Sex Determination used Sex Determining Region Y Gene on the Y-chromosome of Human Teeth (사람 치아 Y염색체상의 sex determining region Y(SRY)유전자를 이용한 성별감정)

  • 최근 중합효소연쇄반응을 이용한 분자생물학적 유전자분석기술의 발달로 성염색체상의 유전좌위 증폭을 통한 성별감정이 활발히 이루어지고 있다. 그중 사람 Y염색체상에 존재하는 남성 고환의 형성을 유도하는 sex-determining region Y(SRY) gene이 규명되어 유전질환의 조기 발견이나 예방 및 태아의 성별판정 등에 응용되고 있다. 그러나, 치아는 외부 환경에 대한 저항성이 가장 높은 장기로 성별감정 등 법의치과학적 개인식별에 널리 이용되고 있음에도 불구하고, SRY 유전자를 이용하여 치아에서의 성별감정에 대한 연구는 시도된 바 없다. 따라서, 본 연구에서는 사람 치아에서 중합효소연쇄반응법을 이용한 SRY 유전자를 검출하여 성별판정에 용용하고자 하였다. 남녀 각각 20개 치아의 치수와 상아질에서 DNA를 추출하여 중합효소연쇄반응 을 시행하고 SRY 유전자를 검색한 결과, 남성에서는 치수 13개중 8개, 상아질 7개중 4개에서 SRY 유전자가 검출되었고, 여생에서는 검출되지 않았다. 이러한 결과는 중합효소연쇄반응법을 이용하여 사람 치아에서 SRY 유전자를 검색할 때, 남성판별에 유용하고 치아를 이용한 성별감정시 기존의 성별감정에 이용되고 있는 다른 유전자와 함께 SRY 유전자를 검색함으로써 성별감정의 신뢰도를 높힐 수 있을 것으로 사료된다.

Daily Serum Collection after Acellular Dermal Matrix-Assisted Breast Reconstruction

  • Background The acellular dermal matrix (ADM)-assisted breast reconstruction technique is widely known, but discouraging results due to early postoperative complications have been reported. As the literature identifies seroma as the most common issue after breast surgery without identifying its pathogenesis, we aimed to report the trend of postoperative daily serum collection after ADM-assisted breast reconstruction and compare it with data in the literature in order to discover more about this little-known topic. Methods A retrospective study on 28 consecutive patients who received ADM-assisted breast reconstruction between February 2013 and February 2014 was performed. In order to reduce the number of variables that could affect serum production, only one brand of ADM was used and all tissues were handled gently and precisely. The daily drainage volume was recorded per patient during the first four days of hospitalization. Likewise, postoperative complications were noted during routine follow-up. Results In total, five (17.9%) bilateral and 23 (82.1%) unilateral ADM-assisted breast reconstructions (33 implants) were performed. The mean age, body mass index, and length of hospital stay were 53.6 years, $21.3kg/m^2$, and 4.5 days, respectively. One major complication led to implant loss (3.0%), and nine minor complications were successfully treated with ambulatory surgery (27.3%). Serum collection linearly decreased after 24 hours postoperatively. Conclusions Daily drainage decreased following the theoretical decline of acute inflammation. In concordance with the literature, daily serum production may not be related to the use of ADM.

  • The wide use of chairside CAD/CAM restorations has increased the diversity of the restorative material. For the practitioner, the selection of the appropriate material is difficult amongst the variety offered by the market. Information on the characteristics of the products can be difficult to assess due to the lack of up-to-date classification and the lack of reliability of manufacturer's advertising. The purpose of this article is to structure the data on restorative materials provided by various sources in order for the practitioner to choose the product most suited to the clinical situation. The objective is to classify chairside CAD/CAM materials and to define their characteristics and indications.

  • SOX9과 SRY 유전자는 척추동물에서 남성고환의 형성을 유도하는 요소로 알려졌다. SOX9 유전자는 SRY related HMG box gene중 하나로 유전질환의 XY성전환 및 성을 결정하는 데에 관여하며 성결정시기에 그 양에 따른 성전환 발생등 연구가 진행되고 있다. 그러나 이 유전자가 성별판정에 유용할 지는 확실치 않다. 반면 SRY 유전자는 포유동물에서의 배형성시기 고환형성을 결정하는 Y염색체 유전자로 남성에만 존재하고 여성에는 존재 않는다. 현재까지 이을 이용하여 법의학적 검체에서 남성판별에 유용하게 사용되고 있다. 본 실험에서는 X, Y와 같은 성염색체가 아닌 상동염색체상에 있으면서 SRY 유전자와 더불어 남성고환을 결정하는 또다른 요소로서의 기능을 가진 SOX9 유전자를 치아에서 검출하여 법의학적 성별판정에 유용할 수 있는지 알아보고자 본 연구를 수행하였다. 남녀각각 5개의 치아에서 치수와 상아질을 분리한 후 DNA를 추출하여 SOX9과 SRY 유전자의 특이적인 시발체를 제작하고 중합효소연쇄반응을 시행하여 증폭하고 전기영동을 시행하였다. 그 결과 SOX9 유전자는 남녀모두에서 유전자가 검출되었고, SOX9 유전자산물과 SRY 유전자를 혼합하여 사용시 남자에서만 유전자가 검출되었다. 이는 법의치과학적 성별판정에 있어 SOX9 유전자는 사람의 치아에서는 남녀 모두 존재하며 남녀 구별을 위한 성별판정에는 이용할 수 없으며 SRY 유전자와 함께 적용시 남성 특이적 SRY 유전자 검사중 발생할 수 있는 가성 음성 반응여부를 확인하는 데 유용할 것으로 사료된다.

  • Forensic odontology can be defined in many ways. One of definitions is simply that forensic odontology represents the overlap between the dental and the legal professions. The most important function of forensic odontology to determine of human identity from dental character. We had found 7 unidentified human bodies in Busan metro politan city, 2007 and we have identified 5 bodies among them.

  • Forensic odontology in individual identification has been progressing rapidly and its significance has been increasing. But not all individual identification can be done single method solely. Therefore, understanding the advantage and disadvantage of each method is necessary to achieve forensic individual identification properly in each case for it to be applied promptly, economically and precisely. Now 4 cases of forensic odontology are studied. 2 cases are age identification and 2 cases is forensic confirmation.

Serum level changes of long chain-polyunsaturated fatty acids in patients undergoing periodontal therapy combined with one year of omega-3 supplementation: a pilot randomized clinical trial

  • Purpose: We aimed to investigate the impact of nonsurgical periodontal treatment combined with one-year dietary supplementation with omega (${\omega}$)-3 on the serum levels of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), docosapentaenoic acid (DPA), and arachidonic acid (AA). Methods: Fifteen patients with chronic generalized periodontitis were treated with scaling and root planing. The test group consisted of seven patients ($43.1{\pm}6.0$ years) supplemented with ${\omega}$-3, consisting of EPA plus DHA, three capsules, each of 300 mg of ${\omega}$-3 (180-mg EPA/120-mg DHA), for 12 months. The control group was composed of eight patients ($46.1{\pm}11.6$ years) that took a placebo capsule for 12 months. The periodontal examination and the serum levels of DPA, EPA, DHA, and AA were performed at baseline (T0), and 4 (T1), and 12 (T2) months after therapy. Results: In the test group, AA and DPA levels had been reduced significantly at T1 (P<0.05). AA and EPA levels had been increased significantly at T2 (P<0.05). The ${\Delta}EPA$ was significantly higher in the test compared to the placebo group at T2-T0 (P=0.02). The AA/EPA had decreased significantly at T1 and T2 relative to baseline (P<0.05). Conclusions: Nonsurgical periodontal treatment combined with ${\omega}$-3 supplementation significantly increased the EPA levels and decreased the AA/EPA ratio in serum after one year follow-up. However, no effect on the clinical outcome of periodontal therapy was observed.

Orthodontic appliances and MR image artefacts: An exploratory in vitro and in vivo study using 1.5-T and 3-T scanners

  • Purpose: The aim of this study was to assess the artefacts of 12 fixed orthodontic appliances in magnetic resonance images obtained using 1.5-T and 3-T scanners, and to evaluate different imaging sequences designed to suppress metal artefacts. Materials and Methods: In vitro, study casts of 1 adult with normal occlusion were used. Twelve orthodontic appliances were attached to the study casts and scanned. Turbo spin echo (TSE), TSE with high readout bandwidth, and TSE with view angle tilting and slice encoding for metal artefact correction were used to suppress metal artefacts. Artefacts were measured. In vivo, 6 appliances were scanned: 1) conventional stainless-steel brackets; 2) nickelfree brackets; 3) titanium brackets; 4) a Herbst appliance; 5) a fixed retainer; and 6) a rapid maxillary expander. The maxilla, mandible, nasopharynx, tongue, temporomandibular joints, and cranial base/eye globes were assessed. Scores of 0, 1, 2, and 3 indicated no artefacts and minor, moderate, and major artefacts, respectively. Results: In vitro, titanium brackets and the fixed retainer created minor artefacts. In vivo, titanium brackets caused minor artefacts. Conventional stainless-steel and nickel free brackets, the fixed retainer, and the rapid maxillary expander caused major artefacts in the maxilla and mandible. Conventional stainless-steel and nickel-free brackets caused major artefacts in the eye globe (3-T). TSE with high readout bandwidth reduced image artefacts in both scanners. Conclusion: Titanium brackets, the Herbst appliance, and the fixed retainer caused minor artefacts in images of neurocranial structures(1.5-T and 3-T) when using TSE with high readout bandwidth.
