Different Physiological Activity of Selected Rice Cultivars to Diphenylether Herbicide, Oxyfluorfen - III.   • This study was conducted to investigate the inhibition of protox activity and the PPIX accumulation of the oxyfluorfen-tolerant and-susceptible rice cultivars with barnyardgrass, a typical susceptible weed in accordance by oxyfluorfen treatment. The susceptible rice cultivars and barnyardgrass showed more inhibition of protox activity due to the treatment of oxyfluorfen than the tolerant rice cultivars. Especially in the concentration at $10^{-6}$M treatment, protox activity of the susceptible rice cultivars and barnyardgrass were the completely inhibited but the tolerant rice cultivars kept 32~59% of activity compared to the control. As the treatment concentration increased, the content of PPIX accumulation increased and it increased untill four hours of light exposure but it tended to decrease these after. The content of PPIX accumulation by the treatment of oxyfluorfen was more pronounced in the light condition than in the dark. Under the light and dark conditions, the susceptible rice cultivars and barnyardgrass showed more PPIX accumulation than the tolerant rice cultivate. Especialiy the susceptible barnyardgrass had more than the rice. With the treatment of GC and DA, tetrapyrrole biosynthesis inhibitor, the herbicidal activity by oxyfluorfen was inhibited, and the susceptible rice cultivars and barnyardgrass tended to have less effective than the tolerant rice cultivars and the content of chlorophyll or PPS accumulation tended to be similar.

Herbicidal Selective Activity of Oxyfluorfen to the Selected Rice Cultivars and Major Paddy Weed Species (벼품종(品種) 및 주요(主要) 논잡초종(雜草種)에 대한 Oxyfluorfen의 선택활성(選擇活性) 연구(硏究))

  • The experiment was carried out to find the feasibility of using Oxyfluorfen in the paddy fields by investigating the difference of selective activity of Oxyfluorfen among rice cultivars and major paddy weed species. The dosage of Oxyfluorfen that show selective activity between rice cultivars and weed species ranged from 0.1 to 0.4kg ai/ha. The degree of growth inhibition was in order of whole-plant soaking application > root soaking application > stem bandage application, and in that case $10^{-5}$M Oxyfluorfen was treated after emergence. Especially the growth inhibition of rice cultivars and Cyperus serotinus was low, among others. Photosynthesis was severely inhibited at the Oxyfluorfen level above $10^{-4}$ M in all the tested weeds, but inhibition of respiration was not to be seen. Isolated single cells of two rice cultivars and Cyperus serotinus were tolerant to $10^{-5}$M Oxyfluorfen,but those of Echinochloa crus-galli and Sagittaria pygmaea were susceptible comparatively. The growth inhibition of suspension cultured rice cell induced by the increments of Oxyfluorfen concentration, and the degree of inhibition was higher in C.V. Mushakdanti than in C.V. Aichiasahi.

Phytotoxic Response of Some Fruit Trees to Oxyfluorfen (Oxyfluorfen 처리(處理)에 따른 과수종(果樹種) 및 품종간(品種間)의 약해반응(藥害反應))

  • Spary and vapor drift injuries of apple, pear, and peach seedling caused by soil-applied oxyfluorfen were studied in a greenhouse. Bud bursting rate of all fruit trees was reduced by both spray and vapor drifts of oxyfluorfen, but reduction in bud bursting rate of pear and peach was greater than that of apple trees. Reduction in the number of leaves per shoot of apple and peach was greater than that of pear trees. Leaf injury of pear was most severe and occurred earliest, but leaves of peach were least injured, Leaf injuries of pear and apple were caused by both spray and vapor drifts, but leaves of peach was injured largely by vapor drift. Reduction in shoot growth of and pear was greater than that of peach trees. Shoot growth of pear was more rapidly retared compared with apple trees. In the field, oxyfluorfen delayed the time of bud bursting in young apple trees. Oxyfluorfen applied between initiation and completion of bud bursting delayed bud bursting more than earlier application although ultimate number of bursted buds was similar to control. The number of leaves per shoot and total length of shoots were lower than control until 40 days after application of oxyfluorfen, and then were similar to control because of vigorous growth after May.

  • The tolerant and susceptible rice cultivars to oxyfluorfen were selected from 400 rice cultivars in a laboratory, and tested in comparison with barnyardgrass, a typical oxyfluorfen susceptible weed. The responses to oxyfluorfen in the different levels of calli, cells and protoplasts of the rice cultivars were investigated. $I_{50}$ value of the tolerant rice cultivars was about $10^{-4}M$, whereas that of the susceptible rice cultivars and barnyardgrass was about $10^{-6}M$, showing significant difference between the two groups. The growth rate of calli segregated from the tolerant rice cultivars was higher than that from the susceptible rice cultivars and barnyardgrass by treatment of oxyfluorfen to the calli. The growth rate of suspension-cultured cells of the tolerant rice cultivars was higher than that of the susceptible rice cultivars and barnyardgrass after treatment of oxyfluorfen. The viability of protoplasts from the tolerant rice cultivars was higher than that from the susceptible rice cultivars and barnyardgrass after one or two hours of oxyfluorfen treatment. The intactness of protoplasts from the tolerant rice cultivars was also higher than that form the susceptible rice cultivars and barnyardgrass.

  • Seven oxyfluorfen formulations were tested for control of 8 weed species and rice injury under four different water depths with various ages of rice seedlings. Among formulations tested, rice injury was slight by formulations of Elvan, Coal Slag, Chitosan and Bentonite B under 0 cm or shallow water depths, and by those of Elvan and Coal Slag under deep water conditions. Weed control was high by Bentonite A and B, and Chitosan, and was low by Elvan. Coal Slag and Sand coated oxyfluorfen, if the target weeds of oxyfluorfen are annual species, further development of Elvan, coal slag, chitosan and Bentonite A would be controlled to increase control efficacy or to decrease rice injury.

  • Growth and anatomical responses of rice and barnyardgrass(Echinochloa crus-galli) to oxyfluorfen were examined under various growing conditions in a greenhouse. With foliar application at 0.08kg ai/ha 7 days after seeding or transplanting, oxyfluorfen completely reduced the shoot and root growth of barnyardgrass under all growing conditions. However, oxyfluorfen reduced the growth of direct seeded and transplanted rice by 0~64% and 0~12%, respectively. Phytotoxic effect on rice growth, with respect to plant height and shoot fresh weight, was more pronounced under water(53~63%) than under dry-condition(16~33%), and in broadcast-seeded(34~62%) than in drill-seeded rice(16~33%). Anatomical changes of epidermal and vascular bundle sheath cells of rice leaves were not induced by oxyfluorfen, but mesophyll cells were partially ruptured and total leaf thickness was reduced. Oxyfluorfen, however, constricted and ruptured mesophyll and vascular bundle sheath cells and severely reduced leaf thickness of barnyardgrass under all growing conditions examined. These anatomical changes were more severe under dry- than under water-seeded condition.

  • Responses of tobacco(Nicotiana tabacum cv. Xanthi) leaves of different age to diphenyl ether herbicide oxyfluorfen were evaluated with respect to cellular leakage, chlorophyll loss, and membrane lipid peroxidation. When tobacco leaves of different age were incubated under light condition at $25^{\circ}C$ following 12hr dark incubation. Significant electrolyte leakage from the treated tissues into the bathing medium occurred. The change of electrolyte leakage was proportional to the oxytluorfen concentration and the duration of light exposure to the tissues. Electrolyte leakage from the tissues treated with oxyfluorfen was highly dependent on the leaf age. From the tissues of younger age, more electrolyte leakage occurred and lag period was greatly reduced. Chlorophyll loss and membrane lipid peroxidation, as measured by malondialdehyde production, caused by oxyfluorfen treatment were also dependent an the age of treated leaf tissues. In conclusion, physiological responses of tobacco leaves to oxyfluorfen greatly varied with the age of treated tissues, and thus tobacco plants could be used as appropriate materials for studying the mechanisms of tolerance to diphenyl ether herbicides.

  • In this study, the relationships between sensitivity to oxyfluorfen, absorption of the herbicide, protoporphyrin IX(Proto IX) accumulation and activities of antioxidative enzymes were examined to identify the tolerance mechanism against oxyfluorfen in various rice cultivars having different level of tolerance to this herbicide. Absorption of oxyfluorfen in tolerant rice cultivars was slower than in susceptible cultivars. Proto IX accumulation in various rice cultivars treated with oxyfluorfen was higher in susceptible cultivars than in tolerant ones. In susceptible cultivars especially, Proto IX accumlated rapidly during the herbicide treatment in the dark. Large amounts of Proto IX accumulation were considered to cause membrane lipid peroxidation in the light. However, among the tested rice cultivars, there was little relationship between their tolerance to oxyfluorfen and the activities of antioxidative enzymes. Therefore, it is assumed that differential susceptibility of rice cultivars to oxyfluorfen was due to difference in their capability to absorb the herbicide and to subsequently accumulate Proto IX.

Weed Control and Safety of Transgenic Rice Event, CPPO06 in Direct-Seeding Flooded Rice Field (담수직파에서 형질전환 CPPO06이벤트 벼의 안전성 및 잡초방제효과)

  • This study was conducted to evaluate the efficacy of weed control and phytotoxicity of oxyfluorfen using a transgenic rice line (CPPO06 event) resistant to protoporphyrinogen oxidase (Protox) inhibiting herbicides in the direct-seeding flooded rice. Five annual weeds including Echinochloa oryzoides and two perennial weeds were occurred in the test field. Oxyfluorfen at 120 g a.i. $ha^{-1}$ in the application timing of two days before sowing and 0 and five days after sowing showed more than 90% weed control value except for Juglans mandshurica. Total weed control value was more than 95% in any application timing indicating a highly effective herbicide in the direct-seeding flooded rice. When compared with untreated control, no visual injuries were detected at single and double dosage of oxyfluorfen. The agronomic characteristics and yield components of CPPO06 event was reasonable in any time of application in this study. Based on these data, oxyfluorfen application before and after sowing can be applied to provide effective weed management in the direct-seeding flooded field.

Expression Site of Protoporphyrinogen Oxidase Influences on Herbicide Resistance in Transgenic Rice (형질전환 벼에서 Protoporphyrinogen Oxidase의 발현 위치가 제초제 저항성에 미치는 영향)

  • The effect of Protox expression site on herbicidal resistance was investigated in wild-type and transgenic rice plants imposed by peroxidizing herbicide oxyfluorfen. The transgenic rice systems involved the plastidal expression of Arabidopsis protoporphyrinogen oxidase (Protox; AP line) and the dual expression of Myxococcus xanthus Protox in chloroplasts and mitochondria (TTS line). The oxyfluorfen-treated TTS4 line showed the lower levels of cellular leakage and malonyldialdehyde and the sustained capacity of 5-aminolevulinic acid synthesis, compared to the oxyfluorfen-treated AP and wild-type lines. During oxyfluorfen action, the TTS4 line had greater herbicide resistance than the AP1 line, indicating that the dual expression of M. xanthus Protox in chloroplasts and mitochondria prevented the accumulation of photodynamic protoporphyrin IX more effectively than the expression of Arabidopsis Protox only in chloroplasts. These results suggest that the ectopic expression of Protox in mitochondria greatly contributes to the herbicidal resistance in rice plants.
