Traditional cell culture(2-dimensional) is the method that provide a nutrient and environment on a flat surface to cultivate cells into a single layer. Since the cell characteristics of 2D culture method is different from the characteristics of the cells cultured in the body, attempts to cultivate the cells in an environment similar to the body environment are actively proceeding in the industry, academy, and research institutes. In this study, we will develop a technology to fabricate micro-structures capable of culturing cells on surfaces with various curvatures, surface shapes, and characteristics. In order to fabricate the hemispheric plastic structure(thickness $50{\mu}m$), plastic preform mold (hereinafter as "preform mold") corresponding to the hemisphere was first prepared by injection molding in order to fabricate a two - layer structure to be combined with a flat plastic film. Then, thermoplastic polymer dissolved in an organic solvent was solidified on a preform mold. As a preliminary study, we proposed injection molding conditions that can minimize X/Y/Z axis deflection value. The effects of the following conditions on the preform mold were analyzed through injection molding CAE, [(1) coolant inlet temperature, (2) injection time, (3) packing pressure, (4) volume-pressure (V/P). As a result, the injection molding process conditions (cooling water inlet temperature, injection time, holding pressure condition (V / P conversion point and holding pressure size)) which can minimize the deformation amount of the preform mold were derived through CAE without applying the experimental design method. Also, the derived injection molding process conditions were applied during actual injection molding and the degree of deformation of the formed preform mold was compared with the analysis results. It is expected that plastic film having various shapes in addition to hemispherical shape using the preform mold produced through this study will be useful for the molding preform molding technology and cast molding technology.
Experimental design for ecological restoration approach was investigated from the P' company's slag dumping area with higher pH value. The degree of pH recovery was measured by the injection of carbon dioxide from the LNG exhaust gas, and the residual carbon, for example, calcium and carbonate which can be controlled by artificial seaweed beds. The degree of adaptability from 3 algaes (Undaria pinnatifida, Sargassum horneri and Ecklonia stolonifera) and uptake nutrient function of Ecklonia stolonifera chosen in the first treatment were measured in the laboratory to determine the transplanting algae in artificial seaweed beds. The higher value of pH was decreased to 7.0~8.5 by injection of LNG exhaust gas with flow rate $20\;m^3/min$. In the experiment design at laboratory, the upper part of frond of Undaria pinnatifida and Sargassum horneri began to decay, and the color changed after 10 days. However, those of Ecklonia stolonifera were after 14~20 days. The uptake rate of nutrient from Ecklonia stolonifera was higher than those of other algaes, and those was similar pattern in the control (e.g. seawater); The DIN concentration uptake of Ecklonia stolonifera was $1.88\;{\mu}g/L/hr$ avg. in leachate, but $2.19\;{\mu}g/L/hr$ avg. in seawater. However, the ${PO_4}^3$-P concentration uptake was $0.18\;{\mu}g/L/hr$ avg. in leachate, but $0.31\;{\mu}g/L/hr$ avg. in seawater. These result indicated Ecklonia stolonifera uptaked these nutrients in the leachate as well as in seawater, and it may suggested for this species to transplant for constructing artificial seaweed beds.
This study was conducted to evaluate the effects of dietary supplementation of plant flavonoid (quercetin) on immune parameters, growth performance, and nutrient digestibility in growing pigs challenged with Escherichia coli lipopolysaccharide (LPS). A total of 40 crossbred ([Landrace × Yorkshire] × Duroc) growing pigs; initial body weight (BW) of 26.95 ± 1.26 kg were used in a six-week experimental trial. Pigs were randomly allocated into one of four treatment groups in a 2 × 2 factorial arrangement with the following factors; without LPS challenge and with LPS challenge (day 21) supplemented with or without 0.1% flavonoid according to BW (2 replicate pens per treatment with 2 gilts and 3 barrows per pen). The single-dose LPS (100 ug / kg BW) injection showed trends tended to be increased in interleukin-6 (IL-6) after 2 h and 6 h of challenge compared with unchallenged pigs. However, other measured immune indices (white blood cell, immunoglobulin G, lymphocyte, and tumor necrosis factor), growth performance, and nutrient digestibility were not significantly different between challenged and non-challenged animals. The supplementation of flavonoid significantly increased (p < 0.05) average daily gain (ADG) during day 0-21, tended to increase dry matter and nitrogen digestibility, significantly reduced IL-6, increased Ig-G and WBC concentrations and increased lymphocytes percentage regardless of LPS challenge.
The purpose of this study is to solve the problem of nutrient removal due to rapid hydraulic retention time (HRT) variation in small size private sewage treatment facility and to have better effluent quality by using natural zeolite as a coagulant aid. The experiments were conducted by applying different conditions of HRT, coagulant, and zeolite dosages. As an experimental result, $TBOD_{5}$, TSS, and TP removal efficiencies were 64.9%, 94.5%, and 98.8% at co-injection with zeolite. Overall removal efficiencies of $TBOD_{5}$, TSS, and TP have improved 15.7, 28.7, 6.1%, respectively. This result shows that zeolite addition could improve flocculation, increase setting velocity, and thus have better treatment. Therefore the use of zeolite as a coagulant aid can be useful in small size private sewage treatment facility, where especially extra nutrient removal is required to meet the legal standard for discharge.
본 연구는 기능성 성분으로 알려져 있는 게르마늄을 '신고' 배 과원에 처리 방법을 달리 했을 때 토양과 엽내 무기성분과 엽 특성에 어떠한 영향을 미치는지에 대해서 2004년에 조사하였다. 게르마늄 처리는 엽면살포, 토양 내 관주, 수간주입, 그리고 앞의 세 가지 처리를 혼용한 복합처리를 포함하였다. 게르마늄 처리는 대조구에 비해 토양 중 질산태질소와 칼륨과 칼슘 농도를 오히려 감소시켰다. 게르마늄 처리는 대조구에 비해 토양 중 게르마늄을 약 50배 이상 증가시켰으며, 엽면적, 엽건물중, 그리고 엽비중을 통계적으로 유의성 있게 증가시켰다. 대조구는 게르마늄 처리구에 비해서 엽내 칼륨농도를 유의성 있게 증가시켰고, 반대로 칼슘은 게르마늄 처리구에서 높은 경향이 나타났다. 엽내 게르마늄은 게르마늄 관주처리를 제외하고 대조구보다 현저히 높게 나타났고, 과실 내 게르마늄은 모든 처리구에서 대조구 보다 높게 나타났다.
본 연구는 신체에 유익한 기능성 성분으로 알려져 있는 셀레늄을 '원황' 배나무에 처리방법을 달리했을 때 어떤 처리방법이 토양과 수체에 셀레늄 흡수를 증진시키는 것에 대해서 조사하였다. 셀레늄 처리는 엽면살포, 토양 내 관주, 그리고 수간주입을 포함하였다. 토양 내 인산과 칼륨 및 마그네슘 함량은 대조구와 셀레늄 관주 처리간에 비슷한 경향을 보였고, 토양 내 칼슘은 셀레늄 처리로 인한 양이온 경쟁작용으로 대조구에서 오히려 높았고 셀레늄 함량은 관주 처리구에서 높게 나타났다. 셀레늄 처리에 따른 엽형질은 별다른 영향을 받지 못했다. 엽중 칼륨과 칼슘은 모든 처리구 간에 별다른 영향이 없었고, 마그네슘은 셀레늄 처리구에서 대조구보다 높은 경향이 나타났다. 엽과 과실 내 셀레늄 함량은 셀레늄 엽면살포와 수간주입 처리구에서 대조구와 관주 처리구보다 유의적으로 높게 나타났다.
When microorganism is injected into porous medium such as soils, biomass retained in the pore. Bacteria within these microcolonies produced large amounts of exopolysaccharides and formed a plugging biofilm. Soil pore size and shape are varied from the initial condition as a result of biofilm formation, which make hydraulic conductivity reduced and friction rate between soil aggregates increased. In this research, hydraulic conductivity reduction was measured after microorganism are inoculated and cultured with synthetic substrate and nutrient. Also, pore sand of before and after biofilm formation compared with scanning electron microscopy. Hydraulic conductivity of Sand and Poorly Graded Sand was decreased approximately 1/10∼1/100 after biomass inoculation and cultivation. Biofilm attached on soil aggregates is resistant to acidic or basic condition.
고기능성 채소 생산을 위해 항산화효과가 뛰어난 셀레니움을 식물체내로 주입시 배양액내에서 발생할 수 있는 황산이온과의 상호작용은 식물체의 생장은 물론 식물에 의한 적정 수준의 셀레니움의 흡수에 영향을 미칠 수 있다. 따라서 본 실험은 양액내의 셀레니움과 황산이온이 Afemisia속 식물에 미치는 영향을 알아보고자 수행되었다. 벨기에의 채소연구소에서 개발한 허브 양액을 이용하여 황산이온 농도를 0. 0.5, 1, 2, 3mM 로 변형시킨 후 각각에 $Na_2$SeO$_4$2mg/$\ell$를 첨가하였다. 생육초기에는 $Na_2$SeO2mg/$\ell$$_4$ 처리시 3mM 황산이온 농도에서 가장 좋은 생육을 보여주었다. 그러나 생육 후기로 갈수록 셀레니움과 황산이온과의 길항작용의 효과는 점차 감소하여 생육에 있어서 유의적인 차이를 보이지 않았다. $Na_2$SeO$_4$2mg/$\ell$처리시 SO$_4$$^{2-}$ 처리에 따른 셀레니움 흡수에 있어 배양액내의 SO$_4$$^{2-}$ 의 농도가 높아질수록 두 이온간의 길항작용으로 인하여 셀레니움의 흡수는 감소하였으며 생육단계에 의한 반응은 생육초기에는 셀레니움 흡수감소의 폭이 적었으나 생육후기로 갈수록 흡수감소의 폭은 현저하였다. $Na_2$SeO$_4$2mg/$\ell$처리시 SO$_4$$^{2-}$ 처리에 따른 황산이온의 흡수는 배양액 내의 황산이온 농도의 증가에 따라 흡수도 증가하였다. 또한 생육후기로 갈수록 그 증가의 폭은 더 컸으나 3mM의 높은 농도에서는 황산이온의 흡수가 급격히 감소하였다.었다.간영향을 어떻게 분리해 낼 것인지에 대한 문제점이 남는다. 예를 들어 화분분석의 경우 1차적인 환경변화에 따른 식생조성변화를 반영한다. 인간이 활발한 농경활동을 하게 되면서 주변식생을 제거하게 되고, 제거된 나대지에는 재배작물과 잡초들이 자라게 된다. 그리고 이들 화분은 주변 소택지에 퇴적물과 함께 퇴적되어 화분분석 결과 벼과(科)(Gramineae)와 문화지표수종이라 부르는 여뀌속(屬)(Persicaria), 국화과(科)(Composite), 쑥속(屬)(Artemisia), 쐐기풀속(屬)(Urtica) 등의 화분에 있어 확연한 조성변화를 보여준다. 그러나 화분을 이용한 분석은 토탄층이나 유기질 퇴적물이라는 한정된 토양에서만 분석이 가능하며, 조성변화에 있어서 역시 기후와 인간활동의 영향 모두를 반영하기 때문에 이를 구분해 주는 명확한 기준이 필요하다. 따라서 본 연구에서는 이러한 다양한 대리자료 중 고고학 유적지에 적용 가능한 동식물 화석 및 미화석, 지리, 지질학적 자료에 어떠한 것들을 적용할 수 있는지를 살피고, 몇 가지 대리자료를 중심으로 선사인의 농경활동에 대해 살펴보고자 한다.avascular injection of the iodinated contrast materials.석 시기중 성공적인 착상성공율은 56%(71/126)였다..8H2O를 사용함으로써 host-guest적인 반응을 유도시키는데 있어 물의 가장 실용적이고 효과적인 수열용매임도 알았다. 본 시험은 Holstein종 수소를 조기에 외과적 거세 후 SR-rbST 투여시 사육 단계별 건물 및 영양분 섭취량 변화에 미치는 영향을 구명하기 위하여 Holstein 수소 32두를 공시 하여 얻어진 주요 결과를 요약하면 다음과 같다. 1. 건물 섭취량은 비거세구와 거세구는 큰 차이 가 없었지만 rbSTl구와 rbST2구는 각각 7.88kg과 7.65kg으로 비거세구와 거세구에 비해 6~9% 정도 건물 섭취량이 적었다. 2. Immune stress is the loss of immune homeostasis caused by external forces. The purpose of this experiment was to investigate the effects of immune stress on the growth performance, small intestinal enzymes and peristalsis rate, and mRNA expression of nutrient transporters in broiler chickens. Four hundred and thirty-two 1-d-old broilers (Cobb500) were randomly assigned to four groups for treatment; each group included nine cages with 12 birds per cage. Group 1 = no vaccine (NV); Group 2 = conventional vaccine (CV); group 3 = lipopolysaccharide (LPS)+conventional vaccine (LPS); group 4 = cyclophosphamide (CYP)+conventional vaccine (CYP). The results demonstrated that immune stress by LPS and CYP reduced body weight gain (BWG), feed intake (FI), small intestine peristalsis rate and sIgA content in small intestinal digesta (p<0.05). However, the feed conversion ratio (FCR) remained unchanged during the feeding period. LPS and CYP increased intestinal enzyme activity, relative expression of SGLT-1, CaBP-D28k and L-FABP mRNAs (p<0.05). LPS and CYP injection had a negative effect on the growth performance of healthy broiler chickens. The present study demonstrated that NV and CV could improve growth performance while enzyme activity in small intestine and relative expression of nutrient transporter mRNA of NV and CV were decreased in the conditions of a controlled rational feeding environment. It is generally recommended that broilers only need to be vaccinated for the diseases to which they might be exposed.
