방향족 아미노산에 의한 대장균 serC-aroA Operon의 발현 억제 (Repression of Escherichia coli serC-aroA Operon by Aromatic Amino Acids)

  • 대장균에서 두 가지 다른 아미노산의 생합성에 관여하는 serC 유전자와 araA유전자는 혼합 operon을 이루고 있다. SerC-aroA 혼합 operon의 발현 조절 현상을 serC-aroA-lacZ fusion plasmid pWH2를 이용하여 측정하였다. serC-aroA 혼합 operon의 발현은 L-tyrosine, L=phenulalanine 및 L-tryptophan 등 방향족 아미노산들에 의하여 억제되었다. 방향족 아미노산에 의한 억제 효과는 $tyrR^-$ 균주 혹은 $trpT^-$ 균주에서는 감소하였다. 또한, 방향족 아미노산은 cyclic AMP에 의한 이 operon의 발현 상승 효과를 감소시키기도 하였다. 이들 결과로부터 대장균 serC-aroA 혼합 operon의 발현은 방향종 아미노산들에 의해 억제된다고 추정하였다.

  • In order to achieve poly-beta-hydroxybutyric acid (PHB) production using recombinant DNA in various host bacterial cells, the isolation of genes for PHB biosynthesis was attempted. As a result, a 5.2kb DNA fragment containing phbCAB operon of Alcaligenes eutrophus was isolated by colony hybridization using synthetic oligodeoxyribonucleotides as probes. The constructed recmbinant plasmid pSK(+)-phbCAB operon was transferred to Escherichia coli, and the obtained transformant accumulated considerable amount of PHB.

Effects of R100 Mutant MerR on Regulation of mer Operon from Shigella flexneri

  • An amino-terminal 14 amino acids deletion and three site-directed mutations were created to investigate the mechanism of induction and repression of MerR regulatory protein in R100 mer operon from gramnegative Shigella flexneri. The amino-terminal 14 amino acids deletion, Cysl17Ser, and Cys126Ser mutations abolished the inducibility of the mer operon and the Hisl18Ala mutation resulted in the reduction of inducibility (about 9.1 % remaining) in complementation experiment in the presence of $Hg^{2+}$ at subtoxic level ($1\mu M$). The complementation experiment with $Hg^{2+}$ absent showed that Hisl18Ala, Cys126Ser, and wild-type MerR could repress the operon but Cysl17Ser could not, and the amino-terminal deletion mutant could neither induce nor repress the R100 mer operon.

Regulation of Expression of the Bacillus caldolyticus Pyrimidine Biosynthetic Operon by pyrR Gene, an Autogenous Regulator

  • The pyrR gene of the pyrimidine biosynthesis (pyr) operon of the thermophile Bacillus caldolyticus, encoding a uracil phosphoribosyltransferase (UPRTase), turned to rely as a pyr operon regulator. It has been proposed that PyrR mediates transcriptional termination-antitermination at three intercistronic regions of the par operon (S.-Y Ghim and J. Neuhard, J. Bacteriol.,176, 3698-3707, 1994). In this research, a plasmid carrying the pyrR region of B. caldolyticus could restore a pyrimidine regulation in a pyrR mutant of B. subtilis. Expression of pyrR was found to increase 6-7 fold during pyrimidine starvation. Additionally, a highly conserved nucleotide sequence which may constitute the binding site for a PyrR protein (PyrR-binding loop) in transcript was staggested. Alternative antiterminator and terminator structures involving three conserved motifs in front of the pyrR, pyrP and pyrB genes, respectively, are proposed to account for the observed regulation pattern.

  • Salmonella typhimurium possesses a cad operon, which contributes to an adaptive response against an acidifying environment. In Escherichia coli, the activation of the cad operon is dependent on cadC, which is located upstream of the operon. However, the activator of cad operon in S. typhimurium has not been known until now. In this study, we selected a putative cadC mutant by trasposon mutagenesis and cloned the cadc of S. typhimurium. Moreover, the cadC mutant was complemented by cadC clone. The cadC gene from S. typhimurium LT-2 consists of 1539 bp encoding a polypeptide ob 512 amino acids, and shows sequence similarity to cadC of E. coli with 53% identity and 67% similarity. The hydrophobicity profile of th S. typhimurim CadC sequence is very similar to E. coli CadC.

  • The groESL bicistronic operon of a restricted facultative methylotrophic bacterium Methylovorus sp. strain SS1 DSM 11726 was cloned and characterized. It was found to consist of two ORFs encoding proteins with molecular masses of 11,395 and 57,396 daltons, which showed a high degree of homology to other bacterial GroES and GroEL proteins. The genes were clustered in the transcription order groES-groEL. Northern blot analyses suggested that the groESL operon is transcribed as a bicistronic 2.2-kb mRNA, the steady-state level of which was markedly increased by temperature elevation. Primer extension analysis demonstrated one potential transcription start site preceding the groESL operon, which is located 100bp upstream of the groES start codon. The transcription start site was preceded by a putative promoter region highly homologous to the consensus sequences of Escherichia coli ${\sigma}^{32}$-type heat shock promoter, which functioned under both normal and heat shock conditions in E. coli. Heat shock mRNA was maximally produced by Methylovorus sp. strain SS1 approximately 10min after increasing the temperature from 30 to $42^{\circ}C$. The groESL operon was also induced by hydrogen peroxide or salt shock.

Transcriptional Regulation of Escherichia coli serC-aroA Operon : Further Support for cAMP-Dependent Expression

  • The Escherichia coli mixed-function serC-aroA operon encodes biosynthethic enzymes for unrelated pathways leading to the syntheses of serine and aromatic amino acids. It has been proposed that the operon is expressed in a cAMP-dependent manner. In this work experiments were performed to investigate the cAMP-dependent expression of the operon. Exogenous cAMP increased ${\beta}$-galactosidase synthesis in the $cya^+$ and cya strains harboring the serC-aroA-lac fusion plasmid. This enhancement was more dramatic in the $cya^-$ strain grown in a minimal medium. In a dot blot assay the serC-aroA mRNA content increased in a concentration-dependent pattern after the addition of exogenous cAMP. The activity of phosphoserine aminotransferase, encoded by the serC gene, apparently increased in E. coli cells after the addition of cAMP. All results obtained confirmed that the expression of the E. coli serC-aroA operon is positively regulated by cAMP at the level of transcription.

Differential Regulation of the Genes of the Streptococcus pneumoniae dnaK Operon by Ca++

  • DnaK is a major antigen in Streptococcus pneumoniae, and is induced by a minor shift in temperature (30 to $37^{\circ}C$) but not by ethanol shock. Although HrcA in the presence of $Ca^{{+}{+}}$ represses the expression of both groEL and hrcA, the control of transcription of the dnaK operon is not completely understood. In this study, the dnaK operon of S. pneumoniae (5' hrcA-grpE-dnaK-dnaJ) was cloned and analyzed. It contains large intergenic regions in grpE/dnaK and dnaK/dnaJ. Pulse labeling with [$^{35}S$]-methionine and immunoblot analyses revealed the presence of higher levels of DnaK than of HrcA even in the presence of $Ca^{{+}{+}}$ after heat shock suggesting that $Ca^{{+}{+}}$ differentially regulates the heat shock responses of hrcA and dnaK. By blocking de novo mRNA synthesis with rifampin it was shown that neither the hrcA nor the groEL transcripts were stabilized by heat shock even though dnaK transcripts were stabilized. We conclude that S. pneumoniae uses fine regulation of the transcription of the individual genes of the tetracistronic dnaK operon to cope with the various stresses experienced during infections.

Improvement of Bacilysin Production in Bacillus subtilis by CRISPR/Cas9-Mediated Editing of the 5'-Untranslated Region of the bac Operon

  • Bacilysin is a dipeptide antibiotic composed of L-alanine and L-anticapsin produced by certain strains of Bacillus subtilis. Bacilysin is gaining increasing attention in industrial agriculture and pharmaceutical industries due to its potent antagonistic effects on various bacterial, fungal, and algal pathogens. However, its use in industrial applications is hindered by its low production in the native producer. The biosynthesis of bacilysin is mainly based on the bacABCDEF operon. Examination of the sequence surrounding the upstream of the bac operon did not reveal a clear, strong ribosome binding site (RBS). Therefore, in this study, we aimed to investigate the impact of RBS as a potential route to improve bacilysin production. For this, the 5' untranslated region (5'UTR) of the bac operon was edited using the CRISPR/Cas9 approach by introducing a strong ribosome binding sequence carrying the canonical Shine-Dalgarno sequence (TAAGGAGG) with an 8 nt spacing from the AUG start codon. Strong RBS substitution resulted in a 2.87-fold increase in bacilysin production without affecting growth. Strong RBS substitution also improved the mRNA stability of the bac operon. All these data revealed that extensive RBS engineering is a promising key option for enhancing bacilysin production in its native producers.

SOS repair에 관여하는 umuDC 유전자

  • E. coli에서 UV 및 다른 많은 화학 돌연변이원에 의해서 유발되는 돌연변이는 chromosome의 umuDC operon이 필요하며, 이 유전자는 DNA 손상을 복구하고 UV나 화학물질에 대한 저항성을 증가시카는 DNA repair 기능을 활성화 시킨다. umuDC operon은 chromosome에 산재해 있고 16개의 다른 유전자로 구성된 E. coli SOS regulation의 한 유전자인데, umu 유전자는 한 operon에 의해 조절되는 umuD와 umuC로 구성되어 있다. 본고에서는 SOS repair에 관여하는 유전자들 중에서 umuDC 유전자를 중심으로 분자생물학적인 연구과정 및 연구결과를 서술하였다.
