Comparisons of Electrical Conductivity between Polyester/Polyurethane and Nylon/Polyurethane Woven or Knitted Fabrics with Silver Paste Patterns in Elongation-Strain test (폴리에스터/폴리우레탄 및 나일론/폴리우레탄에 은 문양을 입힌 편직물의 신장-변형 시 전기 전도도 비교)

  • The objective of this study was to investigate electrical conductivity of fabrics from polyester (PET) and Nylon (N) containing polyurethane (PU), with silver paste patterns screen-stenciled in three directions. The PET/PU and N/PU fabrics knitted or woven were uniaxially strain-recovered up to 22.5% in three times when each change in electrical resistance was simultaneously measured. This study established four variables that complexly affected electrical conductivity of these specimens; fabric structures, components, cover factors, and the percolation of silver particles. The woven or knitted fabric structures did not distinctively cause the changes in electrical resistance, however, the woven fabrics with the diagonal patterns showed their relatively high electrical resistance. The PET/PU fabrics with increasing the PET proportion generally presented the opposite propensity to its electrical conductivity. The changes in electric resistance of the PET/PU 85/15 2/1 twill and double plain fabrics instantaneously responded to the rate of elongation. The PET/PU group exhibited a reverse correlation between its cover factor and electrical resistivity. The highest electrical conductivity of the PET/PU 95/5 interlock fabric, with very few fluctuations, was attributed to the deep percolation of the silver particles that bridged the gaps between one loop and another. On the other hand, the occurrence of the silver cracks along with the elongated direction led to the immeasurably high change in electrical resistance as the strain increased.

  • By Using various grinding mill in powder cosmetics manufacturing process; screen milt and pin mill, jet mill, properties of the powder and grinding mills were studied; talc, mica, nylon powder, silica, titanium dioxide. Besides, the experiments fur evaluation of grinding were performed by using iron oxides those are tracers. In powders of plate shape, they were grinded more vertically than horizontally at the screen mill and pin mill, although were all grinded vertically and horizontally at the jet mill. The spheric powders became the primary particles or aggregation by electrostatic interaction at the screen mill and pin mill. But, at the jet mill, they resulted the agglomeration or transformation or damage up to 2bar. Titanium dioxides became the primary particles by all grinding mill. Pin mill has an excellent result in experiments which is a change of the tone of color by grinding. From these results, suggest that the jet mill is used to pre-treat of powders of plate shape in practical cosmetic manufacturing process, and the screen mill and pin mill are used to match the color of powder cosmetics. If industrial process condition is taken into consideration, suggest that 4times of grinding is excellent on grinding effect by the screen mill, and twice grinding by the pin mill and grind air pressure of 1bar by the jet mill.

  • A newly developed contact angle measurement instrument by laser beam projection allows for rapid and direct determination of contact angles. The instrument may have a possibility to characterize newly developed nano-fibers. When the laser beam impinges on an edge of an interface of liquid and solid, projected beam were split across and made two straight lines on a tangent screen. From the result, it could measure the contact angle directly by reading the angle between two split beams. The purpose of this study was to prove reliability and reproducibility of the contact angle measurement instrument by laser beam projection compare to the conventional one by microscope through the comparative experiment and questionnaire. Test samples were selected by consideration of hydrophilic and hydrophobic, such as nylon 6 and polypropylene, respectively. The laser contact angle measurement has accurate, fast and convenient method to measure contact angle, and it can be a unique method to characterize nano-fibers.

EVALUATING TWO METHODS FOR FINGERPRINTING GENOMES FOR STREPTOCOCCUS MUTANS IN CHILDREN : A COMPARISON WITH AP-PCR AND SOUTHERN BLOT RFLP (유전자형에 따른 Streptococcus mutans의 subtyping: Southern blot RFLP와 AP-PCR을 이용한 비교)

  • The arbitrary primer polymerase chain reaction(AP-PCR) and Southern blot restriction fragment length polymorphism(RFLP) were used to genotype the cariogenic pathogen S. mutans in children. Following the morphologic chracteristics of colony on selective medium for S. mutans, total genomic DNA from 155 strains was extracted by conventional methods. Among 155 strains, 143 strains (92.3%) were confirmed S. mutans by PCR with dexA gene and 114 strains were used in this study. Three random sequence 10-base oligonucleotide primers were chosen for AP-PCR. The amplified DNA products were separated electrophoretically in a 2% agarose gel containing ethidium bromide and the banding patterns were compared among different strains. For RFLP analysis, DNA was digested with EcoRI and BamHI, separated on a 0.7 % agarose gel and transferred to a nylon membrane. The membrane was probed with a previously characterised 1.6 kilobases (kb) DNA fragment cloned from gtf B gene of S. mutans. The probe was labeled with isotope[$^{32}P-{\alpha}CTP$], and hybridized fragments were detected with intensifying screen. AP-PCR produced 4-8 DNA bands in the 0.25-10 kb regions and distinguished 9, 10 or 12 genotypes, depending on the specific primer used. Southern blot RFLP analysis revealed 2 hybridization patterns consisting of 1 DNA fragments 450, 500 bp. These results indicate that AP-PCR is more discriminative method for genotyping of S. mutans.

Sampling Efficiency of Organic Vapor Passive Samplers by Diffusive Length (확산길이에 따른 수동식 유기용제 시료채취기의 시료채취성능에 관한 연구)

  • Passive samplers have been used for many years for the sampling of organic vapors in work environment atmospheres. Currently, all passive samplers used in domestic occupational monitoring are foreign products. This study was performed to evaluate variable parameters for the development of passive organic samplers, which include the geometry of the device and diffusive length for the sampler design. Four prototype diffusive lengths; A-1(4.5 mm), A-2(7.0 mm), A-3(9.5 mm), A-4(12.0 mm) were tested for adsorption performances to a chemical mixture (benzene, toluene, trichloroethylene, and n-hexane) according to the US-OSHA's evaluation protocol. A dynamic vapor exposure chamber developed and verified by related research was used for this study. The results of study are as follows. The results in terms of sampling rate and recommended sampling time test indicate that the most suitable model was A-3 (9.5 mm diffusive lengths on both sides) for passive sampler design in time weighted average (TWA) assessment. Sampling rates of this A-3 model were 45.8, 41.5, 41.4, and 40.3 ml/min for benzene, toluene, trichloroethylene, and n-hexane, respectively. The A-3 models were tested on reverse diffusion and conditions of low humidity air (35% RH) and low concentrations (0.2 times of TLV). These conditions had no affect on the diffusion capacity of samplers. In conclusion, the most suitable design parameters of passive sampler are: 1) Geometry and structure - 25 mm diameter and 490 $mm^2$ cross sectional area of diffusion face with cylindrical form of two-sided opposite diffusion direction; 2) Diffusive length - 9.5 mm in both faces; 3) Amount of adsorbent - 300 mg of coconut shell charcoal; 4) Wind screen - using nylon net filters (11 ${\mu}m$ pore size).

Cloning of a Potentially Strain-Specific DNA Probe of prevotella intermedia ATCC 25611 by Inverted Dot Blot Hybridization Screening Method

  • The purpose of this study was to isolate a specific DNA probe for the strain ATTC 25611 of the species Prevotella intermedia by using a new rapid screening mothod. The whole-genomic DNA of P. intermedia ATCC 25611 was isolated and purified. The HindIII-digested genomic DNAs from the strain were cloned by the random cloning method. To screen the strain-specific DNA probe, inverted dot blot hybridization tests were performed. In this assay, 20 ng of recombinant plasmids containing the HindIII-digested genomic DNA fragment were boiled and blotted onto a nylon membrane, and hybridized with digoxigenin-dUTP labeled genomic DNAs in a concentration of 100 ng/ml. Southern blot analysis was performed in order to confirm the results of the inverted dot blot hybridization tests. The data showed that a Pi34 probe (2.1 kbp; 1 out of 32 probes) was specific for P. intermedia strain ATCC 25611 and could be useful for the detection and identification of the strain, particularly in epidemiological studies of periodontal disease.

  • Microarray technology allows the simultaneous analysis of gene expression patterns of thousands of genes, in a systematic fashion, under a similar set of experimental conditions, thus making the data highly comparable. In some cases arrays are used simply as a primary screen loading to downstream molecular characterization of individual gene candidates. In other cases, the goal of expression profiling is to begin to identify complex regulatory networks underlying developmental processes and disease states. Microarrays were originally used with ceil lines or other simple model systems. More recently, microarrays have been used in the analysis of more complex biological tissues including neural systems and the brain. The application of cDNA arrays in neuropsychiatry has lagged behind other fields for a number of reasons. These include a requirement for a large amount of input probe RNA In fluorescent-glass based array systems and the cellular complexity introduced by multicellular brain and neural tissues. An additional factor that impacts the general use of microarrays in neuropsychiatry is the lack of availability of sequenced clone sets from model systems. While human cDNA clones have been widely available, high qualify rat, mouse, and drosophilae, among others are just becoming widely available. A final factor in the application of cDNA microarrays in neuropsychiatry is cost of commercial arrays. As academic microarray facilitates become more commonplace custom made arrays will become more widely available at a lower cost allowing more widespread applications. in summary, microarray technology is rapidly having an impact on many areas of biomedical research. Radioisotope-nylon based microarrays offer alternatives that may in some cases be more sensitive, flexible, inexpensive, and universal as compared to other array formats, such as fluorescent-glass arrays. In some situations of limited RNA or exotic species, radioactive membrane microarrays may be the most practical experimental approach in studying psychiatric and neurodegenerative disorders, and other complex questions in the brain.

Analyzing Drift Patterns of Spray Booms with Different Nozzle Types and Working Pressures in Wind Tunnel (풍동실험에 의한 붐식 살포 농약의 노즐형태와 분사압력에 따른 비산 특성 분석)

  • With rising concerns about pesticide spray drifts, this study analyzed the drift patterns of two typically-used nozzles, XR nozzle and AI nozzle, concerning their working pressures and wind speeds by wind tunnel experiments. AI nozzle showed low drift potential with larger droplet sizes compared to XR nozzle. Airborne and deposition drifts of XR nozzle were two times higher than those of AI nozzle under high wind speeds (≥2 m s-1). In all cases, higher working pressures decreased the droplet sizes, thereby increasing the airborne and deposition drifts. Higher wind speeds also resulted in more airborne drifts, while ground deposition was increased under lower wind speeds. These effects of working pressures and wind speeds on the airborne and deposition drifts were observed at leeward distances less than 4 m from the nozzles. However, the airborne and deposition drifts were barely affected by the working pressures and wind speeds at leeward distances more than 11 m. The measurements were fitted to regression models of the drift curve with acceptable R2 values greater than 0.8, demonstrating that further studies will be useful to settle domestic issues of spray drifts.

Effects of Heat Treatment of Three Animal by-products on Ruminal Degradation Characteristics and Intestinal Availability of Crude Protein (동물성 부산물 사료 세 종류에 대한 열처리가 조단백질의 반추위내 분해특성 및 하부장기내 이용성에 미치는 영향)

  • In order to investigate the effects of heat treatment of three animal by-products(feather meal, tallow meal, viscera meal) on in situ ruminal degradation characteristics and gastrointestinal availability of dietary crude protein(CP), three ruminally and duodenally cannulated dry Holstein cows were employed. Cows were fed a diet containing 60% concentrate and 40% orchard grass hay, and had free access to water and mineral block. Experimental feeds were processed for 4 hr at 149$^{\circ}C$ in a forced-air oven, and were passed through a 1-mm screen. Degradation kinetics of feed protein in the rumen were fitted to an exponential type model, and intestinal availability was estimated by the mobile nylon bag technique. Effective CP degradabilities in the rumen for feather meal, tallow meal and viscera meal were 30.2%, 75.0% and 56.4% at 5% passage rate per hour(k=0.05), respectively. In addition, heat treatment increased effective ruminal CP degradability on feather meal and viscera meal treatments, whereas decreased in tallow meal treatment(P$<$0.05). Gastrointestinal CP disappearances of feather meal, tallow meal and viscera meal were 56.2%, 18.6%, and 37.9%, respectively. In addition, heat treatment decreased the gastrointestinal CP disappearance on feather meal and viscera meal treatment, but increased in tallow meal treatment(P$<$0.05). Intestinal availability of rumen undegradable protein(A-UDP) was 80.4% for feather meal, 83.8% for tallow meal and 86.9% for viscera meal. In addition, heat treatment increased A-UDP on feather meal and tallow meal treatment, 94.0% and 91.3%, respectively, but decreased on viscera meal treatment, 76.5%(P$<$0.05).

Gene Expression Profiles in Cervical Cancer with Radiation Therapy Alone and Chemo-radiation Therapy (자궁경부암의 방사선치료 및 방사선항암화학 병용치료에 따른 유전자발현 조절양상)

  • Purpose : To analyze the gene expression Profiles of uterine ceulcal cancer, and its variation after radiation therapy, with or without concurrent chemotherapy, using a CDNA microarray. Materials and Methods :Sixteen patients, 8 with squamous ceil carcinomas of the uterine cervix, who were treated with radiation alone, and the other 8 treated w14h concurrent chemo-radiation, were Included in the study. Before the starling of the treatment, tumor biopsies were carried out, and the second time biopsies were peformed after a radiation dose of 16.2$\~$27 Gy. Three normal cervix tissues were used as a control group. The microarray experiments were peformed with 5 groups of the total RNAs extracted individually and then admixed as control, pre-radiation therapy alone, during-radiation therapy alone, pre-chemoradiation therapy, and during-chemoradlation therapy. The 33P-iabeled CDNAS were synthesized from the total RNAs of each group, by reverse transcription, and then they were hybridized to the CDNA microarray membrane. The gene expression of each microarrays was captured by the intensity of each spot produced by the radioactive isotopes. The pixels per spot were counted with an Arrayguage, and were exported to Microsoft Excel The data were normalized by the Z transformation, and the comparisons were peformed on the Z-ratio values calculated. Results : The expressions of 15 genes, including integrin linked kinase (ILK), CDC28 protein kinase 2, Spry 2, and ERK 3, were increased with the Z-ratio values of over 2.0 for the cervix cancer tissues compared to those for the normal controls. Those genes were involved In cell growth and proliferation, cell cycle control, or signal transduction. The expressions of the other 6 genes, Including G protein coupled receptor kinase 5, were decreased with the Z-ratio values of below -2.0. After the radiation thorapy, most of the genes, with a previously Increase expressions, represented the decreased expression profiles, and the genes, with the Z-ratio values of over 2.0, were cyclic nucleotlde gated channel and 3 Expressed sequence tags (EST). In the concurrent chemo-radiation group, the genes involved in cell growth and proliferation, cell cycle control, and signal transduction were shown to have increased expressions compared to the radiation therapy alone group. The expressions of genes involved in anglogenesis (angiopoietln-2), immune reactions (formyl peptide receptor-iike 1), and DNA repair (CAMP phosphodiesterase) were increased, however, the expression of gene involved In apoptosls (death associated protein kinase) was decreased. Conclusion : The different kinds of genes involved in the development and progression of cervical cancer were identified with the CDNA microarray, and the proposed theory is that the proliferation signal stalls with ILK, and is amplified with Spry 2 and MAPK signaling, and the cellular mitoses are Increased with the increased expression oi Cdc 2 and cell division kinases. After the radiation therapy, the expression profiles demonstrated 4he evidence of the decreased cancer cell proliferation. There was no sigificant difference in the morphological findings of cell death between the radiation therapy aione and the chemo-radiation groups In the second time biopsy specimen, however, the gene expression profiles were markedly different, and the mechanism at the molecular level needs further study.
