  • This study was conducted to establish mass propagation system from the axillary bud culture of chrysanthemum zawadskii H. which was used as material of medicinal plants. Shoot egeneration was better on MS medium with NAA and BA. The optimum concentraions of growth regulator for shoot regeneration differed depending on accessionsof C. Zawadskii. Shoot regeneration in Keungucheolcho was better on MS Medium with NAA 0.01mg/1 and BA 0.1mg/1 while Hyangrobonggucheocho was better with NAA 0.1mg/1and BA 0.3mg/1. Addition of NAA into medium was effective for induction of root from shoots regenerated. Shoot multiplcation was more effective when 10mg/1 spermine was added into medium than when other polyamines were treated ino medium . Randomly and specifically amplified polymorphic DAC banding patterns based on polymerase chain reaction (PCR) analysis were used to assess the genetic variation of plants regenerated from in vitro culture.

Production of Protease Inhibitor from Streptomyces sp. SK-862 (방선균이 생성하는 단백질 가수분해효소 저해물질의 생산)

  • A inhibitor acting on substrate proteolytic enzyme was isolated from culture broth of Streptomyces sp. SK-862, which had been isolated from soil in Wonju City, by using the colloidal agar medium. The optimum culture temperature and initial pH for the production of the protease inhibitor was 28$^{\circ}C$ and pH 8.5, respectively. The optimum culture medium was composed of 1.5% glucose, 0.5% peptone, 0.1% K2PHO4, 0.05% CaCO3 and initial pH 8.5. The inhibitor production was maximum when the strain was incubated in shaking incubator at 70 strokes for 60 hours.

$\alpha$-Glucosidase Inhibition by Culture Broth of Streptomyces sp. NS15 (Streptomyces sp. NS15 배양액에 의한 $\alpha$-Glucosidase 저해)

  • For the production of nonprotein $\alpha$-glucosidase inhibitor from the Streptomyces sp. NS15 strain, effects of initial optimum pH, nitrogen sources, carbon sources, cationic metal ions, agitation speed and aeration rate were investigated. Initial optimum pH of medium was 7.0. The most effective nitrogen and carbon sources were soybean meal 2.0%(w/v) and glucose 1.6%(w/v), respectively. The cationic metal ins had no stimulating effect on inhibitory activity of $\alpha$-glucosidase except Fe2+. Agitation speed and aeration rate were effective at 400rpm and 1vvm, respectively. In the jar-fermenter cultivation for 4 days under optimal culture conditions, the culture broth showed the inhibitory acitivity of 3,200units/ml, which is 25 times higher than that of basic medium (CYM) for porcine intestinal $\alpha$-glucosidase. The inhibitory activity of $\alpha$-glucosidase reached about 3,200units/ml after 4 days of cultivation and decreased gradually for a further two days.

  • The optimum conditions for the submerged mycelial culture of Lentinus edodes SR-1 were elucidated to be incubation temperature of 25C, initial pH 4.0, agitation of 300 rpm, inoculation of 10.0%(v/v), and aeration of 1.0 v/v/m in TGY medium. The optimum c/n ratio and economic yield coeffcient for the submerged mycelial culture were 13.1:1 and 0.45 respectively. As the plant growth hormones test, SCM medium containing 0.5ppm of 2,4-dicholorophenoxyacetic acid increased mycelial yield in 1.1%, but 6-benzylaminopurine was not effective.

Producton of Yeast Spores from Rice Wine Cake. (주박으로부터 효모포자의 생산)

  • Rice wine cake (RWC) is the solid waste obtained after rice wine fermentation. For the mass production of the spores of yeast Saccharomyces from RWC, the optimum pretreatment condition of RWC, the optimum composition of culture medium, and the optimum culture condition were examined. For sporulation, yeast cells were grown in the pre sporulation medium (PSM), transferred into sporulation medium (SM) containing 1 % potassium acetate, and incubated in a rotary shaking incubator at $25^{\circ}C$ for 4 days. The supernatant of the mixture of RWC and water was used as the presporulation medium (PSM). The optimum temperature and time for the pre-incubation of the mixture of RWC and water (1:2) to obtain maximum sporulation yield were $V^{\circ}C$ and 24 hr, respectively, and optimum culture time in PSM was 48 hr. Using these optimum conditions, the asci number obtained was 0.72$ 1.06${\times}$10^{8}$$m\ell$. The addition of wheat coat koji into SM increased the final number of asci to beTEX>$10^{8}$ $m\ell$. Spores were formed in the SM with the initial pH of 7-11, but no spores were formed in the SM with the initial pH of 5. To save the time and effort to pretreat the RWC, 2% and 0.5% RWC without any pretreatment were directly added into PSM containing 1 % brown sugar and SM, respectively, and the maximum asci number of $1.27${\times}$10^{8}$ /$m\ell$ was obtained.

Yeast Cell Cultivation of Produce Active Dry Yeast with Improved Viability (생존능이 증진된 활성 건조효모 생산을 위한 효모세포배양)

  • Optimum conditions for vacuum-drying ad cultivation of yeast cells for the production of active dry yeast were examined. At lower temperature, more drying time was required to dry the yeast pellet to reach the desirable water content(8%). Optimum temperature of vaccum oven and time for drying was 63$^{\circ}C$ and 90 min, respectively. Optimum medium composition for flask culture using cane molasses as the substrate were 0.25% sugar, 0.013% $K_2$HPO$_4$, 0.1% $K_2$HPO$_4$. and 0.125% (NH$_4$)$_2$SO$_4$. Culture temperature $25^{\circ}C$ gave the highest survival rate of dired yeast. After finishing fed-batch culture and the culture was left in the fermentor without adding any sugar or nutrient, survival of the dried yeast harvested from the fermentor increased to 86.0% after 36 hr. It was also observed that the yeast cells with higher budding rates showed lower survival rate.

Development of a New Synthetic Medium Composition for the Submerged Culture of Phellinus linteus (Phellinus linteus의 액체배양을 위한 새로운 합성배지의 개발)

  • A new synthetic medium was developed for the submerged mycelial cultures of Phellinus linteus. The medium for maximum mycelial growth of Phellinus linteus (3 days incubation, 28$^{\circ}C$, pH 5) consisted of (per 1 L): glucose, 90 g peptone, 10 g soluble starch, 10 g yeast extract, 3 g KH2PO4, 1 g MgSO4.7H2O, 1 g and CaCl2, 0.1 g. The concentrations of glucose, peptone, yeast extract, KH2PO4, MgSO4.7H2O, and CaCl2 were examined in the ranges of 10~90 g/L, 0~10 g/L, 0~15 g/L, 0~2 g/L, 0~1 g/L, and 0~0.5 g/L, respectively. The dry weight of mycelium in 3 days increased to 16.79 mg/mL using the new synthetic medium. The optimum temperature for mycelial growth of Phellinus linteus was 28$^{\circ}C$. The concentrations of KH2OP4, CaCl2, and yeast extract, which gave the maximum mycelial growth of Phellinus linteus, existed in the concentration ranges examined in this study. But, in the cases of other compositions (MgSO4.7H2O, peptone, and glucose), the mycelial growth of Phellinus linteus increased with the concentration in the ranges.

  • This study was carried out to obtain the basic data on artificial culture of Hohenbuehelia petaloides. The optimum medium are glucose peptone medium (GP), Hennerberg medium, Phellinus igniarius medium (PIM), Lentinus edodes medium (LEM), Czapek dox medium. The optimum condition for the mycelial growth was $30^{\circ}C$ and pH 6.0. The carbon sources such as dextrine, fructose and lactose were favorable to mycelial growth. The optimal concentrations of carbon sources are 10% dextrin and fructose. As nitrogen sources, tryptone, casamino acid and histidine appeared to be favorable. The optimal concentrations of nitrogen sources are 1% soy tone and 0.3% ammonium nitrate. The optimal concentration of yeast extract is 0.4%. The mineral nutrients of $KH_2PO_4$, $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were effective and the optimal concentrations were 0.046, 0.1 and 0.05%, respectively.

Fungi-rice bran based Fermentation of Coptis Chinensis and Curcuma Longa Root and its Influence of Silk Dyeing

  • This study examined the dye-properties of natural fabrics dyed with Coptis chinensis and Curcuma longa root fermented with fungi. The optimum culture conditions for the fermentation of microorganisms, the relationship between natural dye color and fermentation conditions were investigated. Two different medical herbs (ground to 80-100 mesh in size) were used as a natural dyeing source. Phellinus linteus (P. linteus), which can grow in different media, such as Agarmedium (only agar containing medium), maltose extract agar (MA) and potato dextrose extract agar (PDA) culture media, were isolated from the medium. P. linteus was confirmed to be the optimum microorganism for the fermentation of Coptis chinensis and Curcuma longa, and the MA medium was confirmed to be the best for culturing. When using the microorganism as the fermenting agent, $32^{\circ}C$ was found to be the optimum fermenting temperature for both natural colorants. Regarding the dyeing property of the fermented natural dye, silk was dyed quite darkly in an appearance by naked eye estimation and the K/S value in the color strength of silk reached a high level of 16 after the fermenting process. The washing fastness of dyed silk after treatment washing was reduced from 4 to under4 and indicates that dyed silk with fermented plant was not unsubstantial. The light fastness was 1 to 2, showing intended to maintain due to the fermentation process.

Condition of Exo-polysacchride Production from Submerged Mycelial Culture of Ganoderma lucidum by Using Air-lift Fermenter System (Air-lift Fermenter System을 이용한 Ganoderma lucidum 균사체의 심부배양에 의한 세포외 다당류의 생산 조건)

  • For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.
