분자량이 다른 키토산이 흰쥐의 지방대사에 미치는 영향 (Effects of CHitosan of Different Molecular Weights n Lipid Metaboism in Rats)

  • This study was conducted to investigate the effects of chitosan, chitosan oligomer and beef tallow of different levels on lipid metabolism in rats, Seventy male rats of the Sprague-Dawley strain were blocked into 10 groups according to body weight , and were raised for 4 weeks. Dietary fat levels were 20% en and 40% en, and chitosan and chitosan oligomer were given at levels of 05, 3%, and 5%(wt/wt) of diet. The results are summarized as follows, chitosan oligomer supplement decreased serum total lipid, as chitosan 양, and exhibited a tendency to decrease serum total cholesterol. Chitosan oligomer supplement tended to increase the HDL cholesterol ; total cholesterol ration as chitosan did. Liver total lipid and triglyceride concentration were lower in high fat groups than in low fat groups. Liver total lipid concentrations was decreased slightly by chitosan and sigfnificantly by chitosna ligomer. Epididymal fat pad total lipid. total cholesterol and triglyceride concentration were slightly decreased by chitosan and chitosan oligomer supplement, especially in high fat groups. fat absorptivity was decreased by low fat level, chitisan and chitosan oligamer supplement. Fecal excretion of total lipid and triglyceride were increased by high fat level, chitosan and chi샌무 oligomer supplement. However, fecal excretion of total cholesterol was increased by high fat level and chitosan supplement. This indicated that chitosan and chitosan oligomer were effective in interfering with lipid and triglyceride absorption, In conclusion, chitosan oligomer at levels of 3% and 5% has more effective lipid, cholesterol and triglyceride lowering activity than chitosan.

UV-Vis Spectra of N, N'-Bis(4′-Aminophenyl)-1,4-quinonenediimine Doped With $H_{4}SiW_{12}O_{40}$

  • We have synthesized aniline oligomer composites by using heteropoly acid (H$_4$SiW$_{12}$O$_{40}$ ) as a dopant. The doping and dedoping processes of the aniline oligomer composites were investigated with the aid of UV- Vis spectra. The bands of the aniline oligomer at 572 nm weakened or disappeared, and the bands at 268, 412, and 771 nm appeared, after the aniline oligomer was doped. When the solution of the aniline oligomer doped with H$_4$SiW$_{12}$O$_{40}$ was kept at lower values of pH, the aniline oligomer could not be dedoped by dilution. The turning point of doping and dedoping occurred at pH 5.5. The band at 771 nm shifted towards longer wavelengths when the aniline oligomer composites were synthesized using acetone as the solvent. This observation indicates that the molecular chain became stretched. In addition, we also investigated the change of the electronic absorption spectra of the composites with respect to the time laid up.id up.

  • The sodium ${\alpha}-sulfo$ fatty acid vinyl ester oligomers, which are oligomer type surfactants were prepared by polymerization with fatty acid vinyl acetate. The ${\alpha}-sulfonation$ of fatty acid vinyl ester oligomers were carried by direct addition of sulfur trioxide. The dispersing performance of oligomer type anionic surfactants and sodium dodecyl sulfate(SDS) in the aqueous suspension of iron oxide and titanium dioxide particles was evaluated by particle size distribution and zeta-potential measurement. As results, the particles of iron oxide and titanium dioxide were flocculated by addition of small amount of oligomer type anionic surfactants and sodium dodecyl sulfate(SDS), then the flocks redispersed by more addition of oligomer type anionic surfactants and SDS. The flocculation, redispersion process was observed in lower concentration range of oligomer type anionic surfactants than SDS. Especially, the dispersing action of sodium ${\alpha}-sulfo$ palmitic acid vinyl ester oligomer was better than sodium ${\alpha}-sulfo$ lauric acid vinyl ester oligomer.

초음파를 적용한 PET 직물의 알칼리 가수분해에 관한 연구(II) - 기공특성과 올리고머 분석 - (A Study on the Alkali Hydrolysis of PET fabrics with Ultrasonic Application(II)- Surface Porosity and Oligomer Analysis -)

  • In order to give a silk-like touch to PET fabrics, the PET fabrics were treated with NaOH alkaline solution in various conditions. In alkaline treatment, the liquor flow type pilot weight reduction apparatus with magnetostrictive ultrasonic transducer was used for the study. The effects of ultrasonic application, treatment time and temperature at NaOH 4% and 6"A solution on the decomposition rate of PET fabrics. From the results of the decomposition rate of PET fabrics, the qualitative and quantitative analysis of oligomer after decomposition of PET fabrics carried out by the HPLC. On the other hand, the surface pore characteristics of decomposition PET fabrics measured by porosimetery. The pore characteristics on the surface of treated PET fiber depended on the decomposition rate and did not depend on the ultrasonic cavitation. The pore diameter of alkaline untreated PET fiber were 15A and those of treated PET fibers were 5~6$\AA$ at the maximum pore volume. The average pore sizes of fiber before and after treatment were 141 h and 160h, respectively. Total amount of oligomer of the untreated PET fibers were 1.70wt% and 67.7% of total oligomer occupied with PET cyclic trimer and PET cyclic tetramer. Total amount of oligomer of fiber with 26.9% and 48.0% of weight loss without ultrasonic application were 1.78wt% and 1.79wt%, respectively. Also total amount oligomer of fibers which were reduced 27.7% and 48.2% of weight loss with ultrasonic application were 1.74wt%. This result showed that the removal rate of oligomer in the process of alkaline hydrolysis with ultrasonic higher than that of without ultrasonic application.tion.

N,N-디아실, O-아실 키토산 올리고머의 물리화학적 특성에 관한 연구 (Study of Physico-Chemical Properties of N,N-Diacyl, O-Acyl Chitosan Oligomer)

  • 천연 고분자인 키토산의 1차 아민기를 아질산 나트륨에 의해 oxidative-deamination 반응을 일으켜 reducing end에 알데히드기를 갖는 키토산 올리고머를 얻었으며, 이를 수소화붕소산나트륨으로 환원하여 2,5-anhydro-D-mannitol로 전환하였다. 얻어진 피토산 올리고머의 평균 중합도는 겔 투과 크로마토그래피를 통해 2정도임을 알 수 있었고, 키토산에 비해 친유성 유기 용매에 대한 높은 용해성을 나타내었다. 키토산 올리고머에 아실클로라이드를 4-디메틸아미노피리딘 촉매하에서 반응시켜 N,N-디아실, O-아실 키토산 올리고머를 합성하였다. 시차주사열량 분석을 실시한 결과 N,N-디라우로일, O-라우로일 키토산 올리고머의 경우 액정 영역을 나타내었고, 편광현미경을 통해 이를 확인할 수 있었다. X-선 회절분석 실험결과 키토산 올리고머와 달리 N,N-디라우로일, O-라우로일 키토산 올리고머는 높은 결정성 구조를 나타냄을 알 수 있었다.

The Effect of Oligomer Blending on the Flow Properties of Polycarbonate

  • We successfully prepared high-flow polycarbonate (PC) by blending commercial PC with a low molecular weight PC oligomer. The oligomer was synthesized by the addition of a large quantity of mono functional phenol groups, and the chain end group was reacted with p-tertiary butyl phenol (PTBP) to block the reactivity. The viscosity average molecular weight ($M_v$) for the oligomer was about 4,000-5,000 g/mol, compared to ~19,000 g/mol for the PC blend obtained by blending 10 wt% of the prepared oligomer with the commercial grade PC ($M_v$ of 21,000 g/mol). The blended PC had a melt flow index of 45, which is 2.5 times higher, and a processing temperature that was $20^{\circ}C$ lower, than that of commercial grade PC having a similar $M_v$.

효소 가수분해를 통한 매생이 유래 β-Glucan 형태의 Oligomer 생산 및 분석 (Production and Characterization of β-Glucan Type Oligomer Produced with Enzymatic Hydrolysis of Capsosiphon fulvescens)

  • ${\beta}$-Glucan type oligomers which have angiotensin I converting enzyme (ACE) inhibitory activity were isolated and characterized from Capsosiphon fulvescens. After C. fulvescens was hydrolysis with Alcalase at $50^{\circ}C$, supernatant was harvested and separated with ultrafiltration membrane (MWCO 2 kDa). Oligomers which were less than 2 kDa of molecular weight were harvested for characterization. The nutrient composition of Alcalase hydrolysate was 89.9% carbohydrate, 4.2% protein and 5.9% sulfate. After ultrafiltration, the nutrient composition of oligomers was changed to 99.88% carbohydrate, 0.07% protein and 0.05% sulfate. The carbohydrate composition of oligomer was glucose (97.2%) and mannose (1.5%). The ACE inhibitory activities of Alcalase hydrolysate and oligomer were 72.1% and 82%, respectively. The molecular weight of oligomer was about 1 kDa. The oligomer was analyzed with FT-IR, $^1H$-NMR and methylation. The oligomers were ${\beta}$-1,3-glucans with ${\beta}$-(1,3)-linked glucose units.

  • Allylaliphatic carboxylate oligomers were prepared from polymerization giving allyl aliphatic carboxylates in the presence of potassium persulfate in methanol and the ${\alpha}-sulfonation$ of these allyl aliphatic carboxylates oligomers were carried by direct addition of dry sulfur trioxide. The dispersing performance of oligomer type anionic surfactants and SDS in the aqueous suspension of $Fe_{2}O_{3}$ and $Tio_{2}$ particles were evaluated by particle size distribution and ${\zeta}-potential$ measurement. As results, the particles of $Fe_{2}O_{3}$ and $Tio_{2}$ were flocculated by addition of small amount of oligomer type anionic surfactants and SDS, then the flocks redispersed by more addition oligomer type anionic surfactants and SDS. The dispersion and flocculation were observed in lower concentration range of oligomer type anionic surfactants than SDS.

($A{\beta}-oligomer$로 유도된 Neuro2A 세포주에서 용담사간탕(龍膽瀉肝湯)의 치매 억제 효과 (A Study on the Inhibitory Effect of Yeongdamsagantang on Alzheimer in $A{\beta}-oligomer-induced$ Neuro 2A Cell Lines)

  • Objective: To investigate the effects of Yeongdamsagantang (YDGT) on apoptosis of neuronal cells that can result in dementia. Method: The water extract of the YDGT was tested in vitro for its beneficial effects on neuronal survival and neuroprotective functions, particularly in connection with $A{\beta}$ oligomer-related dementias. $A{\beta}$ oligomers derived from proteolytic processing of the ${\beta}-amyloid$ precursor protein (APP), including the $amyloid-{\beta}$ peptide $(A{\beta})$, play a critical role in the pathogenesis of Alzheimer's disease. A neuroblastoma cell line stably expressing an $A{\beta}$ oligomerassociated neuronal degeneration was used to investigate if YDGT inhibits formation of $A{\beta}$ oligomer. To measure the ATP generating level in mitochondrial membrane, luciferin/luciferase luminescence kit (Promega) and luminator was used, and to survey the protein's apparition, confocal microscopy was used. Result: $A{\beta}oligomer$ had a profound attenuation in the increase in CT105 expressing neuro2A cells from YDGT. Experimental evidence indicates that YDGT protected against neuronal damage from cells, but its cellular and molecular mechanisms remain unknown. We demonstrated that YDGT inhibited formation of $amyloid-{\beta}$ $(A{\beta})$ oligomers, which were the behavior, and possibly causative, features of AD. The decreased $A{\beta}$ oligomer in the presence of YDGT was observed in the conditioned medium of this $A{\beta}oligomer-secreting$ cell line under in vitro. In the cells, YDGT significantly attenuated mitochondrion-initiated apoptosis. Conclusion: (i) a direct $A{\beta}$ oligomer toxicity and the apoptosis initiated by the mitochondria; and (ii) multiple cellular and molecular neuroprotective mechanisms, including attenuation of apoptosis and direct inhibition of $A{\beta}$ oligomer aggregation, underlie the neuroprotective effects of YDGT.

LMK02의 품질규격화와 $A{\beta}$ 올리고머에 의해 유도된 희주해마 H19-7세포주에 미치는 항치매효과 (Standardization of Quality and Inhibitory Effect of Alzheimer in $A{\beta}$ Oligomer-induced H19-7 Cells by LMK02)

  • For standardization of LMK02 quality, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelica gigas Nakai in the constituents of LMK02 were estimated as indicative components. From LMK02 water extract, has been used in vitro test for its beneficial effects on neuronal survival and neuroprotective functions, particularly in connection with APP-related dementias and Alzheimer's disease (AD). $A{\beta}$ oligomer derived from proteolytic processing of the ${\beta}$-amyloid precursor protein (APP), including the amyloid-${\beta}$ peptide ($A{\beta}$), play a critical role in the pathogenesis of Alzheimer's dementia. We determined that oligomer amyloid-${\beta}$ ($A{\beta}$) have a profound attenuation in the increase in rat hippocampus H19-7 cells from. Experimental evidence indicates that LMK02 protects against neuronal damage from cells, but its cellular and molecular mechanisms remain unknown. Using a hippocampus cell line on $A{\beta}$ oligomer-induced neuronal cytotoxicity, we demonstrated that LMK02 inhibits formation of $A{\beta}$ oligomer, which are the behavior, and possibly causative, feature of AD. In the Red Ginseng, the average amounts of Ginsenoside Rg3 were $47.04{\mu}g/g$ and $42.3{\mu}g/g$, 90 % of its weight were set as a standard value. And, in the Angelica gigas Nakai, the average amounts of Decursin were 2.71 mg/g and 2.44mg/g, 90 % of its weight were also set as a standard value. The attenuated $A{\beta}$ oligomer in the presence of LMK02 was observed in the conditioned medium of this $A{\beta}$ oligomer-induced cells under in vitro. In the cells, LMK02 significantly activated antiapoptosis and decreased the production of ROS. These results suggest that neuronal damage in AD might be due to two factors: a direct $A{\beta}$ oligomer toxicity and multiple cellular and molecular neuroprotective mechanisms, including attenuation of apoptosis and direct inhibition of $A{\beta}$ oligomer, underlie the neuroprotective effects of LMK02 treatment.
