  • Bacteriocin-producing lactic acid bacteria, Lactococcus sp. HY 449, was isolated from dairy products. Using response surface methodology, the various concentrations of medium compo- nents (tryptone, glucose, yeast extract, tween 80, and initial pH) were tested to find the optimum conditions for maximum bacteriocin production and growth of Lactococcus sp. HY 449. Central composite design was used to control the concentrations of medium components in the experiment. Bacteriocin production and cell growth of Lactococcus sp. HY 449 were most affected by glucose and yeast extract. Estimated optimum growth conditions of Lactococcus sp. HY 449 were as follows; tryptone 1.08%, glucose 1.129%, yeast extract 0.674%, tween 80 0.11%, and initial pH 7.19. Also estimated optimum conditions for bacteriocin production were tryptone 0.937%, glucose 1.108%, yeast extract 0.163%, tween 80 0.09%, and initial pH 6.98.

Optimum Condition of Marine Actinomycetes, Streptomyces sp. NS 13239 for Growth and Producing Antibiotics

  • In previous study, marine actinomycetes producing the antibiotics were investigated to invent new antibiotics from east coast of Korea. The optimum growth conditions of Streptomyces sp. NS 13239 were $28^{\circ}C$, pH 7.0 and $3\%$ of NaCl concentration in various media. Streptomyces sp. NS 13239 showed strong antimicrobial activity against gram-positive bacteria, specially Methicillin resistant Staphylococcus aureus (MRSA), but just weak antimicrobial activity against yeasts and mold. On the other hand, it did not show antimicrobial activity against gram-negative bacteria. The optimum conditions for producing antibiotics were almost consistent with optimum growth conditions except carbon source and nitrogen source.

Lead Tolerance Profile of Pseudomonas Stuzeri in Liquid Culture

  • Pseudomonas stutzeri strain KCCM 34719 was used in this experiment to determine the effects of increasing Pb(II) concentrations on its growth rate. To obtain optimum growth conditions, strain KCCM 34719 was cultivated in nutrient broth under various conditions, such as temperature, pH, and NaCl concentration. Optimal conditions for cell growth were $30^{\circ}C$ of temperature, 8.0 of pH, and 3% of NaCl concentration, respectively. Growth response of bacterial cell to Pb(II) showed tolerance to concentrations ranging from 10 to 100 mg ${\ell}^{-1}$ in liquid culture, following a growth pattern similar to the control. Growth rate was greatly inhibited at 200 mg ${\ell}^{-1}$ of Pb(II).

  • This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.

  • A morphologically different form of Ganoderma lucidum was isolated from a cultivator's farm, and its optimum growth conditions were determined. A major difference in their morphology was color of fruit bodies. Fruit bodies of the mutant were white wherase those of normal Ganoderma lucidum were red. Spores of the mutant were global and mycelia were thin. Mycelial growth of this white mutant was favorable on potato sucrose agar medium, and optimum pH of the medium was 5.5.

  • For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.

  • The effect of fermentation parameters and medium composition on the simultaneous mycelial growth and exo-polymer production from submerged cultures of Ganoderma applanatum was investigated in shake-flask cultures. The optimum initial pH for mycelial growth and exo-polymer production was 5.0 and 6.0, respectively. The optimum temperature was $25^{\circ}C$ and the optimum inoculum content was 3.0% (v/v). The optimal carbon and nitrogen sources were glucose and corn steep powder, respectively. After 12 days fermentation under these conditions, the highest mycelial growth was 18.0 g/l and the highest exo-polymer production was 3.9 g/l.

  • The cultural conditions of the osmophilic red color yeasts (Strain L$_{1}$, L$_{2}$, L$_{3}$ and L$_{4}$) isolated and identified in the previous report were examined and the results obtained were as follows ; 1. The optimum medium for growth of these osmophilic red color yeasts was soy sauce medium. 2. These strains were grown exceedingly well on the medium containing 3 percent of NaCl but somewhat restrained on the medium containing 6% or more. 3. The optimum temperautre for growth of these strains was 25.deg.C and their lethal temperature was 68.deg.C(treatment for 5 minutes). 4. The optimum pH for growth of these strains was 6.0.

  • The media compositions and culture conditions were optimized for mycelial growth of Coriolus versicolor and Lentinus edodes. Media composition for optimal growth of Coriolus versicolor was 2.0% glucose 0.4% peptone and 0.6% yeast extract. Media composition for optimal growth of Lenttnus edodes was 2.0% glucose 2.0% starch 0.4% bacto-soytone and 0.6% yeast extract. The media supplemented with KH2PO4, 0.046% KH2PO4 0.1% and MgSO4, .7H2O 0.05% supported better mycelial growth than the media without mineral salts. The optimum temperature for mycelial growth ranged from $25^{\circ}C$-28$^{\circ}C$. The optimum pH range for mycelial growth of Coriolus versicolor was 5.2~5.6 while that of Lentinus edodes appeared to be 5.75.

  • Seasonal changes of symbiotic nitrogen-fixation activity and growth characteristics of four different natural populations of kummerowia striata were quantitatively analyzed during the growing period. the nitrogen-fixation activity of root nodules attained the maximum rates of 148, 132, 102 and 100$\muM\;C_2H_4\;\cdot\;g\;fw\;nodule^{-1}\;\cdot\;hr^{-1}$, respectively for sunny, multibranched, shade and unibranched populations at the optimum growth conditions. and the seasonal changes showed fluctuations by environmental conditions such as light, temperature, nutrient contents, water stress and plant ages, etc. The multibranched plant showed the greater amount of leaf and root nodule biomass, and the higher nitrogenase activity than the unibranched plant. the optimum conditions of leaf chloropht11 and water content of each organ indicated the active growth and the maximum fresh biomass of 4 different populations were 1.92, 1.85, 0.97 and 0.56 g $fw\cdotplant^{-1}$ for shade multibranched, sunny and unibranched populations, respectively.
