Microbial Quality and Safety of Fresh-Cut Broccoli with Different Sanitizers and Contact Times

  • This study was conducted to investigate the effects of different sanitizers and contact times on storage quality and microbial growth in fresh-cut broccoli. Fresh broccoli samples were cut into small pieces, washed each for 90 s and 180 s in normal tap water (TW), $100\;{\mu}/l$ chlorinated water (CL, pH 7), electrolyzed water (EW, pH 7.2) containing $100\;{\mu}/l$ free chlorine, or $2\;{\mu}/l$ ozonated water ($O_3$). Then, samples were packaged in 30-${\mu}m$ polyethylene bags and stored at $5^{\circ}C$ for 9 days. No significant differences were observed in gas composition and color parameters ($L^*$, $a^*$, $b^*$, and hue angle) among different sanitizers with contact times. No off-odor was detected during the storage. A longer contact time was not effective in reducing microbial population, except with $O_3$ washing. $O_3$ with 90 s was not much effective in reducing microbial population compared with Cl or EW. However, samples washed with $O_3$ for 180 s observed the lowest numbers of total aerobic and coliform plate counts. The result suggested that, a longer contact time of ozone can be used as a potential sanitizer to maintain the microbial quality and safety of fresh-cut broccoli.

  • The pickling brine generated from the salting process of kimchi production is difficult to treat biologically due to very high content of salt. When pickling brine is treated and discharged, it cannot satisfy the criteria for effluent water quality in clean areas, while resources such as the salt to be recycled and the industrial water are wasted. However, sterilization by ozone, UV and photocatalyst is expensive installation costs and operating costs when considering the small kimchi manufacturers. Therefore there is a need to develop economical process. The study was conducted on the sterilization efficiency of the pickling brine using electrochemical processing. The electrochemical treatment of organic matters has advantages over conventional methods such as active carbon absorption process, chemical oxidation, and biological treatment because the response speed is faster and it does not require expensive, harmful oxidizing agents. This study were performed to examine the possibility of electrochemical treatment for the efficient processing of pickling brine and evaluated the performance of residual chlorine for the microbial sterilization.

Properties of Bleachability of Paper Mulberry Pulp by Hydrogen Peroxide and Ultrasonication Bleaching System (과산화수소와 초음파 표백 시스템에 따른 닥나무 펄프의 표백 효율 특성)

  • Nowadays, the concern on the environmental load of bleaching process gave rise to the process of ECF(elemental chlorine free) and TCF(total clorine free). These sequences are based on oxygen-derived compounds such as oxygen, ozone, and hydrogen peroxide which is used as a typical eco-friendly bleaching agent. In this study, paper mulberry pulp was bleached with hydrogen peroxide and some bleaching process were accompanied with ultrasonication in order to increase the bleaching efficiency. The best bleaching efficiency of paper mulberry pulp was obtained in the condition of hydrogen peroxide and ultrasonication(20 kHz) bleaching system at $45^{\circ}C$ for 30 min. The brightness and kappa number of paper mulberry pulp were gained to 5.09% and 3.52 respectively. and yield was slightly loosed to 2%. Therefore, the efficiency of hydrogen peroxide and ultrasonication bleaching system of paper mulberry pulp was superior to the conventional hydrogen peroxide bleaching system. Magnesium sulfate acted as a bleaching stabilizer for the increasement of yield. As a result, the yield and viscosity were increased to 2.2% and 12% respectively.

Control of Soybean Sprout Rot Caused by Pythium deliense in Recirculated Production System

  • A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.

A Study on the Variation of HAA Precursors by Ozonation of Phenol (페놀의 오존 산화시 관찰된 HAA 전구물질 변화에 관한 연구)

  • The purpose of this study was to find out the effect of oxidation by-products for the formation of haloacetic acid (HAA) during ozonation. The phenol was used as a model precursor of HAA, and its oxidation by-products, such as hydroquinone, catechol, glyoxal, glyoxylic acid and oxalic acid were investigated to find out how much HAA formation potential (HAAFP) they have. As the result, among the phenol and its oxidation by-products, the highest reactivity with chlorine was found from the phenol, showing the highest HAAFP. Even though the tested by-products had a lower HAAFP than phenol, it was confirmed that all of them can act as the precursor of HAA. From the ozonation of phenol-containing water, it was found that the efficiency of ozone in controlling of HAAs can be reduced due to the oxidation by-products. In addition, the ozonation of HAAFP was performed under the both pH conditions (acid and base), and the result indicates that OH radical play a important role to decrease HAAFP.

Epidemiological Studies on Giardia Infection Associated with environmental Pollution (Giardia에 의한 환경오염(環境汚染)과 감염(感染)에 관한 역학적(疫學的) 연구(硏究))

  • Giardia lamblia is a pathogenic flagellate causing intestinal disorders such as diarrhea, abdominal pain and malabsorption of nutrients. Giardia is mainly infected by the ingestion of contaminated foods per os. Craun (1979) has recently reported that mass infection of this flagellate through the contaminated water supply systems is one of public health hazards. Also, so-called traveller's diarrhea is sometimes caused by Giardia infection (CDC, U.S.A., 1971). However, a few epidemiological studies figuring out the mode of infection or control measures of Giardia infection has been done so far in Korea. The present study was aimed to know the prevalence of Giardia infection in several Korean populations, detectability of this flagellate in water systems and the viability of the cysts against sewages and disinfectants applying to drinking water. In the present study, 388 stool specimens from orphanage children in Chun-joo, Chung-joo, On-yang and Chun-an areas and 538 stool specimens from inhabitants in Woo-do, In-chon, and Chun-joo were examined by formalin-ether concentration technique to detect out Giardia cysts. On the other hand, water samples from 14 sites of Han River and its tributaries were collected in May through July, 1984. Fifty liter of water sample in each sampling site was then filtered through water filtering system deviced by U.S. Environmental Proutection Agency and the sediments rinsed out from the thread rolls, a part of water filtering system, were examined to detect out the Giardia cysts. In order to observe the viability of Giardia cysts in the sewage samples, the cysts were treated in it at $4^{\circ}C$ or $25^{\circ}C$ for 7 through 28 days. For this purpose, the cysts were also exposed to various concentrations of disinfectants such as chlorine, iodine and ozone gas for proper time intervals. After treatment, the viability test of the Giardia cysts were carried out by method of Rice and Schaefer (1981) with minor modification. The results obtained in this study were as follows : 1) The detection rates of G lamblia cysts in the stool specimens were 18.3% in orphans and 4.3% in general examinees. 2) The prevalences of Giardia Infection were higher in the young age groups than in-adults. The highest positive rate was 18.4% in the age group less than 10. 3) Of 14 water specimens sampled from Han River system and its tributaries around the Seoul area, the Giardia cysts were detected from 4 samples, and no cyst was found in the water supply systems. 4) The cysts treated in the sewage survived for 28 days at $4^{\circ}C$ and for 13 days at $25^{\circ}C$. 5) The cysts were completely destroyed within 60 minutes by exposure to 8 mg/l of residual chlorine at 4g and within 30 minutes by exposure to the same concentration of chlorine at $25^{\circ}C$. 6) The cysts were all dead when exposed to 1 mg/1 of iodine for 60 minutes at $4^{\circ}C$ or $25^{\circ}C$. 7) The cysts were destroyed after 10 minute exposure in 0.15 mg to 0.25mg of residual ozone gas per liter. Summarizing the above results, it is considered that Giardia infection is regarded as water-borne disease and the cysts are able to be controlled by the application with the disinfectants in the water supply systems.

Control Indian meal moth Plodia interpunctella by gas treatment

  • The Indian meal moth, Plodia interpunctella, is one of the most important pests of stored food in the food processing industry worldwide. To control the Indian meal moth, methyl bromide, phosphine, high carbon dioxide, sulfuryl fluoride and plant essential oil fumigation have been considered. However, these treatments have disadvantages. For example, depleting the ozone layer, showing resistance in insect, low control efficacy or need high cost for treatment. Chlorine dioxide ($ClO_2$) is strong disinfectant and insecticide. The gas caused a malfunction in enzymes. The oxidative stress induced by $ClO_2$ gas treatment damaged to a physiological system and all life stages of P. interpunctella. The gaseous $ClO_2$ is a convincing alternative to methyl bromide for controlling P. interpunctella. The gaseous $ClO_2$ was generated by a chlorine dioxide generator (PurgoFarm Co., Ltd., Hwasung, Korea). It generated highly pure $ClO_2$ gas and the gas blown out through a vent into a test chamber. Gas entry to the chamber was automatically controlled and monitored by a PortaSene II gas leak detector (Analytical Technology, Collegeville, PA, USA). The properly prepared eggs, larvae, pupae, and adults of P. interpunctella were used in this experiment. Data were analyzed using SAS 9.4. Percentage data were statistically analyzed after arcsine-root transformation. Analysis of variance was performed using general linear model, and means were separated by the least significant difference test at P < 0.05. Fumigation is an effective management technique for controlling all stages of P. interpunctella. We found that $ClO_2$ gas treatment directly effects on egg, larvae, pupae and adults of P. interpunctella. The gas treatment with proper concentration for over a day achieved 100 % mortality in all stages of P. interpunctella and short time treatment or low concentration gas treatment results showed that the egg hatchability, pupation rate, and adult emergency rate were lowered compare with untreated control. Also, abnormal pupae or adult rate were increased. Gaseous $ClO_2$ treatment induced insecticidal reactive oxygen species (ROS), and it resulted in fatal oxidative stress in P. interpunctella. Taken together, these results showed that exposure proper concentration and time of the gas control all stages of P. interpunctella by inducing fatal oxidative stress. Further studies will be required to apply the gas treatment under real-world condition and to understanding physiological reaction in P. interpunctella caused by oxidative stress.

The Antibiotic Resistant Gene Pollutant Controls using Chlorine or Ozone disinfection (염소 또는 오존을 이용한 항생제 내성 유전오염물질 제어)

  • The aim of this study was to examine ozonation disinfection efficiency for Escherichia coli DH5alpha removal, containing the multi-resistance plasmid pB10 as well as chlorination disinfection efficiency. In addition, plasmid pB10 removal rates were estimated by ozonation and chlorination. The removal efficiency of pB10 via ozonation was about 2 to 4 times higher than chlorination. High removal efficiency of pB10 is likely due to OH radical produced during ozonation. These results suggest that integration of advanced oxidation process such as ozonation (or photocatalytic oxidation) with conventional disinfection such as chlorination may be needed for effective control of antibiotic resistant bacteria and genetic materials.

Fates and Removals of Micropollutants in Drinking Water Treatment (정수처리 과정에서의 미량오염물질의 거동 및 제거 특성)

  • Micropollutants emerge in surface water through untreated discharge from sewage and wastewater treatment plants (STPs and WWTPs). Most micropollutants resist the conventional systems in place at water treatment plants (WTPs) and survive the production of tap water. In particular, pharmaceuticals and endocrine disruptors (ECDs) are micropollutants frequently detected in drinking water. In this review, we summarized the distribution of micropollutants at WTPs and also scrutinized the effectiveness and mechanisms for their removal at each stage of drinking water production. Micropollutants demonstrated clear concentrations in the final effluents of WTPs. Although chronic exposure to micropollutants in drinking water has unclear adverse effects on humans, peer reviews have argued that continuous accumulation in water environments and inappropriate removal at WTPs has the potential to eventually affect human health. Among the available removal mechanisms for micropollutants at WTPs, coagulation alone is unlikely to eliminate the pollutants, but ionized compounds can be adsorbed to natural particles (e.g. clay and colloidal particles) and metal salts in coagulants. Hydrophobicities of micropollutants are a critical factor in adsorption removal using activated carbon. Disinfection can reduce contaminants through oxidation by disinfectants (e.g. ozone, chlorine and ultraviolet light), but unidentified toxic byproducts may result from such treatments. Overall, the persistence of micropollutants in a treatment system is based on the physico-chemical properties of chemicals and the operating conditions of the processes involved. Therefore, monitoring of WTPs and effective elimination process studies for pharmaceuticals and ECDs are required to control micropollutant contamination of drinking water.

Variation of Natural Organic Matter Characteristics through Water Treatment Processes (정수공정별 천연유기물질의 특성 변화)

  • Natural organic matter (NOM) which occurs ubiquitously in both surface and ground waters, consists of both humic (i.e., humic and fulvic acids) and nonhumic components. NOM in general as well as certain constituents are problematic in water treatment. From a regulatory perspective, concerns focus upon the role of NOM constituents as disinfection byproduct (DBP) precursors. The fractionation of NOM through water treatment processes can provide insight into treatment process selection and applicability. Problematic NOM fractions can be targeted for removal or transformation. Significant source-related differences in NOM were observed among various source waters. This study found that bulk Dissolved Organic Carbon (DOC) concentration was hardly removed by oxidation process. Oxidation transformed high Molecular Weight (MW) hydrophobic fraction into low MW hydrophilic fraction. Ozone reduced s-pecific Ultraviolet Absorbance (SUVA) value more than chlorine. High MW hydrophobic fraction was effectively removed by coagulation process. About 50% of Trihalomethane Formation Potential (THMFP) was removed by coagulation process.
