  • A survey of the free amino acids and organic acids in the shoot of Phyllostachys edulis was made by means of amino acid autoanalyzer and gas chromatograph. The results of the survey are summerized as follows. 1. Eighteen amino acids found in bamboo shoot were lysine, histidine, arginine, tryptophan, aspartic acid, glutamic acid, threonine, serine, proline, glycine, alanine, valine, leucine, isoleucine, methionine, tyrosine and phenylalanine, and an unknown was found. Serine showed the highest amount and more than about 44% of total free amino acids. 2. Oxalic acid was the major organic acid, and formic acid, acetic acid, maleic acid, succinic acid, fumaric acid, citric acid, tartaric acid and sorbic acid were determined, and two unknown were found.

Effects of Some Organic Acids on Shelf Life and Textural Properties of Cooked Noodle (유기산이 숙면의 저장성 및 물성에 미치는 영향)

  • The effects of some organic acids on the shelf life of cooked noodle and the change in textural properties were studied. Organic acids used were vinegar, lactic, citric and malic acid. The pH of noodle was adjusted to $4.7{\pm}0.25$ by dipping it in each solution for 30 seconds. Total microbial count and turbidity of the treated samples were measured storage for 4 days at $15^{\circ}C$. The total count was high in order of control, malic, citric, lactic acid and vinegar. High turbidity was observed in order of control, lactic, malic, citric acid and vinegar. Hardness, adhesiveness, cohesivenes and springiness were measured for 30 days at $35^{\circ}C$ using Rheometer along with concurrent sensory evaluation. The acid treated samples showed higher values in hardness and cohesiveness than control but lower in adhesiveness and springiness. After 30 days storage, the malic or citric acid treated sample led to a somewhat higher gumminess than control. Based on the sensory evaluation the malic acid treated noodle significantly exhibited the highest score followed by citric, lactic acid, control and vinegar.

  • In decontamination process to remove radioactive materials of reactor cooling system, the metal ions dissolved by organic acids in decontamination solution are separated by use of ion exchange resin in the column. However, organic acids in decontamination solution decrease the apparent affinity of the resin to metal ions. In light of this, some experiments were carried out on the Amberlite IRN-77 cation resin with cobalt and iron to gain a better understanding of the complexation effects on the ion exchange process. Experimental results showed that EDTA among organic acids used as chemical decontaminants predominantly caused reduction of ion exchange capacity of cobaltous ion to resin since this reagent formed the complex with the cobaltous ion stronger than that with the ferrous ion. In contrast, the effects of oxalic acid and citric acid were found to be negligible. And, single and two-component nonlinear equilibrium relationships of the metal ions were established using experimental data.

  • The focus of this study was the influence of organic acids such as acetic, citric and malic acid on the dissolution of calcium (Ca), magnesium (Mg), and phosphorus (P) on chicken thigh bone. As the concentration (0, 0.5, 1, 2, 4%) of acetic, citric, and malic acid increased, the resultant contents of calcium, magnesium, and phosphorus were higher than that of the control. When the boiling time (2, 4, 6, 8, 12 hours) was increased, dissolved amounts of several minerals from the chicken thigh bone increased. Calcium dissolved the most when chicken stock was boiled for 12 hours with 4% of malic acid added. In addition to minerals, amino acids and proteins were further extracted by adding organic acids. The soup stock which contains minerals such as calcium, can be obtained by boiling the chicken thigh bone for 12 hours with an organic acid.

Antibacterial activity of supernatant obtained from Weissella koreensis and Lactobacillus sakei on the growth of pathogenic bacteria

  • This study was carried out to obtain basic data for the industrial use of Weissella koreensis and Lactobacillus sakei. The antibacterial activity of supernatants obtained from W. koreensis and L. sakei were tested against pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, Salmonella typhimurium KCCM 40253, and Salmonella typhimurium KCCM 15. The supernatant of L. sakei showed antibacterial activity against E. coli KCCM 11234, S. enteritidis KCCM 12021, and S. typhimurium KCCM 15, while the supernatant of W. koreensis showed antibacterial activity against E. coli KCCM 11234 and S. enteritidis KCCM 12021. The effect of pH changes and heat treatment on antibacterial activity of the supernatants was examined using the sensitive pathogenic bacteria (E. coli KCCM 11234, S. enteritidis KCCM 12021 and S. typhimurium KCCM 15). Antibacterial activity against sensitive pathogenic bacteria was maintained under heat treatment at all temperatures, but there was no antibacterial activity associated with pH modification. Furthermore, it was confirmed that the antibacterial activity of the supernatants obtained from W. koreensis and L. sakei was a result of organic acids including, lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acids. Therefore, the present study showed that the organic acids produced by L. sakei and W. koreensis exhibited a strong antibacterial activity against pathogenic bacteria. Moreover, in the food industry, these organic acids have the potential to inhibit the growth of pathogenic bacteria and improve the quality of stored food.

Changes of Organic Acids and Volatile Flavor Compounds in Kimchis Fermented with Different Ingredients (재료의 종류에 따른 김치의 유기산 및 휘발성 향미 성분의 변화)

  • The changes of the content of organic acids, carbon dioxide, alcohols and carbonyl compounds of the various Kimchis which were made of cabbage with green onion, garlic, ginger or red pepper and fermented at $12^{\circ}C-16^{\circ}C$ were investigated. Nonvolatile organic acids identified were lactic, succinic, fumaric and malic acid. Volatile organic acids identified were acetic, formic, propionic, butyric, valeric, n-caproic and n-heptanoic acid. Carbonyl compounds identified were acetaldehyde and acetone. The content of lactic acid was increased with fermentation, and higher in Kimchis containing red pepper, garlic and green onion. The content of acetic acid was increased with fermentation, especially in Kimchi containing garlic. The content of carbon dioxide was higher in Kimchi containing garlic. Alcohols identified in all Kimchis was only ethanol. Carbonyl compounds had no direct effect on off-flavor of Kimchi.

Effects of organic acids on prevention against S. Typhimurium in weaning pigs (이유자돈에서 유기산제의 S. Typhimurium에 대한 효과)

  • Salmonella (S.) Typhimurium infection in pigs is a major source of human foodborne salmonellosis. Recently, the swine industry in Korea has been suffered from salmonellosis causing severe economic loss to farms. Organic acids have antibacterial activity which prevents bacteria from multiplying by reducing the pH in the gastrointestinal tract. The aim of the present study is to evaluate whether $Salstop^{(R)}$ mixed with organic acids is able to have influence on growth performance and whether it prevents against S. Typhimurium in weaning pigs. Four experimental treatments were examined: general diet added with 0.3% of $Salstop^{(R)}$ after S. Typhimurium ($1.0{\times}10^{10}CFU/ml$) challenge, group A; commercial feed after Salmonella ($1.0{\times}10^{10}CFU/ml$) challenge, group B; commercial feed and $Salstop^{(R)}$, group C; commercial feed, Group D. At the end of the study, no significant differences in daily body weight gain and feed intake were observed between groups treated with $Salstop^{(R)}$ and groups treated without, whereas feed conversion ratio (FCR) tended to be improved in groups treated with $Salstop^{(R)}$ between days 1 to 14. Serological and microbiological evolution of the infection were examined by ELISA and microbiological culture from serums and fecal samples, respectively. In groups that challenged by S. Typhimuriums, the group without $Salstop^{(R)}$ showed higher prevalence and bacterial shedding compared to the groups treated with $Salstop^{(R)}$. Our results suggest that the administration of $Salstop^{(R)}$ could be used to promote feed efficiency and to reduce the prevalence of salmonellosis in weaners.

Antimicrobial Effects of Organic Acids and Ethanol on Several Foodborne Microorganisms (식중독 미생물에 대한 유기산 및 에탄올의 항균활성 비교연구)

  • The antimicrobial effects of ethanol and organic acids(acetic, citric, lactic. propionic, tartaric acid), either alone or in combination against four foodborne microorganisms (Listeria monocytogenes, Salmonella typhimurium, Staphylococcus aureus and Escherichia coli O157:H7) in tryptic soy broth were determined. Area under the growth curve, minimum generation time, maximum growth rate, and detection time were measured by using automated turbidometer Bioscreen(Labsystem, Finland), for 24 hr at $30^{\circ}C$. All microorganisms were not grown at 7% ethanol in the media. The 0.1% propionic acid showed the strongest inhibitory effects against S. aureus, L. monocytogenes and E. coli O157 : H7 compared with other organic acids, whereas 0.01% organic acids did not show significant inhibitory effect against microorganisms tested (p > 0.01) except S. aureus. The combination of 1% ethanol and 0.01% organic acids were significantly more effective than alone on growth of S. aureus and L. monocytogenes(p < 0.01).

Changes in Free Sugars and Organic Acids of Banana Fruit at Various Storage Temperatures (저장온도에 따른 바나나의 유리당과 유기산)

  • Banana purchased from local market was stored at 5, 15 and $25^{\circ}C$ for 10 days after which free sugars and organic acids were determined. The amount of glucose, fructose and sucrose was $0.30{\sim}0.36%$ at the initial time of storage. The changes of free sugar was influenced by storage temperature. At $5^{\circ}C$, little change in the content of each free sugar was measured and sucrose had a slightly higher value. Glucose and fructose showed a predominant increase while sucrose content decreased at 15 and $25^{\circ}C$. The values of total sugar, reducing sugar, glucose, fructose and free sugar contents at $25^{\circ}C$ were higher than at any other storage temperature. Citrate, malate, lactate and acetate in Banana were identified as organic acids. The total amount of organic acid decreased from $305{\sim}373.3$ mg% at the initial time to $45.9{\sim}67.9$ mg% after 9 days of storage. The reduction of organic acids contents was relatively high at $25^{\circ}C$.

Characteristics of Fluorescent Organic Matter and Amino Acids Composition in the East Sea (동해의 용존유기물 형광특성 및 아미노산 조성에 관한 연구)

  • Fluorescence characteristic and amino acids composition of organic matter were determined from extracted seawater samples at eight stations in the East Sea of Korea. Organic compounds have been extracted onto C-18 Sep-Pak cartridges. Three dimensional excitation/emission fluorescence contouring of extracts showed two markedly distinct characterized fluoroscopies representing protein-like biomacromolecule and humic-like geomacromolecule. Protein-like biomacromolecule showing fluorescence maxima at 280 nm/330 nm (excitation/emission) were abundant in the surface mixed layer and then apparently decreased below the thermocline at most stations. It suggests that source of biomacromolecule is comely related with vigorous biological synthetic activity in the surface layer and bacteria decompose its biologically labile components near the thermocline and in the deeper layer. On the other hand, humiliate geomacromolecule showing fluorescence maxima at 330 nm/430 nm (excitation/emission) were low in the surface mixed layer implying photochemical oxidation and then increased below the thermocline at most stations. It suggests that geomacromolecule might be transformed by condensation of bio-refractoryorganic fraction after decomposition of biomacromolecule and particulate organic carbon derived from the surface mixed layer. HPLC measurements of amino acids showed similar composition between seawater and extracted organic macromolecule after hydrolysis. Glycine, serine and alanine were predominant, accounting for more than 50% of total amino acids. Dissolved free amino acids of seawater were more abundant in the surface layer(0.7∼1.8 uM) than the deeper layer (0.2∼0.4 uM). D/L racemic ratio of alanine of extracted organic matter showed lower value in the surface layer than the deeper layer. It suggests that biomacromolecule predominant in the surface layer is relatively young, rapidly recycling and biologically labile.
