황금탕이 Streptozotocin으로 유발된 당뇨병 백서의 혈당 및 당뇨병합병증에 미치는 영향 (Effects of Hwanggeum-tang Aqueous Extracts on Streptozotocin-induced Rat's Diabetes and Related Complications)

  • The object of this study was to observe the effects of Hwanggeum-tang (HGT) aqueous extracts on Streptozotocin-induced rat's diabetes and related complications. Three different dosages of HGT extracts were orally administered oncea day for 28 days from 3 weeks after Streptozotocin treatment (60 mg/kg, single intraperitoneally administered). All the rats were checked at 3 weeks after Streptozotocin treatment as follows. Changes on the body weight, blood glucose level, kidney weight, serum BUN and creatinine level, liver weight, serum AST and ALT level, serum LDL, HDL, triglyceride and total cholesterol level were observed with changes on the pancreatic MDA content and GSH content. The results were compared with a potent antioxidant silymarin 100 mg/kg in which the effects on Streptozotocin-induced diabetes and related complications were already confirmed. As results of Streptozotocin-injected diabetes and related complications, dramatical decreases on the body weight, increase of the kidney and liver weight, increase of serum BUN, creatinine, AST, ALT, LDL, triglyceride, total cholesterol level and decreases of serum HDL level were detected in streptozotocin control as compared with intact control. In addition, marked increases of pancreatic MDA content and decreases of GSH content were also detected in streptozotocin control as compared with intact control. However, these diabetes and related complications, and inhibition of antioxidant effects induced by Streptozotocin were inhibited by 28 days continuous treatment of 50, 100 and 200 mg/kg of HGT extracts in the present study. HGT have favorable effects on the diabetes and various diabetic complications. Therefore, more detail mechanism studies should be conducedin future with the efficacy tests of individual herbal composition of HGT and the screening of the biological active compounds in herbs.

이진탕가미방(二陳湯加味方)의 항산화 효과를 통한 역류성 식도염 억제효과 (Protective Effects of Yijin-tang-gamibang Aqueous Extracts on Reflux Esophagitis Mediated by Antioxidant Defense Systems)

  • The object of this study was to observe the protective effects of Yijin-tang-gamibang (YJGMB), Yijin-tang has been traditionally used in the Korean Medicine for treating various digestive diseases. The study showed that it is effective on reflux esophagitis induced by pylorus and forestomach ligation in rats. Three different dosages of YJGMB extracts, 200, 100 and 50 mg/kg, were orally pretreated, once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the present study. The results were compared with omeprazole, antioxidant and proton pump inhibitor, 30 and 10 mg/kg treated group. YJGMB 200 mg/kg were showed similar protective effects as compared with 30 mg/kg of omeprazole but more favorable effects were observed in 200, 100 and 50 mg/kg of all YJGMB treated rats as compared with omeprazole 10 mg/kg in the present study. In addition, YJGMB 200 mg/kg were showed more favorable antioxidant effects as compared with 30 mg/kg of omeprazole in the present study. Detail mechanism studies should be conduced in future with the screening of the biological active chemical compounds in herbs.

Collagen으로 유발된 마우스의 관절염에 대한 의이인탕(薏苡仁湯) 추출물의 억제 효과 (Suppress Effects of Euiiin-tang(yìyĭrén-tāng) Aqueous Extracts on Collagen Induced Arthritic(CIA) DBA/1 Mice)

  • Objectives : The object of this study was to observe the favorable anti arthritic effects of Euiiin-tang($y{\grave{i}}y{\breve{i}}r{\acute{e}}n-t{\bar{a}}ng$) aqueous extract(EIITe), has been traditionally used in Korean medicine for treating rheumatoid arthritis on collagen induced arthritic(CIA) DBA/1 mice. Methods : In the present study, effects of EIITe on the releases of human tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$, matrix metalloproteinase(MMP)-13 and production of Nitric oxide(NO) were observed by in vitro. In addition, to observe the effects on the CIA mice, three different dosages of EIITe, 300, 150 and 150 mg/kg were orally administered once a day for 18 days from 24hrs after antigen challenges(type II collagen) on 21 days after immunization using Type II collagen Freund's complete adjuvant. Six groups, each of 8 DBA/1 mice per group were used in the present study as follows. Changes on the body weights, macroscopic arthritis scores, splenic weights, splenic TNF-${\alpha}$ and IL-6 contents, articular cartilage(femur and tibia) collagen and glycosaminoglycans-chondroitin sulphate, sulphate and hyaluronic acid contents, histopathological observations(microscopic arthritis scores, thicknesses of femur and tibia cartilage thicknesses were monitored, compared to that of dexamethasone, a potent anti inflammatory agents, 1 mg/kg treated mice. Results : As results of collagen challenges, marked decreases of body weights and gains, articular cartilage collagen and glycosaminoglycan - chondroitin sulphate, sulphate and hyaluronic acid contents were observed with increases of macroscopic arthritis scores, splenic weights, splenic TNF-${\alpha}$ and IL-6 contents, articular cartilage(in the both femur and tibia) loss and damages. However, these CIA signs were significantly and dosages dependently inhibited by treatment of EIITe 300 and 150 mg/kg as compared with CIA control, respectively. In addition, the releases of TNF-${\alpha}$, IL-$1{\beta}$, NO and MMP-13 were markedly and dose dependently inhibited by treatment of EIITe, invitro. Although CIA were more favorably inhibited by treatment of dexamethasone 1 mg/kg as compared with EIITe 300 mg/kg, marked decreases of body weights were detected in dexamethasone 1 mg/kg treated mice. Conclusions : The results obtained in this study suggest that over 150 mg/kg of EIITe showed favorable anti arthritic effects on the CIA mediated by immunomodulatory and/or anti oxidative effects. However, detail mechanism studies should be conduced in future with the screening of the biological active compounds in this herb. lthough CIA were more favorably inhibited by treatment of dexamethasone 1 mg/kg as compared with EIITe 300 mg/kg, marked decreases of body weights were detected in dexamethasone 1 mg/kg treated mice, in the present study.

해상교통안전진단제도의 운영현황과 향후 정책방향에 관한 연구 (A Study on the Present Status and Future Directions of Maritime Safety Audit)

  • 최근 선박의 고속화, 대형화와 더불어 국가경제발전 및 경쟁력 향상을 위해 해상횡단교량, 항만건설 및 해상풍력단지 등 사회기반 시설의 확충사업을 적극 추진중이다. 국토해양부에서는 이렇게 선박통항로에 설치되는 각종 시설물의 설치 보수공사 등이 선박의 안전항행에 미치는 정도를 사전에 평가하여 잠재적 위험요인을 미연에 제거하기 위해 해상교통안전법 개정을 통해 해상교통안전진단제도를 도입하였다. 본 논문에서는 해상교통안전진단제도 도입 및 운영을 통해 도출된 약 2년간의 성과와 문제점의 고찰을 통해 제도개선 및 발전을 위한 정책방향을 제시하였다. 진단대상사업 및 진단항목 결정 등의 객관성 및 효율성을 높이기 위해 스크리닝 및 스코핑 도입 검토 및 안전진단제도를 국토해양부 중심의 협의형 제도에서 벗어나 사업계획 지원형 제도로 발전할 수 있도록 관련법령 개정 등 정책 개선사항을 제시하였다. 이를 통해 안전진단제도에 대한 올바른 이해를 유도하고, 제도관련 하위법령 개정시 기초 자료로 활용하고자 한다.

지면 투과 레이더(GPR) 기반의 지뢰 탐지 시스템을 위한 표적 후보 검출 기법 (A Preprocessing Method for Ground-Penetrating-Radar based Land-mine Detection System)

  • 최근 금속을 포함하여 다양한 재질의 지뢰를 탐지할 수 있는 지면 투과 레이더 (Ground Penetrating Radar, GPR)가 지뢰 탐지 분야에서 주목 받고 있으며 지면 투과 레이더를 이용한 지뢰 탐지 기술이 활발하게 연구되고 있다. 본 논문에서는 1차원 지면 투과 레이더 배열을 이용하여 지뢰가 존재할 가능성이 높은 후보 지점을 검출하는 방법을 제안하였다. 제안 기법은 먼저 지면 투과 레이더에 수신되는 신호 세기의 비선형적 감쇠 특성을 반영하여, 지연 시간에 따른 지면 투과 레이더 응답 신호의 세기를 효과적으로 나타낼 수 있는 회귀 모델을 추정하였다. 이를 토대로 지면 투과 레이더 응답 신호를 분석해보면, 토양 신호들은 주로 회귀 모델의 근처에 밀집되어 있고, 지뢰일 가능성이 있는 후보 신호들은 회귀 모델로부터 멀리 떨어져서 분포하는 특성을 보인다. 이러한 사실을 기반으로, 지면 투과 레이더의 측정값과 회귀 모델의 예측 값에 대한 예측 오차를 계산하여 토양 신호로부터 지뢰일 가능성이 있는 후보 신호들을 분리하였다. 후보 신호에는 잡음과 표적 신호가 섞여 있으므로 표적 신호가 갖는 고유한 기하학적 특성을 이용하여 최종적으로 표적 신호를 검출하였다. 성능 검증을 위해 실내 환경에서 획득한 지면 투과 레이더 데이터를 이용하여 실험을 수행한 결과, 제안하는 전처리 기법의 지뢰 탐지율 대비 오경보율이 낮은 것을 확인할 수 있었다.

공공기관의 사내벤처제도 개선방안;A사 사례연구 (Study on Internal Corporate Venture Business in Public Companies;Based on A Public Company)

  • 본 논문은 공공기관의 사내벤처제도 개선방안에 대하여 분석하여 보았다. 연구방법은 문헌조사, 국내외 기업 사례조사 및 A공사의 사내벤처제도 운영실태에 대한 종업원의 설문조사와 인터뷰조사 방법을 사용하였다. 분석결과 A공사의 사내벤처제도 문제점은 다음과 같다. 첫째, 사내벤처제도에 대한 CEO의 강력한 추진의지와 이를 수행할 수 있는 조직분위기 형성의 지속성이 미흡하다. 둘째, 실제 사업을 수행할 종업원의 역량개발 노력이 부족하다. 셋째, 사업아이템 발굴 및 타당성 평가에 있어 종업원의 수용성과 객관성 제고가 필요하다. 넷째, 사내벤처팀의 성과측정을 위한 명확한 기준 및 측정시스템이 미흡하다. 다섯째, 성과에 비례한 보상이 제한적인 범위 내에서 이루어진다. 마지막으로, 사내벤처팀이 별도법인으로 분사 시 모기업의 지분 출자 및 자금지원이 이루어 지지 못하고 있다. 이러한 문제점에 대한 개선방안은 다음과 같다. 첫째, CEO의 강력하고도 일관성 있는 제도 후원 의지 천명과 함께 종업원들이 주도가 된 도전적이고 창의적인 조직문화의 확산이 필요하다. 둘째, 공사는 종업원의 역량제고를 위하여 창업관련 지식 및 개인의 역량개발 프로그램 제공과 이를 공유할 수 있는 정보공유 시스템을 갖추어야 한다. 셋째, 사업아이템 발굴 및 타당성 평가에 있어 외부 전문가의 컨설팅 제공 및 타당성 평가결과에 대한 명확한 feed-back을 통하여 수용성 제고와 함께 종업원의 참여의지를 제고하여야 한다. 넷째, 사내벤처팀의 성과측정을 위하여 자율성과 독립성을 전제로 한 명확한 목표부여 및 벤처팀 특성에 맞는 세부 평가시스템을 마련하여야 한다. 다섯째, 사내벤처 참여자에 대하여 성과에 비례하는 보상을 강화하여 실질적인 동기부여가 될 수 있도록 하여야 한다. 마지막으로, A공사는 벤처사업에 대한 사후관리 강화 차원에서 기존의 기술개발과제 지원제도 활용 및 중소기업 창업투자조합 결성을 통하여 분사하는 사내벤처팀에 대한 지분 출자 및 자금지원을 할 수 있도록 하여야한다.

역류성식도염 유발 흰쥐에 대한 이진탕가미방(二陳湯加味方)의 효과 (Effects of Yijintang-gamibang on Reflux Esophagitis Induced by Pylorus and Forestomach Ligation in Rat)

  • Purpose : The object of this study was to observe the suppressive effects of Yijintang-gamibang (YJGMB), Yijintang with Atractylodis Rhizoma, Massa Medicata Fermentata, Hordei Fructus Germiniatus, and Coptidis Rhizoma. YJGMB has been traditionally used in Korean medicine for treating various digestive diseases. We tested it on the rat reflux esophagitis (RE) induced by pylorus and forestomach ligation in rats as compared with omeprazole, a well-known proton pump inhibitor. Method : Three different dosages of YJGMB 200, 100 and 50mg/kg, were orally pretreated once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the study. Six hours after pylorus and forestomach ligation, changes of the stomach and esophagus lesion areas, gastric volumes, acid and pepsin outputs, invasive lesion percentages, fundic mucosa and total thicknesses were measured as histomorphometry. The results were compared with omeprazole, antioxidant and proton pump inhibitor, and 30 and 10mg/kg treated groups in which the effects on RE were already confirmed. Results : As results of pylorus and forestomach ligation, marked increases of esophageal and gastric mucosa lesion areas, gastric volumes, acid outputs, pepsin outputs were observed with histopathological changes of RE, such as hemorrhages, ulcerative lesions and edematous changes on the fundic mucosa. However, these pylorus and forestomach ligation-induced RE were dose-dependently inhibited by treatment of 200, 100 and 50mg/kg of YJGMB. YJGMB 200mg/kg showed similar protective effects as compared with 30mg/kg of omeprazole in the present study, and more favorable effects were observed in 50mg/kg of YJGMB treated rats as compared with omeprazole 10mg/kg in the present study. Conclusion : The results obtained in this study suggest that YJGMB has favorable protective effects on the RE induced by pylorus and forestomach ligation. Therefore, it is expected that YJGMB will also show favorable effects on RE corresponding well to the suggestion of traditional Korean medicine. However, more detailed mechanism studies should be conducted in future with the screening of the biological active chemical compounds in herbs.

  • Expressed sequence tags (EFTs) are the partial segments of cDNA produced from 5 or 3 single-pass sequencing of cDNA clones, error-prone and generated in highly redundant sets. Advancement and expansion of Genomics made biologists to generate huge amount of ESTs from variety of organisms-human, microorganisms as well as plants, and the cumulated number of ESTs is over 5.3 million, As the EST data being accumulate more rapidly, it becomes bigger that the needs of the EST analysis tools for extraction of biological meaning from EST data. Among the several needs of EST analyses, the extraction of protein sequence or functional motifs from ESTs are important for the identification of their function in vivo. To accomplish that purpose the precise and accurate identification of the region where the coding sequences (CDSs) is a crucial problem to solve primarily, and it will be helpful to extract and detect of genuine CD5s and protein motifs from EST collections. Although several public tools are available for EST analysis, there is not any one to accomplish the object. Furthermore, they are not targeted to the plant ESTs but human or microorganism. Thus, to correspond the urgent needs of collaborators deals with plant ESTs and to establish the analysis system to be used as general-purpose public software we constructed the pipelined-EST analysis system by integration of public software components. The software we used are as follows - Phred/Cross-match for the quality control and vector screening, NCBI Blast for the similarity searching, ICATools for the EST clustering, Phrap for EST contig assembly, and BLOCKS/Prosite for protein motif searching. The sample data set used for the construction and verification of this system was 1,386 ESTs from human intrathymic T-cells that verified using UniGene and Nr database of NCBI. The approach for the extraction of CDSs from sample data set was carried out by comparison between sample data and protein sequences/motif database, determining matched protein sequences/motifs that agree with our defined parameters, and extracting the regions that shows similarities. In recent future, in addition to these components, it is supposed to be also integrated into our system and served that the software for the peptide mass spectrometry fingerprint analysis, one of the proteomics fields. This pipelined-EST analysis system will extend our knowledge on the plant ESTs and proteins by identification of unknown-genes.

뇌졸중 환자의 운전재활 시기 결정을 위한 K-MBI의 임상적 유용성: 예비 연구 (Clinical Usefulness on K-MBI for Decision of Driving Rehabilitation Period in Patients with Stroke: A pilot study)

  • 연구배경 및 목적: 수정된 바델지수(MBI)는 지역사회 재활세팅 및 재활 병원에서 흔히 사용되어지는 기본적인 일상생활 평가 도구이다. 선행 연구들에 의하면 노인 혹은 뇌졸중 환자들의 운전 능력과 일상생활활동의 수준은 유의미한 상관성이 있음을 보고된 바 있다. 그러나, 뇌졸중 환자의 운전능력을 예측함에 있어 임상에서 흔히 사용되는 일상생활 활동 기본 평가인 수정된 바델지수의 유용성을 밝힌 연구는 미비하다. 본 연구에서는 한국형 수정된 바델 지수(K-MBI)룰 활용하여 뇌졸중 환자의 운전 가능성을 알아봄으로서 운전 수행 능력을 예측함에 있어 한국형 수정된 바델 지수의 임상적 유용성을 알아보고자 하였다. 연구방법: 서울 소재 재활병원에서 재활치료를 받고 있는 뇌졸중 환자 48명이 연구에 참여하였다. 모든 연구 대상자에게 K-MBI 검사를 통해 기본적인 일상생활 활동 수준을 측정하였고 가상현실 운전시뮬레이터를 이용하여 운전 수행능력을 측정하였다. 운전 시뮬레이터상의 운전 수행 검사의 합격/불합격에 따른 K-MBI 점수의 예측 타당도를 ROC curve를 이용하여 검증하였다. 결과: 뇌졸중 이후 운전재활을 권고할 만한 기본적인 일상생활 활동 수준은 K-MBI 점수가 최소 86.5점인 경우가 적절한 것으로 나타났다. 결론: 본 연구는 뇌졸중 환자의 운전 재활을 권고하는 시기를 예측함에 있어 유용한 기초 자료가 될 것으로 판단된다.

Adjuvant 유발(誘發) 류마티스 관절염(關節炎)에 대한 소활락단(小活絡丹)의 억제(抑制) 효과(效果) (Suppressing Effects of Sowhalrack-dan(Xiǎohuóluò-dān) on Freund's Complete Adjuvant Induced Arthritic Rats)

  • Objectives : The object of this study is to observe the favorable anti arthritic effects of Sowhalrack-dan($Xi\check{a}ohu\acute{o}lu\grave{o}-d\bar{a}n$)(SWRD), which has been traditionally used in Korean medicine to treat rheumatoid arthritis on Freund's complete adjuvant(FCA) induced arthritic Wistar rats. Methods : Rheumatoid arthritis was induced by intradermal injection of FCA(10 mg in 1 ml paraffin oil 0.1 ml/rats). Each of 8 rats showing regular ankle circumferences per group were selected in 14 days after FCA treatment to confirm the induction of rheumatoid arthritis. 300, 150 or 75 mg/kg of SWRD was orally administered once a day for 14 days from 14 days after FCA treatments. Dexamethasone was intraperitoneally administered 15 mg/kg, once a day for 14 days from 14 days after FCA treatments. Rats were sacrificed after 14 days of continuous oral treatment of SWRD or intraperitoneal administration of dexamethasone, and changes were observed; the body weight, knee circumferences, gross arthritis score, inflammatory tissue $prostaglandin(PG)E_2$ levels and cartilage collagen, glucosaminoglycans compositions - chondroitin sulphate, heparin sulphate and hyaluronic acid in the present study. Results : As results of FCA treatment, classic rheumatoid arthritis featuring dramatical decreases on the body weights, cartilage collagen contents and bone glucosaminoglycans - chondroitin sulphate, heparin sulphate and hyaluronic acid contents. Also, it increases the knee circumferences, gross arthritis scores and inflammatory tissue $PGE_2$ levels. However, these changes from FCA induced rheumatoid arthritis were clearly reduced due to the dexamethasone and both two different dosages of SWRD, 300 and 150 mg/kg in the present study. Although FCA induced arthritis were more favorably inhibited by treatment of dexamethasone 15 mg/kg compared to SWRD 300 mg/kg, marked decreases of body weights were detected in dexamethasone 15 mg/kg treated rats. Conclusions : The results obtained in this study suggest that over 150 mg/kg of SWRD showed favorable anti-arthritic effects on the FCA induced arthritis mediated by suppression of $PGE_2$. However, detailed mechanism studies are needed with the screening of the biological active compounds in SWRD. Although FCA induced arthritis were more favorably inhibited by treatment of dexamethasone 15 mg/kg compared to SWRD 300 mg/kg, marked decreases of body weights were detected in dexamethasone 15 mg/kg treated rats, in the present study.
