Synthesis of Pentadentate Nitrogen-Oxygen(N3-O2) Ligands with Substituents and the Determination of Stability Constants of Their Heavy(II) Metal Complexes (치환기를 가진 질소-산소(N3-O2)계 리간드 합성과 중금속(II)이온 착물의 안정도상수 결정)

  • A new $N_3-O_2$ pentadentate ligand, H-BHPT, was synthesized. Hydrochloric acid salts of Br-BHPT, Cl-BHPT, $CH_3O$-BHPT and $CH_3$-BHPT, having Br-, Cl-, $CH_{3-}$ and $CH_3O-$ substituents at the para position of the phenol hydroxyl group of H-BHPT were synthesized. Hydrochloric acid salts of 3OH-BHPT and 4OH-BHPT, having different position of the phenol hydroxyl group of H-BHPT were also synthesized. The synthesis of each ligand was confirmed by C. H. N. atomic analysis and $^1H$ NMR, $^{13}C$ NMR, UV-visible, and mass spectra. The calculated proton dissociation constants ($log{K_n}^H$) of the phenol hydroxyl group and secondary amine group of the synthesized $N_3-O_2$ ligands showed five steps of the proton dissociations. The order of the overall proton dissociation constants ($log{\beta}_p$) of the ligands was Br-BHPT < Cl-BHPT < H-BHPT < $CH_3O$-BHPT < $CH_3$-BHPT. The order agreed with that of Hammett substituent constants (${\delta}_p$). However, dissociation steps of 3OH-BHPT were four and that of 4OH-BHPT was three. The calculated stability constants ($logK_{ML}$) between the ligands and transition metal ions agreed with the order of $log{\beta}_p$ values of the ligands. The order of the stability constants between the transition metal ions with the synthesized ligands was Co(II) < Ni(II) < Cu(II) > Zn(II) > Cd(II) > Pb(II). The order agreed well with that of the Iriving-Williams.

Treatment of Organic Waste by Vermicomposting (Vermicomposting 에 의한 유기성 폐기물의 처리)

  • A study was made to investigate the possibility of treating various organic waste of cattle manure, swine manure, goat manure, apple pomace, chinese medicine residue and coffee less by feeding these to earthworms and then by studying the growth and reproductive efficiency of earthworms, and the chemical composition of worm cast and its production. The results are summarized as follows. 1. When the feed for earthworms were in good condition which were cattle manure, swine manure and apple pomace, the reproductive efficiency of earthworms was improved however in worse condition the feed of which were goat manure and chinese medicine residue, the increasing rate(IR) became faster. 2. Despite the high content of organic matter(OM) in coffee lees, the earthworms fed coffee lees showed significantly lower reproductive efficiency and increasing rate(IR) than those fed other organic waste due mainly to its lower pH and lower total nitrogen(TN) content. Therefore, when coffee lees is considered to be fed to earthworms, it is believed necessary to be mixed with addiveves or other organic waste in order to improve the feed condition. 3. pH in swine manure, goat manure, apple pomace, chinese medicine residue and coffee lees became neutralized by being fed by earthworms. 4. Available P2O5 and exchangeable cation(EC) of earthworm cast were a little increased compared to those of feed. The contents of Mn, Fe and Cu in earthworm cast were a little decreased to compared to those of feed. 5. The contents of Mn, Fe and Cu in earthworm cast were a little a decreased to compared to those of feed. 6. Mean fresh weight of earthworm at final time(FW2) was negatively correlated with number of young worms(NY)(P<0.01), but positively with increasing rate(IR) (P<0.001) and C/N ratio(P<0.05) respectively. Number of cocoons (NC) and fresh weight of cocoons(WC) were positively(P<0.01) correlated with fresh weight of young worms (WY) and digested matter(DI) but negatively(P<0.001) correlated with residual matter(WY) and digested matter(DI) but negatively(P<0.001) correlated with residual matter(RW). Number of young worms(NY) and fresh weigh of young worms(WY) were negatively(P<0.001) correlated with increasing rate(IR), however increasing rate(IR) of earthworm was positively(P<0.05) correlatedd with C/N ratio.

  • Various catalysts of $Cu_xH_3-{_{2x}}PMo_{12}O_{40}{\cdot}_nH_2O$ with different x-values have been prepared and characterized by thermal analysis, X-ray powder diffraction, infrared spectroscopy, BET surface-area measurement, electron microscopy, and temperature programmed desorption of ammonia. The properties of these catalysts in acrolein oxidation have been investigated in a continuous-flow fixed-bed reactor. The catalysts lost their water of crystallization at about $200^{\circ}C$ and their constitutional water between 300 and $400^{\circ}C$. The Keggin structure of the catalysts was identified by infrared spectroscopy. The decomposition of Keggin anion, $(PMo_{12}O_{40})^{3-}$, was increased with the increase of substituted copper content and identifiable $MoO_3$ and $P_2O_5$ as decomposition products were observed. The conversion of acrolein decreased with the increase of x probably due to the decrease of specific surface area and of total amount of acid sites. But specific reaction rate and selectivity to acrylic acid were maximized at x=1.0, and it showed specific acid site distributions.

  • The EL lamp have been fabricated by screen printing method. the thickness of BaTiO$_3$ dielectric layer and ZnS:Cu phosphor layer was 20 $\mu$m and 40 $\mu$m, respectively. The threshold voltage of green El lamp was 50 $V_{p-p}$ and the maximum brightness was 13.5 $\mu$ W/cm$^2$ at frequency of 700 Hz and the input voltage of 250 $V_{p-p}$. Also when the Rodamin G6 of 0.02 g was doped, the threshold voltage of white EL lamp was 70 $V_{p-p}$ and the maximum brightness was 34 $\mu$W/cm$^2$.

  • In this study, the biochemical characterization of strong proteolytic bacteria and their extracellular pretense isolated from fermented fish paste were experimented for the purpose of industrial large scale-production by accelerated fermentation. The results obtained were as follows: Among 4 strains isolated from fermented fish paste, B. subtilis p-4 and B. licheniformis p-5, which grow well at $40^{\circ}C$, pH 7.0 and $1\%$ of salt contents, were the best proteolytic bacteria and were shown $0.48hr^{-1}$, $0.49hr^{-1}$ of specific growth rate in TPY medium, respectively. Maximum enzyme activity of B. subtilis p-4 was 335n mole-Tyr/min.ml after 30 hrs and that of B. licheniformis p-5 was 300n mole-Tyr/min.ml after 28 hrs of shaking culture. Purified pretense produced by B. subtilis p-4 and B. licheniformis p-5 showed maximum activity at $50^{\circ}C$, pH 7.0 and molecular weight were estimated to be 18,000, 30,000 by sephadex G-100 gel filtration, respectively. These were supposed to be a kind of metal chelator sensitive neutral pretense from the result of high sensitivity against EDTA, o-phe-nanthroline and metal ions such as $Cu^{2+},\;Ni^{2+},\;Zn{2+}$.

Characterization of NAD(P)H-nitroreductase Purified from the TNT-degrading Bacterium, Stenotrophomonas sp. OK-5 (폭약 TNT 분해세균 Stenotrophomonas sp. OK-5에서 분리된 NAD(P)H-nitroreductase의 정제 및 특성 연구)

  • The purpose of this work was to perform the characterization of NAD(P)H-nitroreductase isolated from Stenotrophomonas sp. OK-5 capable of degrading 2,4,6-trinitrotoluene (TNT). Initially, NADP(H)-nitroreductase by a series of purification processes including ammonium sulfate precipitation, DEAE-sepharose, andQ-sepharose was prepared. From samples harvested from fraction collector, three different fractions (I, II & III)having the enzyme activity of NAD(P)H-itroreductase were detected. Specific activities of three fractions I, II,and III of NAD(P)H-nitroreductase were determined to approximately 5.06 unit/mg, 4.95 unit/mg and 4.86 unit/mg, and concentrated to 10.5, 9.8, and 8.9-fold compared to crude extract, respectively. Among these three fractions,the fraction I of NAD(P)H-nitroreductase demonstrated the highest specific activity in this experiment. Several factors affecting on the enzyme activity of NAD(P)H-nitroreductase (fractions I, II & III) were investigated.The optimum temperature of all NAD(P)H-nitroreductase (fractions I, II & III) was 30oC, and the optimal pH was approximately 7.5. Metal ions such as Ag+, Cu2+, Hg2+ inhibited approximately 80% enzyme activity of all NAD(P)H-nitroreductase, and the enzyme activities were decreased about 30-40% inhibition in the presence of Mn2+ or Ca2+. However, Fe3+ showed stimulatory effect on the enzyme activity. The molecular weights of NAD(P)H-nitroreductase (fractions I, II & III) were measured about 27 kDa on the SDS-PAGE.

Synthesis and Characterization of Low-Dimensional Chalcogenide Compound via a Molten Salt Method (용융염법을 이용한 저차원 구조의 금속 칼코겐 화합물의 합성 및 구조 특성연구)

  • The reaction of Cu metal with mixed alkali metal polyselenide flux ($KNaSe_x$) produced large plate-like crystals of $KCu_4Se_3$. The structure of $KCu_4Se_3$, determined with X-ray single crystal diffraction techniques, is tetragonal (P4/mmm, a=4.013(1))${\AA}$, c=9.712(1))${\AA}$, z=1, R=6.7%). The structure is composed $[Cu_4Se_3]_n^{n-}$double layers which are made of fused anti PbO-type Cu2Se2 layers. Temperature variable resistivity measurement on single crystal of $KCu_4Se_3$ shows metallic behavior ranging from $1.8{\times}10^{-4}{\Omega}{\cdot}cm$ (at 300 K) to $1.0{\times}10^{-6}{\Omega}{\cdot}cm$ (at 20 K).

DNA 템플릿을 활용한 전이금속 칼코겐화합물 트랜지스터 기반 바이오센서 연구

  • Field effect transistors (FETs)를 기반으로 한 바이오센서는 빠른 응답속도, 저비용, label-free 등을 이유로 각광받고 있다. 그러나 3D 구조를 기반으로 한 FETs 바이오센서의 낮은 sensitivity의 한계점을 지니며, 이를 극복하기 위해 1D 구조의 나노튜브 등을 활용하였으나 여전히 높은 sensitivity의 확보는 힘들다. 최근에는 이러한 문제점을 극복하기 위해 이차원 반도체 물질 중 하나인 Transition metal dichalcogenide (TMD)를 이용하여, 700 이상의 sensitivity를 지니는 pH센서 및 100 이상의 sensitivity를 지니는 바이오센서가 보고되었다. 하지만 이보다 더 높은 정확성 및 반응성을 높이기 위한 연구는 부족한 실정이다. 우리는 DNA 템플릿을 이용하여, TMD FET 기반 pH 및 바이오센서의 반응성을 극대화시키는 연구를 선보인다. DNA는 7~8정도의 유전상수 (K)를 가지는 물질로 기존 $SiO_2$(K=3.9)보다 높은 유전상수를 가지며 두께를 0.7 nm로 매우 얇게 형성할 수 있는 장점이 있다. 이는 FET 기반 바이오센서의 표면 캐패시턴스를 높여 sensitivity를 극대화할 수 있으며, 기존에 사용된 high-k 기반 바이오센서와 비교하여도 약 10배 이상의 sensitivity 향상을 노릴 수 있다. 또한, TMD 물질로 우리는 $WSe_2$를 선택하였으며, pH 용액의 receptor로써 우리는 3-Aminopropyltriethoxysilane (APTES)를 활용하였고, 템플릿으로 사용된 DNA는 DX tile 및 Ring type의 두 가지를 사용하였다. 추가로, DNA의 phosphate backbone을 중성화시키고 DNA의 base pairing의 charge 안정화를 위해 구리 이온($Cu^{2+}$) 및 란타넘족($Tb^{3+}$)을 추가하였다. 완성된 바이오센서의 pH 센싱을 위해 우리는 pH 6,7,8의 표준 용액을 사용하였으며, 재현성 및 반복성의 확인하였다.

Cloning and Expression of pcbAB Genes from Pseudomonas sp. DJ-12 in Escherichia coli (Pseudomonas sp. DJ-12 pcbAB 유전자의 Escherichia coli에서의 클로닝 및 발현)

  • The pchAB genes of Pseudomonas sp. DJ-12 produce the enzymes of 4-chlorobipheny] (4CB) dioxygenase and dihydrodiol dehydrogenase which act on the first and second steps in degradation of 4CB and biphenyl. The genes were cloned in E coli XLI-Blue. The pcbAB genes of about 2.2 kb in size were contained in the pCUlO1 hybrid plasmid in the cloned cell of CUIOI. The genes were found to have their own promoter and three restriction sites for HindlII. 2,3-dihydroxybiphenyl was detected by the resting cell assay, as the metabolite transformed from biphenyl by the cloned cell of CUIOI. This means that the pcbAB genes are well expressed in E. coli. But dechlorination was unlikely involved in the pchAB gene expression but was believed to occur by functioning on 4CBA produced after ring-cleavage of 4CB.

Effect of Oxygen Radicals and Aeration on Carotenogenesis and Growth of Phaffia rhodozyma(Xanthophyllomyces dendrorhous)

  • Mn(II)+succinate decreased the carotenoid formation of the yeast Phaffia rhodozyma, probably by scavenging $O_2$. When duroquinone (DQ), an internal and external $O_2$ generator, was added to medium, P. rhodozyma produced more amount of carotenoids. The increased carotenoid production was destroyed by oxygen radical (OR) scavengers, ascorbate+Cu(II) and dimethylsulfoxide. When sub-lethal concentrations of $H_2O_2$ , an external OR source, and antimycin, an internal OR inducer, were used, the effect of $H_2O_2$ on carotenoid formation and composition was less significant than that of antimycin. Addition of superoxide dismutase, an external OR remover, rescued cells from death caused by the high concentration of DO. In this condition, the yeast culture showed an increase in carotenoid content. Addition of DQ into P. rhodozyma culture in the stationary phase did not increase carotenoid production. Therefore, carotenoid formation was stimulated by internal ORs in the growing yeast. It was probably due to release of catabolite repression on carotenogenesis in the yeast. Aeration was important for carotenoid production but was not as effective as the internal OR producer, DQ.
