Changes of Volatile Components of Pleurotus sajor-caju During Storage (여름 느타리 버섯 (Pleurotus sajor-caju) 저장중의 향기성분 변화)

  • Volatile aroma concentrates of Pleurotus sajor-caju which stored at $20^{\circ}C$ incubator and cold room were obtained by simultaneous distillation-extraction method. The volatile components were identified by combined capillary gas chromatography-mass spectrometry and comparison of retention data on GC of those authentic compounds. Storage at room temperature resulted in significant decreased in their total aroma contents, whereas little changes at cold storage foured, even though appreciable changes were observed in GC pattern between the control and stored samples. Of 38 components identified, ${\delta}-decalactone$ increased while rest of the volatiles decreased rapidly during storage at room temperature, but at cold storage 2-methyl butanal, 1-octen-3-ol, cis,cis-1,5-octadien-3-ol and pentadecanoic acid decreased while 3-octanone, 1-octen-3-one, 3-octanol and 5-undecanol increased with the storage time. The weight losses and organoleptic properties changed extremely by the storage under room temperature, on the other hand, slight changes were found in cold storage.

Optical Resolution of Racemic Ibuprofen by Candida Rugosa Lipase Catalyzed esterification (Candida Rugosa Lipase에 의한 Ibuprofen 에스테르화 반응과 광학분할)

  • The enantioselective esterification of racemic ibuprofen catalyzed by a Candida rugosa lipase was studied according to reaction conditions such as a lipase concentration, reaction temperature, alcohol chain length and alcohol concentration. The S-(+)-ibuprofen alkyl esters prepared were converted to S-(+)-ibuprofen by hydrolysis with sulfuric acid as a catalyst. High conversions in the esterifications were obtained at 60$^{\circ}C$ and an equimolar ratio of octanol to ibuprofen. The initial reaction rate of the esterification decreased with increasing octanol concentration. Conversion and initial reaction rate increased with increasing alcohol chain length. Values of enantiomeric excess(ee) according to esterification reaction conditions did not change below 60$^{\circ}C$. On the other hand, values of conversion and ee for the chemical hydrolysis of S-(+)-ibuprofen alkyl esters were independent of alcohol alkyl chain length. Optical resolution of racemic ibuprofen was achieved by lipase catalyzed esterification and chemical hydrolysis. The separation method provided a high yield and enantioselectivity for the production of S-(+)-ibuprofen from racemic ibuprofen.

Enhancement of $NH_3$ Bubble Absorption Performance in Binary Nanofluids (이성분 나노유체에서의 암모니아 기포 흡수 성능 향상)

  • The objectives of this paper are to study the absorption characteristics of $NH_3$ bubbles in the binary nanofluids and to quantify the effects of surfactants and nano-particles on the bubble absorption performance. 2-Ethyl-1-Hexanol, n-Octanol, and 2-Octanol are used as the surfactants and nano-sized $Al_{2}O_3$ and Cu particles are added to make the binary nanofluids into $NH_3/H_{2}O$ solution. The concentration of $NH_3$ solution ($x_s$), the concentration of surfactants ($x_{SA}$), and the mass fraction of nano-particles ($w_{np}$) are considered as key parameters. The experimented ranges of $x_s,\;x_{SA},\;and\;w_{np}$ are $0{\sim}17.92\%,\;0{\sim}1,500\;ppm\;and\;0{\sim}0.2\%$, respectively. The absorption rates are calculated by measuring initial and final weights of test section and exposed time. In addition, the bubble absorption processes are visualized using the shadow graphic method. The results show that the absorption performance is significantly enhanced up to 4 times by adding the surfactants and up to 3 times in the binary nanofluids.

Analysis of the Volatile Flavor Compounds Produced during the Growth Stages of the Shiitake Mushrooms (Lentinus edodes)

  • Volatile flavor components, produced during the young (P-1), immature (P-2), mature (P-3) and old (P-4) growth stages, of shiitake mushrooms (Lentinus edodes), were extracted by simultaneous steam distillation and extraction (SDE), using a mixture of n-pentane and diethyl ether (1:1, v/v) as the extraction solvent. Analyses of the concentrates, by capillary gas chromatography (GC) and gas chromatography-mass spectrometry (GC/MS), led to the identification of 129, 129, 111 and 120 components in the P-1, 2, 3 and 4 stages, respectively. The major volatile compounds were l-octen-3-o1, 3-octanol, 3-octanone and 4-octen-3-one. Ethanol and ethyl acetate were also detected in large amounts. The characteristic volatile compounds found in shiitake mushrooms, such as dimethyl disulfide, dimethyl trisulfide and 1, 2, 4-thiolane, were at low concentrations in all samples. The amount of l-octen-3-o1 decreased as growth progressed, but concentrations of 3-octanone increased. The amount of 4-octen-3-ol decreased from P-1 to P-3, but was at a high concentrations in P-4. The concentration of 3-octanol gradually increased and reached its highest concentration in P-3, but decreased in P-4. The C8-compounds comprised 70.91, 64.09, 64.29 and 60.01 % in the P-1, 2, 3 and 4 stages, respectively, so decreased gradually with growth. The S-compounds were found in the highest concentrations in P-3.

Monitoring of Volatile Flavor Components and Amino Acids in Fresh Mushrooms (Agaricus bisporus) Associated with Shelf-Life Extension (양송이의 숙도지연에 다른 휘발성 향기성분과 아미노산의 변화)

  • Some chemical constituents were monitored to evaluate the biochemical and nutritional aspects of ${\gamma}-irradiated$ mushrooms associated with shelf-life extension. Volatile components identified by GC and GC-MS were composed primarily of 1-octen-3-ol(68%)). benzaldehyde(13%), 3-octanone(8%), benzyl alcohol(5%), 3-octanol(2%)). 1-octen-3one(1%). etc. Treatment with 2kGy-irradiation and subsequent storage for 17 days at $(9{\pm}1^{\circ}C\;and\;80{\pm}7%$ RH resulted in appreciable changes In their contents. even though negligible changes were observed in GC patterns between the nonirradiated and 2 kGy-irradiated samples. Most of the amino acids were resistant to ionizing energy of 2 kGy, while sulfur-containing free amino acids were affected significantly by ${\gamma}-irradiation$.

  • A study on the solvent extraction of hexavalent chromium from aqueous solutions has been investigated. The results showed that the extraction efficiency of chromium increased in proportion to extractant concentration. Aliquat 336 of quarternary amine effectively extracted hexavalent chromium and was superior to Alamine 336. And the stabilizers like polyhydric alcohols of octanol, decanol and dodecanol showed the similar effect fur prevention of third phase and a little promotion of extraction efficiency in comparison with non-use of polyhydric alcohols. On the other hand, in solvent extraction of hexavalent chromium from aqueous solutions, the hexavalent chromium was completely extracted at pH range lower than 7 with 1% Aliquat 336 as extractant and 5% decanol as stabilizer and the hexavalent chromium in extractant was completely stripped with 1M sodium hydroxide solution in the stripping step.

Air-Soil Partitioning of PCBs in Rural Area

  • The soil concentrations of polychlorinated biphenyls (PCBs) were measured at 12 sites in Ansung, Kyonggi province, Korea. Correlation coefficient (r) between total PCBs and organic matter content (OM) was significant (r=0.562, p< 0.05). It suggests that organic matter may be a key factor of soil absorption of PCBs. The PCB concentrations of low chlorinated congeners with high vapor pressure were relatively abundant in air but high chlorinated congeners with low vapor pressure were mainly dominated by soil. The results indicated the influence of physicochemical properties of PCBs such as vapor pressure, octanol - air partition coefficient ( $K_{OA}$ ). The calculated soil/air fugacity quotients suggested that the soil may be a source of heavier molecular PCBs (>penta-CBs) to the atmosphere, where lighter molecular PCBs appear to be affected by a movement from air to soil, especially tetra-CBs. Therefore, PCB homologs with low vapor pressure might have been influenced by revolatilization from soil.

  • A study of the POSE (predispersed solvent extraction) for succinic acid by colloidalliq­uid aphrons was conducted. The organic phase contaning TOA (tri-n-octylamine) and 1-octanol permits a selective extraction of succinic acid from its aqueous solution. There was no difference of the extractability of POSE and that of conventional mixer-settler type extraction. Taking into account the no mechanical mixing in POSE, it was concluded that the POSE process is more adaptive than the conventional mixer-settler type extraction process. From mass transfer analysis at the various concentration of TOA in counter-current continuous operation, the concentration of TOA had no influence on the mass transfer coefficient. The loading values in continuous POSE were almost same as those in batch operation.

Pharmacokinetic Study on DWC-751, a New Cephalosporin, in Rats and Mice (신규 세파로스포린계 항생물질 DWC-751의 흰쥐 및 생쥐 체내동태)

  • The distribution and excretion of DWC-751, a new cephalosporin, were examined in rats and mice following a single intravenous administration. DWC-751 in plasma and urine was determined by both HPLC and microbiological assay. The plasma concentration of the drug declined biexponentially. The initial and terminal half lives of the drug were 3.0 and 28.3 min, respectively. Binding of the drug to plasma proteins was 42.3%. The distribution volume at steacly-state ($Vd_{ss}$) was only 0.341 ι/kg, which is well correlated with the low n-octanol/water partition coefficient of the drug ($K_{o/w{\cong}0$) Actually, the drug was distributed to liver, kidney and lung with very low organ/plasma concentration ratio. The drug, was excreted mainly via renal excretion, i.e., the total($CL_T$) and apparent renal($CL_{R}$) clearances of the drug were 10.8 and 7.5 ml/min/kg, respectively.

Studies on Oral Absorption of Piperacillin Pivaloyloxymethyl Ester (피페라실린피발로일옥시메칠에스텔의 경구흡수에 관한 연구)

  • Pivaloyloxymethyl ester of piperacillin was synthesized by reacting sodium piperacillin with chloromethyl pivalate, and its chemical structure was determined by infrared and $^1H$ nuclear magnetic resonance spectroscopic methods. The pharmaceutical properties of the ester were investigated to assess its potential as a novel prodrug of piperacillin. The interface transfer of piperacillin and the ester was studied in a two-phase in vitro system composed of aqueous pH buffers and n-octanol. The ester was more lipophilic, and less water soluble above pH 4.0 than piperacillin. Significant antibacterial activity was not observed in the ester in vitro, but the ester was hydrolyzed into the parent drug in the rat liver homogenate. The serum levels of orally administered ester suspension containing 0.1% Tween 80 were measured in rabbits. It was found that the ester showed higher blood level, comparing with no observation of piperacillin in serum, but the time reaching the maximum serum concentration was 5 hr.
