Fluorescent detection of bacteria associated with gingival sulcus epithelium (DNA 형광 염색을 이용한 치은열구상피부착 세균에 관한 연구)

  • Purpose: The aim of this study was to compare the number of live and dead bacteria attached to, or within, the stratified squamous epithelium lining the tissue side of the gingival sulcus. Materials and Methods: A total of 50 patients was examined and classified into healthy or diseased sites according to inflammatory status of the gingival tissue. The surface of stratified squamous epithelium was removed by gentle scraping of the gingival sulcus with curettes. The cells were processed in the laboratory by density-gradient centrifugation to separate the epithelial cells from the loose bacteria and debris. The LIVE/$DEAD^{(R)}$ $BacLight^{TM}$ Bacterial Viability Kit was applied and the specimens were observed by an epifluorescent microscope and the number of bacteria was counted. Results: Live and dead bacteria were stained to green and red, irrespectively. Generally, the number of total bacteria in the diseased sites was significantly higher than in the healthy sites. The mean number of detected bacteria in the diseased sites was $58.6{\pm}36.0$ (red bacteria $10.4{\pm}9.2$ / green bacteria $48.2{\pm}30.5$), while it was $1.5{\pm}1.7$ in the healthy sites (red bacteria $0.1{\pm}0.3$ / green bacteria $1.4{\pm}1.5$). The percentage of red bacteria was $17.5{\pm}11.2%$ in the diseased sites and $2.0{\pm}5.8%$ in the healthy sites. Conclusion: The total number of bacteria in the diseased sites was significantly higher than that of the healthy sites. The ratio and the number of red bacteria were also significantly higher in the diseased sites.

Changes of Index Microorganisms and Lactic Acid Bacteria of Korean Fermented Vegetables (Kimchi) during the Ripening and Fermentation-Part 1 (김치의 숙성 및 발효중 오염지표미생물과 유산균의 변화-제1보)

  • This study was undertaken to investigate the changes of index microorganisms and lactic acid bacteria of traditional Korean fermented vegetables (kimchi) during the ripening and fermentation period. A type of kimchi, baechoo-kimchi, was prepared and stored at $10^{\circ}C$ for 8 days. The numbers of the total aerobic bacteria, psychrotrophilic bacteria, coliform bacteria, and Escherichia coli in the kimchi and also in raw materials of the kimchi (Chinese cabbage, green onion, ginger, garlic, and red pepper) were counted using appropriate media. The highest number of aerobic bacteria was detected from ginger, then red pepper, then garlic, then Chinese cabbage, and lowest number from green onion. The highest number of psychrotrophilic bacteria was detected from red pepper, then Chinese cabbage, then garlic, then ginger, and the lowest number from green onion. Coliforms and E. coli were not detected from all of the raw materials of kimchi. Total aerobic bacteria and lactic acid bacteria of the kimchi showed gradually increasing during ripening and fermentation. The number of psychrotrophilic bacteria showed a similar level in the kimchi. Coliform bacteria were detected at the 3rd, 4th, and 5th day of the kimchi fermentation period, although they were not detected from the raw materials of the kimchi. However, the bacteria were not detected in the kimchi after 6 days. E. coli was not detected in all kimchi samples. The pH value of the kimchi gradually decreased, and acidity increased over fermentation period. This study indicates that there was contamination of coliform bacteria during the process of kimchi preparation, and lactic acid bacteria proliferated in the kimchi during fermentation inhibited the growth of coliforms. More research is needed to evaluate the inhibitory effects of each raw materials of kimchi.

Bacteriological contamination of toothbrushes by dental plaque acidogenicity and related behaviors to toothbrush use (치면세균막 산생성도 및 칫솔관리행동별 칫솔 세균오염도)

  • One of the best way to prevent dental caries and periodontal disease is removing dental plaque. Toothbrushing is also one of the crucial ways to gel rid of dental plaque and improve dental hygiene. The purpose of th is study was to examine how many bacteria were attached to toothbrushes used by dental palque acidogenecity and related behaviors to toothbrush use. Subjects in this study were 30 college students in their twenties, on whom a survey was conducted to find out how they cleansed their toothbrushes, how long they cleaned toothbrushes after brushing, and how to keep them. And then dental palque acidogenecity by caries activity test was implemented and the number of bacteria attached to their toothbrushes was tested. The findings of the study were as follows : 1. Concerning correlations between dental palque acidogenecity and the number of remaining bacteria in the toothbrushes used by students, the number of bacteria was lower in the group of students with no caries activity than in the group with mild caries activity. 2. As for relations between the way of cleansing toothbrushes and the number of bacteria, there was no statistically significant gap between the group of students who wetted their toothbrushes with water and rubbed the heads of them and the group of students who just only wetted their toothbrushes with no further actions. 3. Regarding the relationship of toothbrush cleansing time to the number of bacteria, the number of bacteria was lower in the group of students who cleansed their toothbrushes for 15 seconds or more. 4. As to the influence of the number of toothbrush used by students, there was no significant difference between the group using just one toothbrush and the other group using two or more toothbrushes. 5. As for correlations between the way of keeping toothbrushes and the number of bacteria, no significant gap existed between the group thai kept their toothbrushes separately and the other that didn't.

Association of periodontitis-related bacteria complex with socio-demographic and oral health condition among the elderly in a rural area (일부 농촌지역 노인의 일반 특성 및 구강상태와 치주염유발세균의 관련성)

  • Objectives: The objectives of this study were to investigate the distribution and level of periodontopathic bacteria with the general characteristics and oral health condition of the elderly. Methods: A total of 335 elderly individuals aged 65 years or older who lived in Ganghwa-gun, Incheon, were included in the study. Oral examination, investigation through a questionnaire, and collection of saliva were carried out. The collected saliva was analyzed for the distribution and levels of bacteria (red and orange complex bacteria) by real-time polymerase chain reaction. Statistical analyses were performed using chi-square test, t-test, one-way analysis of variance, and Pearson's correlation coefficient with SAS statistical software version 9.4. Results: Among the general characteristics, there were significant differences in the distribution of Porphyromonas gingivalis, Treponema denticola, and Parvimonas micra depending on sex, age, and dental visits (p<0.05). The number of remaining teeth and denture use were related to the distribution of periodontopathic bacteria, except T. denticola (p<0.05). Additionally, periodontitis was related to the distribution of P. gingivalis (p<0.05). As the number of remaining teeth increased, the copy number of red and orange complex bacteria also increased (p<0.05). Those individuals who did not use dentures and had periodontal disease had more periodontopathic bacteria (p<0.05). Conclusions: The distribution and copy number of periodontopathic bacteria in the elderly were more related to oral health condition than to general characteristics. In particular, the distribution and copy number of periodontopathic bacteria were higher in subjects with multiple remaining teeth, no dentures, and periodontal disease.

A Study of Summer Socks on Their Properties of Hygiene and Comfortableness (여름양말의 위생성과 쾌적성에 관한 연구)

  • Tho purpose of this study was to investigate hygienic and comfortable properties of socks. Materials are nine summer socks either frequently being worn or new products recently introduced to market. Three female and three male adults participated in this study. Through wearing experiment, the numbers of microbes on foot and sock were counted and subjective sensation was measured. The microbes were isolated and identified based on growth physiological characteristics. Nine different socks had smaller number of bacteria of sock than that of foot. The number of bacteria of sock was significantly related with that of foot in cotton socks, in piled cotton socks, in mesh cotton socks, in cotton+ nylon+ linen blended socks, in nylon socks. Total number of bacteria of tv cut finished socks was most small and total number of bacteria was increased in the order of ultra fresh finished socks, untreated cotton socks, nylon socks, cotton + nylon+ linen blended socks, mesh cotton socks, polyester+ nylcn+ linen blended socks, piled cotton socks, cotton socks. Total number of bacteria of cotton socks and piled cotton socks were significantly different from that of uv cut finished socks. Finished socks and .jocks has high air permeability had significantly small number of bacteria. Comfortable sensation in nylon socks and polyester+nylon+linen socks was significantly uncomfortable. The way socks finished and air permeability of .jocks affected theirs hygienic property, while fiber type of them affected comfortablene, is. Bacteria identified were Staphylo coccus aureus, S. au rice larir, S. cahn ii, S. ep ids midis, S. haemo Iyticus, S. h am in 2's. S.fapraphyticus, S. warnery, 1 cinetobater calcoaceticus bio. anitratus, p.reudomonas mendocina, p. paucimobilis, Flavimonas Q ryzihabitans (CDC Group VE-2), and Xanthomanas maltophina. Fungi isolated were Spicaria sp., Thrichoderma sp., Fusarium sp., Aspergillus sp., Epicoccum sp., Cladosporium sp., and Penicillium sp..

  • Biological treatment of benzene and toluene contaminated soil was investigated in laboratory microcosm of 16 different types for degrading benzene and toluene by indigenous bacteria. At the experimental conditions of the microcosms fast degrading benzene and toluene, moisture contents were 30% and 60% in a soil gap and content of powdered-activated carbon(PCA) for adhesion of benzene and toluene-degrading bacteria was 1% in total soil mass. At the conclusion of the shifted bacteria community, Case 6 and case 7 were operated until 10 days, and then the total cell number and the number of benzene and toluene degrading bacteria were investigated. The total cell number of Case 6 and Case 7 increased 488 fold and 308 fold of total indigenous cell, respectively. The number of benzene and toluene degrading bacteria increased and maintained the percentages occupied in pre-operating microcosm. Species of benzene and toluene degrading bacteria in microcosm changed from species of Gram negative bacteria to Gram positive bacterial species after soil exposed to benzene and toluene.

Production of Polyphenols and Flavonoids and Anti-Oxidant Effects of Lactic Acid Bacteria of Fermented Deer Antler Extract

  • The deer antler has been used as a major drug in oriental medicine for a long time. Recently, the demand for easy-to-take health functional foods is increasing due to economic development and changes in diet. As part of research on the development of functional materials for antlers, lactic acid fermentation of antler extract was performed. It was intended to develop a functional material with enhanced total polyphenol and flavonoid content and enhanced antioxidant activity. Lactic acid bacteria fermentation was performed by adding 4 types of lactic acid bacteria starter products, B. longum, Lb. Plantarum, Lb. acidophilus and mixture of 8 types of lactic acid bacteria to the antler water extract substrate, respectively. During the fermentation of lactic acid bacteria, the number of proliferation, total polyphenol and total flavonoid content, DPPH radical scavenging and antioxidant activity were quantified and evaluated. As a result of adding these four types of lactic acid bacteria to the antler water extract substrate, the number of lactic acid bacteria measured was 2.04~5.00×107. Meanwhile, a protease (Baciullus amyloliquefaciens culture: Maxazyme NNP DS) was added to the antler extract to decompose the peptide bonds of the contained proteins. Then, these four types of lactic acid bacteria were added and the number of lactic acid bacteria increased to 2.84×107 ~ 2.21×108 as the result of culture. The total polyphenol contents were 4.82~6.26 ㎍/mL in the lactic acid bacteria fermentation extracts, and after the reaction of protease enzyme and lactic fermentation, increased to 14.27~20.58 ㎍/mL. The total flavonoid contents were 1.52~2.21 ㎍/ml in the lactic acid bacteria fermentation extracts, and after the protease reaction and fermentation, increased to 5.59 ~ 8.11 mg/mL. DPPH radical scavenging activities of lactic acid bacteria fermentation extracts was 17.03~22.75%, but after the protease reaction and fermentation, remarkably increased to 32.82~42.90%.

Enumeration and Activity of Methanogenic Microorganisms of th Anaerobic Digestion Process

  • The anaerobic digester with sludge from sewage treatment plant was operated in the laboratory for two year to investigate the enumeration and activity of methanogenic microorganisms. In this experimental study, the effects of HRT on the degradation characteristics of organic materials and on the number of methanogenic bacteria produced were investigated. By making the media with the repeated wxperiment, the number and activity of methanogenic bacteria were measured. The increase of the removal rate of organic acid in the digester was oberved at HRT of 2 days. The total number of methane forming bacteria estimated by the MPN method showed 2.3 $\times$ $ at HRT of 3 days, 7$\times$$ of 5 days and 7.9$\times$$ $/ml of 10 days. The optimum incubation time for measuring the number of methanogenic bacteria was found as more than four weeks. The PMA revealed 161ml CH$/l day at HRT of 10 days and the PUA 290mg COD/l day. At the incubation time 4.3 days, the maximum value of CH$ *59.1%) was found. At this time, $ was found as 15.3% and $ 25.6%.

  • Purpose: This study was examined the characteristics of bacteria isolated from the dental stone that is made ??in the dental laboratory. Methods: 104 dental stones samples were collected from the 4 dental laboratory. Characteristics of bacteria were investigated by microorganism isolation culture method using a Blood Tryptic Soy Agar(TSA) medium. Results: The detected various bacteria was confirmed as pathogenic bacteria, non-pathogenic bacteria and natural bacteria. The isolated bacterial number was confirmed $2.9{\times}10^3CFU$ and maxium bacterial number of $3.0{\times}10^4CFU$. Conclusion: Therefore, infection prevention education is required, it must be to live up the hand-washing and wear protective clothing to protect themselves when working in a dental laboratory.
