Anti-oxidative effects of exogenous ganglioside GD1a and GT1b on embryonic developmental competence in pigs

  • Gangliosides are glycolipids in which oligosaccharide is combined with sialic acids. Our previous studies have suggested an interplay between ganglioside GD1a/GT1b and meiotic maturation capacity in porcine oocyte maturation. Furthermore, ganglioside GD1a and GT1b are known for its antioxidant activity, but it is still unclear whether possible antioxidant role of GD1a and GT1b is involved in porcine embryos development competence during in vitro culture (IVC). Here, the effects of ganglioside GD1a and GT1b on the embryonic developmental competence during in vitro culture of porcine were investigated. The effects of ganglioside GD1a and GT1b on the expression of ST3GAL2 were confirmed during embryos development (2-cell, 4-cell, 8-cell and blastocyst) using immunofluorescent staining (IF). As a result, the fluorescent expression of ST3GAl2 was higher in embryos at 4-8 cells stage than blastocysts. Blastocyst development rate significantly increased in only 0.1 μM GD1a and GT1b treated groups compared with control group. To investigate the cellular apoptosis, we analyzed TUNEL assay. In case of only 0.1 μM GD1a and GT1b treated groups, the total number of cells in blastocyst compared with control group, but there was no significant difference in the rate of apoptotic cells. We identified the intracellular ROS levels using DCF-DA staining. According to the result, ROS production significantly decreased in blastocysts derived from the 0.1 μM GD1a and GT1b treated groups. These results suggest that ganglioside GD1a and GT1b improve the developmental competence of porcine embryos via reduction of intracellular ROS during preimplantation stage.

  • 본 연구는 Aliquat 336 상이동 촉매하에서 디페닐메탄을 산화시켜 벤조페논을 합성하는 반응의 메카니즘을 고찰한 것이다. 벤조페논의 생성속도는 Aliquat 336 촉매와 potassium tert-butoxide의 양이 많을수록 증가하였고, 반응의 부산물인 tert-butyl alcohol은 반응을 억제하였다 반응차수는 디페닐메탄과 산소의 농도가 낮을 때는 각각에 대하여 1차로 나타났고, 이들의 농도가 높을 때는 0차로 나타났다. 실험으로 측정한 벤조페논의 초기반응속도는 반응 메카니즘으로부터 유도한 다음의 반응 속도식과 비교적 잘 일치하였다. $$({\gamma}_{BP})_0={\frac{k_1k_3k_5[QCI]_0[DPM]_0[PTB]_0[O_2]_0}{k_2k_4[TBA]_0+k_2k_5[O_2]_0+k_3k_5[O_2]_0[DPM]_0}}$$

기체-액체 혼합 방전에 의한 화학적 활성종 생성 특성 (Generation of Chemically Active Species in Hybrid Gas-Liquid Discharges)

  • 고전압 방전극이 기체상에 위치하고 접지 전극이 수중에 설치된 기체-액체 혼합 방전에 의한 화학적 활성종의 발생 특성에 관해 실험실 규모 실험을 수행하였다. 실험된 전극 구조는 기존의 연구에서 사용해왔던 일반적 전극 배열에서보다 높은 전계 강도(electric field strength)를 형성하고 짧은 폭을 지닌 펄스들을 생성시킴으로써 방전에 의해서 일어나는 화학반응의 에너지 효율성을 높일 수 있는 것으로 나타났다. 방전에 의해 기체상에 생성되는 오존 농도는 실험된 전압 범위의 중간 값인 45 kV 조건에서 가장 높은 것으로 관찰되었다. 용액 전도도가 낮을수록 액체상을 통한 전기 저항이 증가하여 기체상에서 높은 전계 강도가 형성되므로 오존 생성을 촉진시키는 것으로 나타났다. 인가전압이 증가할수록 높은 전계 강도가 형성되어 강한 방전이 이루어지므로 과산화수소 생성속도가 증가하는 것으로 나타났다. 낮은 전압에서는 용액 전도도가 증가하면 과산화수소 분해속도가 증가하기 때문에 과산화수소 생성 속도가 감소하며 높은 전압에서는 용액 전도도가 증가하면 자외선 조사 등에 의해 과산화수소 발생의 중간 생성물인 OH 라디칼의 발생이 촉진되므로 과산화수소 생성 속도가 증가하는 것으로 나타났다. 산소와 아르곤의 혼합기체가 공급될 때, 강하고 안정한 방전이 이루어져 과산화수소 생성속도가 증가하는 것으로 나타났다.

Effects of Combination of Nitrate with ${\beta}$1-4 Galacto-oligosaccharides and Yeast (Candida kefyr) on Methane Emission from Sheep

  • The objective of the present study was to determine whether ${\beta}$1-4 galacto-oligosaccharides (GOS) and Candida kefyr combined with nitrate as manipulators could suppress rumen methanogenesis without nitrate poisoning in sheep. Four rumen fistulated wethers were allocated to a $4{\times}4$ Latin square design. Nitrate (1.3 g $NaNO_3$ $Kg^{-0.75}$body weight) with and without GOS and Candida kefyr were administered into the rumen through fistula as a single dose 30 min after the morning meal. GOS and Candida kefyr were supplemented by sprinkling onto the feed and through rumen fistula, respectively. The four treatments consisted of saline, nitrate, nitrate plus GOS and nitrate plus GOS plus Candida kefyr. Physiological saline was used as the control treatment. Compared to saline treatment, the administration of nitrate alone resulted in a very marked decrease in rumen methanogenesis and an increase in rumen and plasma nitrite production and blood methaemoglobin formation consequently causing a decline in oxygen consumption, carbon dioxide production and metabolic rate. When compared to nitrate alone, the simultaneous administration of nitrate with GOS decreased nitrite accumulation in rumen and plasma and nitrate-induced methaemoglobin, while retaining low methane production. However, GOS could not fully restore metabolic parameters reduced by nitrate. When compared to the simultaneous administration of nitrate with GOS, the simultaneous administration of nitrate with GOS plus Candida kefyr lowered rumen methanogenesis to a negligible level, but did not decrease rumen and plasma nitrite accumulation as well as blood methaemoglobin formation. Thus, these results suggest that combination of nitrate with GOS may be a potent manipulator to suppress rumen methanogenesis with abating the hazards of nitratenitrite toxicity in ruminants.

  • 환원.확산법에 의해 $Sm_{2}Fe_{17}N_{x}$ 계 희토류 영구자석을 제조하기 위한 기초연구로서, 우선 $Sm_{2}Fe_{17}$ 금속간화합물의 제조를 위하여, 금속 Ca에 의한 $Sm_2O_3$의 환원반응과 Fe분말중에 Sm의 확산반응을 검토하였다. 그 결과 전자는 $1000^{\circ}C$이상의 고온의 경우에 매우 빠르게 완료되지만, 후자의 Fe분말의 중심까지 Sm의 확산반응의 완료(완전한 균질화조건)는 $1100^{\circ}C$에서 3h 정도의 R-D 반응이 필요하며, 이 확산반응이 전체반응에 있어서 율속단계임을 알았다. Sm-Fe 계의 금속간화합물들의 성장은$ 1000^{\circ}C$이하에서는 $SmFe_2$, $SmFe_3$,$Sm_{2}Fe_{17}$금속간화합물의 3개의 상이 관찰되었으나, $1100^{\circ}C$에서는 $Sm_{2}Fe_{17}$ 금속간화합물의 상만이 관찰되었다. 본 연구에서 얻어진 최종시료의 산소 및 Ca량은 각각 0.72wt% 및 0.11wt%이었다.

미세조류 유래 astaxanthin의 항염증 및 항산화 효과 (Anti-Inflammatory and Antioxidant Effect of Astaxanthin Derived from Microalgae)

  • Astaxanthin (ATX)은 다양한 생명체에서 생성되는 카로티노이드 색소이다. 본 연구에서는 ATX가 RAW264.7 cell에서 LPS에 의한 inducible nitric oxide synthase (iNOS), nitric oxide (NO), 염증성 사이토카인, nuclear factor-kappa B (NF-${\kappa}B$)와 reactive oxygen species (ROS)의 생성을 억제 시키는 지 또한, superoxide radical 소거능이 있는 지를 조사하였다. iNOS와 NF-${\kappa}B$는 immunoblot analysis로, interleukin (IL)-6와 tumour necrosis factor-${\alpha}$ (TNF-${\alpha}$)는 ELISA 법으로 분석하였다. NO 양은 nitrite의 양을 측정하였고, ROS는 2',7'-dichlorodihydrofluorescin diacetate (DCFH-DA) 법으로 superoxide radical 소거능은 superoxide radical scavenging activity assay로 검증하였다. 100 ${\mu}M$의 ATX 농도에서 LPS로 유도된 NO, IL-6 및 TNF-${\alpha}$ 같은 염증성 사이토카인의 생성 뿐만 아니라 iNOS 및 NF-${\kappa}B$의 발현도 억제되었다. 특히, IL-6 및 TNF-${\alpha}$ 생성에 있어 ATX의 최대 억제율은 각각 65.2% 및 21.2% 이었으며 LPS로 유도된 NF-${\kappa}B$의 전사활성을 억제하였다. 이러한 현상은 세포질에서 핵으로 NF-${\kappa}B$의 전위를 억제하는 것과 관련이 있다. 또한, 25-100 ${\mu}M$의 ATX 농도에서 세포 내 ROS 생성을 억제하였으며, 5 mg/ml 농도의 ATX는 동일농도의 ${\alpha}$-tocopherol에 비해 superoxide radical 소거능이 1.33배 높았다. 이러한 결과들은 ATX가 대식세포에서 ROS 생성 및 NF-${\kappa}B$ 활성을 저해하므로 iNOS의 발현, NO 및 염증성 사이토카인의 생성을 억제하며, 또한 우수한 superoxide radical 소거능을 보유한다는 것을 나타내었다. 결론적으로, ATX가 항염증제 및 항산화제로서 유용하게 사용될 수 있을 것으로 사료된다.

색도물질과 옥살산의 오존분해를 위한 고효율 Jet Loop 반응기의 적용 (Application of High-performance Jet Loop Reactor for the Decolorization of Reactive black 5 and Mineralization of Oxalic Acid by Ozone)

  • As an ozone contactor, we newly adopted HJLR (High-performance Jet Loop Reactor) for the decolorization of Reactive black 5 and the mineralization of oxalic acid, which has been applied exclusively in biological wastewater treatments and well-known for high oxygen transfer characteristics. The ozonation efficiency for organic removals and ozone utilization depending on the mass transfer rate were compared to those of Stirred bubble column reactor, which was controlled by varing energy input in the HJLR and Stirred bubble column reactor. The results were as follows; first, the decolorization rate of Reactive black 5 in the HJLR reactor was nearly proportional to the increasing $k_La$. When the $k_La$ was increased by 25 % from $13.0hr^{-1}$ to $16.4hr^{-1}$, 30 % of the k' (apparent reaction rate constant) was increased from 0.1966 to $0.2665min^{-1}$ (Stirred bubble column; from 0.1790 to $0.2564min^{-1}$). Ozone transfer was found to be a rate-determining step in decolorizing Reactive black 5, which was supported by that no residual ozone was detected in all of the experiments. Second, the mineralization of oxalic acid was not always proportional to the increasing $k_La$ in the RJLR reactor. The rate-determining step for this reaction was OH(OH radical) production with ozone transfer, because residual ozone was always detected during the ozonation of oxalic acid in contrast with Reactive black 5. This result indicates that the increase of $k_La$ in the HJLR reactor is beneficial only when there are in ozone transfer limited regions. In addition, regardless of $k_La$, the mineralization of oxalic acid was nearly accomplished within 60 minutes. It was interpreted as that the longer staying of residual ozone by whirling liquid in the HJLR reactor contributed to an high ozone utilization(83-94%), producing more OR radicals.

Arthrospira platensis 추출물의 항산화 및 UVB에 의해 유도된 활성산소 생성에 미치는 영향 (Antioxidant Effect of Arthrospira platensis Extract and Effect on UVB-induced Free Radical Production)

  • 지구상에서 가장 오래된 해양 미세조류로 알려진 스피룰리나는 인체에 필요한 영양성분을 대부분 함유하고 있는 것으로 알려져 있다. 그 구성성분으로는 phycocyanin, chlorophyll, β-carotene 과 같은 다양한 물질을 다량 함유한다고 보고되고 있으며, 노화 및 미백효과가 있다고 알려져 있다. 본 연구에서는 스피룰리나 정제수 추출물의 UVB로 유도된 활성산소종 ROS (reactive oxygen species) 소거능과 항산화능을 확인하였다. 스피룰리나 정제수 추출물 0.05, 0.10, 0.50 1.0 mg/mL의 DPPH 라디칼 소거능, FRAP 환원능 및 ABTS+ 라디칼 소거능을 측정하여 항산화효과를 확인하였다. 대체실험동물모델인 Zebrafish를 이용하여 스피룰리나 정제수 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리하여 응고율, 부화율, 심장독성을 측정하였으며, 스피룰라나 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리한 후, DCFH-DA로 염색하여 UVB로 유도된 ROS 저해효과를 확인하였다. 항산화효과 측정 결과 양성대조군인 ascorbic acid와 비교시 DPPH, Frpa, ABTS 모두 농도 의존적으로 항산화 효과가 있음을 확인하였다. 대체 실험동물인 Zebrafish를 이용하여 응고율과 부화율, 심박수를 측정한 결과 0.5 mg/mL을 제외한 0.05, 0.10 mg/mL에서는 대조군과 비교 시 독성이 없음을 확인 하였다. UVB로 유도된 Zebrafish의 ROS 소거능은 양성대조군에 비해 높은 ROS 감소 효과를 나타내었다. 본 연구의 결과는 스피룰라나 정제수 추출물이 자외선 및 피부 보호 화장품 소재로 사용가치가 있음을 시사한다.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. 초단파조사 처리된 잉여슬러지와 하수의 병합처리가 활성슬러지공법의 성능에 미치는 영향 (Effects of the Co-treatment of Municipal Wastewater with Microwave-Irradiated Excess Sludge on the Performance of the Activated Sludge Process)

  • 본 연구는 초단파조사 처리된 잉여슬러지를 하수와 함께 활성슬러지공법으로 처리할 때 하수처리효율과 잉여슬러지 발생량에 미치는 영향을 파악하기 위하여 실시되었다. 온도가 $20^{\circ}C$이고 MLSS농도가 약 2,000 mg/L인 250 mL의 잉여슬러지를 발진주파수가 2,450 MHz이고 정격고주파출력이 700 W인 전자렌지(microwave oven)로 초단파조사 처리한 결과 조사시간이 1분 증가함에 따라 수온이 약 $20^{\circ}C$씩 증가하였으며, 슬러지의 SCOD, TKN 및 T-P 농도는 모두 $40\sim130$ sec의 조사시간에서 가장 많이 증가하였다. 그리고 혼합액 미생물의 산소섭취율을 측정한 결과 130 sec 이상의 조사시간($65^{\circ}C$ 이상)에서는 혼합액의 미생물이 거의 사멸하는 것으로 나타났다. 잉여슬러지를 초단파조사 처리한 다음 하수와 혼합시켜 활성슬러지공법으로 처리하였을 때 유입수 및 처리수의 pH 및 알칼리도에는 거의 영향이 없었으나, 유입수의 SS, COD, T-N 및 T-P 농도는 증가하였으며, 처리수의 SS, BOD 및 T-P 농도는 거의 영향을 받지 않았으나 COD 및 TKN 농도는 증가하였다. 그리고 초단파 조사비가 1 g SS/g SS-day 증가할수록 미생물성장계수 값은 0.91 g SS/g COD removed의 율로 감소하였다.
