This study was conducted to examine the condition of in vitro culture system and the viability after embryo transfer of in vitro matured-in vitro fertilized (IVM-IVF) bovine embryos. The in vitro development to the blastocyst stage was enhanced by supplying bovine serum albumin(BSA) to co-culture medium with bovine oviduct epithelial tissue(BOET) compared with that in medium supplemented with fetal bovine serum(FBS) (41.2% vs. 26. 3%, P<0.05). After transfer of IVM-IVF blastocysts into the uterine horn of recipient females (Aberdeen Angus), one was pregnant to term and produced a head of male Korean native calf. These results confirm that the in vitro development of IVM-IVF bovine embryos is affected with different protein source in co-culture with BOET, and IVM-IVF embryos can develop to term after in vitro culture and embryo transfer.
Successful techniques of in vitro fertilization(IVF) are valuable for studying the process of fertilization and for developing economical procedures for gene and nuclear transfer in farm animals. To date, bovine IVF system has been developed with oocytes in vitro or vitro, but the resulting zygotes exhibit limited embryonic development after in vitro culture. Even though in vitro matured oocytes achieved high fertilization and cleavage rates, these embryos appear extremly low rate of pregnancies when transferred to synchronized recipients. Development of early bovine embryos in vitro is generally arrested at the 8-to 16-cell stage. However, recent use of somatic cells such as trophoblastic vesicle, granulosa and oviduct epithelial cell for co-culture with early bovine embryos has proven effective for development of embryos, matured and fertilized in vitro, past the in vitro cell blocks. These factors clearly indicate the value of the co-culture system in promoting development of bovine oocytes matured and fertilized in vitro to morula or blastocyst stage in vitro. In addition, co-culture system may beome a tool for evaluation of viability of ova that have been manipulated by procedures such as splitting, microinjection and nuclear transfer.
The imbalance between reactive oxygen species(ROS) production and total antioxidant capacity reproduction system is correlated with infertility. Therefore, this study was designed to investigate the expression patterns of peroxiredoxin(Prx), a member of antioxidant family, in reproductive system that includes testis, epididymis ovary, oviduct, and uterus. Follicular fluid influxed into the oviduct during ovulation may affect movement of sperm for fertilization Thus, in this study, the effect of follicular fluid, obtained from follicles of l0mm in diameter, on number and quality of sperm recovered by swim-up separation was investigated and sperm-movement stimulating components extracted from follicular fluid with methanol and isooctane were separated by gel filtration with Sepadex G-1O, G-25 and G-1OO gels, and were isolated by electrophoresis with SDS-PAGE mini gel. The results obtained were as follows; 1. Diluted follicular fluid stimulated sperm movement. 2. Sperm-movement stimulating factors were in methanol extract. 3. Sperm-movement stimulating effect of methanol extract appeared in fraction I among fractions recovered after gel filtration. And the fraction I contained proteins indicating 4 major bands as about 47, 43, 25 and 14 kilodaldons and 5 minor bands as about 67, 58, 23, 22 and 21 kilodaldons. 4. The fraction I recovered from G-100 gel showed significantly low percentage of motile sperm and had no protein indicating the band of 67 kilodaldons among the minor bands.
Eight species of bithyniids, Bithynia leachi, B. tentaculata, B. siamensis, B. misella, B. kiusiuensis, B. striatula, Bithynia sp. from Nepal and Gabbia australis were collected from 1957 to 1988. The samples were relaxed with pentobarbital and fixed with 70% ethyl alcohol. There was no significant difference in internal structure of soft body, except the male reproductive system. The alimentary canal of the bithyniids consisted of mouth, buccal mass, esophagus, stomach, intestine, rectum and anus. The heart consisted of an auricle and a ventricle, located at left side of the stomach in the visceral hump. The ctenidium comprised 40-70 subtriangular lamellae. Eight ganglia were located around upper esophagus. The female reproductive system comprised ovary, oviduct, spermaaheca, and albumin gland. The male reproductive system was composed of testis, seminal vesicle, prostate gland, vas deferens, penial duct, and penis in order. Male was distinguished externally from the female by a bifid penis.
Two experiments were conducted to study the production of in vitro fertilized bovine embryos and the viability of blastocysts cryopreserved by vitrification. In experiment 1, production rate of in vitro matured bovine oocytes after fertilization in medium containing bovine oviduct epithelial cells (BOEC), cumulus cells and granulosa cells to blastocysts were 18.4, 14.6 and 13.1%, respectively. Developmental percentages of blastocysts produced at day 6, 7 and 8 were 8.5, 10.6 and 15.2% respectively. Hatching rate of bovine embryos produced was 60.0%. In experiment 2, post-thawed surviving embryos in a vitrification solution consisting of 7.15M ethylene glycol, 2.5 mM ficoll and 0.3 M sucrose were 36.4% (56/154). Also, survival rate of bovine embryos after exposed to vitrification solution at 1, 2, 3, 4 and 5 min were 84.0, 88.0, 71.0, 48.0 and 24.0% respectively.
This study was conducted to examine the effect of culture supernatant of bovine oviductal epithelial cell(BOEC) on in vitro development of mouse embryos. To obtain the culture supernatant, ampullary epithelial cell, ithmic epithelial cell and ciliated eptithelial cell of bovine oviduct were cultured in Ham's F-10 su, pp.emented with 10% FCS. The development rates of mouse embryos to blastocyst stage were significantly(P<0.05) higher in BOEC-culture supernatant(72.3∼82.3%) than in Ham's F-10(50.7%). The proportions of embryonic development into hatched blastocysts were significantly(P<0.05) higher in ampullary cell supernatant(43.2%), ithmic cell supernatant(48.4%) and ciliated cell supernatant (27.7%) than in Ham's F-10(14.4%). On the other hand, the effect of ciliated cell supernatant was lower than those of other cell supernatants(P<0.05). And there was no difference between ampullary cell supernatant and ithmic cell supernatnat.
This study investigated full-term development potential of ultrarap idly frozen and thawed mouse 2-cell embryos. Mouse 2-cell embryos, dehydrated by exposure to freezing medium, were directly immersed into liquid nitrogen and thawed in 37$^{\circ}C$ water. The embryos that were frozen and thawed were cultured in uitro and transferred to foster mothers to examine there developmental potential. As a result, the frozen-thawed 2-cell embryos developed to blastocysts in vitro as a similar rate as control 2-cell embryos did(in vitro 2-cell, 86.4%; in vivo 2-cell, 90.9%; solution control, 89.9%; control, 89.7%). Normal live young were obtained from transfer of frozen-thawed embryos to the oviduct and uterus of pseudopregnant recipients (3l.4~56.7%).
This study was carried out to investigate the aggregation rate of isolated mouse 2-, 4- and 8-cell stage blastomeres in phytohemagglutinin(PHA) solution. The morphologically normal embryos were collected from the oviduct of superovulated female mouse by flushing with M2 and the zona pellucida of embryos were removed with 0.5% pronase. The blastomeres were isolated by pipetting after plunging into Ca++-Mg++free PBS for 20 min. The result showed that aggregation rate in 0.5% (84.9~93.1%) was higher than that in 1.0% PHA(76.0~82.1%). Optimal aggregation time was 60min (83.9~100.0%) when compared with 30min (78.8~87.5%). Developmental to blastocyst in recombinated blastomeres was higher under conditions of 0.5% PHA solution and 60-min aggregation than that under other conditions.
