Study on the Pretreatment of Seafood for the Determination of Total Arsenic using Wet Ashing Method (습식 분해법을 이용한 해산물 중 총 비소 분석을 위한 전처리 방법 연구)

  • Pretreatment method for the determination of total arsenic in seafood sample was studied. NIST SRM 1566a oyster tissue ($14.0{\pm}1.2mg\;As/kg$) as a standard arsenic compound in order to establish the decomposition method. We confirmed that the best way for pretreatment of seafoods to analyze total arsenic content precisely was $HNO_3-H_2SO_4-HCIO_4$ method by comparison of two methods which are dry ashing and wet ashing methods.

Factors Affecting Concentration of Polycyclic Aromatic Hydrocarbons in Oyster, Laver and Rockfish (굴, 김 및 조피볼락에서 다환성방향족탄화수소(Polycyclic Aromatic Hydrocarbons, PAHs)의 잔류에 미치는 요인에 대한 연구)

  • A total of 15 different residues of polycyclic aromatic hydrocarbons (PAHs) in each 20 samples of Pacific oysters, dried laver and rockfish obtained from seafood markets were analyzed. The prevalence of samples in which more than one PAH residues were found was 75% in oyster, 35% in rock fish hepatopancreas, 0% in rockfish muscle and laver, respectively. To estimate factors contributing to this residue level difference among organisms, tissue concentrations were analyzed after exposing three organisms to phenanthrene, a representative PAH, with concentration of 0.01 or $0.1{\mu}g/mL$ for 2 weeks. Phenanthrene levels after exposure were higher in the oyster digestive gland, laver and rockfish hepatopancreas, but were lower in the oyster whole meat or rockfish muscle. This finding disproved that any close relationship between the residue difference of market samples and concentrating properties of PAHs. The second possible factor analyzed was total lipid contents in the three organisms. Although higher lipid level in hepatopancreas of rockfish may contribute accumulation of PAH residues in the rockfish, lipid factor did not affect to PAH levels in other organism samples. Activity of 7-ethyoxyresorufin-O-deethylase (EROD), a kind of cytochrome $P_{450}$ enzyme, was measured to evaluate the eliminated amount of PAHs through metabolism. The higher EROD activity in rockfish, compared to that in oyster, was likely to contribute to the lower PAH residues in the rockfish. More factors, such as different exposure history, organisms' ability to escape, ingestion through prey organisms, and post-harvest loss, should be studied in the future.

Changes of Mortality, Oxygen Consumption Rate and Organ Structure in the Oyster Crassostrea gigas Exposed to Lead (납 (Pb) 에 노출된 굴 Crassostrea gigas의 사망률, 산소소비율 및 기관계 구조의 변화)

  • This study was conducted to find out biological response of the oyster Crassostrea gigas exposed to lead (Pb). Experimental period was four weeks. Experimental groups were composed of one control condition and three lead exposure conditions (0.20, 0.40 and 0.80 mg/L). The results of the study confirmed that lead induces reduction of survival rate and oxygen consumption rate and degeneration of organ structure of the bivalve. Oxygen consumption rate was observed exposure groups lower than control decline by 15%-55%. Histological analysis of organ system illustrated expansion of hemolymph sinus, degeneration of epithelial layer and connective tissue layer of the mantle. Also, epithelial degeneration and disappearance of cilia band are recognized in the gill filament and it was observed disruption of epithelial layer and degeneration of basophilic cell and epithelial cell in the digestive tubules.

Accumulation and Depuration of Paralytic Shellfish Poison in Marine Organisms (수산생물 종류별 마비성 패류독소 축적 및 정화)

  • To compare the accumulation of paralytic shellfish poison (PSP) in different marine organisms, the occurrence and variation of PSP were surveyed in blue mussel Mytilus edulis, oyster Crassostrea gigas, short neck clam Ruditapes philippinarum, bay scallop Argopecten irradians, and warty sea squirt Styela clava collected from Jinhae Bay, Korea, in 2005 and 2006 year. We also investigated the ability of the blue mussel to detoxify PSP by relaying and depuration (via the water flow or water circulation system). In the marine organisms examined, PSP levels were the highest in blue mussel, followed in order by bay scallop, oyster, short neck clam, and warty sea squirt. Comparing the maximum PSP levels in the bivalve species examined in 2005 and 2006, PSP in blue mussel was 1.6-2.0, 4.0-5.9, and 5.1-6.0 times higher than in bay scallop, oyster, and short neck clam, respectively. Therefore, blue mussel could be useful as a bioindicator for PSP monitoring. With the increasing PSP levels in blue mussel in 2006, the proportion of PSP in its digestive gland increased to 95.1% when the maximum level was detected from the whole tissues of blue mussel on May 29. Subsequently, the PSP proportion in the digestive gland decreased as the PSP level in whole tissue decreased. The detoxification of PSP in blue mussel was greatest with relaying, followed by the water flow, and water circulation systems. Relaying decreased the PSP level below the regulatory limit of $80{\mu}g$/100 g after 2 days in low toxic sample with $124{\mu}g$/100 g, and after 7 days in high toxic sample with $401{\mu}g$/100 g. During depuration in the blue mussel with $401{\mu}g$/100 g via the water flow system, the PSP amounts in the digestive gland decreased by about 50% after 1 day, and about 77% after 7 days. In contrast, the PSP amounts in the soft body, gill, and mantle did not change significantly with depuration.

Simplified Procedure for Detection of Poliovirus and Norovirus in Oysters (굴로부터 오염된 폴리오바이러스 및 노로바이러스의 검출)

  • Simplified procedure was developed for concentrating and detecting poliovirus and norovirus in oysters. Viruses were seeded into oyster tissue homogenates and concentrated through polyethylene glycol (PEG) precipitation, chloroform or Freon extraction, with additional PEG precipitation. Amount of viruses was evaluated using poliovirus plaque assay. Virus recovery during concentration procedure was approximately 16.4-26.0%. For defection, viral RNAs in oysters were examined using one-step RT-PCR after extraction with Trizol. Dilution or capturing of viral RNA using silica gel membrane allowed viruses to be detected by RT-PCR, whereas viruses could not be removed using $QIAshredder^{TM}$ Homogenizer, which is effective in removing RT-PCR inhibitors in lettuce and hamburgers. Freon extraction, generally used to concentrate viruses found in food, could be substituted with chloroform extraction using this procedure; no difference could be observed between detection limits of whole oyster extracts and digestive organ extracts indicating that RT-PCR inhibitors were distributed evenly throughout whole tissues. Nested PCR greatly improved efficiency of this procedure. Overall, this procedure could remove sufficient amount of inhibitors to allow detection of norovirus in oysters.

Irradiation Preservation of Korean Shellfish (방사선(放射線) 조사(照射)에 의한 한국산(韓國産) 패류(貝類)의 품질보존(品質保存)에 관(關)한 연구(硏究))

  • Pacific oyster, hard clam and mussel were irradiated at doses up to 0.5 Mrad, the optimum dose rather than the maximum permissible was sought for in each species and post-irradiation storage characteristics studied at $0^{\circ}\;and\;5^{\circ}C$. No shellfish meat irradited at doses as high as 0.5 Mrad produced any adverse odor. However the organoleptic quality of each sample irradiated at lower doses was superior to those irradiated at the higher during the early storage period. The optimum dose was determined to be 0.2 Mrad for Pacific oyster and mussel and 0.1 Mrad for hard clam. By irradiating at the optimum dose, the storage life of Pacific oyster cculd be extend from less than 14 days to 35 days at $0^{\circ}C$ and from only 3 days to 21 days at $5^{\circ}C$. A similar storage extension was observed with mussel. The storage life of hard clam was extended from 7 days to 14 days at $0^{\circ}C$ and from 3 days to 12 days at $5^{\circ}C$. The hard clam meats were particularly susceptible to tissue softening by irradiation; an earlier onset and more extensive softening were observed with increasing dose.

  • Development of a rapid method possessing the requisite sensitivity and specificity for virus monitoring is necessary for protection of the shellfish-consuming public. Oysters tissue usually contains virus particles in relatively small concentrations along with various other substances that can interfere with detection steps. Therefore, the critical point concerning the detection of viruses in shellfish tissues resides in the processing of samples. The current study demonstrated the possibility of purifying small amounts of virus particles at the interface of a 10/50% sucrose gradient after a single round of sucrose gradient ultracentrifugation. We could detect HAV and poliovirus simultaneously from oyster tissues by using two different sets of primer. Furthermore, the method showed a high level of virus recovery rate (>95%) as determined by plaque assays of the final samples. Taken the advantages of the simple and sensitive methods, it was possible to detect 2 pfu of HAV in 5 g of oyster digestive tissues within 24h.

Data intercomparison and determination of toxic and trace elements in Algae using Instrumental Neutron Activation Analysis (중성자방사화분석에 의한 Algae중의 독성미량원소의 정량 및 실험실간 비교검증)

  • For the non-destructive multi-elemental analysis of environmental and biological materials, instrumental neutron activation analysis (INAA) was applied for the determination of toxic and trace elements in a set of three Algae samples provided by the International Atomic Energy Agency (IAEA). The analytical quality control was evaluated by comparing the analytical results of two standard reference materials of the National Institute of Standards and Technology (NIST); Oyster Tissue (SRM 1566a) and Citrus Leaves (SRM 1572). According to given analytical procedure, the concentration of 15-25 elements including spiked elements such as As, Cd, Cr and Hg in Algae samples were determined. To identify and validate these results, a data intercomparison program using more than 35 analytical methods in 150 laboratories was carried out and the estimated statistical data are summarized. Result of INAA is favorable, therefore, it is illustrated that can be applied for routine analysis of essential and toxic elements in algae samples as well as analytical quality assurance.

  • Levels of sterols including ${\Delta}^7$-dehydrogenase isolated from the tissues of marine animal products (20 species) were determined on 1.5% OV-17 columm of gas-liquid chromatography. The composition showed that the mussels and clams contained various sterols in their tissues : cholesterol, brassicasterol. 24-methylenecholesterol with some minor components such as 22-trans-norcholesta-5,22-dien-3${\beta}$-ol, 22-cis-dehydrocholesterol, 22-trans-dehydrocholesterol, desmosterol, 7-dehydrocholesterol, campesterol, stigmasterol, ${\beta}$-sitosterol, isofucosterol, and 7-dehydrocholesterol which could be converted into vitamin $D_3$ in the skin tissue of animal was present in the muscle of oyster, Crassostrea gigas. On the other hand, the others including gastropoda were predominantly composed of cholesterol. The minor sterols such as 24-methylenecholesterol, stigmasterol and ${\beta}$-sitosterol in the fish intestines are supposed to be derived from dietary plankton. Cholesterol ${\Delta}^7$-dehydrogenase which could convert cholesterol into ${\Delta}^7$-dehydrogenase was present in the pickles of Tricurus haumela intestine.

Analysis of polychlorinated biphenyls in shellfish collected from a market

  • We collected seven species of shellfish; all originating from the southern coastal areas of Korea, from a market every three months from Dec. 2001 to Sept. 2002, and determined the total polychlorinated biphenyl(PCB) levels by the sum of 26 individual congener levels. A GC-ECD system was applied for identification and quantification of these PCB congeners. Mussel showed the highest level in Sept. 2002 at 34.5 ng/g dry weight(d.w.). All species except mussel showed the lowest total PCB level in Dec. 2001 and their levels in tissue ranged from 0.6 to 5.5 ng/g d.w. The total PCB levels ranged from 0.8 to 17.3 ng/g in Mar. 2002, 2.2 to 9.5 ng/g in June 2002, and 1.8 to 34.5 ng/g d.w. in Sept. 2002. The principal congener group was penta-CBs, which accounted for 32% of the total PCBs, followed by hexa-CBs at 23%, and tetra-CBs at 21%.
