Design, Optimization and Verification of 16S rRNA Oligonucleotide Probes of Fluorescence in-situ Hybridization for Targeting Clostridium spp. and Clostridium kluyveri

  • Fluorescence in-situ hybridization (FISH) is a common and popular method used to investigate microbial communities in natural and engineered environments. In this study, two specific 16S rRNA-targeted oligonucleotide probes, CLZ and KCLZ, were designed and verified to quantify the genus Clostridium and the species Clostridium kluyveri. The optimal concentration of hybridization buffer solution for both probes was 30% (w/v). The specificity of the designed probes was high due to the use of pellets from pure reference strains. Feasibility was tested using samples of Chinese liquor from the famed Luzhou manufacturing cellar. The effectiveness of detecting target cells appears to vary widely in different environments. In pit mud, the detection effectiveness of the target cell by probes CLZ and KCLZ was 49.11% and 32.14%, respectively. Quantitative analysis by FISH technique of microbes in pit mud and fermented grains showed consistency with the results detected by qPCR and PCR-DGGE techniques, which showed that the probes CLZ and KCLZ were suitable to analyze the biomass of Clostridium spp. and C. kluyveri during liquor fermentation. Therefore, this study provides a method for quantitative analysis of Clostridium spp. and C. kluyveri and monitoring their community dynamics in microecosystems.

Cable-pulley brace to improve story drift distribution of MRFs with large openings

  • This study aims to introduce a new bracing system by which even super-wide frames with large openings can be braced. The proposed system, hereafter called Cable-Pulley Brace (CPB), is a tension-only bracing system with a rectilinear configuration. In CPB, a wire rope passes through a rectilinear path around the opening(s) and connects the lower corner of the frame to its opposite upper one. CPB is a secondary load resisting system with a nonlinear-elastic hysteretic behavior due to its initial pre-tension load. As a result, the required energy dissipation would be provided by the MRF itself, and the main intention of using CPB is to contribute to the initial and post-yield stiffness of the whole system. Using a stiffness calibration technique, optimum placement of the CPBs is discussed to yield a uniform displacement demand along the height of the structure. A displacement-based design procedure is proposed by which the MRF with CPB can be designed to achieve a uniform distribution of inter-story drifts with predefined values. Obtained results indicated that CPB leads to significant reductions in maximum and residual deformations of the MRF at the expense of minor increase in the maximum base shear and developed axial force demands in the columns. In the case of a typical 5-story residential building, compared to SMRF system, CPB system reduces maximum amounts of inter-story and residual drifts by 35% and 70%, respectively. Moreover, openings of the frame are not interrupted by the CPB. This is the most appealing feature of the proposed bracing system from architectural point of view.

  • This paper applies an optimization algorithm for the elastic truss structures. The acceleration technique utilized in this study is the geometric programming method developed by the Operation Research or the applied methematics. The applicability and the efficiency of the algorithm applied in this study are tested for four different trusses. Test results show that the optimum solutions are obtained after only one or seven iterations which is very small compared with other techniques and no oscillation is needed for the convergency. Test rusults also show that the Geometric Programming Method is also effective algorithm for the convergency of the Optimum Solution in case of only being compared with the number of iteration.

Hemorheology and clinical application : association of impairment of red blood cell deformability with diabetic nephropathy

  • Background: Reduced deformability of red blood cells (RBCs) may play an important role on the pathogenesis of chronic vascular complications of diabetes mellitus. However, available techniques for measuring RBC deformability often require washing process after each measurement, which is not optimal for day­to-day clinical use at point of care. The objectives of the present study are to develop a device and to delineate the correlation of impaired RBC deformability with diabetic nephropathy. Methods: We developed a disposable ektacytometry to measure RBC deformability, which adopted a laser diffraction technique and slit rheometry. The essential features of this design are its simplicity (ease of operation and no moving parts) and a disposable element which is in contact with the blood sample. We studied adult diabetic patients divided into three groups according to diabetic complications. Group I comprised 57 diabetic patients with normal renal function. Group II comprised 26 diabetic patients with chronic renal failure (CRF). Group III consisted of 30 diabetic subjects with end-stage renal disease (ESRD) on hemo-dialysis. According to the renal function for the diabetic groups, matched non-diabetic groups were served as control. Results: We found substantially impaired red blood cell deformability in those with normal renal function (group I) compared to non-diabetic control (P = 0.0005). As renal function decreases, an increased impairment in RBC deformability was found. Diabetic patients with chronic renal failure (group II) when compared to non-diabetic controls (CRF) had an apparently greater impairment in RBC deformability (P = 0.07). The non-diabetic cohort (CRF), on the other hand, manifested significant impairment in red blood cell deformability compared to healthy: control (P = 0.0001). Conclusions: The newly developed slit ektacytometer can measure the RBC deformability with ease and accuracy. In addition, progressive impairment in cell deformability is associated with renal function loss in all patients regardless of the presence or absence of diabetes. In diabetic patients, early impairment in RBC deformability appears in patients with normal renal function.

Preparation and Characterization of Antioxidant Peptides from Fermented Goat Placenta

  • The goat placenta was fermented by Bacillus subtilis and the optimal fermentation parameters of strongest antioxidant capacity of peptides were obtained using response surface methodology (RSM). The effects of fermentation time, initial pH value and glucose content on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity of the goat peptides were well fitted to a quadric equation with high determination coefficients. According to the data analysis of design expert, the strongest DPPH radical scavenging capacity value was obtained with the following conditions: content of glucose was 2.23%, initial pH value was 7.00 and fermentation time was 32.15 h. The DPPH radical scavenging capacity commonly referring antioxidant activity showed a concentration dependency and increased with increasing peptide concentration. The effects of temperature and pH were assessed to determine the stability of antioxidant peptides prepared from goat placenta. Antioxidant peptides showed good stabilities when temperature was lower than $70^{\circ}C$. However, the antioxidant peptides lost antioxidant activities rapidly under alkaline and excessive acid condition. Ultrafiltration technique was performed to separate fermentation broth with different Mw (molecular weight). It was found that peptides in the range of < 3 KDa mainly accounted for the antioxidant activities.

  • Gas springs of the television is to control the piston speed when operating under along stroke(200~300 mm, television elevation)is possible. User by this principle is capable of elevation adjustment. First carried out a flow analysis of the piston. A piston speed adjustment technique for precise pipe type cross-section was examined. The piston structure for flow rate control and elevation action is proposed. This study is the development of a gas spring of more than 50 inch television with a large television stand. Hollow piston rod for optimal control(the outer diameter 19.9 mm, the inner diameter 13.9 mm) was injected into the nitrogen gas(0.3 mm/s) in. As a result, the flow rate the pressure drop of the piston rod as the increase was increased without any change of the external force. As a result, control of the displacement via the gas spring is possible.

  • The absorption characteristics of itraconazole, which is an antifungal agent, from intestinal segments in the anesthetized rat i1l situ were investigated in order to design an effective oral drug delivery system. The pH-solubility profile of itraconazole, the rate and extent of absorption of itraconazole, the optimal absorption site(s) of itraconazole and the absorption enhancing effect of sodium cholate on itraconazole were examined in the present study. In situ single-pass perfusion method and recirculating perfusion technique using duodenum(D), jejunum(J) and ileum(I) were employed for the calculation of apparent permeability(Pe) and apparent first-order rate constant(Kobs). respectively. The results of this study were as follows; (1) Itraconazole showed appreciable aqueous solubility only at pH values of below 2.0. (2) pe(cm/sec) decreased in the following order: $D(10.24{\pm}1.78{\times}10^{-4})>J(8.86{\pm}0.79{\times}10^{-4})>I(3.78{\pm}0.13 X 10^{-4})$. (3) $Kobs(min^{-1})$ decreased in the following order: $J(17.12{\pm}3.19{\times}10^{-3})>D(13.37{\pm}0.6{\times}10^{-3})>I(11.05{\pm}0.91{\times}10^{-3})$. (4) The solubility of itraconazole markedly increased with the increase of the concentration of sodium cholate. (5) The addition of 10 mM sodium cholate significantly increased the apparent first-order rate constant of itraconazole in the ileum by a factor of 6.8.

Design of High Performance Multi-mode 2D Transform Block for HEVC (HEVC를 위한 고성능 다중 모드 2D 변환 블록의 설계)

  • This paper proposes the hardware architecture of high performance multi-mode 2D forward transform for HEVC which has same number of cycles for processing any type of four TUs and yield high throughput. In order to make the original image which has high pixel and high resolution into highly compressed image effectively, the transform technique of HEVC supports 4 kinds of pixel units, TUs and it finds the optimal mode after performs each transform computation. As the proposed transform engine uses the common computation operator which is produced by analyzing the relationship among transform matrix coefficients, it can process every 4 kinds of TU mode matrix operation with 35cycles equally. The proposed transform block was designed by Verilog HDL and synthesized by using TSMC 0.18um CMOS processing technology. From the results of logic synthesis, the maximum operating frequency was 400MHz and total gate count was 214k gates which has the throughput of 10-Gpels/cycle with the $4k(3840{\times}2160)@30fps$ image.

Development of a High-Resolution Electrocardiography for the Detection of Late Potentials (Late Potential의 검출을 위한 고해상도 심전계의 개발)

  • Most of the conventional electrocardiowaphs foil to detect signals other than P-QRS-T due to the limited SNR and bandwidth. High-resolution electrocardiography(HRECG) provides better SNR and wider bandwidth for the detection of micro-potentials with higher frequency components such as vontricular late potentials(LP). We have developed a HRECG using uncorrected XYZ lead for the detection of LPs. The overall gain of the amplifier is 4000 and the bandwidth is 0.5-300Hz without using 60Hz notch filter. Three 16-bit A/D converters sample X, Y, and Z signals simultaneously with a sampling frequency of 2000Hz. Sampled data are transmitted to a PC via a DMA-controlled, optically-coupled serial communication channel. In order to further reduce the noise, we implemented a signal averaging algorithm that averaged many instances of aligned beats. The beat alignment was carried out through the use of a template matching technique that finds a location maximizing cross-correlation with a given beat tem- plate. Beat alignment error was reduced to $\pm$0.25ms. FIR high-pass filter with cut-off frequency of 40Hz was applied to remove the low frequency components of the averaged X, Y, and Z signals. QRS onset and end point were determined from the vector magnitude of the sigrlaIL and some parameters needed to detect the existence of LP were estimated. The entire system was designed for the easy application of the future research topics including the optimal lead system, filter design, new parameter extraction, etc. In the developed HRECG, without signal averaging, the noise level was less than 5$\mu$V$_rms RTI$. With signal averaging of at least 100 beats, the noise level was reduced to 0.5$\mu$V$_rms RTI$, which is low enough to detect LPs. The developed HRECG will provide a new advanced functionality to interpretive ECG analyzers.

Enantioselective electrophoretic behavior of lipoic acid in single and dual cyclodextrin systems

  • Capillary electrophoresis (CE) is an effective technique to study chiral recognition because it offers flexibility in adjusting vital factors. Currently, various available cyclodextrins (CDs) can be employed for the chiral separation of numerous analytes. Herein, we investigate the enantioseparation behavior of lipoic acid enantiomers in various types of single and dual CD systems through CE. Additionally, several impacted CE parameters were optimized through the systematic investigation based on the design of experiment (DoE) concept for a single system comprising a heptakis (2,3,6-tri-O-methyl)-β-CD and a dual system containing the combination of the single CD with a sulfated-β-CD. Consequently, absolute enantioresolution was obtained within 15 min on a common standard bare fused-silica capillary (64.5/56 cm in total/effective length, 50/365 ㎛ inner/outer diameter), maintained at 15 ℃ and at an applied voltage of 24 kV. The optimal background electrolyte consisted of 6 mM heptakis (2,3,6-tri-O-methyl)-β-CD dissolved in the solution of 58 mM borate buffer at pH 10. Furthermore, the results of apparent binding constant experiments indicated that the S-enantiomer-heptakis (2,3,6-tri-O-methyl)-β-CD complex exhibited a stronger affinity than its R-enantiomer counterpart. The obtained electrophoretic mobility values could be utilized to interpret the resolution achieved at various CD concentrations and the mobility behavior of the complexes elucidated the migration order of the enantiomers in an electropherogram.
