTase activity.
Adenophora stricta Miq. (Campanulaceae) is an annual herb, which has been used as a traditional medicine in Korea, Japan and China to treat bronchial asthma, tonsillitis, and hypertension. In this study, 12 compounds were isolated from the roots of A. stricta and isolates were identified to be methyl adenophorate (1), decursidin (2), L-tryptophan (3), D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4), vanillic acid 4-O-${\beta}$-D-glucopyranoside (5), syringic acid 4-O-${\beta}$-D-glucopyranoside (6), vanillin (7), vanillic acid (8), p-hydroxybenzaldehyde (9), p-hydroxybenzoic acid (10), p-hydroxyacetophenone (11) and linoleic acid (12). Decursidin (2) and D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4) is firstly reported from A. stricta in current study.
Phenolic compounds present in upland weeds (Artemisia asiatica Nakai, Capsella bursa-pastoris (L.) Medik, Portulaca orleracea L. and Trifolium repens L.) which have shown allelopathic activity were determined using paper chromatography (PC) and high performance liquid chromatography (HPLC). Effect of the determined phenolic compounds on germination and post-germination growth of test plants was also investigated. Kinds of phenolic compounds determined by PC in the four weed species were greater in the aqueous extract than in the methanol extract. Ferulic acid was found in both extracts of the weeds studied, whereas benzoic acid was that the weeds commonly contained hydroquinone, p-hydroxybenzoic, ferulic and cinnamic acids. Out of the phenolic compounds determined by PC and HPLC p-hydroxybenzoic, cinnamic and ferulic acids inhibited germination and post-germination growth of radish and sesame. Inhibition of the phenolic compounds on the radicle growth was greater than on the germination of the test plants.
20 bacterial strains capable of growing on phenol minimal medium were isolated from soil and wastewater by the enrichment culture technique, and among them, one isolate which was the best in the cell growth was selected and identified as Bacillus sp. SH3 by its characteristics. Strain SH3 could grow with phenol as the sole carbon source up to 15 mM, but did not grow in minimal medium containing above 20 mM of phenol. The optimal conditions of temperature and initial pH for growth and phenol degradation were 30$^{\circ}$C and 7.5, respectively. This strain could grow on various aromatic compounds such as catechol, protocatechuic acid, gentisic acid, o-, m-, p-cresol, benzoic acid, p-hydroxybenzoic acid, anthranilic acid, phenyl acetate and pentachlorophenol, and the growth-limiting log P value of strain SH3 on organic solvents was 3.1. In batch culture, strain SH3 degraded 97% of 10 mM phenol in 48 hours. In continuous culture under the conditions of 20 mM of influent phenol concentration and 0.050 hr$^{-1}$ of dilution rate, the treatment rate of phenol was 94%.
Hemicellulosic hydrolysates contain not only sugars but also several kinds of ethanol fermentation inhibitory substances such as carboxylic acids, furans and phenolic compounds. In this work, emulsion liquid membrane (ELM) was chosen as a separation technology to remove the inhibitors. A basic simulated hemicellulosic hydrolysate was composed of xylose as sugar, dilute sulfuric acid solution as solvent, and acetic acid as carboxylic acid, and furfural as furan derivative or p-hydroxybenzoic acid(HBA) as phenolic compound was added to the hydrolysate when necessary. Acetic acid and HBA as weak acid could be selectively removed from the hydrolysates in all the ELM systems considered here, but furfural as aldehyde was quite hard to remove. Also, when HBA was added to the basic simulated hemicellulosic hydrolysate, both of acetic acid and HBA in the feed phase could be selectively removed up to 99% in an ELM system with tributyl phosphate as extractant.
Fungal cytochrome P450 (CYP) enzymes catalyze versatile monooxygenase reactions and play a major role in fungal adaptations owing to their essential roles in the production avoid metabolites critical for pathogenesis, detoxification of xenobiotics, and exploitation avoid substrates. Although fungal CYP-dependent biotransformation for the selective oxidation avoid organic compounds in yeast system is advantageous, it often suffers from a shortage avoid intracellular NADPH. In this study, we aimed to investigate the use of bacterial glucose dehydrogenase (GDH) for the intracellular electron regeneration of fungal CYP monooxygenase in a yeast reconstituted system. The benzoate hydroxylase FoCYP53A19 and its homologous redox partner FoCPR from Fusarium oxysporum were co-expressed with the BsGDH from Bacillus subtilis in Saccharomyces cerevisiae for heterologous expression and biotransformations. We attempted to optimize several bottlenecks concerning the efficiency of fungal CYP-mediated whole-cell-biotransformation to enhance the conversion. The catalytic performance of the intracellular NADPH regeneration system facilitated the hydroxylation of benzoic acid to 4-hydroxybenzoic acid with high conversion in the resting-cell reaction. The FoCYP53A19+FoCPR+BsGDH reconstituted system produced 0.47 mM 4-hydroxybenzoic acid (94% conversion) in the resting-cell biotransformations performed in 50 mM phosphate buffer (pH 6.0) containing 0.5 mM benzoic acid and 0.25% glucose for 24 h at $30^{\circ}C$. The "coupled-enzyme" system can certainly improve the overall performance of NADPH-dependent whole-cell biotransformations in a yeast system.
The phytochemical study for the extract of Nelumbo nucifera (Nymphaceae) seeds has led to the isolation of ten compounds including five simple phenolic compounds, two indole derivatives, a flavonoid glycoside, two abscisic acid derivatives. The interpretation of 1D and 2D NMR and ESI-Q-TOF-MS spectroscopic data revealed the chemical structures of isolates to be p-hydroxybenzoic acid (1), protocatechuic acid (2), (E)-p-coumaric acid (3), (E)-ferulic acid (4), (E)-sinapate-4-O-${\beta}$-$\text\tiny{D}$-glucopyranoside (5), tryptophan (6), 3-indoleacetic acid (7), isoschaftoside (8), dihydrophaseic acid (9), dihydrophaseic acid 3'-O-${\beta}$-$\text\tiny{D}$-glucopyranoside (10). To the best of our knowledge, 1 - 5 and 7 were identified for the first time from N. nucifera seeds, and the presence of dihydrophaseic acid (9) and its glucoside (10) were demonstrated secondly in this plant.
To develop a simple, rapid and simultaneous analytical method of phenolic compounds using gas chromatography (GC) and gas chromatography/mass spectrophometer (GC/Ms), this experiment was carried out to search the retention times of capillary columns and the characteristics of fragment ions in electron impact mass spectra. Most of trimethylsilyl derivatives and underivatized phenolic compounds were separated very well on three kinds of capillary columns(HP-1), Ultra-2 and HP-35). Quantitiative determination of phenolic compounds was achieved by internal standards (p-hydroxybenzoic acid iopropyl ester, p-hydroxybenzoic acid ethyl ester). Calibration plts were linear in the investigated range, and the limits of detection were about 5 ng at split mode method. When analyzed by three columns, theseparation times were fairly constant on two nonpolar columns, but a few compounds showed slightly different separation order by the itnermediate polar HP-35 column. The important characteristic patterns of TMS derivatives of phenolic compounds on the EI/MS spectrra appeared at the base peak of [M-15]+ ion and presented at high abundance in most TMS derivatives of phenoloc compounds. [M]+, [M-CH3-COO]+, [M-Si(CH3)4]+ and [M-Si(CH3)4 -CH3]+ also observed in mass spectra of these compounds . Although several compounds have the same retention times on GC column, it might be possible to identify these compounds by the different patternsof mass frgement ions. The TMS derivatives, thus , provide additional information for identification of phenolic compounds in biological systems.

Water stress obstructs rice growth mainly by oxidative damage in biological cells to cause a reduction of leaf photosynthesis and evapotranspiration processes. In this study, exogenous application of vanillic acid (VA) and p-hydroxybenzoic acid (PHBA) to improve drought tolerance of two Oryza sativa cultivars, Q2 and Q8 was tested. The drought evaluation based on leaf phenotypes to show that both Q2 and Q8 resulted in remarkable water-stress tolerance induced by leaf spraying pretreatment of mixed solution of $50{\mu}M\;VA+50{\mu}M\;PHBA$. The mixtures of $25{\mu}M\;VA+25{\mu}M\;PHBA$ and $50{\mu}M\;VA+50{\mu}M\;PHBA$ treated on Q2 and Q8 in water deficit condition also indicated that total phenols, total flavonoids, and DPPH radical scavenging activity were significantly greater their controls. In general, the accumulation of individual phenolic acids was increased in exogenous phenolic treatments, as compared with controls. Particularly, Q2 obtained a considerable amount of endogenous PHBA after application of $50{\mu}M\;VA$, $25{\mu}M\;VA+25{\mu}M\;PHBA$, and $50{\mu}M\;VA+50{\mu}M\;PHBA$ (0.18 mg/g DW, 0.71 mg/g DW, and 1.41 mg/g DW, respectively); and a negligible content of VA (0.003 mg/g DW) appeared uniquely in the treatment of $50{\mu}M\;VA$. Similarly, Q8 also absorbed a significant quantity of PHBA in $50{\mu}M\;PHBA$, $25{\mu}M\;VA+25{\mu}M\;PHBA$, and $50{\mu}M\;VA+50{\mu}M\;PHBA$ treatments (0.15 mg/g DW, 0.15 mg/g DW, and 0.22 mg/g DW, respectively). In addition, the spraying $50{\mu}M\;PHBA$ and $25{\mu}M\;VA+25{\mu}M\;PHBA$ on Q8 leaves induced similar amount of drought tolerance of Q2 and Q8 were improved, paralleled with the increased amounts of endogenous phenolics revealed that VA and PHBA played an important role to enhance drought tolerance in rice.
