Glycosyltransferases (GTs) from microbes are an emerging and rich source for efficient glycol-transformation of natural/unnatural compounds. Here, we probed the catalytic capability and substrate promiscuity of BmmGT1 from marine-derived Bacillus methylotrophicus B-9987. The regioselectivity of BmmGT1 on macrolactin A (1) was explored by optimization of the reaction conditions, in which a series of O-glycosylated macrolactins (1a-1e) were generated, including two new di/tri-O-glucosyl analogs (1b and 1e). Furthermore, BmmGT1 was able to catalyze the glycosylation of the thiol (S-) or amine (N-) sites of phenolic compounds (2 and 3), leading to the generation of N- (2a) or S-glycosides (3a and 3b). The present study demonstrates that BmmGT1 could serve as a potential enzyme tool for O-, N-, or S-glycosyl structural diversification of compounds for drug discovery.
In a previous study, the sequential optimization and regulation of environmental parameters using the PhotoBiobox were demonstrated with high-throughput screening tests. In this study, we estimated changes in the biovolume-based composition of a polyculture built in vitro and composed of three algal strains: Chlorella sp., Scenedesmus sp., and Parachlorella sp. We performed this work using the PhotoBiobox under different temperatures (10-36℃) and light intensities (50-700 μmol m-2 s-1) in air and in 5% CO2. In 5% CO2, Chlorella sp. exhibited better adaptation to high temperatures than in air conditions. Pearson's correlation analysis showed that the composition of Parachlorella sp. was highly related to temperature whereas Chlorella sp. and Scenedesmus sp. showed negative correlations in both air and 5% CO2. Furthermore, light intensity slightly affected the composition of Scenedesmus sp., whereas no significant effect was observed in other species. Based on these results, it is speculated that temperature is an important factor in influencing changes in algal polyculture community structure (PCS). These results further confirm that the PhotoBiobox is a convenient and available tool for performance of lab-scale experiments on PCS changes. The application of the PhotoBiobox in PCS studies will provide new insight into polyculture-based ecology.
Dietary fibers (DF) have been used as functional food components due to the various health promoting activities. Dietary fibers have been separated from the peels of Korean tangerine by employing ultrafiltration (UF) membranes. Optimum conditions in a batch type ultrafiltration unit using YM100 (molecular weight cut-off, MWCO=100,000), YM 10 (MWCO=10,000) and YM1 (MWCO=1,000) membranes were : transmembrane pressure 7.5 psi, temperature of the peel extracts $35^{\circ}C$, and pH of the peel extract 3.0, respectively. The flux in YM 10 membrane unit was higher than that in YM 10 or YM 1 membrane unit. However, YM 100 membrane was superior to YM 10 or YM 1 membrane with respect to the recovery of the retentate and the contents of DF The contents of DF in the tangerine peel extract, in the 170 mesh retentate, and in the YM 100 retentate were shown to be 33.4%, 18.5% and 8.4% based on dry matter, respectively. Most dietary fibers were recovered at the separation stages of 170 mesh and YM 100.
The purpose of this study is to obtain basic data on the optimization of firing process conditions for enamel coating in chemical heat exchanger. The method of increasing the firing temperature in order to apply enamel coating to shell & tube type heat exchanger was examined. The temperature distribution of the heat exchanger in the firing kiln was numerically calculated using a commercial CFD program. The structural safety of the heat exchanger was confirmed by thermal stress analysis using the FSI method. Numerical analysis and experimental results show that there is a problem of safety due to temperature difference when the heat exchanger at room temperature is directly put into a firing kiln at 860℃. Therefore, a preheating process is need to reduce the temperature difference. As in Case2 with fewer firing steps, the first stage preheating temperature of 445℃and the second stage firing temperature of 860 ℃are considered to be most suitable.

In this paper, we studied some design methodologies of typical FIR filters based on the peak-error constrained least square criterion which was first introducedd by Adams in 1991. This method is a mixed type of the classical least squared error method(LSM) and the so-called min-max error method (MMM). And by considering both the least squared error as well as the maximum error, the solution, i.e. the impulse response of the filter, can be found only when the restrictions on maximum gain, transition bandwidth, and the squared error are satisfied simultaneously under some trade-off conditions. We used the multiple exchange algorithms for optimization procedure and applied the design methodology to the cases of the multiband filter, the differentiator, and the Hilbert transformer by taking the balance of two design criteria into account. The results show that the peak-error constrained least weighted square error design method(PLEM) is superior in performance to the existing LSM and MMM from both the squared error and the maximum error standpoints. And it is verified that PLEM can be applied to not only the case of simple low pass filter, but also to various types of FIR filters.

A novel method was developed for the speciation of chromium in natural water samples based on homogeneous liquid-liquid extraction and determination by flame atomic absorption spectrometry (FAAS). In this method, Cr(III) reacts with a new Schiff's base ligand to form the hydrophobic complex, which is subsequently entrapped in the sediment phase, whereas Cr(VI) remained in aqueous phase. The Cr(VI) assay is based on its reduction to Cr(III) by the addition of sodium sulfite to the sample solution. Thus, separation of Cr(III) and Cr(VI) could be realized. Homogeneous liquid-liquid extraction based on the pH-independent phase-separation process was investigated using a ternary solvent system (water-tetrabutylammonium ion ($TBA^+$)-chloroform) for the preconcentration of chromium. The phase separation phenomenon occurred by an ion-pair formation of TBA and perchlorate ion. Then sedimented phase was separated using a $100\;{\mu}L$ micro-syringe and diluted to 1.0 mL with ethanol. The sample was introduced into the flame by conventional aspiration. After the optimization of complexation and extraction conditions such as pH = 9.5, [ligand] = $1.0{\times}10^{-4}\;M$, [$TBA^+$] = $2.0{\times}10^{-2}\;M$, [$CHCl_3$] = $100.0\;{\mu}L$ and [$ClO_4$] = $2.0{\times}10{-2}\;M$, a preconcentration factor (Va/Vs) of 100 was obtained for only 10 mL of the sample. The relative standard deviation was 2.8% (n = 10). The limit of detection was sufficiently low and lie at ppb level. The proposed method was applied for the extraction and determination of chromium in natural water samples with satisfactory results.
The experimental design and response surface methodology (RSM) have been applied to the investigation of the electro-UV complex process for the disinfection of E. coli in the water. The disinfection reactions of electro-UV process were mathematically described as a function of parameters power ($X_1$), NaCl dosage ($X_2$), initial pH ($X_3$) and disinfection time ($X_4$) being modeled by use of the Box-Behnken technique. The application of RSM using the Box-Behnken technique yielded the following regression equation, which is an empirical relationship between the residual E. coli number and test variables in actual variables: Ln (CFU) = 23.57 - 0.87 power - 1.87 NaCl dosage - 2.13 pH - 2.84 time - 0.09 power time - 0.07 NaCl dosage pH + 0.14 pH time + 0.03 $power^2$ + 0.47 NaCl $dosage^2$ + 0.20 $pH^2$+ 0.33 $time^2$. The model predictions agreed well with the experimentally observed result ($R^2$ = 0.9987). Graphical response surface and contour plots were used to locate the optimum point. The estimated ridge of maximum response and optimal conditions for the E. coli disinfection using canonical analysis was Ln 1.06 CFU (power, 15.40 W; NaCl dosage, 1.95 g/L, pH, 5.94 and time, 4.67 min). To confirm this optimum condition, the obtained number of the residual E. coli after three additional experiments were Ln 1.05, 1.10 and Ln 1.12. These values were within range of 0.62 (95% PI low)~1.50 (95% PI high), which indicated that conforming the reproducibility of the model.
The successful replacement of missing teeth has been one driving aim behind the emergence of implant dentistry as both a technology and clinical vocation for over four decades. To date, a multitude of dental implant devices had been designed and utilized in the patient population. Most of these devices have been designed without support of the engineering criteria. The long-term success of any dental implant is dependent upon the optimization of stresses which occurs during oral function and parafunction. Although many studies have examined the biologic interactions between dental implants and living tissue, few studies have been reported on the biomechanical aspects of dental implants. The purpose of this study was to analyze the stress distribution of osseointegrated prosthesis on certain conditions, such as amount of load, location of load, length of fixtures, number of fixtures used, arch shape, bone quality, etc. Three dimentional finite element analysis was used for this study. FEM models were created using commercial software(Super SAP. for IBM 16 bit AT computer. All elements were 8-node brick, isoparametric. Mandible and prosthesis was modeled with 780 elements and 1074 nodes. The results were as follows : 1. In case of cantilever extension, there was a compressive stress at the base of the first implant and a tensile stress at the base of the second implant. 2. The stresses were linearly proportional to the amount of load. 3. The stresses were linearly proportional to the length of cantilever. 4. There was a stress concentration at the neck of the implant and bone under horizontal loads.
We have been making dual dome enclosures which are useful to track artificial space objects at SSNT (Space Science and Technology Lab.) Kyung Hee University. We verified the safety of the dome enclosures using basic design and structure analyses before manufacturing them, and then performed an optimization analysis for economic and safe systems. The dome enclosure has a fully-open type structure to smoothly operate a telescope made in the style of altazimuth mount with very fast tracking. It is also designed to be safe against extreme weather conditions. The general structure of the observatory system consists of the dual dome enclosures at the top of a container. For the structural analyses, we consider the following two methods: (1) gravitational sustain analysis - how the structure supporting the dome withstand the weight of the dome, and (2) wind load analysis that considers the effect of the wind velocity at the region where the observatory is located. The result of overall deformation is found to be less than 0.551mm and the result of equivalent stress is found to be 20.293Mpa, indicating that the dual dome system is reasonably designed. This means structurally to be safe.

In this paper, we presents robust digital redesign (DR) method for observer-based linear time-invariant (LTI) system. The term of DR involves converting an analog controller into an equivalent digital one by considering two condition: state-matching and stability. The design problems viewed as a convex optimization problem that we minimize the error of the norm bounds between interpolated linear operators to be matched. Also, by using the bilinear and inverse bilinear approximation method, we analyzed the uncertain parts of given observer-based system more precisely, When a sampling period is sufficiently small, the conversion of a analog structured uncertain system to an equivalent discrete-time system have proper reason. Sufficiently conditions for the state-matching of the digitally controlled system are formulated in terms of linear matrix inequalities (LMIs).
