Isolation and culture of protoplasts from leaf tissue of Capsicum annnum var. accumnatum Fingerh and C. frutescensL. [Syn. C. minimum Roxb.] (Bird chilli)

  • Isolation and culture of leaf protoplasts from two chilli cultivars (Capsicum annuum var. accumnatum and Bird chilli) were developed to enhance selection process in the somatic hybridization programmes. In order to isolate the protoplasts from leaves of these two chilli cultivars different incubation periods (3, 5 and 10 hours) were tested with combinations of enzyme mixtures containing cellulase and macerozyme. Leaves were incubated on three enzyme mixtures (2% cellulase + 0.4% macerozyme, 1% cellulase + 0.2% macerozyme and 0.5% cellulase + 0.1 % macerozyme in 13% mannitol) at 251oC in the dark. Three hours of incubation using 2% cellulase and 0.4% macerozyme was the best for the protoplast isolation of both chilli cultivars tested. The yield was 5 ${\times}$ 108protoplasts/ml/ g leaf tissue in both chilli varieties. It was found that in the mixed nurse method using Nagata and Takebe (NT) medium supplemented with 1.0mg/12,4-D, NAA and BAP with 0.5M mannitol and 1.2% Sea Plaque agarose is the best medium for protoplast culture. Protoplasts of Capsicum annum var. accumnatum were alive for 14 days forming cell walls and initiating cell division.

A Study on the Somatic Cells in Quarter Milk Samples of Holstein Cows with Suspected Mastitis (유방염으로 의심된 유우 분방유즙의 체세포 연구)

  • This study was conducted to survey the relationship between the somatic cell count (SCC), and California mastitis test (CMT) & mastitis. A total of 328 quarter milk samples were collected from 211 cows suspected to have mastitis; Both SCC and CMT were performed on the samples. Milk smear was stained with Broadhurst and Paley stain and the cells were classified into either epithelial or blood cells. Bacterial isolation was made and antimicrobial susceptibility was tested. Of the 328 quarters, 78 ($23.8{\%}$) were CMT negative with SCC <750,000/ml. As expected, CMT score increased with the increase of SCC. The number of epithelial cells decreased with the increasing number of somatic cells, while the opposite was the case with the number of blood cells. The critical point was when the SCC reached 1,000,000/ml. Up to 1,000,000 cells/ml, the number of epithelial cells was greater than that of blood cells. The results indicate that when epithelia:blood cell ratio is 58.1:41.9, the milking line should be checked and bacterial isolation be performed on the samples in order to identity mastitis.

  • 동양달팽이 (Nesiohelix samarangae)는 동양달팽이속(Nesiohelix)에 속하는 종으로 한국에서 동양?팽이속으로는 유일하게 발견되는 종이다. 본 실험에서는 동양달팽이의 웅성생식기관의 각 부분 즉, 난정소(ovotestis), 양성관(hermaphrodite duct), 수정실(fertilization chamber), 전립선(prostate), 수정관(vas deferens), 상음경(epiphallus), 음경(penis)등의 배열 상태를 관찰하고 이들에 대한 광학현미경적 관찰을 위해 Hematoxylin- Eosin염색과 Mallory 염색을 실시하였다. 난정소(ovotestis)는 많은 수의 선포(acini)로 이루어져 있으며 각 선포는 배상피세포층(germinal epithelial cell layer)과 영양세포(nurse cell), 생식세포(gamete cell)들로 이루어져 있었다. 선포(acini)는 원주상피로 덮여있었다. 양성관(hermaphrodite duct)은 가늘고 긴 관으로 내벽은 섬모입방상피세포와 얇은 결합조직층으로 이루어져 있었으며 내강은 농축된 정자로 가득 차 있었다 . 수정관(fertilization chamber)은 3개의 도관이 결합조직에 의해 둘러싸인 형태로 이루어져 있었다. 도관의 상피세포는 뚜렷한 위중층섬모원주세포로 되어 있었다. 전립선(prostaoe)은 분비기능을 가진 것으로 사료되는 세포들과 실제 분비가 일어나는 실(chamber)로 이루어져 있었으나 다른 염색법 또는 전자현미경적 관찰에 의해 다른 구조들의 존재유무를 확인 해야할 것으로 사료된다. 실(chamber)은 분비포로 보이는 세포들과 접해 있었다. 수정관(vas deferens)은 결합조직으로 이루어진 근육질의 두꺼운 벽을 가지고 있었으며 내강은 돌출부위에 의해 4부분으로 갈라져 있었다. 내강에 연한 상피는 원주세포로 되어 있었다. 상음경(epiphallus)의 벽은 근섬유 다발은 벽의 외곽으로 갈수록 밀도가 높아졌다. 상부에 내강과 연결되는 함입구조(groove)를 가지며, 내강에 연한 상피는 섬모가 없는 원주세포로 이루어져 있었다. 음경(penis)은 근섬유가 산재된 두꺼운 벽을 가졌으며 상음경보다 굵은 원통형이었다. 내강은 많은 돌출부에 의해 복잡하게 나뉘어 있으며 상피세포는 원주 세포로 이루어져 있었고 섬모는 관찰되지 않았다. 내강 내의 분비물과 세포의 형태로 보아 내강상피세포는 분비기능을 가진 것으로 사료된다.

Activation and Recruitment of Regulatory T Cells via Chemokine Receptor Activation in Trichinella spiralis-Infected Mice

  • As most infections by the helminth parasite elicit the recruitment of $CD4^+CD25^+Foxp3^+$ T ($T_{reg}$) cells, many scientists have suggested that these cells could be used for the treatment of immune-mediated inflammation and associated diseases. In order to investigate the distribution and alteration of activated $T_{reg}$ cells, we compared the expression levels of $T_{reg}$ cell activation markers in the ileum and gastrocnemius tissues 1, 2, and 4 weeks after infection. The number of $T_{reg}$ cells was monitored using GFP-coded Foxp3 transgenic mice. In mice at 1 week after Trichinella spiralis infection, the number of activated $T_{reg}$ cells was higher than in the control group. In mice at 2 weeks after infection, there was a significant increase in the number of cells expressing Foxp3 and CTLA-4 when compared to the control group and mice at 1 week after infection. At 4 weeks after infection, T. spiralis was easily identifiable in nurse cells in mouse muscles. In the intestine, the expression of Gzmb and Klrg1 decreased over time and that of Capg remained unchanged for the first and second week, then decreased in the 4th week. However, in the muscles, the expression of most chemokine genes was increased due to T. spiralis infection, in particular the expression levels of Gzmb, OX40, and CTLA-4 increased until week 4. In addition, increased gene expression of all chemokine receptors in muscle, CXCR3, CCR4, CCR5, CCR9, and CCR10, was observed up until the 4th week. In conclusion, various chemokine receptors showed increased expressions combined with recruitment of $T_{reg}$ cells in the muscle tissue.

Automatic Flow Control and Network Monitoring of IV Injection (자동 IV 주사 유량 자동 제어 및 네트워크 모니터링)

  • Intravenous (IV) injection is widely used to supply Ringer solution directly into a vein in hospital. Generally, a passive injection method has been used, which causes the inconsistent flow rate of fluid and inappropriate control of injection time by a patient. It leads to an unnecessary nurse's overwork and decrement of IV injection's effect. To solve these problems, flow control infusion pumps have been developed. But because of relatively heavy weight and high price, its usage has been limited. In the present study, a new automatic IV injection system is developed. It is installed with a small pressing mechanism driven by a small electric motor to regulate the flow rate by pressing tube. Proportional integral derivative (PID) feedback control algorithm is applied to control the electric motor. The system is smaller in size and uses lower power than the existing commercial product. The newly developed system is also installed with networking capability, which enables monitoring the status of several automatic IV injection system at the same time.

Identifying Minimum Data Sets of Oral Mucous Integrity Assessment for Documentation Systematization (구강점막의 통합성 사정기록 체계화를 위한 최소자료세트(Minimum Data Set) 규명)

  • Purpose : The purpose of this study was to identify minimum data sets for oral mucous integrity-related documentation and to analyze nursing records for oral care. Methods: To identify minimum data sets for oral status, the authors reviewed 26 assessment tools and a practical guideline for oral care. The content validity of the minimum data sets was assessed by three nurse specialists. To map the minimum data sets to nursing records, the authors examined 107 nursing records derived from 44 patients who received chemotherapy or hematopoietic stem cell transplantation in one tertiary hospital. Results: The minimum data sets were 10 elements such as location, mucositis grade, pain, hygiene, dysphagia, exudate, inflammation, difficulty speaking, and moisture. Inflammation contained two value sets: type and color. Mucositis grade, pain, dysphagia and inflammation were recorded well, accounting for a complete mapping rate of 100%. Hygiene (100%) was incompletely mapped, and there were no records for exudate (83.2%), difficulty speaking (99.1%), or moisture (88.8%). Conclusion: This study found that nursing records on oral mucous integrity were not sufficient and could be improved by adopting minimum data sets as identified in this study.

The Filter Membrane Culture Procedure with Feeder Cells in Rice Protoplast Culture (Filter membrane과 feeder세포를 이용한 벼의 원형질체 배양)

  • To investigate the response on feeder cell cultures, protoplasts isolated from cell suspensions initiated from mature seed scutellum-derived callus of the Japonica rice variety Taipei 309, were cultured on filter membranes under various conditions. The effects of various factors, such as gelling agents, feeder cell and protoplast densities, species of feeder cells and heat shock treatment, have been investigated to improve protoplast plating efficiencies on filter membranes. Maximum protoplast plating efficiencies were obtained when protoplasts were cultured on KPR medium semi-solidified with Sea Plaque agarose at a density of $5\;\times\;10^{5}\;ml^{-1}$ protoplasts in the presence of Lolium multiflorum as feeder cells (0.5 ml pcv per 10 ml of protoplast culture medium). Pre-culture heat shock treatments for 1 min. and 5 min. to the protoplasts did not give any appreciable increase on the plating efficiency of protoplasts in the presence of feeder cells. Maltose-supplemented medium was superior for plant regeneration from protoplast-derived colonies compared with medium containing only sucrose. The plants were transferred to the glasshouse, flowered and were fertile.

  • The study looked at the effect of body food on stress reduction and NK cell activity in improving natural healing for nurses, and examined whether it is effective in preventing and curing human health. The study was conducted for 30 days on 22 nurses who currently worked for more than a year at a general hospital in Gyeonggi Province. Stress measurements were conducted through pulse wave measuring instrument, and blood tests were conducted on the activity of NK cells. Treatment of collected data was performed by Paired t-testing through the Stat program SPSS 21 and providing body food suitable for the physical quality of participants after the classification of the body based on the internal diameter of the emperor and scholarship. Studies have shown that the effect on reducing the stress of body food has significant positive effects and that body food is effective for the activity of NK cells. This study has the following significance: First, the main health threat factor for modern people is the increased resistance of natural healing through body food to stress, which has the potential to prevent disease. Second, immunodeficiency in disease prevention and treatment is very important, which can increase the natural healing power of the human body by increasing the activity of NK cells through body food.

Assessment of Nutritional Status before TPN Support of Hospitalized Patients (입원환자의 TPN 투여전 영양상태 평가)

  • The purpose of this study was to assessment of hospitalized patients. Nutritional assessment was performed on 353 hospitalized patients before TPN support by ideal body weight. albumin, total lymphocyte count, cholesterol. The data analysis were performed on frequency, percentage, means and standard deviation. The major findings as follows: 1. Everage NPO duration was $6.77\pm5.39$day and $\geq5$days 48.9%, >5days 51.1%. NPO duration of 1M, NL, GS, Others prolonged. 2. %IBW was everge $94.86\pm17.21%$ 43.1% normal. 37.8% low weight. There were low weight IM 44.8%. TS 47.8%, Others 44.0%. 3. Albumin was everage $2.93\pm0.51$g/dl. moderate malnutrition 45.4%, mild malnutrition 30.9%. All session were malnutrition status. 4. Total lymphocyte count was ever age $960.41\pm721.32cell/mm^2$. severe malnutrition 43.9%. moderate malnutrition 29.0%, mild malnutrition 24.4%, normal 2.7%. All session were malnutrition status. 5. Cholesterol was everage $123.02\pm45.67$mg/dl 58.4% low level. 41.2% normal. 0.4% high level. 1M, GS and TS were low level. NL, NS, DR, Others were normal. It was very poor nutritional status before TPN support of patients. The malnutrition decrease immunity, slowly cure injury, cause abnormality organs, increase the complication and obstruct the recovery. Therefore, Evaluating and correcting are very important.

Transovarial Transmissibility and Histopathology on the Gonad Ivfection with Nosema bombycis in the Silkworm, Bombyx mori, (집누에 생식소의 Nosema bombycis 감염과 경란전달성에 관한 조직병리학적 해명)

  • Through the histological and anatomical investigation of silkworm gonad, N. bombycis infection was found to begin from the peripheral region of ovarial sheath or testicular sheath, then, the pathogens spread to the inner portion. Peroral inoculation with purified spores of N. bombycis to 2nd instar larvae at dosages around 106-8/㎤ of artificial diet resulted in the extremely extended larval survival as long as 15 to 22 days of 4th instar. The growth of ovarioles was confirmed in the 10 to 14 day old larvae, oogonia developed into oocytes and nurse cell against heavy infection of the ovary. Gonads rarely obstructed oogenesis and spermatogenesis in the pupae failed in adult eclosion. Light infection of female hosts effected insignificantly on the ovarial development, however, recorded 100% transmission of the pathogens to the progeny populations. Conclusively, ovarial inflection of silkworm induce transovarial transmission begins around 2~3 day old pupae when ovarioles extruded out to hemocoel, and the infection period thought to be continued until the stage of eggs complete shell formation in the ovariole.
