  • Kong chain $\omega$3 & $\omega$6 fatty acids and taurine have been suggested to have structural and /or functional roles in the brain. The purpose of this study was to evaluate the changes in the percentage of fatty acids and taurine concentration in human milk obtained from Korean mothers(n=37) as lactation proceeds from birth to 5wks postpartum , and to determine the correlation between taurine concentration and fatty acid composition in breast milk. Level of fatty acids and taurine in breast milk were measured by GLC and HPLC, respectively. The percentages of medium -chain saturated fatty acids(10 : 0, 12 : 0 & 14: 0) were increased significantly , while percentages of long-chain saturated fatty acids(22 :0 & 24 : 0), and most of the long chain $\omega$6 and $\omega$3 fatty acids(20 : 3$\omega$6 , 20 :4$\omega$6, 22 :4$\omega$6 , 22 :5$\omega$6, 22 :5$\omega$3 & 22 : 6$\omega$3) were decreased significantly in mature milk (5 wks postpartum ) as compared to the colostrum. Taurine concentrations in colostrum and mature milk from Korean mothers were 549 $\pm$58 and 233 $\pm$41nmol/ml , respectively. Taurine concentrations in human milk was negatively correlated with the percentages of medium -chain saturated fatty acids ( 10 : 0 &12 : 0), and positively correlated with the percentages of long-chain saturated fatty acids( 16 :0, 20 : 0, 22 : 0& 24 : 0) and long-chain monounsaturated fatty acids( 20 : 1, 22 : 1 & 24 ; 1). Taurine concentration in human milk was positively correlated with elongation indices of both $\omega$6 and $\omega$3 fatty acids, and negatively correlated with the desaturation index of $\omega$6 fatty acids. These close relationships between fatty acids compositions and taurine level in human milk are worth pursing further investigation, especially with regard to their common roles in retina and brain development.

  • The supply of different fatty acids during the development period has significant effects. This study examined the effects of dietary $\omega$3 and $\omega$6 fatty acid compositions on phospholipids (PLs) of RBC and rat brain subcellular fractions (synaptosome, microsome, mitochondria), and on learning ability of the 2nd generation rat. Rats were fed experimental diets 3-4 wks prior to the conception. Early in the lactation period, the feeding mothers were exchanged. Diets consisted of 10% fat(by weight), which was either safflower oil('S') poor in $\omega$3 fatty acids or computer-searched mixed oil('M') with P/M/S ratio, 1/1.4/1 and $\omega$6/$\omega$3 ratio, 6.1/1. The 'S' and 'M' rats were subdivided further into SS, SM, MS & MM rats according to their lactation stauts. At 3 (weaning) & 9 wks of age, the percentage of total $\omega$3 fatty acids to their lactation status. At 3 (weaning) & 9 wks of age, the percentage of total $\omega$3 fatty acids and the ratios of $\omega$3/$\omega$6 fatty acids in PLs of RBC and brain subcellular fractions in SM and MM groups fed milk from the mixed oil-fed mothers for 2 wks tended to be higher than those in SS and MS groups respectively. In contrast, the concentrations of $\omega$6 fatty acids, especially 22:5$\omega$6 in all fractions, were significantly lower in the SM & MM groups compared to those of the SS & MS groups respectively. In contrast, the concentration of $\omega$6 fatty acids, especially 22:5$\omega$6 in all fractions, were significantly lower in the SM & MM groups compared to those of the SS & MS groups, The values for the DHA$\omega$3/22:5$\omega$6 ratios after the lactation period were markedly higher in the groups (SM & MM) which were reared by mixed oil(MO) fed mothers. In carring out Y-water maze at 9th wk of age, the SM(4.2$\pm$0.5) & MM (5.3$\pm$0.5) groups made significantly less errors compared to the SS(6.2$\pm$0.6, p<0.05 compared with SM) & MM (7.2$\pm$0.5, p<0.05 compared with MM) groups which were lactated by the safflower oilfed mothers. Therefore, by feeding a balanced fatty acid diet from the lactation period up to 9 wks of age as compared with the groups fed $\omega$3 fatty acid-deficient diet regardless of mother's diet given before parturition. The levels of DHA(synaptosome) and 22:5$\omega$3 (mitochondria) were positively correlated not only with these values in RBC but also with visual discriminating ability. The levels of DHA and 22:5$\omega$3 in RBC can, therfore, reflect visual discriminatng ability in the rat.

  • Effect of oral taurine supplementation on plasma total and phospholidpid -fatty acid profiles and their metabolism were evaluated in healthy female adults. Among twenty five female volunteers(23.6$\pm$0.3 years old ) participated in the taruine supplementation program(6g taurine /day), twenty four subjects succesfully completed the 2 week program , and only nine subjects continued to take taurine for another 2 weeks. Levels of plasma fatty acids and taruine were measured by gas-liquid chromatobraphy and an automated amino acid analyzer based on ion exchange chromatography, respectively. Plasma taurine concentration s of the subjects were 108. 7$\pm$3.4 , 184.2$\pm$8.2 and 235.9$\pm$77.0$\mu$emol/L at 0 , 2 and 4 weeks of taurine supplementation. Fatty acid compositions and elongation and desaturation indices of polyunsaturated fatty acids (PUFA) in plasma total lipids were not influenced by oral taurine supplementation. However, fatty acid compositions and their metabolism in plasma phospholipids were significantly affected by taurine supplementation in female adults. Compared to the values for 0 week, the percentage of saturated fatty acids (SFA) in plasma phospholipid was significantly lowered at 2 weeks, but elevated at 4 weeks of taurine supplementation. In contrast , the percentage of phospholipid PUFA significantly increased at 2 weeks and decreased at 4 weeks of taurine supplementation from to the values for 0 weeks. Foru weeks of oral taurine supplementation signifinatly elevated the eongation index(20 : 4$\omega$6 ⇒22 : 4 $\omega$6, p<0.01), and decreased the desaturation index (20 : 3 $\omega$6 ⇒20 : 4 $\omega$6 , p<0.01) of $\omega$6 fatty acids in plasma phospholipids. Plasma taurine concentration was positively correlated with the percentage of 14 : 0 fatty acids and the enlongation index o f$\omega$3 fatty acids(20 : 5 $\omega$3 ⇒22 : 5 $\omega$3), and thenegatively correlated with the percentage of 20 : 0 in plasma phospholipids. These results indicate that oral taurine supplementation for 4 weeks signidicantly elelvated the percentage of SFA, and lowered the percentage of PUFA in plasma phospholipids with no influence on plasm total fatty aicd composition in healthy female adults.

  • This study was conducted to compare the lipid content and fatty acid composition of the Korean breast milk produced at 1, 2 and 3 months postpartum with the infant formula produced in Korea. The content of total lipids of the breast milk was low compared with the other data of Korean as well as foreign breast milk, and the content tended to reduce during the progress of lactation. The composition of fatty acids of the breast milk was characterized by low saturated fatty acids(SFA) and high $\omega$3 series of polyunsaturated fatty acids (PUFA), especially 20:5 and 22:6, so the P/S and $\omega$6/$\omega$3 rations were high. The formulas had more total lipids, triglycerides(TG) and phospholipids and less cholesterol(CHOL) and free fatty acids than the breast milk. The cholesterol content of the formulas was around 50% of that of the breast milk, so the CHOL/TG ratio was low. Although the formulas varied in fatty acid composition, the formulas had more SFA and $\omega$6 series of PUFA, but less monounsaturated fatty acids and $\omega$3 series of PUFA than the breast milk. This study shows that there are some differences in lipid content and fatty acid composition between breast milk and infant formula. Therefore, further studies needed to investigate the physiological effect of this difference on lipid metabolism of infants.

  • Docosahexaenoic acid(DHA), a $\omega$3 series fatty acid and arachidonic acid(AA). a $\omega$6 series fatty acid were found in relatively high concentrations in the phospholipids(PLs) of cell membranes of nerve tissues, and they can be affected by various factors. The present study examined the effects of dietary $\omega$6 and $\omega$3 fatty acid composition on P/M/S and on $\omega$3/$\omega$6 fatty acid ratios in brain PLs of 2nd generation rats. The expeimental diets consisted of 10% fat(by wt), which were computer- searched mixed oil('M') with P/M/S ratio, 1 : 1.4 : 1 and $\omega$6/$\omega$3 ratio, 6 : 1 and safflower oil('S') poor in $\omega$3 fatty acids. The experimental diets were started 3-4 wks prior to conception. During the lactation period, the feeding mothers were switched 1 wk after birth and provided the pups for 2 wks with milk which had compositions different from that of their natural mother. The same diet as their mothers was provided from weaning to 9 wks of age. The 'M'and 'S' rats were again subdivided into MM, MS, SS, SM rats according to diet which their lactating mothers were fed from the begining of the experiment. The relative percentage of P/M/S fatty acids in brain PLs in all experimental groups converged to a very similar value at 9 wks of age, indicating the existence of a control mechanism for the degree of fatty acids, unsaturation. The $\omega$3/$\omega$6 fatty acid ratios of brain PLs converged to about 1.0 in MM & SM groups and to 0.7 in SS & MS groups, suggesting also the existence of some balance between $\omega$3 and $\omega$6 fatty acids in developing rat brain. The concentrations of $\omega$3 fatty acids, especially DHA, in the SM group were increased and became similar to those in MM group at 9 wks of age. The increase in DHA of brain PLs was counterbalanced b)r a decrease in 22 5$\omega$6. Therefore, the ratios of 22 : 6$\omega$6/22 : 5$\omega$6 were higher in both MM & SM groups than those of SS & MS groups at 9 wak of age. Although dietary $\omega$3 and $\omega$6 fatty acids affected 22 : 6$\omega$S and 22 : 5$\omega$6 contained in rat brain PLs reciprocally, the relative percentage of AA did not appear to be significantly influenced by the diet in all groups at 9 wks of age, suggesting that a mechanism for the maintenance of a certain level of AA in brain PLs exists. In conclusion, the $\omega$3/$\omega$6 fatty acid and 22 : 6$\omega$3/22 : 5$\omega$6 ratios, but not P/M/S ratio, of rat brain PLs were affected by the postnatal dietary changes. Futher studies are required to clarify the mechanism(S) of ensuring a certain level of DHA and of maintaining a similar level of AA in rat brain PLs after. weaning(9 wk) regardless of prenatal and postnatal dietary changes. • Studies of the relationship between the composition of serum fatty acids and blood pressure are complex and controversial. Fatty acids, important constituents of biological membranes, could potentially affect vasoreactivities including blood pressure. In this study the compositions of fatty acids in serum phospholipids were compared between three types of hypertensive subjects (men, pre-menopausal women, and post-menopausal women) and their respective nrmotensive controls. Serum lipids were extracted and phospholipids were separated by thin layer chromatography. The percentage of palmitic acid (16 : 0) in serum phospholipids was significantly higher and the percentage of stearic acid (18 : 0) was significantly lower in all three hypertensive groups, compared with their corresponding control groups. Only in the group of post-menopausal women, palmitic acid was closely associated wish increases in both systolic (SBP) and diastolic blood pressure (DBP), while stearic acid was associated with decreases in both SBP and DBP. The polyunsaturated fatty acids in serum phospholipids behaved differently from saturated fatty acids. The ratios of products / precursor fatty acids, such as $\sumLCPUFA\omega6/18 : 2\omega$6, 20 : 4$\omega$6/18 : 2$\omega$6, ∑LCPUFA$\omega$3/18 : 3$\omega$3 and 22 : 6$\omega$3/20 : 5$\omega$3, were all clearly associated with both SBP and DBP in hypertensive, post-menopausal women. Desaturation and elongation in fatty acid metabolism could affect the bioavailability of eicosanoid precursors. Changes in the constituent fatty acids of phospholipids and eicosanoid precursors may also influence fluidity, ionic transport, hormone receptors and enzyme activities in biological membranes. In conclusion, both systolic and diastolic blood pressure in post-menopausal women was positively associated with the level of palmitic acid, and negatively associated with the level of stearic acid, in serum phospholipids. The relationships between serum phospholipid-$\omega$6 and $\omega$3 series fatty acids and blood pressure in women, especially in post-menopausal women, require further investigation by taking into consideration hormonal status and eicosanoid metabolism. Funker study is needed to determine the value of dietary manipulation of fatty acid constituents of serum phospholipids, relating to hypertension in women.

  • In search for several fatty acid with unusual structure in vegetable oils, we have found that unknown peaks were shown on GLC in the analysis of fatty acids of the lipids from the pulp of ripened jujube (Zizypus jujuba var. inermis) fruits. These fatty acids were identified as a series of cis-monoenoic acids with ${\omega}-5$ double bond system such as $C_{14:1{\omega}5}$, $C_{16:1{\omega}5}$ and $C_{18:1{\omega}5}$, including ${\omega}-7$ fatty acid as $C_{16:1{\omega}7}$ and $C_{18:1{\omega}7}$, by GLC, solid-phase extraction silver ion-column chromatographic, GLC-mass spectrometric and IR techniques. First of all, total fatty acid methyl esters were resolved into saturated and branched fatty acid, monoenoic acid, dienoic acid, and trienoic acid fraction, respectively, with 100% dichloromethane (DCM), DCM/acetone (9:1, v/v) 100% acetone, and acetone/ acetonitrile (97:3, v/v) solvent system. Unknown fatty acids were included in the monoenoic fraction and were confirmed to have cis-configuration by IR. Picolinyl esters of monoenoic fatty acids gave distinct molecular ion peak and dominant diagnostic peaks, for example, m/z 317, 220 and 260 fragment for $cis-C_{14:1{\omega}5}$, m/z 345, m/z 248 and 288 fragment for $cis-C_{16:1{\omega}5}$ and m/z 373, m/z 276 and 316 fragment for $cis-C_{18:1{\omega}5}$. In this way the occurrence of $cis-C_{16:1{\omega}7}$ and $cis-C_{18:1{\omega}7}$ could be deduced from the appearance of prominent fragments as m/z 345, 220 and 260, and m/z 373, 248 and 280. Level of total ${\omega}-5$ fatty acids amounted to about 30% in the fatty acid composition with the predominance of $C_{16:1{\omega}5}$ $ (18.7{\sim}25.0%)$, in the semi-ripened and/or ripened samples collected in September 14 ($C_{16:1{\omega}5}$ ; 18.7%, $C_{14:1{\omega}5}$ ; 3.6% and $C_{18:1{\omega}5}$ ; 3.0%), September 22 ($C_{16:1{\omega}5}$ ; 25.0%, $C_{14:1{\omega}5}$ ; 1.4% and $C_{18:1{\omega}5}$ ; 2.6%), and October $7 (C_{16:1{\omega}5}$ ; 24.7%, $C_{14:1{\omega}5}$ ; 7.7% and $C_{18:1{\omega}5}$ ; 2.5%). However, the lipids extracted from unripened jujube in July and August contain these unusual fatty acids as low as negligible. It could be observed that the level of ${\omega}-5$ fatty acids in the pulps increased sharply with an elapse of ripening time of jujube fruits. Other monoenoic fatty acids with ${\omega}-7$ series, $C_{16:1{\omega}7}$ (palmitoleic acid) and $C_{18:1{\omega}7}$ (cis-vaccenic acid) could be detected. And in the lipids of the kernel and leaf of jujube, none of ${\omega}-5$ fatty acids could be detected.

  • The present study was carried out in normal and atherosclerotic New Zealand white(NZW) rabbits, to evaluate the effect of dietary supplementation with Korean pinenut oil, on the total fatty acid composition of red blood cell membranes. Erythrocyte fatty acids of chain lengths ranging fro 12 : 0 to 22 : 6$\omega$3 were identified. In study I, palmitic acid(16 : 0) was found to be the main erythrocyte membranes fatty acid, followed in turn by stearic(18 : 0), oleic(18 : 1), and linoleic acids(18 : 2$\omega$6). There was a large increase in the proportion of 20 : 4$\omega$6 in 10% pinenut oil group on days 40 and 80(compared with control), although the increase was not statistically significant. Most erythrocyte fatty acids were less affected by the types or levels of oils supplemented for periods fo 40 and 80days. In study II,at the end of the dietary treatment, the levels of 12 : 0, 14 : 0, 15 : 0 and 18 : 0 were significantly decreased in the 5% soybean oil/cholesterol group, whereas those of 12 : 0, 14 : 0 and 18 : 0 were significantly decreased, but that of 18 : 2$\omega$6 was significantly increased in the 10% soybean oil/cholesterol group, compared to the cholesterol group. No significant differences in fatty acid composition were seen between the pinenut oil/cholesterol group, whereas the 10% soybean oil/cholesterol group had significantly elevated 18 : 2$\omega$6, compared to the cholesterol group. The proportion of 18 : 4$\omega$6 was significantly raised in the 5% soybean oil/cholesterol and 5% pinenut oil/cholesterol group, and the content of 16 : 1$\omega$7 was significantly lowered in the 5% and 10% soybean/cholesterol groups, compared to the cholesterol group at 10 weeks supplementation. In studies I,II and III, the total proportions of saturated fatty acids were not significantly affected by the types of levels of oils supplemented throughout the study period.

  • The purposes of this study were to 1) examine the dietary patterns of fatty acids and the fatty acid composition in serum, 2) determine selenium (Se) concentrations, and 3) investigate how serum fatty acid compositon affects serum levels of Se in Korean elementary school children. The subjects consisted of 168 school children(82 boys & 86 girls) belonging to 1st through 6th grades, and their dietary assessment was evaluated. The serum fatty acid composition was analyzed by GLC, and the ICP/MS method was employed to measure serum Se concentration. Total average intake of PUFA, MUFA and SFA were 10.5g, 12.7g, 11.2g respectively. Total average dietary P/M/S ratio was 1.10/1.18/1.0. The average intake of $\omega$3 fatty acids and $\omega$6 fatty acids were 1.33g, 9.19g, respectively. The $\omega$6/$\omega$3ratio was 14.2 which is higher than the recommended range. In serum fatty acid composition, PUFA, MUFA, SFA were 40.6%, 28.1% and 31.6% respectively. The average $\omega$3 fatty acid composition was 3.80%, and the mean value of $\omega$6 series was 36.8%. The M/S ratio of girls was significantly higher than boy's in fatty acids intake and in serum composition. Mean Se concentration of total subjects was 416.7ug/l and it was significantly higher in the lower grades(1st-3rd)than in the upper grades(4th~6th, p<0.05). Although the mean serum Se concentration was negatively correlated with PUFA composition in total boys and girls, it was not significant. Serum Se concentration was negatively correlated with P/S ratio and C24:1 concentration(p<0.05). In addition, serum composition of $\omega$6 PUFA and C24:1 showed negative correlations with serum Se in only lower grades girl(1st~3rd), and further studies are needed to clarify in these phenomena. In conclusion, the mean serum Se concentration was significantly lower in upper grades (4th~6th)than in lower grades(1st~3rd) of elementary school children and was negatively correlated with P/S ratio of serum fatty acids. More detailed studies on relationship between Se and fatty acids are required.

  • This study was designed to evaluate the balance of fatty acids for the Korean style broiled beef and pork ingredient of the rate of mixture oil with sesame oil, soybean oil and perilla oil, by self-developed computer program. Each 3 kinds of Korean style broiled beef and pork by the rate of mixure oil with sesame oil, soybean oil and perilla oil, were evaluated by using the self-developed computer program. Contents of calories were 415.6kcal in Korean style broiled beef, 656.3 kcal in Korean style broiled pork. The mean of protein were 41.35g, Korean style broiled beef, 32.66g, Korean style broiled pork. The ratio of C/P/F ratio of Korean style broiled beef and pork, 21/ 40/ 39, 10/ 20/ 70. The mean P/ M/ S of fatty acids in Korean style broiled beef and pork ratio was similar $0.4{\sim}0.5/\;1.4{\sim}1.5/\;1$. The ${\omega}6/{\omega}3$ ratio of fatty acids of Korean style broiled beef and pork using sesame oil was 54.3, 56.9 much higher than desirable $level(4{\sim}8)$. But the ${\omega}6/{\omega}3$ ratio of fatty acids of Korean style broiled beef using mixture oil with sesame oil, soybean oil and perilla oil were 4.6, 4.2 desirable $level(4{\sim}8)$, Korean style broiled pork using mixture oil with sesame oil, soybean oil and perilla oil were 3.0, 6.2. It means the ${\omega}6/{\omega}3$ ratio of fatty acids of Korean style broiled beef and pork was improved of desirable level respectively by using the mixture oil with sesame oil, soybean oil and perilla oil, than using sesame oil.
