First Record of Three Pleuromamma Species (Copepoda: Calanoida: Metridinidae) in Korea

  • Three Pleuromamma copepods were newly collected from the South Sea of Korea in the summer when the Tsushima Warm Current was strong. They were identified as Pleuromamma abdominalis(Lubbock, 1856), P. indica Wolfenden, 1905, and P. xiphias (Giesbrecht, 1889). Although these species have been reported in world oceans, they were first reported from Korean waters. So far, four species of Pleuromamma (P. borealis Dahl, 1893, P. gracilis Claus, 1863, P. piseki Farran, 1929, and P. robusta (Dahl, 1893)) have been recorded in Korean waters. We provided re-description of these three species insufficiently described by previous authors and key characters for species identification in addition to species hitherto known in Korea.

Korean Red Ginseng water extract inhibits COX-2 expression by suppressing p38 in acrolein-treated human endothelial cells

  • Cigarette smoke is considered a major risk factor for vascular diseases. There are many toxic compounds in cigarette smoke, including acrolein and other ${\alpha},{\beta}$-unsaturated aldehydes, which are regarded as mediators of inflammation and vascular dysfunction. Furthermore, recent studies have revealed that acrolein, an ${\alpha},{\beta}$-unsaturated aldehyde in cigarette smoke, induces inflammatory mediator expression, which is known to be related to vascular diseases. In this study, we investigated whether Korean Red Ginseng (KRG) water extract suppressed acrolein-induced cyclooxygenase (COX)-2 expression in human umbilical vein endothelial cells (HUVECs). Acrolein-induced COX-2 expression was accompanied by increased levels of phosphorylated p38 in HUVECs and KRG inhibited COX-2 expression in HUVECs. These results suggest that KRG suppresses acrolein-induced COX-2 expression via inhibition of the p38 mitogen-activated protein kinase signaling pathway. In addition, KRG exhibited an inhibitory effect on acrolein-induced apoptosis, as demonstrated by annexin Vepropidium iodide staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling assay. Consistent with these results, KRG may exert a vasculoprotective effect through inhibition of COX-2 expression in acrolein-stimulated human endothelial cells.

Effects of Enhanced pCO2 and Temperature on Reproduction and Survival of the Copepod Calanus sinicus

  • We tested the combined effects of increased partial pressure of $CO_2$ ($pCO_2$) and temperature on the reproduction and survival of the copepod Calanus sinicus from Asan Bay, the Yellow Sea under laboratory conditions to understand the impact of acidification on copepods. Egg production rate, survival rate, and fecal pellet production of C. sinicus were not affected by 1305 ppm $pCO_2$ or with combined treatments of temperature and $pCO_2$, including $8^{\circ}C$ and 289 ppm $pCO_2$ (ambient), $8^{\circ}C$ and 753 ppm $pCO_2$ (high $pCO_2$), $12^{\circ}C$ and 289 ppm $pCO_2$ (high temperature), and $12^{\circ}C$ and 753 ppm $pCO_2$ (greenhouse), for 5 or 10 d of exposure. However, egg hatching success of C. sinicus decreased significantly in the greenhouse treatment compared with the ambient or the high $pCO_2$ treatments. These results suggest that a combined treatment ($pCO_2$ and temperature) affected egg viability more than a single treatment($pCO_2$).

간호사의 희망, 감사성향, 일터영성이 조직시민행동에 미치는 영향 (Effects of Hope, Grateful Disposition, Workplace Spirituality on Organizational Citizenship Behavior among Nurses)

  • 본 연구는 간호사의 희망과 감사성향, 일터영성이 조직시민행동정도를 파악하고, 조직시민행동에 미치는 영향요인을 분석하여 바람직한 간호조직문화를 형성 및 증진시켜 간호조직의 효율적이고 경쟁력 있는 인적자원관리방안을 제시하고, 중재방안을 마련하는데 기초자료를 제공하고자 시도되었다. 서울 및 경기도 소재 종합병원 이상의 4개 병원에서 근무하는 간호사 147명을 대상으로 설문조사하였고, 수집된 자료는 SPSS 18.0 프로그램을 이용하여 t-test와 ANOVA, Person's correlation coefficients, multiple stepwise regression으로 분석하였다. 연구결과 간호사의 희망(${\gamma}=.22{\sim}.36$, p<.001), 감사성향(${\gamma}=.38$, p<.001), 일터영성(${\gamma}=.38{\sim}.54$, p<.001), 조직시민행동 간에는 정적 상관관계를 보였다. 또한, 일터영성(${\beta}=.54$, p<.001), 이직희망(${\beta}=.12$, p<.001), 희망(${\beta}=.09$, p<.001), 감사성향(${\beta}=.08$, p<.001)은 조직시민행동에 유의한 영향을 주었으며, 설명력은 41.6%이었다. 본 연구는 간호조직에서 아직 초기단계의 연구가 진행되고 있는 일터영성에 대한 기초이론을 체계화하였고, 간호사의 조직시민행동을 유발시키기 위해 간호사의 희망, 감사성향, 일터영성을 높일 수 있는 다양한 교육 프로그램과 지원 제도가 요구된다.

LPS에 의해 자극된 RAW264.7 대식세포에서 L-AHG에 의한 M1 분극화 및 친염증 신호 경로의 억제 (L-AHG-mediated Suppression of M1 Polarization and Pro-inflammatory Signaling Pathways in LPS-stimulated RAW264.7 Macrophages)

  • 아가로오스의 효소가수분해산물로 부터 확보할 수 있는 3,6-무수갈락토오스(L-AHG), 네오아가로비오스(NA2), 네오아가로테트라오스(NA4) 및 네오아가로헥사오즈(NA6)의 항염증 활성을 규명하고자, 마우스 대식세포주 RAW264.7 세포를 대장균 유래 지질다당류(LPS)로 자극할 때 세포표면의 TLR4 수용체의 역할을 통해 유도되는 친염증성 M1 분극화 및 이에 따른 친염증성 반응에 미치는 영향을 조사하였다. 그 결과로서, 이들 아가로오스 분해산물들 중에서 단당류인 L-AHG만이 유일하게, LPS 자극에 의해 RAW264.7 세포에서 유도될 수 있는 M1 분극화의 대표적인 마커인 iNOS 효소의 발현과 이에 따른 NO 생성을 농도의존적으로 현저하게 저해하였고 염증 매개체인 프로스타글란딘 E2의 생성을 촉매하는 COX-2의 발현 수준도 LPS 자극후에는 증가하였지만, L-AHG의 존재에 의해서는 그 증가 수치가 다소 저해되는 것으로 나타났다. 이때 RAW264.7 세포에 대한 L-AHG의 세포독성은 확인되지 않았다. 또한 L-AHG는 LPS로 처리된 RAW264.7 세포내에서 유도되는 친염증 신호전달경로에 있어서 초기 신호전달 단계인 TAK1 활성화 단계까지는 별 영향을 나타내지 않았으나 TAK1의 촉매작용에 의한 JNK 및 p38 MAPK의 활성화 인산화(activating phosphorylation)는 현저하게 저해되었다. 특히, 대식세포의 친염증 신호전달경로에서 TAK1 활성화는, 그 하류 단계에서 NF-kB의 활성화가 성공적으로 일어날 수 있도록 해주며, 또한 TAK1에 의한 하류 신호분자인 p38 MAPK 활성화는 NADPH 활성화 및 이에 따른 친염증성 ROS 분자들의 생성을 유발하기 때문에, LPS에 의해 자극된 대식세포내의 친염증 신호전달경로에 있어서 TAK1-JNK/p38 MAPK 경로의 L-AHG에 의한 저해는 대식세포의 M1 분극화 및 친염증 반응을 억제하는 효과적인 기전이 되며, 그 활용성이 크게 기대된다. 아울러 L-AHG가 LPS와 RAW264.7 세포의 표면분자인 TLR4와 상호작용을 방해할 수 있는지에 대해서도 유세포분석기와 형광표지가 된 FITC-LPS를 이용하여 조사한 결과, L-AHG는 대식세포의 표면에서 이루어지는 LPS-TLR4 상호결합은 방해하지 않는 것으로 확인되었다. 이는 L-AHG의 항염증 작용의 표적이 LPS가 TLR4 수용체를 통해 유도하는 세포 내 신호전달경로에 있음을 지지해 준다. 이상의 연구결과들은 아가로오스 유래 희귀 단당류인 3,6-무수갈락토오스(L-AHG)가 LPS 자극에 의해 유발되는 RAW264.7 마우스 대식세포의 M1 분극화 및 염증 반응에 대해, TLR4의 친염증 신호전달경로의 TAK1-JNK/p38 MAPK 단계를 저해하는 항염증 활성을 효과적으로 발휘할 수 있음을 시사한다.

상기생과 봉독이 간암 세포주 Hep G2에 대해 미치는 항암 기전 비교 (Comparative Study of Korean Mistletoe Lectin and Bee Venom on the Anti-Cancer Effect and Its Mechanisms of Action in Hepatocellular Carcinoma Cells)

  • Background and Objectives : Korean mistletoe lectin (Viscum album coloratum agglutinin, VCA) and bee venom (BV) have been reported to induce apoptosis in various cancer cell lines in vitro and to show antitumor activity against a variety of tumors in animal models. However, the comparative effect of VCA and BV on the anti-cancer effect and mechanisms of action has not been determined. In this study, the effect in a human hepatocellular carcinoma cell line, Hep G2 cells, was examined. Methods : Cytotoxic effects of VCA and BV on Hep G2 cells were determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay in litro. The apoptotic cell death was then confirmed by propidium iodide staining and DNA fragmentation analysis. The mechanisms of action were examined by the expression of anti-apoptotic proteins and activation of mitogen-activated protein kinases. The involvement of kinase was examined in VCA or BV-induced apoptosis by using kinase inhibitors. Results : VCA and BV killed Hep G2 cells in a time and dose-dependent manner. Treatment of Hep G2 cells with VCA activated poly (ADP-ribose) polymerase-1 (PARP-1) known as a marker of apoptosis, and mitogen-activated protein kinases signaling pathways including MAPK/ERK, p38 MAPK and JNK. BV also activated PARP-1, MAPK/ERK. and p38 MAPK but not JNK. The expression level of anti-apoptotic molecule, Bcl-X, was decreased by VCA treatment but not by BV. Finally, the phosphorylation level of ERM proteins involved in the cytoskeleton homeostasis was decreased by both stimuli. VCA-induced apoptosis was partially inhibited by in the presence of JNK and p38 inhibitor, but BV only by p38 inhibitor. Conclusions : VCA-induced apoptosis is dependent on the activation of p38 and JNK. while BV-induced apoptosis is mediated by p38 activation in Hep G2 cells.

Redox Factor-1 Inhibits Cyclooxygenase-2 Expression via Inhibiting of p38 MAPK in the A549 Cells

  • In this study, we evaluated the role of apurinic/apyrimidinic endonuclease1/redox factor-1 (Ref-1) on the tumor necrosis factor-$\alpha$ (TNF-$\alpha$) induced cyclooxygenase-2 (COX-2) expression using A549 lung adenocarcinoma cells. TNF-$\alpha$ induced the expression of COX-2 in A549 cells, but did not induce BEAS-2B expression. The expression of COX-2 in A549 cells was TNF-$\alpha$ dose-dependent (5~100 ng/ml). TNF-$\alpha$-stimulated A549 cells evidenced increased Ref-1 expression in a dose-dependent manner. The adenoviral transfection of cells with AdRef-1 inhibited TNF-$\alpha$-induced COX-2 expression relative to that seen in the control cells ($Ad{\beta}gal$). Pretreatment with $10\;{\mu}M$ of SB203580 suppressed TNF-$\alpha$-induced COX-2 expression, thereby suggesting that p38 MAPK might be involved in COX-2 expression in A549 cells. The phosphorylation of p38 MAPK was increased significantly after 5 minutes of treatment with TNF-$\alpha$, reaching a maximum level at 10 min which persisted for up to 60 min. However, p38MAPK phosphorylation was markedly suppressed in the Ref-1-overexpressed A549 cells. Taken together, our results appear to indicate that Ref-1 negatively regulates COX-2 expression in response to cytokine stimulation via the inhibition of p38 MAPK phosphorylation. In the lung cancer cell lines, Ref-1 may be involved as an important negative regulator of inflammatory gene expression.

Evidence for the Association of Ce11u1ar Iron Loss in Nitric Oxide-induced Apoptosis of HL-60 Cells: Involvement of p38 Kinase, c-Jun N-terminal Kinase, Cytochrome C Release, and Caspases Pathways

  • Nitric oxide has high affinity for iron, and thus it can cause intracellular iron loss. We tested the idea that intracellular iron can be the primary target of NO toxicity by comparing the signaling mechanisms involved in cell death caused by iron depletion and that caused by NO. Treatment of HL-60 cells with a NO donor, S-nitroso-N-acetyl-DL-penicillamine (SNAP), decreased the intracellular iron level rapidly as that observed with the iron chelator deferoxamine (DFO). Iron chelators such as DFO and mimosine could induce death of human leukemic HL-60 cells by a mechanism requiring activation of p38 kinase, c-Jun N-terminal kinase, caspase-3 and caspase-8. DFO and SNAP also caused release of cytochrome c from mitochondria. Inhibition of p38 kinase by a selective inhibitor, SB203580, abolished the NO and DFO-induced cell death, release of cytochrome c, and activation of caspase-3 and caspase-8, thus indicating that p38 kinase lies upstream in the cell death processes. In a parallel situation, the cells that are sensitive to NO showed similar sensitivity to DFO. Moreover, simultaneous addition of ferric citrate, an iron-containing compound, inhibited the SNAP and DFO-induced activation of caspases and also blocked the NO-mediated cell cycle arrest at $G_1$ phase. Collectively, our data implicate that the NO-induced cell death of tumor cells including HL-60 cells is mediated by depletion of iron and further suggest that activation of p38 kinase lies upstream of cytochrome c release and caspase activation involved in this apoptotic process.
