산침지와 가열 살균이 떡볶이 떡의 형상 변화와 품질 특성에 미치는 영향 (Effect of Acid Soaking and Thermal Sterilization on the Shape and Quality Characteristics of Tteokbokki Rice Cake)

  • Effects of acid soaking (AS) and thermal sterilization (TS) on the shape and quality characteristics of Tteokbokki rice cake (TRC) were investigated. The F-value of a sequential process (SP) of the combination of AS and TS was quantitatively determined with a reference microorganism of Bacillus cereus. F-values were evaluated according to the minimum and the maximum D-value of B. cereus and the reduction exponent (m=12). The heat penetration curves at cold point (CP) of TRC (400 g) were used to estimate the TS time at 121, 100, and $95^{\circ}C$. F-values of the SP were revised according to the adjusted m values after AS at different pH. The non-uniformity parameters (NUP) of TRC had no significant changes during AS but it dramatically increased after applying TS at a low pH by AS. The result of solubilized starch contents (SSC) demonstrated that the shape changes during SP are related to SSC. The texture characteristics and the whiteness were significantly influenced by a low pH condition (3.5) (p<0.05). Sensory analysis showed that a lower pH and a longer thermal processing time influenced negatively on the acceptability. This study showed that pH 4.0 and $95^{\circ}C$ was an optimum condition for the SP.

METHODS TO IMPROVE UTILIZATION OF RICE STRAW III. EFFECT OF UREA AMMONIA TREATMENT AND UREA MOLASSES BLOCKS SUPPLEMENTATION ON INTAKE, DIGESTIBILITY, RUMEN AND BLOOD PARAMETERS

  • In two separate experiments with crossbred bulls (Sahiwal $\times$ indigenous) the effect of access to a urea-molasses lickblock (MOL-U-MIN) on straw diets was studied. The animals were given either untreated (US) or urea treated (TS) rice straw with or without lickblock supplementation. In experiment 1, individual dry matter intake (DMI) and dry matter digestibility (DMD) were measured, while in experiment 2 in addition to the above rumen (pH, ammonia, minerals) and blood (protein, minerals and haemotological) parameters were also measured. With both experiments urea treatment did not effect DMI, but lickblock supplementation significantly (p < 0.05) increased DMI. The DMD values obtained in both experiments for TS were significantly (p < 0.05) higher than for US, but lickblock supplementation did not effect the DMD of either US or TS fed animals. Both urea treatment (6.97 vs 6.93) and lickblock supplementation (6.98 vs 6.92) significantly (p < 0.001) reduced the rumen pH. Urea treatment and lickblock supplementation increased the rumcn $NH_3-N$ concentration (mg/100 ml) from 8.7 to 11.9 and 9.2 to 11.4, respectively. Both US and TS diets fed with or without lickblock increased the molar ratio of Na : K in saliva. Phosphorus content in blood plasma was significantly (p < 0.01) increased due to lickblock supplementation, whereas the Fc content in blood was significantly increased (p < 0.01) by urea treatment. Haemoglobin content in blood ranged from 11.3 to 11.7 g/dl, and was not influenced by urea treatment or lickblock supplementation. Lickblock significantly reduced the number of red blood cells, but increased the mean corpuscular volume. It is concluded that feeding urea treated straw with proper mineral supplementation could be a more economical alternative to lickblock supplementation.

  • STA1 유전자의 promoter 와 분비신호서열을 이용하여 B. subtilis 의 CMSase 를 분비하는 재조합 효모균주를 육성하였다. STA1A 유전자의 promoter, 분비신호서열, TS region 및 mature glucoamylase N-말단부위의 아미노산 98개와 B. subtilis 의 CMCase 구조유전자가 차례로 연결된 재조합플라스미드 pYESC24 를 제작한후 효모에 형질전환하였으나 CMCase 가 세포외로 분비되지 않았다. 반면에 STA1 의 TS region 및 mature glucoamylase N-말단 아미노산 98 개를 제거하여 CMMase 구조유전자갸 STA1 의 분비신호서열에 바로 연결된 재조합 플라스미드 pYESC11 에 의한 효모형질전환 균주는 CMCase 분비능이 아주 우수하였다. 이 형질전환 균주를 YPD 배지에서 4 일간 배양한 후 세포부위 별 CMCase 역가를 측정한 결과 배양액 1 m/당 총역가 44.7 unit 존재하였으며 이중 93% 이상이 배양상등액에서 관찰되었다.

타피오카전분의 첨가가 쌀쿠키의 품질특성에 미치는 영향 (Effects of Tapioca Starches on Quality Characteristics of Rice Cookies)

  • The effects of tapioca starch (TS) and tapioca modified starch (TMS) as additives on the quality characteristics of rice cookies were investigated in this study. The amounts of TS and TMS replaced were 5, 10 and 15%(w/w) based on 100 g of rice flour. At the result of rapid viscosity analyser (RVA) properties of rice flour added with starches, peak, trough, and final viscosity of rice flour added with TMS decreased significantly(p<0.05). Hardness of dough and cookies as measured by Texture analyzer (TA) significantly decreased upon addition of TMS, and hardness of cookies significantly decreased upon addition of TS, hardness of cookies TMS lower than TS. L value of dough and cookies increased upon addition of TMS, whereas a value decreased upon addition of TMS. Spreadability of cookies increased upon addition of TMS, 10% TMS is the highest. Bulk density of dough decreased upon addition of TMS, bulk density of cookies decreased upon addition of TS and TMS, and bulk density of TMS cookie lower than TS cookie. In sensory evaluation, 10% TS added rice cookies showed the highest score in overall acceptability, hardness, brittleness, color, softness, taste and flavor, and 15% TS added rice cookies showed the highest score in appearance. And 15% TMS added rice cookies showed the highest score in overall acceptability, taste, brittleness, flavor, and 10% TMS added rice cookies showed the highest score in hardness, flavor, taste and color. It is indicated that the quality characteristics and sensory characteristics of the rice cookie can be improved by adding TS and TMS.

BODY HEAT CONTENT, HEAT PRODUCTION AND RESPIRATION IN SHEEP EXPOSED TO INTERMITTENT COLD

  • Five adult sheep were exposed to intermittent cold for 12 h (18:00-06:00) at an air temperature of $5{\pm}1^{\circ}C$ followed by 12 h (06:00-18:00) at $25{\pm}2^{\circ}C$ over a period of 8 days continuously. Carotid artery blood (Tc), mean skin (Ts) and mean body (Tb = 0.86 Tc + 0.14 Ts) temperatures, heat production rate (HP), respiratory evaporative heat loss, respiration rate (RR) and volume were measured before and after exposure. Tc during the 12 h cold period of intermittent cold exposure was similar to that during the corresponding period in the warm environment, while Tc in the $25^{\circ}C$ of intermittent cold was higher (p < 0.05) than that in the corresponding period in the warm environment. Ts during the cold period markedly decreased (p < 0.001) by about $9^{\circ}C$ when compared with that in the corresponding time period in the warm environment, while Ts during the $25^{\circ}C$ period of intermittent cold recovered to a similar level to that in the warm environment. Tb was lower (p < 0.001) during the cold period of intermittent cold, whereas a slight increase in Tb during the $25^{\circ}C$ period of intermittent cold was significant (p < 0.05) when compared with the value during the similar period in the warm environment. HP was greatly increased (p < 0.001) by cold exposure, followed by an immediate decrease during the first one hour of the 12 h warm period, reaching a similar level to that in the warm environment. A lower (p < 0.05) RR was observed during both the cold and $25^{\circ}C$ period of intermittent cold than during the corresponding periods in the warm environment. The results of the present investigation clearly show that the body temperature of sheep increased during a 12 h warm period following 12 h of exposure to cold. These results suggest that during a warm period of an intermittent cold exposure cycle, heat could be shored in the animal body.

스트랜드/파티클 복합체의 기계적 성질에 관한 연구(I) - 단면구성이 기초물성에 미치는 영향 - (A Study on Mechanical Properties of Strand/Particle Composites(I) - Effect of Layer Constructions -)

  • 본 연구는 미이용 자원인 소경목(小經木) 조생수(早生樹) 등의 저질원료로부터 고성능을 지니는 구조용 보드를 제조하는 기술을 개발하는 것을 목적으로 하여 할렬 스트랜드 (S)/파티클(P) 복합체의 층구조와 S와 P의 혼합비율을 바꾼 단면구성에 따른 기초적 물성을 검토했다. 그 결과 스트랜드층을 포함하고 있는 SP 복합체의 경우, 전체적으로 휨성능(MOR, MOE)이 매우 높았다. 또 S단층 보드는 현저한 이방성을 나타내었지만 층구조가 PB에 가까워질수록 이방성은 감소하여, 7층 구조의 복합체는 이방성이 적었으며 특히 SP7은 직교방향도 휨강도가 높았다. 습윤시 휨강도 성능도 같은 경향을 나타내었다. 박리강도(IB)는 PB가 가장 높았고, 스트랜드층을 가진 복합체는 거의 같은 값을 나타내었다. 두께팽창율(TS) 은 PB가 가장 적은 값을 나타내었으며, 표층이 P층인 구조가 S표층구조보다 적은 경향을 나타내었다. 복합체의 표면특성은 표층 엘리먼트의 영향을 받아, 적고 미세한 엘리먼트인 P표층의 복합체가 크고 두꺼운 S표층 복합체보다 양호하였다. S와 P의 혼합비율의 영향은, SP비(比)를 증가시켜도 강도물성(MOR, MOE, IB)은 향상되지 않으며, 오히려 저하되는 것도 있었다. 두께팽창율(TS)은 SP 비(比) 1:1 이상의 복합체에서는 S만으로 제조한 보드와 같은 정도의 값을 나타내었다.

Quality and Amount of Morning and Evening Milk of the Bangladesh Baghabarighat Milk Shed Area Throughout the Year

  • The Baghabarighat Milk Shed Area (BMSA), in Bangladesh belonging to the Bangladesh Milk Producers Cooperative Union Limited (BMPCUL) was selected to study the qualitative and quantitative aspects of dairy milk and to establish a trend throughout the year (July 1995 to June 1990) of milk collected at morning and evening, and daily. The average fat, solids-not-fat (SNF) and total solid (TS) contents of the morning milk were 4.58, 7.75 and 12.33%, respectively, and evening milk contained 5.41, 7.81 and 13.23%. The values for the quality varied (p<0.01) throughout the year without a specific trend. Higher milk collection occured at morning (52.77%) than evening (47.24%), but total yield of fat from morning milk was lower (48.74%) than evening milk (51.26%) due to a lower fat value (4.58%). Yield of SNF (52.57% morning, 47.37% evening) followed the values of the amount of milk collected due to their similar value at morning (7.75) and evening (7.81). Overall, TS yield was 51 and 49% from morning and evening. There was not a significant (p>0.01) relationship between fat and TS values of morning milk with those of evening milk, but there was a correlation (p<0.01) between milk yield at morning and all other parameters for evening. Solids-not-fat value of morning milk was related with yield and SNF value of milk from evening, but not with fat and TS evening milk. It may be concluded that evening milk contains higher amounts of fat, SNF and TS, but yields were higher at morning, except fat. Milk collected at morning showed a relationship with all the parameters found at evening.

Improvement of Enzymatic Stability and Catalytic Efficiency of Recombinant Fusariumoxysporum Trypsin with Different N-Terminal Residues Produced by Pichiapastoris

  • Fusarium oxysporum trypsin (FOT) is a fungal serine protease similar to mammal trypsin. The FOT could be successfully expressed in Pichiapastoris by engineering the natural propeptide APQEIPN. In this study, we constructed two recombinant enzymes with engineered amino acid sequences added to the N-terminus of FOT and expressed in P. pastoris. The N-terminal residues had various effects on the structural and functional properties of trypsin. The FOT, and the recombinants TE (with peptide YVEF) and TS (with peptide YV) displayed the same optimum temperature ($40^{\circ}C$) and pH (8.0). However, the combinants TE and TS showed significantly increased thermal stability at $40^{\circ}C$ and $50^{\circ}C$. Moreover, the combinants TE and TS also showed enhanced tolerance of alkaline pH conditions. Compared with those of wild-type FOT, the intramolecular hydrogen bonds and the cation ${\pi}$-interactions of the recombinants TE and TS were significantly increased. The recombinants TE and TS also had significantly increased catalytic efficiencies (referring to the specificity constant, $k_{cat}/K_m$), 1.75-fold and 1.23-fold than wild-type FOT. In silico modeling analysis uncovered that the introduction of the peptides YVEF and YV resulted in shorter distances between the substrate binding pocket (D174, G198, and G208) and catalytic triad (His42, Asp102, and Ser180), which would improve the electron transfer rate and catalytic efficiency. In addition, N-terminal residues modification described here may be a useful approach for improving the catalytic efficiencies and characteristics of other target enzymes.

2D 혈관조영술에서 직접 측정한 혈관 직경과 MR 영상에서 단면적 기반 환산 직경의 비교 분석 (Comparative Analysis between Directly Measured Diameter in 2D Angiography and Cross-Sectional Area-Converted Diameter in MR Image)

  • This study aimed to quantitatively compare the diameters measured directly from the coronal plane or sagittal plane of 2D digital subtraction angiography (DSA) and the cross-sectional area-converted diameters calculated from contrast-enhanced MR (CE-MR) imaging. A retrospective analysis was conducted on 20 patients who underwent both 2D DSA and CE-MR imaging. Firstly, the venous diameters of the superior sagittal sinus (SSS) and transverse sinus (TS) were directly measured from 2D DSA. Subsequently, the axial planes for SSS diameter and the sagittal plane for TS in CE-MR imaging were utilized to calculate cross-sectional area-based converted diameters. The numerical values obtained from 2D DSA and CE-MR imaging were compared pairwise at each location. For SSS, the diameter measured by 2D DSA was 27% larger than the conversion-based diameter from CE-MR imaging (9.8±1.4 mm vs. 7.1±1.3 mm, P<0.05). Similarly, for the right TS, the difference was 16% (8.8±3.2 mm vs. 7.4±2.0 mm, P<0.05), and for the left TS, the difference was 22% (8.4±2.8 mm vs. 6.6±1.3 mm, P<0.05). In conclusion, the diameter measured directly in conventional 2D DSA may be larger than the diameter converted based on the cross-sectional area. Therefore, when selecting the size of the stent, it is crucial to make precise determinations while keeping this fact in mind.

$aroP^{-}$변이가 E.coli에서 트립토판 방출에 미치는 영향 (Effects of $aroP^{-}$ mutation on the tryptophan excretion in escherichia coli)

  • 증폭된 재조합 trp operon의 발현을 위한 숙주박테리아 개발의 일환으로 숙수 E. coli에 $aroP^{-}$ 변이를 도입하였다. $aroP^{-}$ 변이의 유도에는 trans po son Tn10을 사용하였으며 P1Kc파아지를 이용하여 숙주박테리아에 형질도입하였다. General aromatic amino acid transport system이 결여된 $aroP^{-}$ 변이주는 $\beta$-thienylalanine ($(2{\times}10^{-4}M)$). p-fluor-phenylalanine ($(2{\times}10^{-4}M)$) 그리고 5-methyltryptophan에 저항성을 가졌다. $aroP^{-}$ 변이주는 $aroP^{-}$ 야생주에 비해서 〔$[^3H]$-tryptophan uptake가 상당히 감소하였다. 또한 NaN, ($(2{\times}10^{-4}M)$)를 처리하였을 때의 ($[^3H]$)-tryptophan uptake 비율은 aroP 변이주가 $aroP^{-}$야생주보다 덜 감소하였다. E. coli $trpR^{-ts}/ColE_1 -trp^+$ 균주에 aroP 형질을 도입하였을 때 트립토판 방출이 $aroP^{-}$ 야생주에 비해서 4 배나 증가하였다.
