  • We study hypersurfaces in the Euclidean space with the following property: the tangential part of the position vector has constant length. As a result, we prove that among the connected and complete hypersurfaces in the Euclidean space, only the hypersphere centered at the origin satisfies the property.

Genetic Analysis of Ancient Bones of Cervidae Animals from Archaeological Site in Jeju, Korea

  • DNA extracted from ancient bones of Cervidae animals was examined to identify the species and to determine the phylogenetic relationships to those from extant cervids. Abundant ancient bones were excavated from Kumsung archaeological site in Jeju Island, Korea, and were identified as Cervidae animals based on morphological features of their antlers and lower mandibles. Their mitochondrial DNA (mtDNA) control region (CR) was partially sequenced and subsequently compared with those previously reported in database. The results confirmed that the ancient sequences are lineage of Cervidae. On the phylogenetic trees constructed using the sequence diversity of the CR sequences of family Cervidae, the ancient DNA sequences were found on distinct clusters. The ancient sequences were located in the subfamily Capreolinae cluster, and six ancient sequences were closely related to those of extant Korean roe deer in Jeju Island and Korean Peninsula. Consequently, the results of this study suggest that the roe deer inhabited Jeju Island in ancient times. However, there is no evidence for the existence of subfamily Cervinae, including Sika deer, while it has been described in several historical records. The results suggest that this finding could contribute to understanding of the origin and phylogenetic relationships of extant and ancient roe deer on Jeju Island.

Effects on the Initiation of Simian Virus 40 DNA Replication by Antisense RNA

  • When DNA replication of simian virus 40 (SV40) is initiated on the replication origin, the regions containing the initiation sites of DNA primase, which participates in the transient RNA primer synthesis for formation of Okazaki fragments in the lagging strand, were chosen as the target sites of antisense RNA for studies of the inhibition of SV40 DNA replication. Four recombinant transcription vectors, pUC-PrI, pUC-PrII, pGEM-PrBS, and pGEM-PrSN, coding antisense RNA, were constructed. Four antisense RNAs (named as I, II, BS, and SN) having the size of 18, 19,58, and 123 nts, respectively, were made from the transcription vectors by in vitro transcription. And then, antisense RNA in the concentration of 2${\mu}m$ were added to COS cells transfected with pATSV-W which is a recombinant plasmid containing the SV40 origin of replication. The inhibitory extent of DNA replication was measured by DpnI resistance and was confirmed by measurement of transient RNA primer synthesis. The result shows that six combinations of antisense RNA (I, II, BS, SN, I+SN, and BS+SN) lead to the inhibition of SV40 DNA replication by up to 85%.

Quantitative Comparison of Ginsenosides and Nitrogen Compounds in Korean Ginsengs and Related Origin (고려인삼류와 유사생약중 ginsenoside 및 질소화합물의 함량 비교)

  • To differentiate the quality of Korean ginseng from those of other habitats, the quantitative analysis of free amino acids(FAA) and total amino acids(TAA) in addition to ginsenoside Rb1 and Rg1 was carried out using amino acid analyzer and HPLC, respectively. FAA pattern in Korean ginseng was much different from that of Panax notoginseng. The difference in total content of FAA was also found that Korean ginseng contained 26.3-39.8mg/g while Panax notoginseng contained 6.5mg/g. This FAA content had a tendency to increase with the age of radix. The contents of FAA and TAA in Korean ginseng(6 years old) from Kumsan were shown to be the highest than other ginseng origins tested. The content in the 6 years Panax ginseng from China was about same with that of 4 years Korean ginseng of Kumsan. However, regarding to gisenoside Rbl and Rg1, which have been accepted as the characteristic components of Panax ginseng-Panax notoginseng showed considerably higher content than those of any other ginseng origin.

Genetic Diversity of mtDNA D-loop Polymorphisms in Laotian Native Fowl Populations

  • Here, we studied the genetic diversity of native fowls in Laos by analyzing a mitochondrial DNA (mtDNA) sequence polymorphism. A 546-bp fragment of the mtDNA D-loop region was sequenced in 129 chickens from the areas of Vientiane, Luang Prabang and Pakse. In total, 29 haplotypes were identified and formed five clades. Haplotype diversity and nucleotide diversity of the native fowls in Laos were $0.85536{\pm}0.0172$ and $0.010158{\pm}0.005555$, respectively. Although the Laotian native fowls were distributed across five clades, most of them were clustered in two main clades (A and B), which were originated in China. The other haplotypes were contained in clades D, F, and I, which originated from continental southeast Asia. These results suggest that multiple maternal lineages were involved in the origin of domestic chicken in Laos. Moreover, there appear to be at least two maternal lineages, one from China and the other from the southeast Asian continent.

THE DISTRIBUTION CHARACTERISTICS OF TRACE ELEMENTS IN AIRBORNE PARTICULATES FROM AN URBAN INDUSTRIAL COMPLEX AREA OF KOREA USING INSTRUMENTAL NEUTRON ACTIVATION ANALYSIS

  • An instrumental neutron activation analysis was used to measure the concentrations of about 24 elements associated with airborne particulates (PM10) that were collected in the most polluted urban region of Daejeon city, Korea from 2000 to 2002. Using the measurement data for various elements, both the extent of elemental pollution in the study area and the seasonality in their distribution characteristics were examined. Examinations of their distribution patterns indicated that most elements with crustal origin tend to exhibit seasonal peaks during spring, while most elements with anthropogenic origin tend to exhibit seasonal peaks during fall or winter. In order to explain the factors regulating their mobilization properties, the data were processed by a factor analysis. Results of the factor analysis suggested competing roles of both industrial and natural source processes, despite that the study site is located at a downwind position of the industrial complex. Based on the overall results of this study, it is concluded that the site may be strongly impacted by man-made sources but the general patterns of elemental distributions in the study area inspected over a seasonal scale are quite consistent with those typically observed from natural environment.

Construction of a High-efficiency Shuttle Vector Containing the Minimal Replication Origin of Bacillus thuringiensis

  • In order to improve the transformation efficiency of the Bacillus thuringiensis (Bt)-Escherichia coli (E. coli) shuttle vector, pHT3101, we intended to minimize replication origin of Bt in pHT3101. For this, two modified shuttle vectors, pHT1K and pHT261, in which 2.9 kb of replication origin of Bt were shortened to 1 kb and 261 bp, respectively as previously reported. Whereas the pHT1K could efficiently transform Bt into the antibiotic resistant, no transformants were obtained with pHT261. Furthermore, pHT1K showed higher transformation efficiency compared to that of parent vector, pHT3101. Therefore, pHT1K might be a very useful Bt-E. coli shuttle vector carrying minimal replication origin of Bt.

  • Faint z ~ 5 quasars with M1450 ~ -23 mag are known to be the potentially important contributors to the ultraviolet ionizing background in the post-reionization era. However, their number density has not been well determined, making it difficult to assess their role in the early ionization of the intergalactic medium (IGM). In this work, we present the updated results of our z ~ 5 quasar survey using the Infrared Medium-deep Survey (IMS), a near-infrared imaging survey covering an area of 85 square degrees. From our spectroscopic observations with the Gemini Multi-Object Spectrograph (GMOS) on the Gemini-South 8 m Telescope, we discovered eight new quasars at z ~ 5 with -26.1 ≤ M1450 ≤ -23.3. Combining our IMS faint quasars with the brighter Sloan Digital Sky Survey (SDSS) quasars, we derive, for the first time, the z ~ 5 quasar luminosity function (QLF) without any fixed parameters down to the magnitude limit of M1450 = -23 mag. We find that the faint-end slope of the QLF is very flat (-1.2) with a characteristic luminosity of -25.7 mag. The number density of z ~ 5 quasars from the QLF gives lower ionizing emissivity and ionizing photon density than those in previous works. These results imply that quasars are responsible for only 10-20% of the photons required to completely ionize the IGM at z ~ 5, disfavoring the idea that quasars alone could have ionized the IGM at z ~ 5.

Comparative Analysis of ITS Sequences from Acer Species (Aceraceae) in Korea

  • Sequences from the internal transcribed spacer (ITS) regions of nuclear ribosomal DNA were determined to assess their potential as a phylogenetic tool for Korean Acer species, including A. okamotoannum and A. takesimense which are endemic on Ullung Island of volcanic origin. Although the genus Acer has been studied by various authors, different infrageneric dispositions have been suggested, and the phylogeny of the genus has been in dispute. The variation of ITS sequences from seven species of Acer was very low among species within the same section, but comparative analysesof the molecular data obtained suggest that ITS sequences may provide enough phylogenetic resolution for sectional relationships in the genus.

Mechanical, rheological, and durability analysis of self-consolidating concretes containing recycled aggregates

  • In the present paper, the effect of recycled aggregates on the rheological and mechanical properties of self-consolidating concrete is investigated experimentally and numerically. Hence, the specimen with two types of recycled aggregates, i.e., known and unknown resistance origins, are utilized for the studied specimens. The experiments in this study are designed using the Box-Behnken method, which is one of the response surface methods. Input variables in mixtures include silica fume in the range of 5-15% as a percentage substitute for cement weight and recycled coarse and fine aggregates in the range of 0-50% for both series of recycled materials as a substitute for natural materials. The studied responses are slump flow, V funnel, compressive strength, tensile strength, and durability. The results indicate that the increase in the amount of recycled aggregates reduces the rheological and mechanical properties of the mixtures, while silica fume effectively improves the mechanical properties. In addition, the results demonstrate that the fine recycled aggregates affect the total response of the concrete significantly. The results of tensile and compressive strengths indicate that the mixtures including 50% recycled materials with known resistance origin demonstrate better responses up to 8 and 10% compared to the materials with unknown resistance origins, respectively. Recycled materials with a specific resistance origin also show better results than recycled materials with an unknown resistance origin. Durability test results represent those concretes containing recycled coarse aggregates have lower strength compared to recycled fine aggregates. Also, a series of mathematical relationships for all the responses are presented using variance analysis to predict mixtures' rheological and mechanical properties.
