Seed Protein Content and Fatty Acid Composition of Black Seeded Soybeans Collected From Southwestern Islands (서남해안 검정콩들의 단백질과 지방산 조성변이)

  • A total of 135 black seeded soybeans genotypes were collected in 45 island locations from January to May 2001. Seeds of 135 genotypes collected were analyzed for crude protein and fatty acid compositions. The crude protein content was averaged to be 40.75%, and was ranged from 34.70% to 44.20%. The average palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid content were 11.41%, 3.93%, 22.75%, 53.55%, and 8.35%, and the ranges of those were 9.00% to 14.40%, 2.90% to 5.00%, 22.75% to 26.50%, 50.30% to 57.20% and 6.7% to 11.20%, respectively. Heritabilities of palmitic acid, oleic acid, linoleic acid, and crude protein were higher, but that of stearic acid and linolenic acid were relatively lower, Crude protein content was correlated positively with oleic acid content, whereas it was correlated negatively with linoleic acid, palmitic acid, stearic acid, and linolenic acid contents.

Tylosin Production by Mutant Resistant to Oleic Acid (Oleic Acid 내성균주로부터 Tylosin 생산)

  • When rapeseed oil as the carbon source was used for tylosin production from Streptomyces fradiae TP-1239 was very sensitive to oleic acid. Cell growth was restrained by adding 0.8 g/l of oleic acid to the culture broth. Mutant strain TM-224-1 resistant to 1.2 g/l of oleic acid was obtained by screening in solid and liquid media containing oleic acid. The uptake rate of oleic acid by TM-224-1 was approximately 3.8 fold higher than the parent strain. For comparing the TM-224-1 and the parent strain, batch cultures were carried out in a jar fermentor. Cell growth of TM-224-1 strain was higher than the parent strain after two days of culturing. However, after four days of culturing, it was similar to that of the parent strain. The amount of rapeseed oil consumed by TM-224-1 and the parent strain were 60.5 and 78.2 g/l, respectively. The production and yield of tylosin was aproximately 2.0 and 3.2 fold higher than the parent strain, respectively. From these results, it was concluded that this mutant, which was resistant to oleic acid, has improved tylosin production.

  • To find esterase inhibitors and in the metabolites of Streptomyces strain DMC-498, two active compounds were isolated from the methanol extract of the mycelia of the strain by Silica gel column chromatography and preparatory argentation TLC. These compounds were proved to show competitive inhibition. Compound B was found to consist of linoleic and oleic acids. Fifty percent inhibition concentration ($lC_{50}$) of linoleic acid was $0.045\mu\textrm{g}/ml$, whereas oleic acid exhibited no inhibitory activity. Associated lipids: isostearic acid, isostearic acid methyl ester, oleic acid methyl ester and linoleic acid methyl ester, were isolated from the same extract, showing no inhibition of the esterase. Compound A was found to be a liquid inhibitor with an alicyclic ring and two or more oxygens, its molecular weight being more than 500.

Unambiguous Evidence for Phase Transitions of Oleic Acid in Pure Liquid State by Near-Infrared Spectroscopy and Pricipan Comaonent Analysis

  • Temperature-dependent changes in near-infrared (NIR) spectra have been measured for oleic acid, and nonanoic acid in the pure liquid state. Particular attention has been paid to the 5400-4800 cm$\^$-1/ region where a number of combination bands appear. The NIR spectra of oleic acid show that a band at 5303 cm$\^$-1/ increases with temperature while that at 5270 cm/sup-1/ decreases. It ha been found from their second derivative spectra that these spectral changes take place stepwisely with two break points at 30 and 53$\^{C}$, which correspond to the phase transition temperatures oleic acid reported previously. Principle component analysis (PCA) has been carried out for the NIR spectra of oleic acid in the 5400-4800 cm$\^$-1/ region measured over a temperature range of 15-80$\^{C}$. core plots of the first and second principal components (PCs) show that the NIR spectra are classified into three groups; the spectra measured in the temperature range of 15-30$\^{C}$, those in the range of 31-53$\^{C}$, and those in the range of 54-80$\^{C}$. These temperature ranges correspond to those for quasi-smectic liquid crystal, disordered liquid crystal, and isotropic liquid of oleic acid in the pure liquid state. In other words, PCA provides unambiguous evidence for the phase transitions. similar studies have been carried out for petroselinic acid and nonanoic acid in the pure liquid states, but they do not show any evidence for phase transitions.

Studies on the Polar Lipids Composition in Brown Rice and Milled Rice (현미와 백미의 극성지질의 조성에 관한 연구)

  • The composition of glycolipid, phospholipid and fatty acid of brown rice, milled rice and bran were studied for four varieties of rice such as Nampung, Milyang # 23, Whasung and Jinhung. The content of esterified steryl glycosides(ESG) in glycolipids was the highest in the brown rice, milled rice and bran. Monogalactosyl diglycerides(MGDG) and steryl glycosides(SG) were the second highest. Of the phospholipids, phosphatidyl cholines(PC) + phosphatidyl serines(PS) were the next. Fatty acid composition of glycolipids were different from total lipids and neutral lipids in brown rice and milled rice. Palmitic acid content was very high and oleic acid content was low in glycolipids. The major fatty acid components of phospholipids in brown and milled rice were also palmitic, linoleic and oleic acid. In the bran, the major fatty acid classes were different from those of milled rice.

Production and Analysis of Oxygenated Unsaturated Fatty Acids from Oleic Acid by Flavobacterium sp. Strain DS5 (Flavobacterium sp. Strain DS5에 의한 Oleic Acid로부터 산화불포화 지방산의 생산 및 분석)

  • Vegetable oils are desirable inexpensive feedstocks for various bioproducts. The content of unsaturated fatty acids such as oleic and linoleic acids are 22% and 55% for soybean oil, 26% and 60% for corn oil, and 61% and 21% for canola oil, respectively. Keto and hydroxy fatty acids are useful industrial chemicals, used in plasticizer, surfactant, lubricant and detergent formulations because of their special chemical properties such as higher viscosity and reactivity compared with other fatty acids. In this study, a microbial isolate, Flavobacterium sp. strain DS5 (NRRL B-14859), was used to convert oleic acid to 10-ketostearic acid (10-KSA) via 10-hydroxystearic acid (10-HSA). Two bioconversion products, 10-KSA and 10-HSA, were quantitatively and qualitatively analyzed using gas chromatography, gas chromatography-mass spectrometry, and $^1H-$ and $^{13}C$-nuclear magnetic resonance. The maximum production of 10-KSA and 10-HSA in flask cultures were 3.4 g/L and 0.5 g/L, respectively. The optimum concentrations of glucose and yeast extract, addition time and volume of oleic acid for 10-KSA production were less than 20 g/L, more than 5 g/L, 18 hand 0.3 ml/50 ml, respectively.

Fatty Acid constituents and Relative Compositions of Reproductive Tract Fluids in Korean Native Cows (한우에 있어서 생식기관액 중 지방산 조성과 조성율)

  • To investigate fatty acid constituents and relative compositions in the fluid of the follicles, oviducts, uterine body and uterine body in Korean native cow, the fluids of the reproductive tract were analyzed using gas chromatography. The samples were taken from various reproductive tract of 23 Korean native cows. q. Caprylic acid (C8:0), myristic acid(C14:0), palmitic acid(C16:0), palmitoleic acid(C16:1), stearic acid(C18:0), oleic acid(C18:1), linoleic acid(C18:2), arachidonic acid(C20:4) were found in the reproductive tracts of the cows, which made 8 kinds of fatty acid in total. 2. Palmitic acid, oleic acid, stearic acid were predominant with 35.67%, 24.98% and 17.52%, respectively. while low levels of fatty acids(<5%) were myristic acid, palmitoleic acid and caprylic acid with 1.75%, 1.28% and 2.69%, respectively. 3. Two kinds of polyunsaturated fatty acids, linoleic acid arachidonic acid were found in the reproductive tracts of cows. 4. Palmitic acid among saturated fatty acids and oleic acid among unsaturated fatty acids were the highest level in all of the reproductive tracts. 5. The Highest level of arachidonic acid was found in the uterine horn. 6. The sum of the palmitic acid and oleic acid were 61.72%, 63.72%, 57.66% and 57.65% for the fluid of follicle, oviduct, uterine horn and uterine body of the cows, respectively. 7. The relative compositions of palmitic acid, palmitoleic acid and caprylic acid were higher during the luteal phase than during the follicular phase. 8. The relative compositions of arachidonic acid was higher during the follicular phase in the fluid of uterine horn and uterine body of the cows. 9. The long chain fatty acid, the palmitic acid, stearic acid, oleic acid and linoleic acidshowed higher relative compositions during the follicular phase(86.49%∼95.51%) than during the luteal phase(85.64%∼88.93%).

  • This experiment was conducted to investigate the oil content, change of fatty acid composition affected by ferlilizer levels in upland and paddy field cultivation of rapeseed, The oil content with the fatty acid such as palmitic, linoleic and linolenic was increased in the winter crop on drained paddy field compared with those of upland field in which oleic and stearic fatty acid was increased. Unsaturated, good quality fatty acid content such as oleic and linoleic acid in the cultivation of upland field was higher by 2-5% than those of paddy field. Oleic and linoleic fatty acid contents showed increased with increment of nitrogen fertilizer up to 15kg/10a, and showed same trend until 80kg/ha fertilization level of phosphate and potassium in upland field but there was no effect in paddy field cultivation.

  • The effect of unreactive coupling agent on mechanical and thermal properties of talc-filled polypropylene (PP) composites was studied. Stearic and oleic acids were introduced as coupling agents, and tensile, flexural, and impact strength, thermal stability and melting transition temperature were measured and analyzed according to the types and concentration of coupling agents. Tensile and flexural strength were enhanced by introduction of coupling agent and the maximum effect was observed at the concentration of 3 wt% of coupling agent. Tensile and flexural strength of PP treated with oleic acid were higher than those of PP treated with stearic acid. but impact strength vice-versa. The reasons for these results were postulated by analyzing morphologies of talc-filled PP observed by SEM.

Quality Test of the Sweet-and-Sour Porks and Deep-Frying Oil from the Chinese Restaurants in Seoul (서울 지역 중화 요리점의 탕수육 및 튀김유의 품질 측정)

  • In this study, we determined the fat content, total fatty acid composition, trans fatty acid (tFA) content, and acid value of twenty samples of sweet-and-sour pork and fifteen samples of used frying oils collected from Chinese restaurants in Seoul. After the extraction of crude fat by the Folch method, the total fat content of the twenty sweet-and-sour porks ranged from 9.93 to 20.04%. The total unsaturated fatty acid content ranged from 50.05 to 81.22%, which mostly consisted of oleic and linoleic acid, while those of total saturated fatty acids were 18.06~49.26%. The tFA content of all of the twenty sweet-and-sour porks tested was less than 0.24 g per 100 g of food. The acid values of the fat extracted from the twenty sweet-and-sour porks ranged from 0.44 to 4.37. In the used frying oils, the ranges of the major compositional fatty acids were as follows: palmitic acid, 4.47~20.28%; oleic acid, 23.43~77.45%; linoleic acid, 5.6~53.06%; stearic acid, 1.81~7.38%. The tFA content in all of the fifteen used frying oils was less than 0.98 g per 100 g of fat, while the acid values of the fifteen used frying oils ranged from 0.27 to 2.41.
