Isolation and Some Properties of Bitter Taste Compounds from Cultured Oyster, Crassostrea gigas (양식산 굴로부터 쓴맛 성분의 분리 및 성질)

  • Five bitter taste compounds (OY-22, OY-23, OY-24, OY-25 and OY-26) were firstly isolated from cultured oyster (Crassostrea gigas) at Gamak Bay, Sourthern coast of Korea, between November, 1989 and January, 1990, and smoked-canned oyster, which were produced by the same oysters. They were presumed as cyclic peptides composed with 6 or 7 amino acids, including sulfur on the basis of NMR and MS spectra. Val and Leu in OY-24, leu and lie in OY-25 and tow leucines in OY-26 were detected from those each compounds, seperately, by the amino acid analysis. Another amino acids were thought as non-constitutional amino acids. They showed non-toxic to mice $(100{\mu}g/20g\;mice i.p.)$ and non actibacterial artivities to Asp. niger and B. subtilis $(10{\mu}g/disk)$. The chemical structures and other biological activities of them are now in studying.

  • Vibrio are become prevailing if superficial temperature of ocean is raised and their activities of area are expanded and most of ocean creatures(fishes, oysters etc) are polluted with vibrio. The one who has taken these poluted fishes and aysters uncooked caused foodpoisoning and diarrhea from Vibrio. Frequencies of these diseases breakout is disposed in westsea shore of Korea. According to ancient and traditional Korean medical book -${\ulcorner}$Bangyak Happyeon${\lrcorner}$(Collection of Local Medicines, 1884) - and their single prescribes, we carried out experiment check the activities of natural medicinal effects on Vibrio parahemolyticus. The prescriptions of trial materials are processed from extraction boiling water and 80% methanol and followed freeze dried and adsorbed to every discs in dosage of 10mg. Gentamycin of 10mg were used for control. The result of compound prescription displayed special diseases in antimicrobial activities of boiling water and MeOH extraction compared with control. In compound prescription, extraction MeOH of Sashinhwan(clear zone : 17mm) presented extraordinaire antimicrobial activity. In single prescription, extraction of boiling water(clear zone : 16mm) and MeOH(clear zone : 18mm) of Fructus Chebulae presented extraordinaire antimicrobial activity. The MBC of Fructus Chebulae extracts was expressed in boiling water(1.28mg/ml) and MeOH(0.64mg/ml).

MICROENVIRONMENT IN OYSTER FARM AREA 1. On the Eutrophication and Raft Density in Geoje Bay (굴 양식장의 미세환경에 관한 연구 1. 거제만의 양식장밀도 및 부영양화에 관하여)

  • After a great mortality owing to abnormal oceanographical condition and fungal disease in 1973 hanging cultch lines per raft has been reduced to 450 from 558, and oyster production per raft has also been decreased. It seems to be result of dense culture of oysters and its resulting accumulation of waste materials on the sea bottom in the farm area. The present study was carried out to investigate the effect of rearing density and the degree of eutrophication in Geoje Bay for 6 months from June through November in 1977. Total area of this bay is about $48.9\;km^2$, and the area of registered oyster farms as of November in 1977 is around $10.9\;km^2$, which is about $22.3\%$ of this bay. Water Quality during summer season was shown as transparency 5.5 m, COD 1.5ppm, degree of oxygen saturation $90.6\%$ in upper and middle layers and $82.2\%$ near bottom, chlorophyll-a 3.0 mg/m^3, and phytoplankton $8.7\times10^4\;cells/l$. In superficial mud in August COD was 35.4 mg/g, total sulphide 0.24 mg/g, and phaeophytin $43.7\;{\mu}g/g$dry mud. These values indicate that water qualify in this bay is so far excellent but quantities of chemical oxygen demend and sulphide in bottom mud show maximum level or a little over eutrophication standard.

Comparison of Food Component of Oyster Drip Concentrates Steamed under Different Retort Pressures (레토르트 열처리 조건에 따른 굴자숙수 농축물의 식품성분 특성 비교)

  • This study was conducted to investigate the food components of cooking drips from shell oysters steamed under various retort pressures. Among the drips from shell oyster steamed under different retort pressures (1.0, 2.0, 2.5, 3.0, 3.5, 4.0 and 4.5 kg/$cm^2$) the oyster drip obtained at the 4.5 kg/$cm^2$ of retort pressure had the highest degree of brix, yield, crude protein, glycogen and trichloroacetic acid soluble-N contents, while its salinity was the lowest. In the results of food safety test, the presence of E. coli (18 MPN/100 g>) and viable cell (30 CFU/g>) in the oyster drip was in acceptable level as a food-stuff. However, the sensory evaluation such as color, flavor and taste, total amino acid and free amino acid contents of cooking drip from shell oyster steamed at 4.5 kg/$cm^2$ were inferior to those of oyster wash water. Differences in the major amino acids of total amino acid and free amino acid between oyster cooking drip and oyster wash water were also found. The results suggested that the effective use methods of oyster cooking drip should be investigated.

Evaluation of the Bacteriological Safety for the Shellfish Growing Area in Hansan.Geojeman, Korea (한산.거제만해역 패류양식장에 대한 세균학적 위생안전성 평가)

  • In Hansan Geojeman area, 2,050 ha of shellfish growing area has been designated as shellfish production area for export. The main shellfish species from the designated area is oysters. For the sanitary management of the designated area established in Hansan Geojeman area, bacteriological examination of sea water and shellfish at the sampling stations inside and outside of the designated area were performed from January 2006 to December 2008. The range of fecal coliform of 756 sea water samples at 21 stations located in the designated area were <1.8~>1,600 MPN/100mL. And the range of geometric mean and the estimated 90th percentile of fecal coliform were 1.8~2.9 and 2.7~15.8 MPN/100mL, respectively. Sanitary conditions of the current designated area in Hansan Geojeman meets the required standards of the Fisheries Product Quality Control and National Shellfish Sanitation Program (NSSP, USA) criteria for the approved area. Also, the sanitary status of the shellfish harvested from the designated area met the Korean Shellfish Sanitation Program (KSSP) fecal coliform criterion (<230 MPN/100g). And the human pathogen such as Salmonella spp. and Shigella spp. were not detected from the examined shellfish samples.

Estimation of Carrying Capacity by Food Availability for Farming Oysters in Goseong Bay, Korea (먹이가용성에 의한 고성만의 굴 양식장 수용력)

  • For the continuous stable production of oyster, estimation of food availability (F) was carried out in Goseong Bay, south of coast Korea. Primary productivity ranged from 0.07 to $0.44gC/m^2/day$ (average $0.25gC/m^2/day$), lowest in July and highest in January. The distribution of primary productivity at Goseong Bay showed the pattern of "high in the south and low in the north." Food availability (F) was $F{\leq}0$, indicating insufficient food supply, from August to November and F > 0 from January to April. Continuous insufficient food supply was observed at 18 oyster farms in the southern part of the bay and 4 in its northern part. Mortality at the oyster farms was 56% on the average, and around 58% of death occurred during November when food supply was insufficient. The optimal population of cultured oyster per unit flow area was calculated to be $110-115indiv./m^2$ (198-201 indiv./string). When the sea area was divided into 3 regions (A, B, C) according to carrying capacity, the carrying capacity of (A) regions was $52-53indiv./m^2$ (93-95 indiv./string), (B) regions was $142-144indiv./m^2$ (255-259 indiv./string), and (C) regions was $198-202indiv./m^2$ (356-363 indiv./string). In particular, (A) regions showed extremely low productivity. For continuous stable oyster farming at Goseong Bay, it is necessary to control point and non-point source pollution through continuous environmental monitoring and to adjust harvest according to the base carrying capacity during the season of high water temperature.

Development of Oyster Culture Industry in Korea (우리 나라 굴양식업의 발상과 발달과정)

  • The author investigated the history of oyster culture in Korea through examining old books and reports dealing with the fishery. Oysters have been produced in the all coastal areas in Korea, especially the center areas of production were the Hwangupo (Hamkyongbuk-do), Younghungman (Hamkyongnam-do), the estuary of Nakdonggang (Kyongsangnam-do), Kwangyangman, Haechangman, the estuary of Yongsangang (Chollanam-do), Yonghoto (Hwanghae-do) and the estuary of Uprockgang, where the large size oyster (more than 40cm in shell height) inhabited in form of groups. It was called by nine different names in the old times, Gul, Seukhwa and Moryeo were the most common names. The oyster was used as 46 kinds of medicinal stuff. The habitat forms of natural oyster can be distinguished to two types; one is that oyster is exposed to the air during the period of low tide (A-type), another is that oyster is not (B-type). The first oyster culture by the old books in Korea was started about 530 years ago as the bottom culture method derived from the A-type of oyster. After long years, the hanging culture method might have been started from the hint of the habitat form of B-type. The oyster culture in Korea developed based on the habitat forms of natural oyster at the biginning of 1900s. After World War II, the Korean government strongly recommded the laver farmers to culture oyster as relief measures. At that time, the stone culture method was mainly adopted. The quantity of oyster production is now about $15{\times}10^6{\sim}19{\times}10^6\;\frac{M}{T}$, and the half of oyster production including seed oyster is exported to the foreign countries.

Effects of Sediment Elutriates on the Early Reproductive Outputs in the Pacific oyster, Crassostrea gigas

  • This is a subsequent study to our previous finding that Pacific oysters, Crassostrea gigas, gained a so-called upper plateau concentration, around 30,000 ng/g dry weight digestive gland for benzo(a)pyrene, showed reproductive behavior but produced their ensuing reproductive outputs damaged. A serial dilution of sediment elutriates from Jinhae Bay, Korea, where pollution was progressive, were exposed to gametes of the Pacific oyster for 0, 5, 10, 20, 30, and 60 min to detail the pollutant effects on very young specimens. There was an apparent critical dilution over which adverse effects are evident. This was $10\%$ of the present sediment elutriate, corresponding to 0.3 ng/g on the basis of total polycyclic aromatic hydrocarbons (PAHs) for the oyster. Within the dilution the embryonic development was not influenced by the duration of exposure to its gamete stage. At higher dilutions over the critical dilution, occurrence of abnormality increased dependent on the pollutant dilution and the duration of exposure. Similar trends were also found in larval mortality. However, overall, the chemical toxicity was more significant to morphogenesis than to survival, suggesting a potential recruitment of the pollutants-induced abnormal larvae in the wild population to threaten the population integrity.

Influence of Environmental Factors on the Prevalence of the Ovarian Parasite, Marteilioides chungmuensis, in Crassostrea gigas, Cultured in Pukman Bay, Tongyeong (양식환경이 통영 북만의 참굴, Crassostrea gigas에 기생하는 난소기생충, Marteilioides chungmuensis 감염에 미치는 영향)

  • Occurrence and prevalence of Marteilioides chungmueasis have been reported in several waters around Tongyeong but no report has been made for Pukman bay. Therefore, we investigated that the prevalence and infection intensities in Pukman Bay at the inside and the outside areas which are hydrographically divided by tidal current. Furthermore, various environmental parameters were investigated in order to elucidate effective parameter for parasitic infection. Infection rates of Marteilioides chungmuensis in adult oysters were ranged 3.3-20.0% at the inside area during September 2002 through January 2003, and 3.3-30.9% at the outside area during August 2002 through January 2003. External manifestation of infected oyster consisted of abnormal egg masses with nodular appearance in the soft tissue. Histopathological symptoms included massive hemocytic infiltration within or around the follicle wall and atrophic epithelium of digestive diverticula. For the environmental parameters, comparative study made differences between two side of the Bay during the infection period: inside > outside for SS while inside < outside for chlorophyll-a. A positive relationship was observed between chlorophyll-a and infection period, which might indicate the difference in food availability between two areas. The prevalence of ovarian parasite Marteilioides chungmuensis, therefore, was highly associated with food availability. Pearson's correlation analysis was made between environmental parameters and infection prevalence. Significance was observed in water temperature (p < 0.05), suspended solids (p < 0.01) and chlorophyll-a (p < 0.05). A principle component analysis showed that infection of the ovarian parasite, Marteilioides chungmuensis, exhibited effects of seasonality (component I = 55.2%) and chemical/physical environmental factors (component II = 24.4%). These results clearly indicate that the infection of ovarian parasite, M. chungmuensis in the Pacific oyster Crassostrea gigas is closely associated with seasonality and food availability.

Antimicrobial Resistance in Escherichia coli Isolated from Shellfish Farms on the West Coast of Korea (서해안 패류양식장에서 분리된 대장균의 항균제 내성)

  • A total of 423 shellfish (Oysters, Crassostrea gigas; short-necked clams, Ruditapes philippinarum and corb shells, Cyclina sinensis) were collected from the west coast of Korea, from which 277 Escherichia coli strains were isolated. The antimicrobial susceptibility patterns of isolated strains were analyzed for 22 antimicrobial agents used in Korea for clinical or veterinary therapy. The resistance of E. coli isolates to ampicillin (37.2%) was highest, followed by cephalothin (21.7%), cefazolin (19.9%), trimethoprim (15.2%). Antimicrobial resistance was present in 56.7% of E. coli isolates against at least one antimicrobial agent. Of 277 isolates, 44 (15.9%) were resistant to multiple antimicrobial agents.
