Study on the Relationship between Polymorphisms in Glutathione S-transferase and Ischemic Cerebrovascular Disease

  • Objective : Glutathione S-transferase polymorphism (GST) were examined in 120 cases with ischemic cerebrovascular disease (ICVD) to test the hyperthesis that GST polymorphisms confer a risk to an individual to develop ICVD. Tobacco smoking is a major cause of both cancer and vascular disease. Methods : therefore We were stratified the subjects with ICVD for smoking status, and then examined whether polymorphisms in this detoxification enzyme gene, GST, influence risk of ICVD Results : Neither GSTM1 nor GSTT1 genotypes in the ICVD group was significantly different from the control group (n=207), even in smokers. We attempted the combined analyses for GSTM1 and GSTT1 genotypes in ICVD for smoking status. No significant association observed between the combined genotypes and ICVD Conclusion : Our observation do not confirm the effect of the GSTM1 and GSTT1 genotypes as a risk factor for ICVD, even in smokers.

  • Six orient varieties of N.tabacum and 15 $F_1$ hybrids, not including reciprocal crosses, were evaluated for the degrees of heterosis and the combining abilities of 6 characteristics in Suweon, Korea, 1981. The values 9f general combining abilities tiers greater than those of specific combining abilities in all traits but significant SCA was also observed for Plant height, days to flowering and yield. In general, CCA effect was large in Samsun, Kavala and Izmir varieties. Large SCA values were observed for number of leaves in Samsun x Izmir, days to flowering in Samsun x Serves, leaf shape in Basma x Xantlli, and yield in Basma x Kavala and Xanthi x Izmir. Heterosis of favorable directions in $F_1$ was generally observed for all traits. Especially the $F_1$ of Samsun x Kavala flowered earlier than its midparents. Large heterosis effects were manifested for the number of leaves in Samsun x Izmir and yield in Samsun x Izmir as well as Xanthix Izmir.

The Analysis of Neutral Volatile Flavor Compounds in Tobacco (잎담배 중 neutral volatile flavor 화합물 분석)

  • This work has been conducted to develop a method for the analysis of neutral volatile flavors and their precursors in tobacco. The neutral volatile compounds and precursors in tobaccos have been investigated by Neutral Volatile scan method(NV scan) using Soxhlet extractor. The method has been used to analyze a range of different tobaccos and tobacco products. Neutral flavor compounds were classified as three sections(1st Volatile Fraction, Breakdown Flavor Products and Cembranoid Precursors). The major components of the First Volatile Fraction were 2-cyclohexene-1-one, 6-methyl-5-hepten-2-one, limonene and phenyl ethanol. The major components of Breakdown Flavor Products were isophorone, solanone, damascenone, 3-hydroxy-$\beta$-damascone, geranyl acetone, $\beta$-ionone, dihydroactinidiolide, norsolanadione, neophytadiene, hexahydrofarnesylacetone, farnesyl acetone and megastigmatrienone. The major cembranoid precursor compounds were dibutyl phthalate, duvatrenediols, 8,12-epoxy-14-labden-13-ol, 11-hydroperoxy-2,7,12(20)-cembratriene-4,6-diol, 12,15-epoxy-12,14-labadien-8-ol, 2,7,11-cembratrien-4,6-diol and 8,13-epoxy-14-labdien-12-ol. The NV scna results of tobacco types(flue-cured, burley and oriental) showed that each tobacco type has a characteristic flavor component profile.

  • Less-volatile organic components in leaves from aromatic tobaccos of different varieties, both Oriental and Korean types were isolated and concentrated using a simple apparatus with fewer manipulations. Each less-volatile concentrate was then subjected to spectrophotometric recording in the visible range, to thin-layer chromatographic group separation, and high-performance liquid chromatographic profile analysis. The methods allow detection of significant quantitative differences in visible absorption spectra, TLC patterns, and high resolution HPLC profiles among varieties.

  • Essential oil components were isolated both from Oriental and- Korean using a modified distillation/solvent extraction method. The essential oils were then analyzed by high-resolution glass capillary gas chromatography, to give characteristic GC profile from each tobacco. Varietal differences were detected from the quantitative comparison of the GC data.

INFLUENCE OF CURING TEMPERATURE ON CHARACTERISTICS OF ORIENTAL-TYPE TOBACCO (향끽미종 연초건조중 온도차가 건조엽의 특성에 미치는 영향)

  • During curing of aromatic tobacco leaves, coloring temperatures were set at 26, 30, 34, 38 and $42^{\circ}C$, and also leaf drying temperatures were established at 35, 40, 45, 50, 55 and $60^{\circ}C$, respectively, to investigate the thermal effect on characteristics of cured leaves. Appearance of yellow color from green was accelerated, the contents of non-volatile organic and higher fatty acids in leaves tended to increase with the higher yellowing temperature. Quality by price per kg was best at 45-$50^{\circ}C$ and score of smoking test was highest at 40-$50^{\circ}C$ during leaf drying stage. The concentrations of non-volatile organic acids and volatile essential oil inclined to decrease, but a reverse in this trend with higher fatty acids as temperature increased during leaf drying stage. The difference of other components was discussed.

Optimization of Headspace Sampling Using Solid Phase Microextraction For Volatile Organic Acids in Different Tobacco Types

  • A Solid-phase micro extraction(SPME) was evaluated as a tool for headspace sampling of tobacco samples. Several experimental parameters (sampling temperature, pH, and type of SPME fibers) were optimized to improve sampling efficiency in two aspects ; maximum adsorption and selective adsorption of volatile organic acids onto SPME fibers. Among four types of SPME fibers such as PDMS(Polydimethylsiloxane), PA(Polyacrylate), Car/PDMS (Carboxen/Polydimethylsiioxane) and PDMS/DVB(Polydimethylsiioxane/Divinylbenzene) which were investigated to determine the selectivity and adsorption efficiency. A variety of tobacco samples such as flue cured, burley and oriental were used in this study. The effect of these parameters was often dominated by the physical and chemical nature (volatility, polarity) of target compounds. This method allowed us to make important improvements in selectivity and sensitivity. The Car/PDMS fiber was shown to be the most efficient at extracting the 10 selected volatile organic acids. The parameters were optimized: $80^{\circ}C$ adsorption temperature, 30 min of adsorption time, $240^{\circ}C$ desorption temperature, 1 min of adsorption time.

  • Methoxylated flavonoids (sinenstin, nobiletin, tangeretin) were isolated and identified by preparative HPLC, mass and NMR. Chung-pi(I) and Chung-pi(II) contained methoxylated flavonoids most richly. Methoxylated flavonoids in Chung-pi obtained from Korean Milgam increased with fruit maturity, while contents of the methoxylated flavonoids gradually decreased after mid of October.
