Improving the Rapidity and Accuracy of Carbapenem-Resistant Enterobacteriaceae Detection by Shortening the Enrichment Duration

  • Identifying carbapenem-resistant Enterobacteriaceae (CRE) is necessary to prevent nosocomial CRE infection outbreaks. Here, a rapid identification method with reduced enrichment time was developed without compromising accuracy. A total of 49 rectal swabs requested for CRE screening at the Department of Diagnostic Medicine at Hospital B in Busan, Korea, were included in this study. Specimens were inoculated on MacConkey and CHROMID Carba media either directly or following enrichment for 3, 6, and 24 h in 100 μl trypticase soy broth containing an ertapenem disk. The enriched cultures were further inoculated on CHROMID Carba or MacConkey media containing an ertapenem disk. In total, 19 CRE and 5 carbapenem-intermediate Enterobacteriaceae isolates were obtained from the 49 swabs. Among the 19 CRE isolates, carbapenemase-producing Enterobacteriaceae constituted 13 strains. Moreover, of the 19 CRE isolates, 16 (81.25%) and 17 (88.24%) were identified from the direct cultures on MacConkey and CHROMID Carba media, respectively. After 3 h of enrichment, the proportions of the CRE identified in the media were: MacConkey medium, 16/19 (81.25%); CHROMID Carba medium, 17/19 (88.24%); and MacConkey medium containing an ertapenem disk, 17/19 (88.24%). The detection rates after 6 h of enrichment were the same for all three media (19/19 strains, 100%), whereas those after 24 h of enrichment were 21, 22, and 24 strains, respectively, but included false positives. These findings suggest that a 6-h enrichment before inoculation on the CHROMID Carba medium is optimal for the rapid and accurate detection of CRE in clinical samples.

  • L.S. Pontryagin's maximum principle is applied to the minimum critical mass problem without any restriction on the ranges of uranium enrichment. For the analysis, two group diffusion equation is adopted for a cylindrical reactor neglecting the vertical axis consideration. The result shows that the three-zoned reactor turns out to be most optimal: the inner and outer zones with the minimum enrichment ; whereas the middle 3one with the maximum enrichment. With the given three-zoned reactor, critical condition is derived, which leads to the calculation of the determinant. By finding the roots of the determinant the numerical calculation of the minimum critical mass is carried out for the case of Kori reactor geometry changing the minimum or the maximum enrichment. It is found from many computed values that the least possible critical mass turns out to be the case of 1.2% maximum enrichment for the middle zone and 0.65% minimum enrichment for the inner and out zones.

  • Reducing carbon dioxide ($CO_2$) exhaust has become a major issue for society in the last few years, especially since the initial release of the Kyoto Protocol in 1997 that strictly limited the emissions of greenhouse gas for each country. One of the primary sectors affecting the levels of atmospheric greenhouse gases is agriculture where $CO_2$ is not only consumed by plants but also produced from various types of soil and agricultural ecosystems including greenhouses. In greenhouse cultivation, $CO_2$ concentration plays an essential role in the photosynthesis process of crops. Optimum control of greenhouse $CO_2$ enrichment based on accurate monitoring of the added $CO_2$ can improve profitability through efficient crop production and reduce environmental impact, compared to traditional management practices. In this study, a sensor-based control system that could estimate the required $CO_2$ concentration considering emission from soil for cucumber greenhouses was developed and evaluated. The relative profitability index (RPI) was defined by the ratio of growth rate to supplied $CO_2$. RPI for a greenhouse controlled at lower set point of $CO_2$ concentration (500 ${\mu}mol{\cdot}mol^{-1}$) was greater than that of greenhouse at higher set point (800 ${\mu}mol{\cdot}mol^{-1}$). Evaluation tests to optimize $CO_2$ enrichment concluded that the developed control system would be applicable not only to minimize over-exhaust of $CO_2$ but also to maintain the crop profitability.

The Optimal Enrichment Condition of Rotifer Brachionus rotundiformis (소화효소 활성으로 본 rotifer Brachionus rotundiformis의 적정 영양강화 조건)

  • The purpose of the study was to suggest the optimal lipid enrichment conditions used digestive enzyme activity of rotifer changing due to water temperature and salinity. The high population growth appeared at the experiment temperature more than 28 degrees highly on the culture temperature(maximum 32 degrees, 1,453 individual/mL). The fecundity was low at high temperature, and the egg ratio was high at low temperature. Population growth of 10 and 15 ppt appeared in most highly, but the fecundity and the egg ratio were high most significantly appeared in natural seawater(32 psu). The digestive enzyme activity by the culture environment mainly showed high activity in natural seawater(amylase exclusion, 15 psu). However, the TAP activity by the water temperature showed highly at the more high temperature, but the amylase and the lipase appeared at low temperature. We carried out the lipid enrichment at 20 degrees and 26 degrees in a condition of the natural seawater. Total protein, the total essential amino acids differed not significantly. The methionine content that was essential amino acids, a total lipid content, unsaturated index of fatty acids, DHA and the DHA/EPA ratio were high significantly each in $20^{\circ}C$ enrichment trial. Therefore, we could suggest the $20^{\circ}C$ and natural seawater for the optimal lipid enrichment condition in aquaculture, because methionine contents, several indexes by the lipid, TG-lipase activity, fecundity and egg ratio are high.

Optimal Enrichment Temperature for Rotifer and Microalgae Selection for Cold-Water Species Culture (한해성 품종 종묘생산을 위한 로티퍼의 적정 영양강화 수온 및 미세조류 선택)

  • The purpose of this study is to establish methods for enhancing the survival and growth of cold-water fish and crustacean larvae based on the nutritional components of zoo and phyto live foods. Rotifers, Brachionus rotundiformis, were cultured with a supplement of freshwater condensed Chlorella vulgaris at $28^{\circ}C$ and enriched with Algamac $2000^{(R)}$ at 16, 20 and $26^{\circ}C$, respectively. Isochrysis galbana, Chaetoceros calcitrans and Chlorella ellipsoidea were centrifuged for component analysis after being cultured for approximately one week with conway medium at $20^{\circ}C$. The crude protein and lipid contents of the rotifers were 58.4% and 10.9%, respectively, before enrichment. After enrichment at each temperature, total protein and essential amino acid contents were increased by reducing the enrichment temperature. However, unsaturated fatty acids and multiple fatty acid index (UI) showed their highest values at $20^{\circ}C$. Mono-unsaturated fatty acid content was highest (72.6%) at $16^{\circ}C$. The total protein contents of C. calcitrans and C. ellipsoidea were higher, 33.0% and 35.2%, respectively, than that of I. galbana, 27.8%. Methionin, leusine and histidine, essential amino acids of C. ellipsoidea, had considerably higher values, 50.2, 287.2 and 68.1 mg/g dry matter, respectively, compared to other microalgae. Total lipids, UI, DHA and n-3 PUFA of I. galbana had higher values, 23.6, 272.0, 12.9% and 45.2%, respectively, than other microalgae. Therefore, for cold-water fish and crustacean larvae that require high n-3 PUFA and DHA contents, enrichment of rotifers is desirable at $20^{\circ}C$. Fish larvae would also need more I. galbana than other microalgae.

Comparison of Common Enrichment Methods for Recovery of Yersinia Enterocolitica from Artificially Inoculated Swine Feed Samples

  • Five different enrichment methods were studied to find an optimal method to recover Yersinia enterocolitica from swine feed samples. When the recovery of Y. enterocolitica GER-C (serotype O:3) strain was studied at 1000 CFU/g feed, phosphate-buffered saline (PBS) enrichment at $4^{\circ}C$ and PBS plus sorbitol and bile salts (PSB) enrichment at $4^{\circ}C$ and $21^{\circ}C$ were not effective (< 22%). In contrast, both irgasan-ticarcillin-potassium chlorate (ITC) and tryptic soy broth plus polymyxin B sulfate and novobiocin (TSBPN) enrichment methods showed a full recovery (100%) at 100-1000 CFU/g feed. At 10 CFU/g feed, both ITC and TSBPN methods still recovered the strain (> 50%). In recovery of ATCC 9610 (serotype O:8) strain, TSBPN method was more sensitive than any other methods (P < 0.05) at 1000 CFU/g feed. Using TSBPN method, the strain was still recovered at 100 CFU/g feed, but not at 10 CFU/g feed. With its sensitivity and relatively simple recipe, TSBPN was most desirable method to recover Y. enterocolitica from swine feed samples.

Production of Cloned Pigs Derived from Double Gene Knockout Cells Using CRISPR/Cas9 System and MACS-based Enrichment System

  • Pigs are considered as optimal donor animal for the successful xenotransplantation. To increase the possibility of clinical application, genetic modification to increase compatibility with human is an important and essential process. Genetic modification technique has been developed and improved to produce genetically modified pigs rapidly. CRISPR/Cas9 system is widely used in various fields including the production of transgenic animals and also can be enable multiple gene modifications. In this study, we developed new gene targeting vector and enrichment system for the rapid and efficient selection of genetically modified cells. We conducted co-transfection with two targeting vectors for simultaneous inactivation of two genes and enrichment of the genetically modified cells using MACS. After this efficient enrichment, genotypic analysis of each colony showed that colonies which have genetic modifications on both genes were confirmed with high efficiency. Somatic cell nuclear transfer was conducted with established donor cells and genetically modified pigs were successfully produced. Genotypic and phenotypic analysis of generated pigs showed identical genotypes with donor cells and no surface expression of ${\alpha}$-Gal and HD antigens. Furthermore, functional analysis using pooled human serum revealed dramatically reduction of human natural antibody (IgG and IgM) binding level and natural antibody-mediated cytotoxicity. In conclusion, the constructed vector and enrichment system using MACS used in this study is efficient and useful to generate genetically modified donor cells with multiple genetic alterations and lead to an efficient production of genetically modified pigs.

Fungal bioconversion of Korean food wastes for the production of animal feed additive enzymes

  • Korean food waste, one of the abundantly available but environmentally problematic organic wastes in Korea, was utilized as solid-substrate by fungal strain Aspergillus niger ATcC 6275 for the production of enzymemixture containing amylase, cellulase and xylanase. The enzyme mixture can be used as high value-added animal feed. Solid-state fermentation method yielded a 84-fold enhancement in xylanase activity compared with submerged fermentation method. The effect of incubation period, incubation temperature, pH of medium, moisture content, inoculum size and enrichment of the medium with nitrogen and carbon sources were observed for optimal production of these enzymes The optimal amylase activity of 33.10 U/g, cellulase activity of 24.41 U/g, xylanase activity of 328.84 U/g were obtained at 8 days incubation with 50%(w/w) soy bean flake, with incubation temperature of $25^{\circ}C$, pH of 6.38, optimal moisture content of 55% and with inoculum size of $3.8{\times}10^6$spore/g. Enzyme activities were enhanced when ImM $CaSO_4$, 2% Malt extract and 2% galactose were added as mineral, nitrogen and carbon enrichment respectively.

Food value or Freshwater Rotifer (Brachionus calyciflorus) for Culture of Sweetfish (Plecoglossus altivelis) Larvae (은어 자어 (Plecoglossus altivelis) 사육에 있어서 담수산 rotifer (Brachionus calyciflorus)의 먹이효과)

  • This study investigated the possibility of salinity acclimation of freshwater rotifers (Brachionus calyciflorus) as live food for sweetfish (Plecoglossus altivelis) larvae, and also examined the optimal salinity for the growth of sweetfish. Freshwater rotifers cultured in 0 and 4 PSU and seawater rotifers (B. rotundiformis) cultured in 33 PSU were supplied to the larvae with four kinds of enrichment material (condensed freshwater Chlorella, $\omega-yeast,$ baker's yeast, Super Selco) and larval growth at 4 PSU was examined. Growth of the freshwater rotifers positively increased from 0 PSU to 6 PSU, but decreased when over 8 PSU was reached. Growth and survival of the sweet fish larvae reared in 0 PSU were significantly lower than those reared in either 4 PSU or 33 PSU. This indicated that the freshwater rotifers (B. calyciflorus) could be used as live food for sweetfish larvae reared in 4 PSU. The body weight of sweetfish larvae fed on freshwater rotifers enriched with Super Selco was the highest at 0.163 mg, but there was no significant difference in survival and body length of the fish fed with the other enrichment materials. The content of n-3 HUFA of the sweetfish larvae fed on the freshwater rotifers enriched with Super Selco and the condensed freshwater Chlorella was higher than that enriched with $\omega-yeast$ and baker's yeast. These results indicated that B. calyciflorus cultured with the condensed freshwater Chlorella could be used for the sweetfish larvae without enrichment, and the most efficient enrichment material for B. calyciflorus is Super Selco.

Examination of Optimal Reaction Mechanism in Oxygen Enriched Condition (산소부화조건에서의 반응기구 검토)

  • Burning velocities of conventional methane flame and oxygen-enriched methane flame were determined by experimentally and numerically at atmospheric pressure in order to examine the validity of various detailed reaction mechanisms in oxygen-enriched flame. The schlieren system was adopted to obtain the burning velocity of flame stabilized on a circular nozzle. Premix code was employed to compute the burning velocity. Three reaction mechnisms were tested at several oxygen enrichment level, whose names are GRI 3.0, MB(Miller and Bowman) and LKY(Lee Ki Yong) reaction mechanism. Sensitivity analysis was also performed to discriminate dominantly affecting reaction on burning velociy. The results showed that conventional reaction mechanisms originally based on methane-air flame were underpredict the burning velocity at high oxygen-enrichment level. The modified GRI 3.0 reaction mechanism based on our experimental results was suggested and shows a good agreement in estimating the burning velocity and the NO number density of oxygen-enriched flame.
