  • The bioconversion of cellulosic biomass hydrolyzates consisting mainly of glucose and xylose requires the use of engineered Saccharomyces cerevisiae expressing a heterologous xylose pathway. However, there is concern that a fungal xylose pathway consisting of NADPH-specific xylose reductase (XR) and $NAD^+$-specific xylitol dehydrogenase (XDH) may result in a cellular redox imbalance. However, the glycerol biosynthesis and glycerol degradation pathways of S. cerevisiae, termed here as the glycerol cycle, has the potential to balance the cofactor requirements for xylose metabolism, as it produces NADPH by consuming NADH at the expense of one mole of ATP. Therefore, this study tested if the glycerol cycle could improve the xylose metabolism of engineered S. cerevisiae by cofactor balancing, as predicted by an in-silico analysis using elementary flux mode (EFM). When the GPD1 gene, the first step of the glycerol cycle, was overexpressed in the XR/XDH-expressing S. cerevisiae, the glycerol production significantly increased, while the xylitol and ethanol yields became negligible. The reduced xylitol yield suggests that enough $NAD^+$ was supplied for XDH by the glycerol cycle. However, the GPD1 overexpression completely shifted the carbon flux from ethanol to glycerol. Thus, moderate expression of GPD1 may be necessary to achieve improved ethanol production through the cofactor balancing.

  • Institutional foodservice is a type of meal provided to workers in industry and affects the health and psychological wellbeing of workers and productivity. Thus, research on customer satisfaction with institutional foodservice is important. In addition to food industry and food culture developments, the requirements of institutional foodservice customers are diversifying due to COVID-19. Therefore, the purpose of this study was to identify factors that affect customer satisfaction with institutional foodservice using a user-based approach. In this study, the quality of institutional foodservice was defined using customer satisfaction or dissatisfaction, and by using in-depth interview and open coding (a qualitative research method), we derived qualities of institutional foodservice from the user's perspective and compared these with those of previous studies. This study is meaningful as the quality of institutional foodservice was analyzed using a user-based approach, in-depth interview, and open coding and compares results with those of previous studies.

  • [Purpose] The goal of this study was to review data on physical activity level (PAL), a crucial index for determining estimated energy requirement (EER), calculated as total energy expenditure (TEE, assessed with doubly labeled water [DLW]) divided by resting metabolic rate (RMR, PAL = TEE/RMR) in female athletes and to understand the methods of assessing athletes' EERs in the field. [Methods] For the PAL data review among female athletes, we conducted a PubMed search of the available literature related to the DLW method. DLW studies measuring TEE and RMR were included for the present review. [Results] Briefly, the mean PAL was 1.71 for collegiate swimmers with moderate training, which was relatively low, but the mean PAL was 3.0 for elite swimmers during summer training camp. This shows that PAL can largely vary even within the same sport depending on the amount of training, and the differences in PAL were remarkable depending on the sport. Aside from the DLW method, there is currently no research tool related to athletes' EERs that can be used in the field. [Conclusion] Briefly, the mean PAL was 1.71 for collegiate swimmers with moderate training, which was relatively low, but the mean PAL was 3.0 for elite swimmers during summer training camp. This shows that PAL can largely vary even within the same sport depending on the amount of training, and the differences in PAL were remarkable depending on the sport. • The population aged 95 years and older in Seoul approximately increased to five-fold over the past 10 years, while nationwide rates increased to three-fold. In order to examine the dietary habit and nutritional status of oldest-old population living in Seoul, we recruited 87 subjects (25 males and 62 females) aged 95 years and older. The prevalence of underweight (BMI < 18.5 $kg/m^2$) and obesity (BMI ${\geq}25kg/m^2$) were 18.2% and 18.2% in males, and 20.8% and 9.4% in females, respectively. In self-assessment of health, only 25.3% answered to be unhealthy. More males exercised regularly and reported a wide range of activities than females. The average of %Kcal from carbohydrate, protein and fat (C : P : F) was 64.9 : 13.8 : 21.2 in males and 68.1 : 14.2 : 17.7 in females. The average daily energy intake was 1,307 kcal in males and 1,304 kcal in females. More than 75% of subjects were taking under estimated average requirements (EAR) for vitamin $B_1$, $B_2$ & C and Ca. The average of mean adequacy ratio (MAR) was 0.66 in males and 0.70 in females, and 28.8% of males and 12.9% of females were in MAR < 0.50. Based on MAR, 32.0% of males and 14.5% females were classified as normal and 16.0% of males and 25.8% of females were classified as malnourished. Our subjects were taking more animal food, especially milk and its products, compared to those living, in rural areas. However, a significant proportion did not meet the EAR for vitamin $B_1$, $B_2$ & C and Ca.

  • According to economical status, malnutrition and overnutrition coexist among preschool children in Korea. Malnutrition have been a serious problem for children in some of rural. Many children have deficiencies of some nutrients such as riboflavin A, and simultaneously they have overcaloric intakes with empty calorie foods. Preschool children have nutritional problems which can affect the growth and development. To investigate whether nutritional intakes can influence on the growth or not, nutritional intakes and anthropometric measurements were determined for infants aged 4~6 years old in three different kinds of preschools. Daily nutrient intakes were calculated by the combination of a direct measurement and 24 hour recall, and the diet consumption patterns were determined by diet quality index. Overall growth of infants in Cheonan area was under the normal growth in Korea. Preschool children in orphanage consumed almost all nutrients much less than the recommended daily allowances, on the other hands, children in private preschool had over-intakes of nutrients when it is compared with the daily requirements. However, the scores of the diet quality index were similar in three preschools since children in private preschool consumed more fat and junk food than those in orphanage. The weight and height of preschool children in orphanage was lower than those in private preschools. This was due to the lower consumption of the important nutrients in infants in orphanage. The consumption of all nutrients except fat did not show any correlation with height or weight. The quality index of fat, cholesterol, grains and protein intakes was higher in children in private preschool than in orphanage. In conclusions, 1)the private preschool children had worse nutrient intake patterns. Better consumption of vitamin $B_2$, calcium and grains were, bigger height and chest circumferences children obtained. 2)Since most of energy intake of preschool children came from junk foods, they needed to consume more supplementation of vitamins and minerals, which is necessary for normal growth. 3)It is desirable for the children to cut down junk foods and empty calorie foods.

  • Twenty eight 3-4 month old castrated Black Bengal kids (Capra hircus) were used to determine the effects of source and level of dietary copper (Cu) concentration on their performance and nutrient utilization. Cu was supplemented (0, 10, 20 and 30 mg/kg diet DM) as copper sulfate ($CuSO_4$, $5H_2O$) or copper proteinate (Cu-P). Kids were fed a basal diet containing maize (19.5%), soybean (17.0%), deoiled rice bran (56.5%), molasses (4.0%), di-calcium phosphate and salt (1.0% each) and mineral and vitamin mixture (0.5% each) supplements at 3.5% of body weight to meet NRC (1981) requirements for protein, energy, macro minerals and micro minerals, excluding Cu. The basal diet contained 5.7 mg Cu/kg, 122.5 mg Fe/kg, 110 mg Zn/kg, 0.26 mg Mo/kg and 0.32% S. $CuSO_4$ or Cu-P was added to the basal diet at the rate of 10, 20 and 30 mg/kg. Kids were housed in a well ventilated shed with facilities for individual feeding in aluminum plated metabolic cages. Blood samples were collected from the jugular vein on d 0, 30, 60 and 90 to determine hemoglobin (Hb), packed cell volume (PCV), total erythrocyte count (TEC), total leukocyte count (TLC) and serum enzymes (alkaline phosphatase, alanine transferase and aspertate transferase). A metabolism trial of 6 days duration was conducted after 90 days of experimental feeding. Statistical analysis revealed that source and level of Cu supplementation improved live weight gain (p<0.04) and average daily gain (p<0.01). No significant contribution of source and level of Cu to alter serum serum enzymes was evident. Goats fed Cu-P tended to have higher Hb, PCV and TEC than with $CuSO_4$ supplementation. Cu-P increased digestibility of ether extract (EE, p<0.02) and crude fiber (p<0.05) and showed an increasing trend (p<0.09) for digested crude protein (CP) and crude fiber (CF). Supplemental dose of Cu linearly improved (p<0.02) digestibilities of dry matter (DM), organic matter (OM), EE and nitrogen free extract (NFE). Though the absorption of nitrogen (N) was not affected (p>0.10) by both source and dose of Cu, N retention was affected (p<0.04) and there was a significant $Source{\times}Dose$ interaction (p<0.05). Final body weight (BW) was not influenced (p>0.10) by the source of Cu but increasing dose of Cu increased (p<0.04) the BW of kids. TDN intake (g/kg $W^{0.75}$) was higher (p<0.05) with the increased dose of Cu and there was a significant $Source{\times}Dose$ interaction. It was concluded that supplementation of Cu from different sources and varying dose level in a concentrate based diet may improve performance, nutrient utilization and plane of nutrition in castrated Black Bengal kids. The effects on performance and nutrient utilization are more pronounced with Cu-P than $CuSO_4$ supplementation. Higher dose of Cu showed better result than lower dose.

  • Whole lupinus albus seeds were pressure toasted at temperatures of 100, 118 and $136^{\circ}C$ for 3, 7, 15 and 30 min to study rumen degradation and post-rumen digestion and to determine optimal heating conditions for the Dutch dairy feed industry. In sacco nylon bag and mobile bag techniques were employed for rumen and intestine incubations to determine ruminal degradation characteristics and intestinal digestion of crude protein (CP) in 4 lactation rumen cannulated and 4 lactating intestinal cannulated Dutch dairy cows fed 47% hay and 53% concentrate according to Dutch dairy requirements. Measured rumen degradation characteristics were soluble fraction (S), undegradable fraction (U), potentially degradable fraction (D), lag time (T0) and rate of degradation (Kd) of insoluble but degradable fraction. Percentage bypass feed protein (BCP), ruminal microbial protein synthesized based on available nitrogen (N_MP) and that based on available energy (E_MP), true protein supplied to the small intestine (TPSI), truly absorbed BCP (ABCP), absorbed microbial protein (AVP) in the small intestine, endogenous protein losses in the digestion (ENDP), true digested protein in the small intestine (TAP or DVE in Dutch) and degraded protein balance (PDB or OEB in Dutch) were totally evaluated using the new Dutch DVE/OEB System. Pressure toasting decreased (p<0.001) rumen degradability of CP. It reduced S (p<0.05) and Kd (p=0.06), increased D (p<0.05) and U (p<0.01) but did not alter T0 (p>0.05), thus resulting in dramatically increased BCP (p<0.001) with increasing time and temperature from 73.7 (raw) up to 182.5 g/kg DM ($136^{\circ}C/15min$). Although rumen microbial protein synthesized based on available energy (E_MP) was reduced, true protein (microbial and bypass feed protein) supplied to the small intestine (TPSI) was increased (p<0.001) from 153.1 (raw) to 247.6 g/kg DM ($136^{\circ}C/15min$). Due to digestibility of BCP in the intestine not changing (p>0.05) average 87.8%, the absorbed BCP increased (p<0.001) from 62.3 (raw) to 153.7 g/kg DM ($136^{\circ}C/15min$). Therefore DVE value of true digested protein in the small intestine was significantly increased (p<0.001) from 118.9 (raw) to 197.0 g/kg DM ($136^{\circ}C/15min$) and OEB value of degraded protein balance was significantly reduced (p<0.001) from 147.2 (raw) to 63.1 g/kg DM ($136^{\circ}C/15min$). It was concluded that pressure toasting was effective in shifting degradation of CP of lupinus albus from the rumen to small intestine without changing intestinal digestion. Further studies are required on the degradation and digestion of individual amino acids and on the damaging effects of processing on amino acids, especially the first limiting amino acids.

  • This study was conducted to re-evaluate the dietary vitamin C requirement in juvenile eel, Anguilla japonica by using L-ascorbyl-2-monophosphate (AMP) as the vitamin C source. Five semi-purified experimental diets were formulated to contain 0 ($AMP_0$), 30 ($AMP_{24}$), 60 ($AMP_{52}$), 120 ($AMP_{108}$) and 1,200 ($AMP_{1137}$) mg AMP $kg^{-1}$ diet on a dry matter basis. Casein and defatted fish meal were used as the main protein sources in the semi-purified experimental diets. After a 4-week conditioning period, fish initially averaging $15{\pm}0.3$ g (mean${\pm}$SD) were randomly distributed to each aquarium as triplicate groups of 20 fish each. One of five experimental diets was fed on a DM basis to fish in three randomly selected aquaria, at a rate of 3% of total body weight, twice a day. At the end of the feeding trial, weight gain (WG) and specific growth rate (SGR) for fish fed $AMP_{52}$ and $AMP_{108}$ were significantly higher than those recorded for fish fed the control diet (p<0.05). Similarly, feed efficiency (FE) and protein efficiency ratio (PER) for fish fed $AMP_{52}$ were significantly higher than those for fish fed the control diet (p<0.05). Broken-line regression analysis on the basis of WG, SGR, FE and PER showed dietary vitamin C requirements of juvenile eel to be 41.1, 41.2, 43.9 and 43.1 (mg $kg^{-1}$ diet), respectively. These results indicated that the dietary vitamin C requirement could range from 41.1 to 43.9 mg $kg^{-1}$ diet in juvenile eel when L-ascorbyl-2-monophosphate was used as the dietary source of vitamin C.

  • Fifteen crossbred cattle bulls of about 22-24 months age (mean body weight ranged from 291.0 to 298.1 kg) were randomly divided into 3 groups of 5 animals following randomized block design. Animals in group 1 were fed concentrate mixture containing wheat bran 97%, mineral mixture 2% and salt 1% plus ad libitum chopped green sugarcane tops (SCT) as a roughage source. Animals in group 2 were fed concentrate mixture containing lentil chuni 97%, mineral mixture 2% and salt 1% while group 3 concentrate contained wheat bran 48.5%, lentil chuni 48.5%, mineral mixture 2% and salt 1% plus ad libitum chopped green SCT to meet their maintenance requirements. During metabolism trial period, the mean total intake and digestibility of DM in groups 1, 2 and 3 was $8.09{\pm}0.41$, $8.41{\pm}0.49$ and $7.86{\pm}0.16kg/d$ and 60.24, 63.24 and 65.05%, respectively. The intake of CP (p<0.05) and EE (p<0.01) was significantly higher in group 2 and group 1, respectively as compared to remaining 2 groups, which were comparable. Digestibility of CP and EE was significantly (p<0.01) higher in group 3 and that of CP was comparable with group 2. The intake and digestibility of total carbohydrates, NDF, ADF, Hemi-cellulose and cellulose were similar among 3 groups. The mean body weight changes in group 1, 2 and 3 were 173.4, 253.4 and 203.4 g/d, respectively, which was significantly (p<0.05) higher in group 2. The total nitrogen (N) and phosphorus (P) intakes were significantly higher in group 2 and group 1, respectively. The total volatile fatty acids, ammonia-N, trichloro-acetic acid precipitable nitrogen (TCA ppt.-N; p<0.05), total-N concentrations and pH were significantly (p<0.01) higher in group 3, however, pH and TCA ppt.-N was comparable with group 2 and 1, respectively. From the results, it may be deduced that the feeding of chopped green SCT supplemented with a concentrate mixture having both wheat bran (48.5%) and lentil chuni (48.5%) has shown a better performance with respect to intake and digestibility of nutrients and growth as well.
