Effect of Sihogayonggolmoryeotang on SPS-induced PTSD in Rats (시호가룡골모려탕(柴胡加龍骨牡蠣湯)이 흰쥐에서 SPS로 유도된 PTSD에 미치는 효과)

  • Objective : To investigate the effect of sihogayonggolmoryeotang (SY) on Single Prolonged Stress(SPS)-induced Post Traumatic Stress Disorder(PTSD). Method : To confirm the effects of SY on SPS-induced PTSD, Changes in body weight, sucrose intake open field test(OFT) and forced swimming test(FST)were observed. After behavioral tests, the plasma corticosterone(CORT) from the abdominal aorta, serotonin(5-HT) from prefrontal cortex, hippocampus, amygdala and striatum, norepinephrine(NE) and dopamine(DA) from hippocampus was measured by ELISA. mRNA expression of brain-derived neurotrophic factor(BDNF) and cAMP response element-binding protein(CREB) in hippocampus was measured by RT-PCR. Result : Weight change and sucrose intakes of rats in 14th day after the administration of SY were significantly increased in the SPS + SY450 group compared to the SPS group (p<0.05). Numbers of crossing in the central zone in the OFT were significantly increased in the SPS + SY450 group (p<0.05) compared with the SPS group. The immobility time of FST was significantly decreased in SPS + SY450 group compared with SPS group (p<0.05). The change of plasma CORT concentration was significantly decreased in SPS + SY450 group compared with that in SPS group (p<0.05). The change of 5-HT concentration was significantly increased in the SPS + SY450 group at hippocampus and amygdala compared with the SPS group (p<0.05). The concentration of DA was significantly increased in the SPS + SY450 group compared with the SPS group (p<0.05). The expression of BDNF and CREB were significantly increased in SPS + SY450 group compared with the SPS group (p<0.05). Conclusion : SY administration lowered the increase of CORT caused by PTSD and increases the 5-HT concentration and reversed the decreased expression of NE and DA and BDNF and CREB by PTSD. It is postulated that SY is effective in treating PTSD by restoring cognitive function, memory impairment, unstable emotional disturbances.

Effects of Fermented Soy Protein on Nitrogen Balance and Apparent Fecal and Ileal Digestibility in Weaned Pigs

  • This study was conducted to evaluate the effects of providing fermented soy protein to weaned pigs on nitrogen balance and apparent fecal and apparent ileal digestibility (AID) of AA. Four weaned ((Yorkshire${\times}$Landrace)${\times}$Duroc) barrows (BW = 6.58${\pm}$0.98 kg), surgically fitted with a simple T-cannula approximately 15 cm prior to the ileo-cecal junction, were fed four diets according to 4${\times}$4 Latin square design. Diets were a basal diet supplemented with one of the following: 3% SDPP (spray dried plasma protein), 5% RBP (soy protein fermented by Lactobacillus spp.), 5% PSP (soy protein fermented by Aspergillus oryzae and Bacillus subtilis), and 2.5% RPP (2.5% RBP+2.5% PSP). No differences were observed in DM and N intakes among treatments. However, the level of urine excretion was greater in the RPP group than in the PSP group. Additionally, fecal DM excretion, fecal N concentration and fecal N excretion were increased in the RBP, PSP and RPP groups when compared with the SDPP group (p<0.05). Furthermore, total excretion was increased in the RPP group when compared with the PSP group (p<0.05). In addition, N absorption and the N absorption ratio were higher in the SDPP group than in the RPP group (p<0.05). Moreover, the DM and N digestibilities were lower in the RBP, PSP and RPP groups than in the SDPP group (p<0.05), and the ash and energy digestibilities were higher in the SDPP and RBP groups than in the PSP and RPP groups (p<0.05). However, no significant differences were observed in the DM, N, Ash, Ca, P or ileal digestibilities among treatments, although the energy digestibility was higher in the SDPP group than the RBP group (p<0.05). In addition, the apparent ileal digestibilities of essential amino acids (Arg, His, Iso, Leu, Lys, Phe, Thr, and Val) were significantly higher in the SDPP group than in the other groups (p<0.05), and the levels of Ala, Cys, Glu and Try were greater in the SDPP treatment group than the RBP, PSP and RPP groups (p<0.05). Additionally, the levels of Asp, Gly and Ser were higher in the SDPP group than the PSP and RPP groups, and the level of Pro was higher in the SDPP group than the RPP group (p<0.05). Finally, total non-essential amino acid and total amino acid digestibility were higher in the SDPP group than in the other treatments (p<0.05). Taken together, the results of this study indicate that animal protein is more bioavailable than plant protein. However, the N absorption ratio and ileal digestibility were found to be similar in the SDPP and RBP groups.

The Effects of Packed Red Blood Cell Washing and Circuit Precirculation-Ultrafiltration on the Production of Cytokines by Open Heart Surgery (충전용 농축적혈구의 세척 및 체외순환로의 전순환-초여과법이 개심수술에 의한 사이토카인 형성에 미치는 영향)

  • Background: The washing of packed red blood cells could remove pro-inflammatory mediators, cell debris, and micro-particles contained in packed red blood cells, and the preci-rculation-ultrafiltration (recirculation and ultrafiltration of circuit itself before cardiopulmonary bypass) could attenuate the initial inflammatory reaction and remove the initial proinflam-matory mediators. This study was performed to evaluate whether the washing of packed red blood cells and precirculation-ultrafiltration can reduce the production of cytokines that have an important role in myocardial reperfusion injury. This study investigated the effects of washing the packed red blood cells and precirculation-ultrafiltration on the production of cytokines during and after cardiopulmonary bypass and open heart surgery. Material and Method: Forty eight infants with VSD undergoing open heart surgery under cardiopulmonary bypass were randomized into control group (group C, n=12), washing group (group W, n= 12), precirculation-ultrafiltration group (group F, n: 12), and combined group(washing and precirculation-ultrafiltration, group WF, n=12). Blood samples were obtained before, during, and after the bypass to assess plasma level of tumor necrosis factor-$\alpha$(TNF-$\alpha$), interleukin-6(IL-6), and interleukin-8 (IL-8). Results: Expressions of TNF-$\alpha$ were significantly reduced in combined group (group WF) compared with group C, group W, and group F (p<0.05). Expression of IL-6 were significantly reduced in group W, group F, and group WF compared with group C (p<0.05), but similar among group W, group F, and group WF (p=0.053). Expression of IL-8 were reduced in group W and group WF compared with group C (p<0.05), but similar among group W, group F, and group WF (p=0.067). Conclusion: In conclusion, the washing of packed red blood cells and precirculation-ultrafiltration blunted the increase of TNF-$\alpha$ , IL-6, and IL-8 during and after open heart surgery with cardiopulmonary bypass. However, the clinical benefits of these treatments remains unproven.

Antioxidant Activities and Quality Characteristics of Pudding Added with Olbyeossal (올벼쌀 첨가 푸딩의 항산화 활성 및 품질 특성)

  • The quality characteristics of pudding products comprising of Korean Olbyossal (traditional steamed rice), added at different percentages, were examined to explore the application of Olbyossal to develop different types of processed food products. Five groups of pudding were produced containing 0%, 25%, 50%, 75%, and 100% of Olbyossal. Evaluation of their quality characteristics and antioxidant activities revealed that the control group (with no added Olbyossal) had the highest moisture content and pH (p<0.001). The sugar content and turbidity was highest in the 100% Olbyossal group and lowest in the control group (p<0.001). The salinity was lowest in the 100% Olbyossal group (p<0.001). In terms of color values, the L-value (p<0.001) and a-value were highest in the control group, while the b-value was highest in the 100% group (p<0.001). Texture properties revealed that hardness (p<0.001), springiness (p<0.01), and chewiness (p<0.001) decreased as the percentage of Olbyossal increased. Increase in adhesiveness was noted with an increase in the percentage of Olbyossal (p<0.01). Based on the sensory evaluation of Olbyossal pudding in terms of appearance (p<0.05), flavor (p<0.05), taste (p<0.01), texture (p<0.001), and overall quality (p<0.001), the 100% group showed the best results. Examination of Olbyossal pudding revealed that antioxidant activities, radical scavenging activities for DPPH and superoxide anion, and reducing power increased as the percentage of Olbyossal increased (p<0.001). Therefore, pudding containing 100% Olbyossal is considered as the ideal product with reference to the high level of preference and product quality.

Effect of the Interval from Calving to First Insemination and Days Open on the Subsequent Reproductive Performance in Dairy Cows (젖소에서 분만 후 첫 수정 간격 및 임신 간격이 이후의 번식능력에 미치는 영향)

  • This retrospective study evaluated the effects of the interval from calving to first insemination (ICFI) and days open (DO) on the reproductive performance in dairy cows. In the first analysis, data from 705 cows were grouped based on the ICFI: short (30-60 days, n=217), medium (61-90 days, n=309), and long (91-150 days, n=179). The occurrence of endometritis was greater in the long group than in short and medium groups (p<0.05). The occurrence of ovarian cysts increased with increasing ICFI (p<0.05), while body condition score (BCS) during the 5 month postpartum period was lower with increasing ICFI (p<0.01). The hazard of pregnancy by 365 days in milk (DIM) was lower (hazard ratio [HR]=0.70, p<0.0001) in the long group, but higher (HR=1.41, p<0.0001) in the short group compared with the medium group. In the second analysis, data from 436 cows were grouped based on the DO: short (30-90 days, n=154), medium (91-180 days, n=183), and long (181-360 days, n=99). The occurrence of a retained placenta was greater in the long group than in the medium group (p < 0.05). Ovarian cysts occurred more frequently in medium and long groups than in the short group (p=0.08). BCS was lower in the short group compared with medium and long groups at month 1 postpartum (p<0.05). Milk yield (kg/day) was greater in the medium group compared with the short group at months 2 to 5 postpartum (p<0.05). The hazard of first insemination by 150 DIM was lower in the long group than in the short group (HR=0.73; p<0.02). The hazard of pregnancy by 365 DIM was lower (HR=0.64, p<0.0001) in the long group than in the medium group, while the hazard did not differ between short and medium groups (p>0.05). Moreover, the culling rate was greater in the long group than in the short group (p<0.05). In conclusion, a longer ICFI and DO resulted in reduced reproductive performance in dairy cows, which was attributable to an increase in postpartum diseases, a lower BCS and a greater milk yield.

Effects of Dietary Lipid Sources and Levels on Lecithin : Cholesterol Acyltansferase Activity and Cholesterol Metabolism in Rats (식이지방의 종류와 수준이 흰쥐의 Lecithin : Cholesterol Acyltransferase 활성 및 콜레스테롤대사에 미치는 영향)

  • This study was carried out to investigate the effects of different sources and level of dietary lipid on lecithin : cholesterol acyltrasferase activity and cholesterol metabolism in male rats of Sprague-Dawley strain. The effects of different lipid sources was compared with sardine oil($\omega$3 EPA and DHA), beef tallow(SFA), perilla oil($\omega$3 linolenic acid) and corn oil($\omega$6 linoleic acid). Diets were formulated in such a way that 10%, 20% and 40% dietary energy were supplied with each of four experimental lipid sources. Control diet contained only non-lipid energy. A total number of 78 rats, equally divided into 13 groups, were fed the experimental diets for a period of 6 weeks. In vitro cultures were also carried out to study the cholesterol synthetic activity in the liver prepared from rats used in feeding trials. The concentration of plasma total cholesterol, HDL-cholesterol, LDL-cholesterol and HDL-C/T/C(total cholesterol) ratio were significantly (p<0.001) influenced by dietary lipid sources. Higher HDL-cholesterol and lower LDL-cholesterol concentration in plasma were obtained in rats fed $\omega$3 fatty acid supplemented diets(sardine oil and perilla oil group) compared to diets containing $\omega$6 and saturated fatty acid(corn oil and beef tallow group). In total cholesterol concentration of plasma, beef tallow group was significantly (p<0.001) higher than other lipid groups, and non-lipid group was significantly(p<0.05) higher than the lipid supplemented groups. The activity of lecithin : cholesterol acyltransferase(LCAT) in plasma was greatly(p<0.001) affected by dietary lipid sources and levels. In LCAT acivity of plasma, lipid supplemented groups were significantly(p<0.05) higher than non-lipid group, vegetable oil groups were significantly (p<0.001) higher than animal fat groups, and sardine oil group were significalylty (p<0.001) higher than beef tallow group. Also perilla oil group was significanlty (p<0.05) higher than corn oil group, and sardine oil group was significantly (p<0.05) higher than perilla oil group. Low lipid group, compared with medium or high lipid group, showed higher activity of LCAT in plasma. In cholesterol synthetic activity of liver tissues culture, sardine oil group($\omega$3 EPA and DHA) was significantly(p<0.001) higher than other lipid groups, non-lipid group was significantly(p<0.001) higher than the lipid supplemented groups, and amimal fat group were significantly(p<0.001) higher than vegetable oil groups, but the synthetic activity was not affected by dietary lipid levels.

Effect of Portulaca Oleracea water Extract on the Changes of Urine Amino acid Contents and Survival Rate by Irradiation in Mice (쇠비름 수침액이 방사선 조사에 의한 생쥐의 생존율과 뇨중 아미노산 조성에 미치는 영향)

  • A study has been conducted to investigate the radioprotective effects of purslane extracts (P) when i.p. injected before irradiation (R). For studying the amino acid composition, three groups of ICR mice (7-week-old) were tested. The first group i.p. injected with purslane extract for 5 days were irradiated with 6 Gy of ${\gamma}$-radiation (P+R). The second group was irradiate with 6 Gy without any pretreatment (R). The third group was non-irradiated control (CT). Each group divided into two groups for the survival rate study, one injected with saline (S) and the other injected with purslane extract (P) for five days and then irradiated with 8 Gy. The amino acid composition of urine samples were analyzed with HPLC for 19 days after irradiation. A few kinds of amino acids such as lysine and methionine in urine from P+R group increased in comparison with those from CT or R group. The survival rate of P group maintained much higher than that of S group during experimental period. The results obtained may support that this plant has radioprotective substances by means of life-lengthening, not of urine amino acid components.

Comparison of Effects among Tai-Chi Exercise, Aquatic Exercise, and a Self-help Program for Patients with Knee Osteoarthritis (슬골관절염 환자를 위한 타이치운동, 수중운동과 자조관리프로그램의 효과비교)

  • Purpose: This study was to compare the effects among Tai-Chi exercise, aquatic Exercise, and a self-help program for knee osteoarthritis patients on symptoms of arthritis, muscle strength, balance, and difficulty of performing activities. Method: There were 50 final subjects50. A non-equivalent pretest-posttest design was used. The collected data was analyzed using SPSS for Window. One-way ANOVA and Scheffe's multiple comparison test were used 8weeks after each program. Result: There were significant differences in joint pain(p=.000), stillness (p=.001), knee extensor peak torque(p=.006), knee flexor(p=.002), and difficult of performing activity (p=.000), but there was no significant difference in balance(p=.648). The Tai-Chi group was significantly different from the self-help group for knee extensor peak torque, knee flexor and stiffness on Scheffe's multiple comparison tests. In addition, the Tai Chi group or aquatic group were significantly different from the self-help group for difficulty of performing activities(p<0.05). Conclusion: There are significant differences in the effects of the nursing intervention among the three groups. The Tai Chi group and aquatic group were significantly different from the self-help group. However, it seems that Tai-chi exercise may be more suitable than aquatic exercise in osteoarthritis exercise programs. Further studies with a longitudinal study are necessary to confirm the longer exercise period.

Effects of Undaria Pinnatifida on Serum and Liver Lipids in Rats (미역첨가급식(添加給食)이 흰쥐의 간(肝) 및 혈청(血淸)의 지질농도(脂質濃度)에 미치는 영향(影響))

  • This study was designed to observe the effect of Undaria Pinnatifida (U. P.) Powder on serum and liver lipids concentration in rats. Adult male rats of Sprague-Dawley strain were divided into four diet groups: one control diet and the other three diets in which U.P. powder was mixed with the control diet at the rate of 1%, 2% and 3% respectively. The results of the above observation were as follows : Serum and liver total lipids in 3% U.P. group were lower an those in control group at the 6th week. There was no difference in serum and liver total cholesterol among the diet groups. Serum HDL-cholesterol and HDL-cholesterol per total cholesterol ratio in 3% U.P. group were higher than those in control group at the 3rd and 6th week. Serum triglyceride in 3% U.P. group was significantly lower than that in control group at the 3rd week. Serum phospholipid in 1% U.P. and 3% U.P. group was significantly lower than that in control group at the 3rd week, but there was no difference among them at the 6th week.

THE CLINICAL AND MICROBIOLGICAL EFFECT OF CHLORHEXIDINE RINSE AND SUPRAGINGIVAL PLAQUE CONTROL ON ADULT PERIODONTITIS (Chlorhexidine용액 구강양치와 치은연상치태 제거의 임상 및 미생물학적 변화에 대한 연구)

  • The purpose of this study was to assess the clinical and microbiological effect of chlorhexidine rinse and supragingival plaque control following scaling and root planing on adult periodontal disease. 14 patients with adult periodontitis were selected for the study . They had not taken antibiotics for 6 months and history of dental treatment for 6 months before the study. Patients received a supragingival scaling and root planing under local anesthesia, plaque control group was subjected to professional plaque control 2 times for a period 2 week, chlorhexidine rinse group were subjected to twice daily 0.2% chlorhexidine rinse for a period 2 week. Clinical examination (plaque index, gingival index, probing pocket depth) and distribution of the bacteria morphology of subgingival plaque were monitored on baseline (0 week), 1 week, 2 week, 4 week and 6 week. The results were as follows : 1. Plaque index in chlorhexidine rinse group , plaque control group and control group was significantly reduced during all weeks (P<0.05). 2. Probing pocket depth was significantly reduced at 2, 4, 6 week (P<0.05) in chlorhexidine rinse group and control group, plaque control group was significantly reduced during all weeks (P<0.05). 3. Gingival index was significantly improved at 2, 4, 6 weeks(P<0.05) in chlorhexidine group and plaque control group, control group was significantly improved at 1, 2, 4 weeks (P<0.05). 4. Percentage of cocci was significantly increased at 1, 2, 4 and 6 weeks in chlorhexidine rinse group and control group, plaque control group was significantly increased at 2, 4 and 6 weeks(P<0.05). 5. Percentage of non-motile rods in all group were not significantly changed when compared with those of baseline (0 week) (P<0.05). 6. Percentage of motile rods was significantly reduced during all weeks (P<0.05) in chlorhexidine rinse group, plaque control group was significantly reduced at 2, 4, 6 weeks and 1, 2 and 4 weeks in control group. 7. Percentage of spirochetes was significantly reduced during all weeks (P<0.05), plaque control group was significantly reduced at 2, 4, 6 weeks and 1, 2, 4 weeks in control group. This results were suggested that clinical and microbiological effect of chlorhexidine rinse and supragingival plaque control following scaling and root planing on periodontal disease
